The Expression of E-Cadherin and Vimentin in Non-Small-Cell Lung Cancer and Their Correlation with Epithelial-Mesenchymal Transition (EMT)

To investigate the expression of E-cadherin and Vimentin protein in non-small-cell lung cancer (NSCLC) and to analyze the relationship between their expression and epithelial-mesenchymal transition (EMT). Immuno-chemical staining with SP method was conducted to detect the protein expression of E-cadherin and Vimentin in 44 lung cancer tissues. E-cadherin was highly expressed in all normal lung tissues, and Vimentin was lowly expressed in all normal lung tissues. The expression rate of E-cadherin and Vimentin was 40.9% and 25.0% in NSCLC cases. The expressions of E-cadherin and Vimentin were not associated with gender, age, histopathological grading, lymphatic metastasis or smoking status (P > 0.05). But they were associated with clinical stage (P < 0.05). In addition, a negative correlation between the E–cadherin and Vimentin was observed (Spearman’s r = ?0.397, p < 0.05). E-cadherin and Vimentin are involved in the transformation of epithelial cells into the mesenchymal cells, which play important role in the metastasis of lung cancer. 展开更多

OBJECTIVE To investigate the expression of TGF-β1,Snail,E-cad-herin and N-cadherin in gastric cancer(GC),and to examine its relationship to malignant features of the tumors. METHODS The expression of TGF-β1,Snail,E-cadherin and N-cadherin proteins was detected in GC and adjacent tissues by immunohistochemical staining,and compared with the clinico-pathological data. RESULTS Positive rates of expression for TGF-β1,Snail,E-cadherin and N-cadherin were 63.5%,83.3%,37.5%and 44.8%in GC,and 28.8%, 41.3%,100%,11.3%in adjacent tissues,respectively.The expression of all four proteins showed a significant difference between the GCs and adjacent tissues(P<0.05).The positive rate of TGF-β1,Snail and N-cadherin,or the negative rate of E-cadherin expression was significantly related to the differentiated degree,histological type,invasion and metastasis of GC.In addition,the expression of N-cadherin was positively related to that of TGF-β1, but negatively related to that of E-cadherin.There was negative correlation between expression of E-cadherin and TGF-β1 and Snail in GC(P<0.05). CONCLUSION The over-expression of TGF-β1 and Snail and decreased expression of E-cadherin and the abnormal expression of N-cad-herin were involved in the process of invasion and metastasis of GC.The data showed that E-cadherin might switch to N-cadherin.TGF-β1 and Snail might play a fundamental role in the process. 展开更多

和胃降逆胶囊对胃腺癌细胞凋亡的影响及其机制

目的以核转录因子-κB/上皮间充质转化(NF-κB/EMT)信号通路为基础,探讨和胃降逆含药血清对胃腺癌AGS细胞增殖和凋亡的影响及作用机制。方法制备不同药物剂量(3,6,12 g/kg)的和胃降逆含药血清,作为含药血清低、中、高剂量组,不含药物的... 目的以核转录因子-κB/上皮间充质转化(NF-κB/EMT)信号通路为基础,探讨和胃降逆含药血清对胃腺癌AGS细胞增殖和凋亡的影响及作用机制。方法制备不同药物剂量(3,6,12 g/kg)的和胃降逆含药血清,作为含药血清低、中、高剂量组,不含药物的血清作为空白对照组。采用不同药物剂量的和胃降逆含药血清分别干预AGS细胞24,48,72 h后,用CCK-8法观察细胞增殖;DAPI染色法检测细胞凋亡;划痕试验观察细胞迁移;实时定量聚合酶链反应法(RT-qPCR)检测NF-κB、EMT标志物相关蛋白E-钙黏蛋白(E-cadherin)、N-钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)mRNA水平;蛋白免疫印迹法(Western blot)检测细胞内NF-κB、E-cadherin、Vimentin相关信号通路蛋白的表达。结果同一时间点,与空白对照组比较,和胃降逆含药血清组细胞活力降低(P<0.01);同一含药血清剂量组48,72 h与24 h比较细胞活力降低(P<0.05)。干预48 h,与空白对照组比较,不同剂量含药血清组AGS细胞凋亡率剂量依赖性增加(P<0.05)。与空白对照组比较,和胃降逆含药血清组划痕愈合速度均减慢,迁移率降低(P<0.05);干预24 h,与空白对照组比较,高剂量组细胞迁移降低(P<0.01),干预48 h,中、高剂量组细胞迁移率降低(P<0.01)。与空白对照组比较,和胃降逆含药血清组AGS细胞中NF-κB、Vimentin的mRNA水平剂量依赖性下调(P<0.05),E-cadherin的mRNA水平剂量依赖性上调(P<0.01)。与空白对照组比较,和胃降逆含药血清组AGS细胞中NF-κB、Vimentin的蛋白表达剂量依赖性下调,E-cadherin的蛋白表达剂量依赖性上调(P<0.05)。结论和胃降逆胶囊可能通过抑制NF-κB活化、调控下游EMT相关蛋白的表达介导胃腺癌细胞AGS的迁移和侵袭,进而抑制AGS细胞增殖并促进其凋亡。 展开更多

Environmental microcystin exposure triggers the poor prognosis of prostate cancer: Evidence from case-control, animal, and in vitro studies 被引量:1

Microcystin-leucine-arginine(MC-LR)is positively linked with multiple cancers in humans.However,the association between MC-LR and the risk and prognosis of prostate cancer has not been conducted in epidemiological studies.No reported studies have linked MC-LR exposure to the poor prognosis of prostate cancer by conducting experimental studies.The content of MC-LR was detected in most of the aquatic food in wet markets and supermarkets in Nanjing and posed a health risk for consumers.MC-LR levels in both prostate cancer tissues and serum were significantly higher than controls.The adjusted odds ratio(OR)for prostate cancer risk by serum MC-LR was 1.75(95%CI:1.21-2.52)in the whole subjects,and a positive correlation between MC-LR and advanced tumor stage was observed.Survival curve analysis indicated patients with higher MC-LR levels in tissues exhibited poorer overall survival.Human,animal,and cell studies confirmed that MC-LR exposure increases the expression of estrogen receptor-α(ERα)and promotes epithelial-mesenchymal transition(EMT)in prostate cancer.Moreover,MC-LR-induced decreased E-cadherin levels,increased vimentin levels,and increased migratory and invasive capacities of prostate cancer cells were markedly suppressed upon ERαknockdown.MC-LR-induced xenograft tumor growth and lungmetastasis in BALB/c nude mice can be effectively alleviated with ERαknockdown.Our data demonstrated that MC-LR upregulated vimentin and downregulated E-cadherin through activating ERα,promoting migration and invasion of prostate cancer cells.Our findings highlight the role of MC-LR in prostate cancer,providing new perspectives to understand MC-LR-induced prostatic toxicity. 展开更多
