DS is a life-threatening complication, characterized ...In acute promyelocytic leukemia, differentiation thera-py based on all-trans-retinoic acid can be complicated by the development of a differentiation syndrome(DS). DS is a life-threatening complication, characterized by respiratory distress, unexplained fever, weight gain, interstitial lung infiltrates, pleural or pericardial effusions, hypotension and acute renal failure. The diagnosis of DS is made on clinical grounds and has proven to be difficult, because none of the symptoms is pathognomonic for the syndrome without any definitive diagnostic criteria. As DS can have subtle signs and symptoms at presentation but progress rapidly, end-stage DS clinical picture resembles the acute respiratory distress syndrome with extremely poor prognosis; so it is of absolute importance to be conscious of these complications and initiate therapy as soon as it was suspected. The radiologic appearance resembles the typical features of cardiogenic pulmonary edema. Diagnosis of DS remains a great skill for radiologists and haematologist but it is of an utmost importance the cooperation in suspect DS, detect the early signs of DS, examine the patients' behaviour and rapidly detect the complications.展开更多
AIM:To explore the apoptosis of ARPE-19 cells after the treatment with different doses of all-trans-retinoic acid(ATRA).METHODS:ARPE-19 cells were used in the in-vitro experiment.Flow cytometry assay was employed to e...AIM:To explore the apoptosis of ARPE-19 cells after the treatment with different doses of all-trans-retinoic acid(ATRA).METHODS:ARPE-19 cells were used in the in-vitro experiment.Flow cytometry assay was employed to evaluate the level of reactive oxygen species(ROS)and apoptosis.The effects of ATRA(concentrations from 2.5 to 20μmol/L)on the expression of endoplasmic reticulum stress(ERS)markers in vitro were evaluated by Western blot and realtime quantitative polymerase chain reaction(qRT-PCR)assays.The contribution of ROS and ERS-induced apoptosis in vitro was determined by using N-acetyl-L-cysteine(NAC)and Salubrinal,an antagonist of NAC and ERS,respectively.RESULTS:Flow cytometry showed that ATRA significantly increased ARPE-19 cell apoptosis and ROS levels in each group(F=86.39,P<0.001;F=116.839.P<0.001).Western blot and qRT-PCR revealed that levels of CHOP and BIP were elevated in a concentration-dependent pattern after the cells were incubated with ATRA(2.5-20μmol/L).The upregulation of VEGF-A and CHOP induced by ATRA could be inhibited by NAC(antioxidant)and Salubrinal(ERS inhibitor)in vitro.CONCLUSION:ATRA induces the apoptosis of ARPE-19 cells via activated ROS and ERS signaling pathways.展开更多

OBJECTIVE Autosomal dominant polycystic kidney disease(ADPKD) is a common.monogenetic disease characterized by progressive development of renal cysts.Thereis still further need for effective therapy.Based on our preci...OBJECTIVE Autosomal dominant polycystic kidney disease(ADPKD) is a common.monogenetic disease characterized by progressive development of renal cysts.Thereis still further need for effective therapy.Based on our precious study that Ganoderma triterpenes(GT),which is the major secondary metabolites of Ganoderma lucidum,is able to attenuate renal cyst development.The aim of this study was to investigate the effect of a monomer,Ganoderic acid A(GA-A) that was purified from the GT,which has been reported to exhibit antinociceptive,antioxidative,hepatoproctive and anti.cancer activities,to have a potent anti-cyst effect in ADPKD.METHODS We first evaluated the potential cytotoxicity of GA-A on MDCK cells using a CCK-8 assay.Then we used MDCK cyst model,cultivated MDCK cells in vitro to form fluid-filled cysts surrounded by monolayer cells.MDCK cells were co-incu.bated with 10 μmol·L^(-1) FSK with or without GA-A(25 μg·mL-1) and equal concentration GT as positive control from day 0 to day 6 to investigate the inhibitory effect of GA-A on cyst formation.And to further investigate the inhibitory effect of GA-A on cyst enlargement,MDCK cysts were treated with different concentration of GA-A(6.25,25 and 100 μg·mL-1) from day 5 to day 12.Next we used an embryonic kidney cyst model,wile-type mice kidneys were taken out on embryonic day 13.5 to form renal cysts stimulated with 8-Br-cAMP to prove the renal cyst inhibition at organ level.Meanwhile,we explored the possible mechanisms underlying GA-A inhibition on renal cyst development using MDCK cells treated with 10 μmol·L^(-1) FSK co-incubated with GA-A(25 μg·mL-1) and equal concentration GT.Several key components of Ras/MAPK pathway was evaluated by Western blot,the protein expression of H-ras,B-raf,p-ERK,Egr-1 and c-fos was evaluated.RESULTS MDCK cell viability was not affected by GA-A that were used ofincreasing concentrations up to 200 μg·mL^(-1).GA-A had no significant influence on cyst formation,but inhibited cyst enlargement dose-dependently and the inhibitory effect is significantly better than that of the same concentration of GT which proved that GA-A may be an effective monomer from GT.And after washing out GA-A on day 8,MDCK cysts regrew to a large size,suggesting that the inhibitory effect of GA-A on MDCK cyst enlargement was reversible.GA-A inhibited embryonic kidney cyst development significantly in a dose-dependent and reversible manner proving GA-A cyst inhibition at organ level,which is also more effective than equal concentration GT.Treatment of MDCK cells with FSK caused a significant elevation of H-ras,B-raf,p-ERK,Egr-1 and c-fos signaling molecules,while treatment with GA-A reduced the level of H-ras,B-raf,p-ERK,Egr-1 and c-fos expression.GA-A downregulated Ras/MAPK signaling pathway could contribute to its inhibitory effect on cyst development.CONCLUSION Ganoderic acid A from Ganoderma lucidum retard ADPKD renal cyst development via down-regulating Ras/MAPK signaling pathway.展开更多

Palynological analysis showe...The aim of this investigation was to analyze two samples of pollen from jandaíra stingless bees (Melipona subnitida) in view of their mineral composition, free amino acids and sugars. Palynological analysis showed that the predominant pollen was monofloral from Senna sp. species (94.5%, pollen 2011) and the second pollen sample showed the presence of two primary species, Chamaecrista sp. (39.2% pollen 2009) and Mimosa tenuiflora (43.5%) (pollen 2009). The highest mineral content was potassium. The bee pollen contained 20.8% and 31.0% of mannitol in samples from 2011 and 2009, respectively. Proline and serine are the predominant amino acids. High content of essential amino acids, minerals and the sugar mannitol confirmed high nutritional value of pollen samples from jandaíra bees.展开更多

This is an important process for the photopro...The formation of triplet chlorophyll and carotenoid by radical pair recombination have been observed in the reaction centers of photosystems of bacteria and higher plants. This is an important process for the photoprotection of the reaction centers, for the dissipation of excessive energy by non_radiative decay of carotenoid triplet. Triplet generation by the same mechanism in an artificial system has rarely been observed, only a few cases were reported in donor_acceptor triad supermolecules. This is probably the first time report of the simulation of the generation of triplet by back electron transfer using dye_sensitized TiO 2 solar cell reaction. Triplet states have been observed in all_ trans _retinoic acid sensitized TiO 2 colloid during the recombination of the trapped electron with the retinoic acid radical cation after photoexcitation. The intermediates were characterized by ns time_resolved spectroscopy.展开更多
