Inhibiting ceramide synthase 5 expression in microglia decreases neuroinflammation after spinal cord injury

Microglia,the resident monocyte of the central nervous system,play a crucial role in the response to spinal cord injury.However,the precise mechanism remains unclear.To investigate the molecular mechanisms by which microglia regulate the neuroinflammatory response to spinal cord injury,we performed single-cell RNA sequencing dataset analysis,focusing on changes in microglial subpopulations.We found that the MG1 subpopulation emerged in the acute/subacute phase of spinal cord injury and expressed genes related to cell pyroptosis,sphingomyelin metabolism,and neuroinflammation at high levels.Subsequently,we established a mouse model of contusive injury and performed intrathecal injection of siRNA and molecular inhibitors to validate the role of ceramide synthase 5 in the neuroinflammatory responses and pyroptosis after spinal cord injury.Finally,we established a PC12-BV2 cell co-culture system and found that ceramide synthase 5 and pyroptosis-associated proteins were highly expressed to induce the apoptosis of neuron cells.Inhibiting ceramide synthase 5 expression in a mouse model of spinal cord injury effectively reduced pyroptosis.Furthermore,ceramide synthase 5-induced pyroptosis was dependent on activation of the NLRP3 signaling pathway.Inhibiting ceramide synthase 5 expression in microglia in vivo reduced neuronal apoptosis and promoted recovery of neurological function.Pla2g7 formed a“bridge”between sphingolipid metabolism and ceramide synthase 5-mediated cell death by inhibiting the NLRP3 signaling pathway.Collectively,these findings suggest that inhibiting ceramide synthase 5 expression in microglia after spinal cord injury effectively suppressed microglial pyroptosis mediated by NLRP3,thereby exerting neuroprotective effects. 展开更多

目的探究母体血清免疫球蛋白G(IgG)抗A/B抗体联合新生儿脐带血总胆红素对新生儿溶血病(HDN)的诊断价值。方法收集2019年4月至2024年4月收治的母婴血型不合所致HDN新生儿82例为观察组,选择同期母婴血型不合健康新生儿77例为对照组。对比... 目的探究母体血清免疫球蛋白G(IgG)抗A/B抗体联合新生儿脐带血总胆红素对新生儿溶血病(HDN)的诊断价值。方法收集2019年4月至2024年4月收治的母婴血型不合所致HDN新生儿82例为观察组,选择同期母婴血型不合健康新生儿77例为对照组。对比2组新生儿母体分娩前血清IgG抗A/B抗体效价及脐带血总胆红素水平。绘制受试者工作特征(ROC)曲线,分析IgG抗体效价、脐带血总胆红素及二者联合对HDN的诊断价值。结果观察组母体血清IgG抗A/B抗体效价≤1∶32、1∶64的占比[3.66%(3/82)、8.54%(7/82)]低于对照组[48.05%(37/77)、24.68%(19/77)],1∶256、≥1∶512的占比[31.71%(26/82)、35.37%(29/82)]高于对照组[5.19%(4/77)、3.90%(3/77)],差异有统计学意义(P<0.05);观察组脐带血总胆红素水平[(34.38±7.28)μmol/L]高于对照组[(23.96±5.73)μmol/L],差异有统计学意义(P<0.05);ROC曲线分析显示,血清IgG抗A/B抗体效价、脐带血总胆红素联合诊断HDN的效能高于单独诊断,其敏感度为90.22%,特异度为89.56%,曲线下面积为0.936。结论母体血清IgG抗A/B抗体联合脐带血总胆红素对HDN的诊断价值较高。 展开更多

采用原位生长法制备了二元异质结光催化剂BiOCl/g-C_(3)N_(4)-Br,运用多种技术手段对其进行了表征,考察了BiOCl/g-C_(3)N_(4)-Br在模拟可见光照射下对罗丹明(RhB)的光催化降解性能。表征结果显示,BiOCl附着在g-C_(3)N_(4)-Br表面,二者... 采用原位生长法制备了二元异质结光催化剂BiOCl/g-C_(3)N_(4)-Br,运用多种技术手段对其进行了表征,考察了BiOCl/g-C_(3)N_(4)-Br在模拟可见光照射下对罗丹明(RhB)的光催化降解性能。表征结果显示,BiOCl附着在g-C_(3)N_(4)-Br表面,二者之间形成了Z-scheme异质结。实验结果表明:BiOCl/g-C_(3)N_(4)-Br催化剂比g-C_(3)N_(4)-Br和BiOCl具有更高的光催化活性,在RhB质量浓度20 mg/L、BiOCl/2%-g-C_(3)N_(4)-Br(g-C_(3)N_(4)-Br质量分数为2%)加入量0.5 g/L的条件下,经日光灯(14 W)照射90 min后,RhB去除率可达98%;经过4次循环实验后,RhB去除率从98%逐渐降低至90%,表现出良好的稳定性和可重复利用性。BiOCl/2%-g-C_(3)N_(4)-Br光催化降解RhB过程中发生的氧化还原反应分别在BiOCl和g-C_(3)N_(4)-Br的表面进行,BiOCl与g-C_(3)N_(4)-Br之间高度紧密的接触面形成了Z-scheme异质结,实现了电子和空穴的快速分离,降低了二者在催化剂内部的复合速率,提高了催化性能。 展开更多

