The spatial variability of soil stiffness is modelled using a variogram and calibrated by high-quality experimental data. Multiple random field samples (RFSs) of soil stiffness are generated using geostatistical analysis and mapped onto a finite element mesh for stochastic analysis of excavation-induced structural responses by Monte Carlo simulation. It is found that the spatial variability of soil stiffness can be described by an exponential variogram, and the associated vertical correlation length is varied from 1.3 m to 1.6 m. It also reveals that the spatial variability of soil stiffness has a significant effect on the variations of retaining wall deflections and box culvert settlements. The ignorance of spatial variability in 3D FEM can result in an underestimation of lateral wall deflections and culvert settlements. Thus, the stochastic structural responses obtained from the 3D analysis could serve as an effective aid for probabilistic design and analysis of excavations.展开更多

文摘Construction and commissioning accidents investigation shows that engineer’s errors in basic and detailed phases are the main causes. It will be gained more efficiency of investment when a relevant method is applied for error identification and risk assessment in basic and detailed phases. Most of risk assessment methods are related to construction phase, meanwhile hidden risks that were happen in previous phases cannot be found by these methods.Although HAZOP study can be applied in detail phase but it can help to find process defections.Importance of identifying risks in basic and detailed phases is to apply preventive actions against birth and grows risks. This risk assessment method can reduce costs and improve environmental protection, safety, and reliability of designs. The main purpose of this study is to identify engineer’s errors and analyse their HSE effects. The method is applied in seven oil and gas engineering projects in Iran, raising the level of engineer’s attention to safety precautions and environmentally-friendly design, reducing the cost of improving the level of safety and environmental protection, improving environmental protection, safety, and design reliability, and improving procedures related to design process and control have been achieved.
文摘Little research can be found in relation to the stability of anisotropic and heterogenous soils in three dimensions. In this paper,we propose a study on the three-dimensional(3D)undrained slopes in anisotropic and heterogenous clay using advanced upper and lower bounds finite element limit analysis(FELA).The obtained stability solutions are normalized, and presented by a stability number that is a function of three geometrical ratios and two material ratios,i.e.depth ratio, length ratio, slope angle, shear strength gradient ratio and anisotropic strength ratio.Numerical results are compared with experimental data in the literature, and charts are presented to cover a wide range of design parameters.Using the multivariate adaptive regression splines(MARS)analysis, the respective influence and sensitivity of each design parameter on the stability number and the failure mechanism are investigated. An empirical equation is also developed to effectively estimate the stability number.
文摘Objective This study aimed to compare 9 perfluoroalkyl sulfonic acids(PFSA)with carbon chain lengths(C4–C12)to inhibit human placental 3β-hydroxysteroid dehydrogenase 1(3β-HSD1),aromatase,and rat 3β-HSD4 activities. Methods Human and rat placental 3β-HSDs activities were determined by converting pregnenolone to progesterone and progesterone secretion in JEG-3 cells was determined using HPLC/MS–MS,and human aromatase activity was determined by radioimmunoassay. Results PFSA inhibited human 3β-HSD1 structure-dependently in the order:perfluorooctanesulfonic acid(PFOS,half-maximum inhibitory concentration,IC50:9.03±4.83μmol/L)>perfluorodecanesulfonic acid(PFDS,42.52±8.99μmol/L)>perfluoroheptanesulfonic acid(PFHpS,112.6±29.39μmol/L)>perfluorobutanesulfonic acid(PFBS)=perfluoropentanesulfonic acid(PFPS)=perfluorohexanesulfonic acid(PFHxS)=perfluorododecanesulfonic acid(PFDoS)(ineffective at 100μmol/L).6:2FTS(1H,1H,2H,2H-perfluorooctanesulfonic acid)and 8:2FTS(1H,1H,2H,2H-perfluorodecanesulfonic acid)did not inhibit human 3β-HSD1.PFOS and PFHpS are mixed inhibitors, whereas PFDS is a competitive inhibitor.Moreover,1–10μmol/L PFOS and PFDS significantly reduced progesterone biosynthesis in JEG-3 cells.Docking analysis revealed that PFSA binds to the steroid-binding site of human 3β-HSD1 in a carbon chain length-dependent manner. All 100μmol/L PFSA solutions did not affect rat 3β-HSD4 and human placental aromatase activity. Conclusion Carbon chain length determines inhibitory potency of PFSA on human placental 3β-HSD1 in a V-shaped transition at PFOS(C8),with inhibitory potency of PFOS>PFDS>PFHpS>PFBS=PFPS=PFHxS=PFDoS=6:2FTS=8:2FTS.

