Changes in Polyphenols and Antioxidant Activities of Yingshan Yunwu Tea during Digestion in Vitro

[Objectives]To explore the change rule of polyphenol content and antioxidant activity of coarse old leaves of Yingshan Yunwu Tea in the process of human digestion.[Methods]The coarse and old leaves of Yunwu tea in Yingshan,Huanggang,Hubei Province were selected as the research object,and their digestion in vitro was simulated.The total polyphenol content was determined by Folin-phenol reagent colorimetric method,and the DPPH radical scavenging activity and total antioxidant activity were determined.[Results]After simulated gastrointestinal digestion in vitro,the polyphenol content and antioxidant activity of coarse old leaf tea soup showed a downward trend.After gastrointestinal digestion,the polyphenol content in tea infusion separately decreased by 31.8%and 8.5%;the scavenging rate of DPPH free radical was 97%before digestion,decreased to 92%after gastric digestion and 65%after intestinal digestion,which decreased by 5%and 27%,respectively;after gastrointestinal digestion,the total antioxidant capacity of tea soup decreased by 4.7%and 3.1%,respectively.[Conclusions]This study provided a reference for the development and application of coarse old leaves of Yingshan Yunwu tea,and provided a reference for the nutritional value evaluation and comprehensive utilization of coarse old leaves,so as to make the best use of coarse tea leaves and reduce the waste of resources. 展开更多

Objective:Polyphenols are complex compounds containing multiple phenolic hydroxyl groups.They are widely distributed in plants and have antioxidant activities.Whether the antioxidant activities of the cultivated varieties of Echinacea are similar to or better than those of the wild ones and the relationship between the accumulation of polyphenols and their antioxidant activities are still not clear.Methods:Folin-Ciocalteu method,high performance liquid chromatography(HPLC),2,2-diphenyl-1-picrylhydrazyl(DPPH)radical scavenging assay,ferric ion reducing antioxidant power(FRAP)assay,2,2'-azino-bis(3-ethylbenzothiazoline-6)-sulfonic acid(ABTS)radical scavenging assay,and Fe^(2+)chelating ability assay were used,respectively,to detect the total polyphenols and 5 kinds of caffeic acid derivatives(chicoric acid,caffeic acid,caftaric acid,chlorogenic acid,and 1,5-dicaffeoylquinic acid)in the roots,stems,leaves,and flowers,and the antioxidant activities of 3 varieties of Echinacea:E.purpurea L.,cultivar E.purpurea'Aloha',and E.purpurea'White Swan'.Results:E.purpurea L.had the highest contents of total polyphenols,5 caffeic acid derivatives and antioxidant activities,followed by E.purpurea'White Swan'and E.purpurea'Aloha',respectively.E.purpurea'White Swan'had the strongest ability to remove the DPPH,ABTS·^(+)and free radicals,and to chelate Fe^(2+);E.purpurea L.had the strongest ability to reduce FRAP.The correlation analyses revealed that the contents of total polyphenols and caffeic acid derivatives of E.purpurea L.and E.purpurea'White Swan'were correlated with their antioxidant activities.Conclusion:E.purpurea L.was the most appropriate material for the development of medicinal plants.E.purpurea'White Swan'could be used as a substitute for E.purpurea L.in terms of its antioxidant activity. 展开更多

Qingke(highland hull-less barley)is a grain replete with substantial nutrients and bioactive ingredients.In this study,we evaluated the effects of boiling(BO),steaming(ST),microwave baking(MB),far-infrared baking(FB),steam explosion(SE),and deep frying(DF)on bioactive components,phenolic compounds,and antioxidant activities of Qingke compared with the effects of traditional roast(TR).Results showed that the soluble dietary fiber,beta-glucan and water-extractable pentosans of Qingke in dry heat processes of TR,SE,MB and FB had a higher content compared with other thermal methods and had a better antioxidant activity of hydroxyl radical scavenging and a better reduction capacity,while those in wet heat processes of BO and ST had a better antioxidant activity of ABTS radical scavenging and a better Fe^(2+) chelating ability.DF-and SE-Qingke had a higher content of tocopherol,phenolic,and flavonoid.Overall,6 free phenolic compounds and 12 bound phenolic compounds of Qingke were identified,and free phenolic compounds suffered more damage during thermal processing.Principal component analysis showed that SE had more advantages in retaining and improving the main biological active ingredients of Qingke,and it may be the best method for treating Qingke. 展开更多

