Results BAFF immunostaining appeared on the cellular membrane and in the cytoplasm of tubal epithelial cells. Both BAFF protein and mRNA in normal fallopian tubes had lower levels than those in inflamed fallopian tubes (P〈0.01). Conclusion BAFF protein and mRNA are present in human tubal tissues. 目的克隆TNF家族B细胞激活因子(B cell activating factor to the TNF family,BAFF)胞外区134~285(sBAFF134-285)氨基酸残基段缺失突变体(s△BAFF)的cDNA,并进行表达及纯化.方法以构建的重组质粒pUC19/sBAFF为模板,采用一步反向...目的克隆TNF家族B细胞激活因子(B cell activating factor to the TNF family,BAFF)胞外区134~285(sBAFF134-285)氨基酸残基段缺失突变体(s△BAFF)的cDNA,并进行表达及纯化.方法以构建的重组质粒pUC19/sBAFF为模板,采用一步反向PCR法,扩增缺失编码sBAFF的142~160位氨基酸的核苷酸序列.经测序证实后,克隆入原核表达载体pQE-80L.经IPTG诱导表达,SDS-PAGE和Western blot检测表达产物,Ni2+-NTA柱层析纯化目的蛋白.结果经一步反向PCR扩增后得到401 bp的DNA片段,该片段序列与GenBank报道的编码人△BAFF的胞外区(s△BAFF)cDNA序列一致.含s△BAFF的表达载体在大肠杆菌中可表达出相对分子质量为18 000的蛋白质并以包涵体的形式存在,经Ni2+-NTA柱层析纯化后得到高纯度的目的蛋白.结论已成功地制备出人s△BAFF蛋白,为其功能的研究创造了条件.展开更多

目的 分析程序性死亡因子-1(programmed death factor-1,PD-1)、程序性死亡配体-1(programmed death-ligand-1,PDL1)、TNF家族B细胞活化因子-R(B cell-activating factor-R belonging to the TNF family,BAFF-R)在弥漫大B细胞淋巴瘤组...目的 分析程序性死亡因子-1(programmed death factor-1,PD-1)、程序性死亡配体-1(programmed death-ligand-1,PDL1)、TNF家族B细胞活化因子-R(B cell-activating factor-R belonging to the TNF family,BAFF-R)在弥漫大B细胞淋巴瘤组织中的差异性表达及临床意义。方法 以延安大学附属医院血液科2019年1月至2020年4月初诊的28例弥漫性大B细胞淋巴瘤组织及瘤旁组织样本作为研究对象,采用免疫组化法检测其PD-1、PD-L1及BAFF-R表达,然后统计分析PD-1、PD-L1及BAFF-R表达与病人临床病理特征的关系。结果 弥漫性大B细胞淋巴瘤组织PD-1、PD-L1及BAFF-R阳性表达积分分别为(5.44±0.79)、(5.58±0.86)及(4.49±0.65)分,瘤旁组织PD-1、PD-L1及BAFF-R阳性表达积分分别为(3.31±0.35)、(3.25±0.34)及(2.77±0.27)分,差异有统计意义(P<0.05);淋巴瘤组织PD-1表达差异与临床病理特征差异无统计学意义(P>0.05);淋巴瘤组织PD-L1表达与Ann Arbor/Costwards分期、病理分型及EBV是否阳性差异有统计意义(P<0.05);淋巴瘤组织BAFF-R表达与Ann Arbor/Costwards分期、病理分型、IPI评分及EBV是否阳性差异有统计意义(P<0.05)。结论 PD-L1及BAFF-R在弥漫性大B细胞淋巴瘤组织中呈高表达,且与弥漫性大B细胞淋巴瘤分期、病理分型及EBV感染密切相关,二者联合检测对病人预后评估可能具有较高价值;对难治性弥漫大B细胞淋巴瘤而言,以PD-L1及BAFF-R的信号通路为靶点,是潜在的免疫治疗方案。展开更多
