Expression of annexin II in gastric carcinoma and its role in gastric cancer metastasis

AIM To investigate the expression of annexin II in gastric carcinoma and its role in the metastasis of gastric cancer.METHODS The expression of annexin II in 51 cases of gastric carcinoma and 24 cases of adjacent tissues was detected by immunohistochemistry. The relationship between annexin II and clinical features of gastric cancer was analyzed. Annexin II specific si RNA was used to inhibit the expression of annexin II in gastric cancer HGC-27 cells, and the effects of annexin II on the migration and secretion of matrix metalloproteinases(MMPs) were observed.RESULTS The positive rate of annexin II protein was 82.4% in gastric cancer tissues and 37.5% in adjacent tissues. There was significant difference between the two groups(P < 0.01); and the positive expression of annexin II was not related to the sex and age of the patients(P > 0.05). The expression of annexin IIprotein was correlated with tumor size, histological differentiation, TNM stage, Lymph node metastasis and other clinical features were significantly correlated, the difference was statistically significant(P < 0.05). Inhibition of annexin II expression, gastric cancer HGC-27 cells migration and secretion of MMPs were significantly decreased, the difference was statistically significant(P < 0.05).CONCLUSION Annexin II is highly expressed in gastric cancer tissues, annexin II protein expression is related to tumor size, histological differentiation, TNM staging, lymph node metastasis and other clinical features were significantly correlated. Annexin II high expression can promote the invasion and metastasis of gastric cancer.

Annexin II、尿激酶型纤溶酶原激活剂在甲状腺癌中的表达及临床意义

目的探究Annexin II、尿激酶型纤溶酶原激活剂（uPA）在甲状腺癌发生发展和淋巴结转移中的作用。方法应用免疫组织化学法检测AnnexinII、uPA蛋白在35例甲状腺乳头状癌组织、10例甲状腺腺瘤组织、16例结节性甲状腺肿组织、7例甲状腺正常组织中的表达。结果Annexin II、uPA蛋白在甲状腺乳头状癌组织中的表达较正常、结节性甲状腺肿和腺瘤组织中的表达显著增高（P〈0．05），且2者在癌组织中的表达呈正相关（P〈0．05）。Annexin II蛋白的表达与肿瘤大小和淋巴结转移有关，uPA蛋白的表达只与淋巴结转移有关。结论Annexin II、uPA蛋白参与了甲状腺乳头状癌的发生发展；并在甲状腺乳头状癌淋巴结转移中发挥促进作用。

Arsenic trioxide downregulates the expression of annexin II in bone marrow cells from patients with acute myelogenous leukemia

Background Most patients with acute myelogenous leukemia （AML） suffer from disordered hemostasis. We have previously shown that annexin II （Ann II）, a high-affinity co-receptor for plasminogen/tissue plasminogen activator, plays a central role in primary hyperfibrinolysis in patients with acute promyelocytic leukemia （APL）. The expression of Ann II in cells from patients with major subtypes of AML and the effect of arsenic trioxide （As203） on Ann II expression in AML cells were investigated to determine whether As203-mediated downregulation of Ann II could restore hemostatic stability. Methods A total of 103 patients （48 females and 55 males; age, 19-58 years） were included. Plasma samples were collected before and after treatment as well as after complete remission. Ann II and plasminogen activation were measured in leukemic cells during treatment with 1 pmol/L As203. Results Before AS203 treatment, Ann II mRNA expression （real-time PCR） was the highest in M3 cells （P 〈0.05）, higher in M5 cells than that in M1, M2, M4, and M6 cells （P〈0.001）, and positively correlated with Ann II protein expression （flow cytometry） （r=0.752, P 〈0.01）. Exposure for up to 120 hours to As203 （1 μmol/L） had no significant effect on Ann II protein in M1 and M2 leukemic cells, but decreased Ann II protein expression twofold within 48 hours of exposure in M3 cells （P 〈0.05） and twofold within 96 hours in M5 cells （P 〈0.05）. The rate of plasmin generation was higher in APL, M5, and M4 cells than in M1, M2, and M6 cells. Conclusions As203 may reduce hyperfibrinolysis in AML by downregulation of Ann 11. Furthermore, As2O3 affects more than one form of AML （APL, M4 and M5）, suggesting its potential role in their management.

