To investigate the dynamic change of heat shock protein 90 (Hsp90) in the genesis and development of tumor, we successfully established tumor animal model using Marek’s disease and then determined the location of Hsp90 in the tumor tissue using immunohistochemistry method, the antibody titer level of Hsp90 in the serum and the expression level in the tissue using enzyme-linked immunosorbent assay (ELISA) method. Our result showed that Hsp90 location in the tumor tissue was signiifcantly associated with the tumor cell and most in the cytoplasm of the tumor cell, and Hsp90 expression level in the tissue and the antibody titer level in the serum was most signiifcantly increased with the development of tumor. This is the ifrst report to show the presence of Hsp90 in tumor tissues induced by the Marek’s disease, with its expression correlated to the tumoral grading. These data may also be valuable for developing new molecular anti-cancer therapies. 展开更多

Monitoring Report of Maternal Antibody of Broiler Avian Influenza Virus H5 Subtype Re-8 Strain

[ Objective ] The paper was to prevent the occurrence of broiler avian influenza virus HS subtype Re-8 strain effectively in the breeding process of broilers. [Method] The maternal antibodies of broilers in Beijing Baochen Hongwang farm were monitored. According to the disappearance law of maternal antibody, the optimal immune time of broiler avian influenza virus H5 subtype Re-8 strain was determined. [ Result] The maternal antibody level of 2-day-old broilers was relatively high, concentrated at 6 log2 -9 log2, and the antibody positive rate was 100%. The maternal antibody level of 8-day-old broilers concentrated at 4 log2 -6 log2, and the antibody positive rate was 100%. The maternal antibody level of 17-day-old broilers concentrated at 0 log2 -3 log2 , and the antibody positive rate was 0. The average maternal antibody level of 24 - 37 days old broilers was 〈 1 log2, and the antibody positive rate was 0. [ Conclusion ] Although the av- erage maternal antibody level of 8-day-old broilers was higher than 5 log2 , 20% of chickens was 4 log2, and maternal antibody could not protect the flock completely. Therefore, the best primary immunization day age of chicks against avian influenza virus was 8 - 10 days of age. 展开更多

This study was conducted to investigate the effect of Se-enriched lactobacillus on antibody of Newcastle Disease Virus(NDV)of broilers.One thousand day-old AA broiler chickens were selected from a chicken farm of Jinghai County,and the chickens were divided randomly into two groups,which respectively had five repeat groups with 100 chickens each,one named basal diet group,and the other named Se-enriched lactobacillus group.Corn soybean meal diet was selected for basal diet group,while 2%se-enriched lactobacillus was added into corn soybean meal diet for the other group.The method of feeding was free diet,drinking,and routine immunization program was adopted.This test period was 42 d.Two chickens were selected randomly in each repeat for slaughtering and collecting serums on the 14th,28th and 42nd d.Assessments on the effect of Se-enriched lactobacillus in antibody titer of NDV of broilers were made by hemagglutination-inhibition test.The results indicated that antibody titer of NDV was increased significantly by Se-enriched lactobacillus. 展开更多

H9N2 avian influenza virus(AIV) has widely circulated in poultry worldwide and sporadic infections in humans and mammals. During our surveillance of chicken from 2019 to 2021 in Shandong Province, China, we isolated 1... H9N2 avian influenza virus(AIV) has widely circulated in poultry worldwide and sporadic infections in humans and mammals. During our surveillance of chicken from 2019 to 2021 in Shandong Province, China, we isolated 11 H9N2AIVs. Phylogenetic analyses showed that the eight gene segments of the 11 isolates were closely related to several sublineages of Eurasian lineage: BJ/94-like clades(HA and NA genes), G1-like clades(PB2 and M genes), and SH/F/98-like clades(PB1, PA, NP and NS genes). The isolates showed mutation sites that preferentially bind to humanlike receptors(HA) and mammalian fitness sites(PB2, PB1 and PA), as well as mutations in antigen and drug resistance sites. Moreover, studies with mice revealed four isolates with varying levels of pathogenicity. The average antibody titer of the H9N2 AIVs was 8.60 log2. Based on our results, the epidemiological surveillance of H9N2 AIVs should be strengthened. 展开更多