The present study was conducted to assay the antioxidant activities of essential oils and ethanol extracts of fennel(Foeniculum vulgare Mill.)seeds from Egypt and China.The major components of the Egyptian fennel essential oil were estragole(51.04%),limonene(11.45%),l-fenchone(8.19%)and trans-anethole(3.62%)by GC–MS analysis.Whereas,the major constituents of Chinese fennel essential oil were transanethole(54.26%),estragole(20.25%),l-fenchone(7.36%)and limonene(2.41%).The fennel seed extracts from Egypt and China contained appreciable levels of total phenolic contents(42.24 and 30.94 mg PE/g,respectively).The extracts exhibited good DPPH radical scavenging capacity with IC50(6.34 and 7.17 mg/g),respectively.A high variation in free radical scavenging activities of essential oils was observed.The Chinese fennel essential oil showed high activity in DPPH radical scavenging with IC50(15.66 mg/g).The Egyptian fennel essential oil showed very low activity with IC50(141.82 mg/g).The results of the present investigation demonstrated significant variations in the antioxidant activities of fennel essential oils and extracts from Egypt and China. 展开更多

Brown algae are well known as a source of biologically active compounds, especially those having antioxidant activities, such as phlorotannins. In this study we examined the antioxidant activities of crude phlorotannins extracts(CPEs) obtained from Sargassum hemiphyllum(SH) and fractionated according to the molecular weights. When CPEs were administrated at a dose of 30 mg/kg to Kunming mice pre-treated with carbon tetrachloride(CCl4), the levels of oxidative stress indicators in the liver, kidney and brain were significantly reduced in vivo. All the components of various molecular weight fractions of CPEs exhibited greater scavenging capacities in clearing hydroxyl free radical and superoxide anion than the positive controls gallic acid, vitamin C and vitamin E. Particularly, the components greater than 30 k D obtained from ethyl acetate phase showed the highest antioxidant capacities. These results indicated that SH is a potential source for extracting phlorotannins, the algal antioxidant compounds. 展开更多

Phenolic compounds and its antioxidant activity of extracts from seven cultivars of Chinese jujubes were investigated by high performance liquid chromatography(HPLC)with standards and different antioxidant evaluation methods,such as phosphomolybdenum assay,superoxide radical scavenging activity(SRSA),hydroxyl radical scavenging activity(HRSA),antihemolytic activity and inhibition of lipid peroxidation in rat liver homogenate,respectively.The results showed the components of the extracts are comprised of total phenols and flavonoids,and its content ranges from 454.3 to 1298.9(GAE mg/100 g dry weight).Phlorizin,catechin,gallic acid,chlorogenic acid,caffeic acid were the predominant phenolic compounds.All the extracts had significant antioxidant activities either in vitro or in vivo.Correlation analysis indicated that the antioxidant capacities of Chinese jujube extracts demonstrated a good positive relationship with some phenolic acids,which was higher in Xiao and Goutou.The results indicated that Xiao and Goutou could be attributed to a potential source of natural antioxidants for food applications. 展开更多

Both ethanol extracts and water extracts had high ability of 1, 1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging, 2, 2'-azinobis(3-ethylbenzthiazoline-6-sulphonic acid)(ABTS) radical scavenging and ferric reducing antioxidant power(FRAP). The ethanol extract of Sichuan pepper showed the highest ability of DPPH radical scavenging. The extract with the highest ABTS radical scavenging effect was that of ethanol extract of cinnamon. Both ethanol and water extracts of cinnamon possessed the high ferric reducing antioxidant power(FRAP) with the values of 4 541.87 and 1 134.52 μmol of Fe(Ⅱ)/g. The extracts with high hydroxyl radical-mediated deoxyribose degradation were all the ethanol extracts as follows: cinnamon, bay leaf, Sichuan pepper, star anise and fennel. The extracts with high antioxidant activities also had the high contents of the phenols. The study indicated that these spices might be potentially be used as natural antioxidants in food. 展开更多