肾透明细胞癌中AnnexinⅡ的表达及其临床意义

目的:探讨AnnexinⅡ在人肾透明细胞癌和癌旁组织样本中的表达情况及其临床意义。方法:采用Western blot方法检测29例新鲜肾透明细胞癌及相应癌旁组织中AnnexinⅡ蛋白的表达;采用免疫组织化学方法检测120例肾透明细胞癌及相应癌旁组织中AnnexinⅡ的表达情况,并将结果与患者的临床病理参数及预后进行相关性分析。结果:AnnexinⅡ在肾透明细胞癌及相应癌旁组织中均有表达,前者的表达水平较后者显著升高(P<0.01);免疫组织化学显示肾透明细胞癌组织中,AnnexinⅡ的阳性表达率高达67.5%,与癌旁组织存在明显差异(P<0.01);AnnexinⅡ在肾透明细胞癌组织中以胞膜表达为主,其表达状况与肾透明细胞癌患者的临床分期(P<0.05),组织学分级(P<0.05),肾被膜受侵(P<0.01)和远地转移(P<0.01)紧密相关;并且AnnexinⅡ的表达状况与肾透明细胞癌患者的5年生存率显著相关。结论:AnnexinⅡ在肾透明细胞癌组织中过表达,其与肾癌的发展密切相关,在肾癌的侵袭及转移过程中可能发挥重要作用。

前列腺癌膜联蛋白Ⅱ微卫星不稳定的荧光复合扩增研究

目的探讨膜联蛋白Ⅱ微卫星的不稳定对前列腺癌早期诊断的作用。方法使用荧光复合扩增的方法对16例前列腺癌组织、16例良性前列腺增生组织及16例正常人血样中的膜联蛋白Ⅱ的三个基因座进行扩增,检测PCR产物,分析其差异与疾病间的关系。结果膜联蛋白Ⅱ的D15S1185,D15S1292两个正常基因座在前列腺癌组织、良性前列腺增生组织以及血液样本中扩增大小无差别;在AN2-di-repeat-NH的二核苷酸重复序列微卫星基因座区域表现差异,以扩增大小134 bp为分界线,前列腺癌组织与正常血液样本存在显著性差异(χ2=9.087,df=1,P=0.003),前列腺癌组织与良性前列腺增生组织之间显著性不明显(χ2=1.997,df=1,P=0.158),良性前列腺增生组织与正常血液样本之间有显著性差异趋势(χ2=3.405,df=1,P=0.065)。结论膜联蛋白Ⅱ二核苷酸微卫星基因座AN2-di-repeat-NH的多态性有可能成为早期前列腺癌的预警指标。

Annexin Ⅱ、NF-κBp65及VEGF-C蛋白在胃腺癌组织中的表达及临床意义

目的探讨膜联蛋白(Annexin)Ⅱ、核因子(NF)-κBp65及血管内皮生长因子(VEGF)-C蛋白在胃腺癌组织中的表达及临床意义。方法采用免疫组化SP法检测55例正常胃黏膜组织、30例慢性萎缩性胃炎组织及55例胃腺癌组织中AnnexinⅡ、NF-κBp65及VEGF-C蛋白的表达,分析其与肿瘤临床病理特征的关系。结果 AnnexinⅡ蛋白在胃腺癌组织中的阳性表达率为83.6%,明显高于慢性萎缩性胃炎组织的46.7%(χ2=12.779,P<0.01)和正常胃黏膜组织的29.1%(χ2=33.266,P<0.01)。NF-κBp65蛋白在胃腺癌组织中的阳性表达率为76.4%,明显高于慢性萎缩性胃炎组织的40.0%(χ2=11.079,P<0.01)和正常胃黏膜组织的16.4%(χ2=39.811,P<0.01)。VEGF-C蛋白在胃腺癌组织中的阳性表达率为72.7%,明显高于慢性萎缩性胃炎组织的36.7%(χ2=10.518,P<0.01)和正常胃黏膜组织的27.3%(χ2=22.727,P<0.01)。淋巴结转移组AnnexinⅡ、NF-κBp65、VEGF-C蛋白的表达率明显高于无淋巴结转移组(P<0.05或P<0.01);与中高分化组相比,低分化组NF-κBp65、VEGF-C蛋白表达率明显增高(P<0.05或P<0.01)。三者表达均与肿瘤浸润深度无关(P均>0.05)。结论 AnnexinⅡ、NF-κBp65和VEGF-C蛋白可能参与了胃癌的发生发展过程,与胃腺癌的生物学行为相关。

钙磷脂结合蛋白Ⅱ在人肝细胞癌组织的表达及其临床意义

目的观察钙磷脂结合蛋白Ⅱ（Annexinll）在人肝细胞癌（HCC）组织中的表达。方法采用荧光实时定量聚合酶链反应（Real—timePCR）方法检测33例HCC患者肝癌组织和配对的癌旁肝组织以及11例正常肝组织中AnnexinⅡmRNA表达；利用免疫组织化学方法检测104例HCC组织和配对的癌旁肝组织中AnnexinⅡ蛋白的表达。结果肝癌组织中AnnexinⅡmRNA表达量高于癌旁肝组织和正常肝组织，是癌旁肝组织的3．38倍（P〈0．01），是正常肝组织的4．42倍（P〈0．01）；AnnexinII蛋白表达明显高于配对的癌旁肝组织（P〈0．05）。年龄≤40岁的患者AnnexinⅡmRNA表达水平是年龄〉40岁患者的1．875倍（P〈0．05）。结论AnnexinII在人HCC患者肝癌组织中表达上调，可能成为肝细胞癌临床诊断的新的候选基因。