Objective To develop a model of SHIV-KB9/Chinese origin rhesus (Ch Rh) macaques for vaccine research and to compare the pathogenesis of SHIV-KB9 in Ch Rh macaques with that reported in Indian rhesus (Ind Rh) macaques. Methods Seven mamu-A*01 negative Ch Rh macaques were inoculated intravenously with 1-10000 MID50 of SHIV-KB9. The monkeys were monitored for viral load, CD4, CDS, SHIV-specific antibody and virus genetic variation. The results were compared with those previously observed in Ind Rh macaques. Results As compared to that observed in Ind Rh macaques, SHIV-KB9 in Ch Rh macaques displayed three identical disease progression patterns. However, the primary pattern was not identical between the two subspecies. The level of plasma viremia differed in SHIV-KB9-infected Ch Rh macaques which exhibited different outcomes from those in Ind Rh macaques. Generally, the values of viral load and the maintenance of CD4^+ T cells were associated with humoral responses. Otherwise, the viral genetic distances (divergence, diversity) were larger in animals (M419, M425) with their CD4^+ T cells profoundly depleted. Conclusion The model of SHIV-KB9/Ch Rh macaques displays a relatively slow progression to AIDS compared with Ind Rh macaques, which may more accurately reflect the potential of candidate vaccines in humans. 展开更多

In order to evaluate effects of Chinese medicinal herb (CMH) on growth and immunization of laying male chicks,eleven CMHs were used in this study,which are Astragalus membranacens (AM),Schisandra chinensis (SC),Ligustrum Lucidum(LL),Codonopsis,Scutellaria baicalensis (SB),Atractylodes macrocephala(AMA),Haw,Ginger,Acanthopanax scenticosns (AS),Angelica and Lycium,added to basal diet as 1%,respectively,with the basal diet supplemented with 50 mg·kg^(-1) bacitracin zinc(BZ) as the control.The body weight (BW) of birds were recorded at 1,21 and 42 days after birth,individually.The birds were vaccinated 0.50 mL against Newcastle disease (ND) with La sota vaccinel (containing mineral oil as adjuvant) by i.m.inoculation at 21 d of age.All of birds was vaccinated with F48E9 NDV by i.m.inoculation at 49 day of age.Blood samples were taken via wing vein from each bird on the day receiving the vaccination (Day-1) and on 14,21,28,30 and 34 d after vaccination (Days 14,21,28,30 and 34). Results showed that bodyweight (BW) and bodyweight gain(BWG) of the trial groups are similar to the control group.No significant differences of relative weight (RW) of bursa and spleen were observed among trial groups except for AS (P<0.05).Serum antibody titers of SC,LL,Codonopsis,SB,AMA and Lycium groups were significantly increased (P<0.05) in contrast to these control group on day-21 or 28 after immunized ND La sota vaccinel,which suggested that SC,LL,Codonopsis,SB,AMA and Lycium could augment antibody formulation.Furthermore,Compared with the control,antibody titers in SC,Codonopsis,AS,Lycium,SB and AMA group were higher (P<0.05) after vaccinated with F48E9 NDV,which suggested that SC,Codonopsis,AS,Lycium,SB and AMA have activity of antivirus.In conclusion,CMH used in the present study have similar effect on BW and BWG of chicks compared with the control.Moreover,these eleven CMH have little influence on RW of immunized organ except AS.However,SC,Codonopsis,LL,Lycium,SB and AMA could augment antibody formulation.Furthermore,SC,Codonopsis,AS,Lycium,SB and AMA have activity of antivirus compared with the control. 展开更多

Experiments were conducted to determine the effects of advanced vaccination and Chinese herbal adjuvants (CHA), containing astragalus polysaccharides (APS) and ginsenosides (GS) on the immune response to rabbit hemorrhagic disease (RHD) vaccine in young rabbits. In experiment 1, 5 New Zealand rabbits of each group at 30, 35, 40, or 45 days of age were injected with 2 mL of inactivated RHD vaccine, respectively. The dynamic changes of antibody titers were tested by the hemagglutination inhibition (HI) method. In experiment 2, 30 New Zealand rabbits at 35 days of age were randomly assigned to 5 treatment groups, representing inoculation with 3 mL of non-adjuvant RHD vaccine, CHA-RHD vaccine, CHA-HA vaccine (half dose antigen), aluminium adjuvant-RHD vaccine, and PBS, respectively. The dynamic changes of peripheral lymphocyte proliferation and serum antibody titers were tested by the MTT method and the HI method. The results showed that the titer of maternal HI antibody in the 35-day-old rabbits was lower than the protective level of 3 log2, while on days 7 to 49 after the vaccination, the antibody titers were higher than 3 log2. The CHA promoted the lymphocyte proliferation and enhanced the serum antibody titer (P〈 0.05). These findings from the two experiments suggested that advanced vaccination and Chinese herbal adjuvants significantly enhanced the immune response to vaccine against RHD, and effectively protected the young rabbits against RHD challenge. 展开更多