In this study, we measured total anthocyanins and flavonols of onions of three colors. The fresh weight(FW) of total anthocyanins were(29.99±1.19),(9.64±1.30) and(0.75±0.40) mg 100 g^–1 fruit in red, yellow and white onions, respectively. Likewise, the FW of total flavonoids were(111.10±5.98),(36.64±3.59), and 0 mg 100 g^–1 in red, yellow and white, respectively. Four types of anthocyanins(delphinidin 3,5-diglycosides, cyanidin 3,5-diglycosides, cyanidin 3-glycosides and cyanidin 3-(6′′-malonyl)-glucopyranoside) and two kinds of flavonoids(quercetin and quercetin 3-glycosides) were identified in two varieties(red and yellow bulb) of onions by HPLC/DAD-ESI/MS. The total polyphenol contents were also measured by means of Folin-Ciocalteu method. Moreover, all the concentrations of anthocyanins, flavonoids and polyphenols showed significantly positive correlations with antioxidant activities measured by DPPH˙, ABTS˙+ and FRAP assays. This study was carried out to characterize total residual catechins and their fractions, polyphenolic contents and antioxidant activities of black teas enriched with high levels of theaflavin-3,3’-digallate. The made teas were processed from eleven selected cultivars. A comparative study was carried out between the processed teas and those from commercially grown Kenyan cultivars in relation to the above chemical parameters. A correlation matrix analysis was also conducted to find out whether a relationship existed between the antioxidant activities and the said chemical parameters. The total residual catechins were found to range between 3.10% and 8.08%. The total polyphenol levels varied between 19.00% and 28.90%, while the antioxidant activities of the teas ranged from 82.70% to 91.70%. There was a significant p < 0.001 correlation between the antioxidant activity and total polyphenols (r = 0.8948). There was also a high correlation p < 0.001 between the antioxidant activity and total catechins (r = 0.8878). Out of the four catechin fractions, the antioxidant activity correlated most with EGCG (r = 0.8774). The total polyphenolic contents and antioxidant activities for most of the cultivars were comparable to those of the green tea reference standard. From the figures obtained, it can be concluded that the most of the newly developed black teas of the selected cultivars have higher quality and enhanced antioxidant activities and that they can be recommended for commercial production. 展开更多

The antioxidant activity of solvent extracts from two main bamboo species, moso bamboo (Phyllostachys pubescens) and madake bamboo (P. bambusoides) in Japan, was first evaluated by scavenging free radical of 1,1-diphenyl-2-picrylhydrazyl (DPPH), the inhibition activity for peroxidation of linoleic acid, and the reduction power. The methanol-extracts of moso bamboo culms and madake bamboo leaves presented stronger antioxidant activity compared with DPPH scavenging activity. Methanol-extract of moso bamboo culms was further fractionated by different solvents and n-butanol soluble fraction exhibited the most significant activity in the DPPH scavenging assay. The fractionation of n-butanol soluble extract was isolated by silica gel column with gradient mixture solvent of chloroform and methanol. The isolated fractions were directed by the antioxidant activity measured by scavenging the stable DPPH free radical. It is of great significance to study the relationship between the excited state intramolecular proton transfer(ESIPT)properties and antioxidant activities of compounds in the field of life sciences.In this work,two novel compounds 5HF-OMe and 5HF-NH2 are designed through introducing a methoxy-and amino-group into the structure of 5-hydroxyflavone(5HF)respectively.The relationship between the ESIPT reaction and antioxidant activities of the three compounds is studied via the density functional theory(DFT)and time-dependent DFT(TD-DFT)methods.The calculated potential energy curves suggest that the rate of ESIPT reaction will gradually slow down from 5HF to 5HF-OMe and 5HF-NH2.In addition,the antioxidant activities of the three compounds gradually enhance from 5HF to 5HF-OMe and 5HF-NH2,which can be seen from the calculated energy gaps and ionization potential values.Interestingly,the above results imply that the rate of ESIPT reaction has a negative relationship with the antioxidant activities of the compounds,i.e.,the slower rate of ESIPT reaction will reflect the higher antioxidant activity of the compound,which will provide valuable reference for detecting the antioxidant activity of compound via the photophysical method. 展开更多