文摘Vibration induced by shield construction can lead to liquefaction of saturated sand.Based on FLAC3D software,a numerical model of tunnel excavation is established and sinusoidal velocity loads with different frequencies are applied to the excavation face. The pattern of the excess pore pressure ratio with frequency,as well as the dynamic response of soil mass under different frequency loads before excavation,is analyzed. When the velocity sinusoidal wave acts on the excavation surface of the shield tunnel with a single sand layer, soil liquefaction occurs. However, the ranges and locations of soil liquefaction are different at different frequencies, which proves that the vibration frequency influences the liquefaction location of the stratum. For sand-clay composite strata with liquefiable layers, the influence of frequency on the liquefaction range is different from that of a single stratum. In the frequency range of 5-30 Hz,the liquefaction area and surface subsidence decrease with an increase in vibration frequency. The research results in this study can be used as a reference in engineering practice for tunneling liquefiable strata with a shield tunneling machine.

文摘[Objectives]The paper was to clone and analyze bioinformatics of vscN gene from Vibrio alginolyticus.[Methods]A pair of specific primers was designed based on the vscN gene sequence of V.alginolyticus HY9901.The full length of vscN gene was amplified by PCR and bioinformatics analysis was performed.[Results]The vscN gene was 1323 bp in total, encoding 440 amino acids, with the theoretical molecular weight of 47.86 kD and the theoretical pI value of 5.89.The online prediction showed that there was no signal peptide and no transmembrane region in VscN.The amino acid sequence had 10 N-myristoylation sites,8 phosphorylation sites(2 protein kinase C phosphorylation sites,6 casein kinase II phosphorylation sites),1 amidation site,11 microbody C-terminal target signal sites,1 ATP/GTP binding site motif A(P ring),and 1 ATPaseαandβsubunit specific site.Homology analysis showed that the VscN protein of V.alginolyticus had high homology with that of V.antiquarius, with a similarity of 95.14%.Phylogenetic tree analysis showed that the VscN of V.alginolyticus was clustered into the same subgroup as that of V.diabolicus and V.antiquarius.Functional domain analysis of VscN protein showed that it had Pfam and AAA domains, and involved in the regulation of bacterial virulence. The three-dimensional structure model of VscN simulated by SWISS-MODEL software was similar to the structure of flagellate-specific ATPase FliH-FliI complex.[Conclusions]The results lay a foundation for further study on the regulatory mechanism of VscN protein on bacterial virulence.

文摘In this study, a three-dimensional (3D) finite element modelling (FEM) analysis is carried out to investigate the effects of soil spatial variability on the response of retaining walls and an adjacent box culvert due to a braced excavation. Thus, the stochastic structural responses obtained from the 3D analysis could serve as an effective aid for probabilistic design and analysis of excavations.
文摘In the construction and maintenance of particle accelerators, all the accelerator elements should be installed in the same coordinate system, only in this way could the devices in the actual world be consistent with the design drawings. However, with the occurrence of the movements of the reinforced concrete cover plates at short notice or building deformations in the long term, the control points upon the engineering structure will be displaced, and the fitness between the subnetwork and the global control network may be irresponsible. Therefore,it is necessary to evaluate the deformations of the 3D alignment control network.Different from the extant investigations,in this paper,to characterize the deformations of the control network, all of the congruent models between the points measured in different epochs have been identified, and the congruence model with the most control points is considered as the primary or fundamental model, the remaining models are recognized as the additional ones.Furthermore, the discrepancies between the primary S-transformation parameters and the additional S-transformation parameters can reflect the relative movements of the additional congruence models. Both the iterative GCT method and the iterative combinatorial theory are proposed to detect multiple congruence models in the control network.Considering the actual work of the alignment,it is essential to identify the competitive models in the monitoring network, which can provide us a hint that, even the fitness between the subnetwork and the global control network is good, there are still deformations which may be ignored. The numerical experiments show that the suggested approaches can describe the deformation of the 3D alignment control network roundly.
文摘This scientific paper is a comparative analysis of two mathematical conjectures. The newly proposed -3(-n) - 1 Remer conjecture and how it is related to and a proof of the more well known 3n + 1 Collatz conjecture. An overview of both conjectures and their respective iterative processes will be presented. Showcasing their unique properties and behavior to each other. Through a detailed comparison, we highlight the similarities and differences between these two conjectures and discuss their significance in the field of mathematics. And how they prove each other to be true.