BACKGROUND Reactivation of hepatitis B virus(HBV)during anticancer treatment is a critical issue.When treating patients with solid tumors,it is unclear whether specific cancer types or treatments affect HBV reactivation in hepatitis B surface antigen(HBsAg)-negative and hepatitis B core antibody(HBcAb)-positive patients,socalled de novo hepatitis B patients.The risk of de novo hepatitis B may vary based on different background factors.AIM To determine the frequency and risk factors for de novo hepatitis B during solid tumor treatment.METHODS This retrospective cohort study comprised 1040 patients without HBsAgs and with HBcAbs and/or hepatitis B surface antibodies(HBsAbs).The patients were treated for solid cancer from 2008 to 2018 at the National Kyushu Cancer Center and underwent HBV DNA measurements.Patient characteristics and disease and treatment information were investigated.HBV DNA measurements were performed using TaqMan polymerase chain reaction(PCR).To identify the risk factors associated with HBV DNA expression,the age,sex,original disease,pathology,treatment method,presence or absence of hepatitis C virus(HCV),and HBsAb and/or HBcAb titers of all subjects were investigated.In patients with HBV DNA,the time of appearance,presence of HBsAgs and HBsAbs at the time of appearance,and course of the subsequent fluctuations in virus levels were also investigated.RESULTS Among the 1040 patients,938 were HBcAb positive,and 102 were HBcAb negative and HBsAb positive.HBV DNA expression was observed before the onset of treatment in nine patients(0.9%)and after treatment in 35 patients(3.7%),all of whom were HBcAb positive.The HBV reactivation group showed significantly higher median HBcAb values[9.00(8.12-9.89)vs 7.22(7.02-7.43),P=0.0001]and significantly lower HBsAb values(14 vs 46,P=0.0342)than the group without reactivation.Notably,the reactivated group showed a significantly higher proportion of cancers in organs related to digestion and absorption(79.0%vs 58.7%,P=0.0051).A high HBcAb titer and cancers in organs involved in digestion and absorption were identified as independent factors for HBV reactivation(multivariate analysis,P=0.0002 and P=0.0095).The group without HBsAbs tended to have a shorter time to reactivation(day 43 vs day 193),and the frequency of reactivation within 6 mo was significantly higher in this group(P=0.0459)than in the other group.CONCLUSION A high HBcAb titer and cancers in organs involved in digestion and absorption are independent factors that contribute to HBV reactivation during solid tumor treatment. 展开更多

The effects on immune function of broilers was studied by adding different doses of Shuanghuanglian propolis oral liquid in drinking water, to provide a theoretical basis for application and promotion of Shuanghuanglian propolis oral liquid. Ninety one-day-old broilers were randomly divided into three groups : group I was designed as control; groups Ⅱ and Ⅲ were added with 0.5 and 1.0 mL/L Shuanghuanglian propolis oral liquid in drinking water, respectively. Compared with group I, all tested indicators in groups Ⅱ and Ⅲ were increased at varying degrees as follows : bursa of fabricius index increased by 5.74% (P 〈 0.05 ) and 7.66% (P 〈 0.05 ) ; thymus index increased by 1.47% (P 〉 0.05 ) and 2.06% (P 〈 0.05 ) ; spleen index increased by 5.88% (P 〉 0.05 ) and 7.84% ( P 〈 0.05 ) ; Newcastle disease virus (NDV) antibody titer increased by 11.99% (P 〈 0.05 ) and 16.94% (P 〈 0.05 ) ; IL-2 content increased by 2.71% (P 〉 0.05) and 4.52% (P 〈 0.05 ) ; IL -6 content increased by 4.04% (P 〉 0.05 ) and 7.26% ( P 〈 0.05 ), respectively. The results showed that adding 0.5 and 1.0 mL/L Shuanghuangllan propolis oral liquid in drinking water could improve the immune function of broiler and immune effect of ND vaccine. 展开更多

It is due to tuboperitoneal lesions mainly because of infection;especially sexually transmitted infection. Worldwide, Chlamydia trachomatis is the main pathogen. In our setting, some studies failed to establish the link between tubal infertility and chlamydia trachomatis. The current study aimed to determine the local data related to chlamydia trachomatis role in tubal infertility and the usefulness of Chlamydia trachomatis antibody titer test (CAT) in discrimination of the patients with and without tuboperitoneal lesions. Patients’ average age was 33.9 ± 4.8 years, average coitarche 19.4 ± 4.4 years and average number of partners: 3.1 ± 1.6. The level of CAT is correlated to the tuboperitoneal severity. CAT was more specific (93.3%;CI 95%: 81.7 - 98.6) than sensitive (72.7% CI 95%: 49.8 - 89.3) and discriminated correctly 89% (AUC = 0.89) of the patients with or without tuboperitoneal lesions. In conclusion, as it is stated worldwide, Chlamydia trachomatis is the most frequent sexually transmitted pathogen associated with tubal infertility. CAT has to be used as a tool to select patients to be submitted to invasive investigation, like laparoscopy. 展开更多