[Objectives]To explore the antimicrobial and antioxidant activities of flavonoid and polysaccharide extracts from bulbs of Lycoris aurea.[Methods]The disc diffusion method was used to evaluate the antimicrobial activity of flavonoid and polysaccharide extracts from the bulbs of L.aurea.The antioxidant activity was determined by scavenging of hydroxyl and 1,1-diphenyl-2-picrylhydrazine(DPPH)free radicals,and reducing ability.[Results]Flavonoids were effective on 7 microorganisms,but polysaccharides had no effect.Flavonoids had better hydroxyl free radical scavenging activity and reducing capacity than polysaccharides,but weaker than ascorbic acid.However,flavonoids had similar DPPH free radical activity to ascorbic acid.The strong antibacterial and antioxidant activities of flavonoids were mainly attributed to the high content of total phenols in the extract,which was(76.71±4.42)mg/g.[Conclusions]The study shows that flavonoids from bulbs of L.aurea could be regarded as a new potential source of natural bacterial inhibitors and antioxidants. 展开更多

Diabetes mellitus is a socially significant disease characterized by chronic hyperglycemia and metabolic disorders of proteins, carbohydrates, and lipids due to reduced function of insulin. Medicinal plants, rich in bioactive components that promote prevention and treatment, are inexpensive and no side effects. <i>Apium graveolens</i>, <i>Cleome gynandra</i>, and <i>Hibiscus sabdariffa</i> from Burkina Faso were investigated for their phytochemical profile and antioxidant activities. The high-performance thin-layer chromatography profile revealed flavonoids, tannins, and sterols in these herbaceous. The <i>Hibiscus sabdariffa</i> methanolic extract exhibited the highest total phenolic (138.4 ± 0.5 mg GAE/g DW) and flavonoid (52.8 ± 0.6 mg RuE/g DW) contents comparatively to <i>Cleome gynandra</i> and <i>Apium graveolens</i>. <i>Hibiscus sabdariffa</i> methanolic extract also presented the highest antioxidant activity (IC<sub>50</sub> = 0.31 ± 0.002 mg/mL) using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity assay. A high correlation between flavonoid contents and hydrophilic antioxidant activities (r = 0.99) was observed, indicating that flavonoids contribute significatively to these herbaceous antioxidant properties. <i>Apium graveolens</i>, <i>Cleome gynandra, and <i>Hibiscus sabdariffa</i> constitute a natural source of phenolic compounds that could be exploited in diabetes mellitus management.</i> 展开更多

More and more Parkia biglobosa plants’ stem barks, and roots are collected for traditional medicine uses. This work aimed to quantify Parkia biglobosa plant parts total polyphenols, total flavonoids contents and their antioxidant activities. The hypothesis was that the plant parts would have different phytochemical contents. So, leaves, stem barks, root barks and mature fruit pulp were collected. Alongside the leaves, the stem and root barks were sliced, air-dried at room temperature for 30 days. Thereafter, the samples were crushed and sieved. Following this, the powders were extracted with distilled water by maceration and decoction. As a result, the decoction was more efficient than the maceration. Leaves had the highest total polyphenol content (14.68), followed by stem barks (11.69), and root barks (9.09 ± 0.43 mg GAE/g) (0.0001 ≤ p ≤ 0.0002). However, for total flavonoid contents, the stem barks were better than the other parts. Indeed, stem barks delivered 0.74 and were followed by leaves for 0.6, and root bark for 0.49 ± 0.02 mg EQ/g (0.0001 ≤ p ≤ 0.0169). Finally, the antioxidant activities were 3.07 and 2.86 ± 0.1 μmol T.E/g, respectively for stem bark, and leaves (p = 0.0532). In conclusion, there is no need to debark the stems, because, in decoction, leaves and stem barks would have the same efficacy. 展开更多

Tea is the most widely consumed beverage throughout the world, after water and is considered as a good antioxidant nutrient against free radical reactions. Lemons are citrus fruits that are very common in many parts of the world and are well known for their health benefits. In this study, different assays were used to evaluate total phenolic content and antioxidant activities of green tea with and without lemon. Our results showed that green tea with lemon has higher phenolic content, stronger reducing power than green tea without lemon;and exhibits a significant inhibition of 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>). However, both samples exhibited high ferrous chelating activity with no significant difference among each other indicating a good antioxidant potential for both. This is a comparative study between green tea and green tea supplemented with lemon. The study contributes to a better understanding about how the addition of supplements like lemon might influence the antioxidant activity of tea. 展开更多