Novel coronavirus,SARS-CoV-2 is responsible for causing a pandemic that has affected individuals worldwide,over 192 million people and about 4.1 million people died so far.The spread is ongoing and the numbers are still increasing.Numerous therapeutic approaches have been explored and developed during this pandemic.Immunotherapy with virus-specific antibodies in convalescent plasma(CP)has shown potential benefits for various pathogenic diseases.In many instances,it is the only available and safe management option for the COVID-19 patients.Here we describe two confirmed cases of COVID-19 from two different geographical areas that were managed with standard treatment modalities initially.Both of the patients were presented with high-grade fever,dry cough,and sore throat.Lab reports showed increased values of D-dimer,serum ferritin,leukocyte count(LC),Lactate dehydrogenase(LDH),and C-reactive protein(CRP).Chest X-ray showed bilateral infiltration(multifocal and bilateral ground-glass opacities and consolidations with peripheral and basal predominance),consistent with the previous reports on COVID-19 infection.The patients received conservative treatment according to the hospital's protocol.The convalescent plasma(from recovered patients)infusion was the last treatment given to both patients.After the convalescent plasma transfusion,both patients showed a reduction of viral load,an increase of anti-SARS-CoV-2 IgG and IgM antibodies,reduction in lung infiltration,with no adverse events.However,further randomized controlled trials are needed to investigate the full scope of safety and efficacy(both short and long-term)of convalescent plasma therapy for COVID-19 and other related infections. 展开更多

Background:Several vaccines have been approved against COVID-19,and 5 have been used in Indonesia.Due to the decrease in antibody levels 3 to 6 months after the second dose of CoronaVac,healthcare workers received the third booster of mRNA vaccine(mRNA-1273)to increase the antibody level.This study aimed to evaluate the risk factors of anti-S-RBD IgG levels differences in healthcare workers.Methods:This study is a retrospective cohort study of 576 healthcare workers without previous SARS-CoV-2 infection who received 2 doses of CoronaVac and the third dose of mRNA-12736 months after the second dose.Blood samples were obtained 2nd,6th,12th,and 24th weeks after the second dose of CoronaVac vaccine administration,with mRNA-1273 booster on week 20.Quantitative measurements of IgG antibodies were performed with Elecsys Anti-SARS-CoV-2 S immunoassay.We identify the baseline factors predicting post-vaccination antibody titers using univariate and multivariate linear regression analysis.Results:This study comprised 576 participants aged 32 years old,72.05%female,and 45.84%from high-risk occupation subgroups.The median antibodies titer level on the 2nd,6th,12th,and 24th weeks after the second vaccine dose administration were 40.99 u/mL,42.01 u/mL,54.78 u/mL,and 23,225 u/mL.Antibody levels trended highest in female and younger age group(20-29 years old).Conclusions:The third dose of vaccine increased the quantitative SARS-CoV-2 spike IgG antibody titers and eliminated differences in antibodies titer by gender. 展开更多

Background:COVID-19 vaccines that trigger a strong secretory antibody response in breast milk may achieve effective passive protection of vulnerable newborns and breastfed infants of immunized mothers.The aim of this work was to investigate the presence of SARS-CoV-2 spike RBD-specific IgA and IgG antibodies in breast milk,5 and 9 weeks after vaccination with 3 doses of the protein subunit vaccine Abdala,compared to those found in breast milk from COVID-19-recovered women,collected at least 40 days after the infection.Methods:SARS-CoV-2 spike RBD-specific IgA and IgG antibodies were semi-quantified by indirect ELISA,using a homemade standard generated by pooling twenty breast milk samples with high absorbance values according to preliminary data.The validity of the standard curves was proved following the European Medicines Agency Guideline.Two breast milk samples from 2 unvaccinated women who had not been infected with COVID-19 were included as negative controls.Potentially neutralizing antibodies was assessed by a SARS-CoV-2 surrogate virus neutralization test.Results:High levels of anti-RBD IgA antibodies were detected in breast milk samples 9 weeks after vaccination and anti-RBD IgG antibodies rise from the fifth to the ninth week.In the post-COVID-19 time that was evaluated,the IgG-type response was notably higher compared to both post-vaccination periods.Neutralizing antibody titers were similar in breast milk from vaccinated and COVID-19 recovered women.Conclusions:This is the first report about the immune response in breast milk after the administration of a COVID-19 protein subunit vaccine,which could provide analogous protection to that conferred by SARS-CoV-2 infection.This implies a potential passive immunity that breastfed infants receive from their mothers vaccinated with Abdala. 展开更多