Paramount concerns encompass spectrum allocation, the harmonization of network architectures, and inherent latency issues in satellite transmissions. Potential mitigations, such as dynamic spectrum sharing and the deployment of edge computing, are explored as viable solutions. Looking ahead, the advent of quantum communications within satellite frameworks and the integration of AI spotlight promising research trajectories. 岩藻多糖调控G3BP1/NF-κB信号通路影响胰腺癌细胞增殖

目的:探讨岩藻多糖对胰腺癌的影响,并分析其机制。方法:MTT法分析细胞增殖抑制率,GEPIA数据库分析G3BP1在胰腺癌组织中的表达水平及与生存率的关系。qRT-PCR分析GTP酶激活蛋白结合蛋白1(Ras-GTPase-activating protein binding proteins... 目的:探讨岩藻多糖对胰腺癌的影响,并分析其机制。方法:MTT法分析细胞增殖抑制率,GEPIA数据库分析G3BP1在胰腺癌组织中的表达水平及与生存率的关系。qRT-PCR分析GTP酶激活蛋白结合蛋白1(Ras-GTPase-activating protein binding proteins,G3BP1)水平,Western blot法分析p-NF-κBp65、NF-κBp65和IκB-α水平。免疫共沉淀检测G3BP1与p-NF-κBp65之间相互作用。敲低或G3BP1过表达,观察其对岩藻多糖调控细胞增殖以及NF-κB信号通路的影响。裸鼠成瘤实验验证岩藻多糖对裸鼠体内瘤体的瘤重、瘤体积及G3BP1、p-NF-κBp65、NF-κBp65和IκBα水平的影响。结果:1~32μg/mL岩藻多糖抑制capan-1细胞增殖,岩藻多糖48 h IC_(50)为7.729μg/mL。G3BP1在胰腺癌肿瘤组织中的表达明显高于正常组织,G3BP1高表达患者的生存率低于G3BP1低表达患者。2、4、8μg/mL岩藻多糖能下调G3BP1、p-NF-κBp65,上调IκBα水平。Co-IP实验发现G3BP1与p-NF-κBp65相互结合,并且岩藻多糖作用后结合能力降低。敲低G3BP1能促进岩藻多糖抑制capan-1细胞增殖,下调G3BP1、p-NF-κBp65,上调IκBα水平(P<0.05);G3BP过表达能下调岩藻多糖抑制capan-1细胞增殖效果,上调G3BP1、p-NF-κBp65,下调IκBα水平(P<0.05)。体内实验显示,敲低G3BP1能促进岩藻多糖减少瘤体体积、瘤体质量,下调瘤体G3BP1、p-NF-κBp65,上调IκBα水平(P<0.05)。结论:岩藻多糖抑制capan-1细胞增殖,对体内移植瘤抑瘤效果显著,其机制与调控G3BP1/NF-κB信号通路有关。 展开更多

目的ATP结合盒B亚家族成员1(ATP binding cassette subfamily B member 1,ABCB1)的异常表达在多种癌症的发生发展中发挥关键作用。然而,G蛋白偶联受体C家族5组A型(G protein coupled receptor family C group5 type A,GPRC5A)调控的ABCB... 目的ATP结合盒B亚家族成员1(ATP binding cassette subfamily B member 1,ABCB1)的异常表达在多种癌症的发生发展中发挥关键作用。然而,G蛋白偶联受体C家族5组A型(G protein coupled receptor family C group5 type A,GPRC5A)调控的ABCB1表达对肺腺癌增殖的影响仍不清楚。本研究探讨了GPRC5A调控的ABCB1表达对肺腺癌增殖的影响。方法我们采用RT-PCR、Western-blot或免疫组化实验,分析ABCB1在肺腺癌细胞系、人肺腺癌组织以及GPRC5A基因敲除小鼠和野生型小鼠的气管上皮细胞和肺组织中的表达。采用细胞计数试剂盒-8(CCK-8)分析GPRC5A基因敲除小鼠气管上皮细胞对化疗药物的敏感性。采用皮下肿瘤形成实验探讨下调ABCB1表达是否可抑制体内肺腺癌增殖。采用免疫荧光和免疫沉淀实验研究GPRC5A和ABCB1之间潜在的调控关系。结果ABCB1在肺腺癌细胞系和人类肺腺癌组织中表达上调。GPRC5A基因敲除小鼠的气管上皮细胞及肺组织的ABCB1表达高于野生型小鼠。与GPRC5A野生型小鼠的气管上皮细胞相比,GPRC5A基因敲除小鼠的气管上皮细胞对塔立奇达和多柔比星更敏感。注射移植细胞28天后,接受ABCB1基因敲除细胞移植的GPRC5A-/-C57BL/6小鼠的肺肿瘤的体积和重量均明显低于野生型细胞移植小鼠(P=0.0043,P=0.0060)。此外,免疫荧光和免疫沉淀实验表明,GPRC5A通过直接结合方式调控ABCB1的表达。结论GPRC5A通过抑制ABCB1表达降低肺腺癌增殖。GPRC5A调节ABCB1表达的途径有待研究。 展开更多

光催化降解技术高效、环保、节能的优点使其在环保领域具有广泛应用前景,为解决环境污染问题提供了新的思路。其中高效催化剂是实现污染物高效光催化降解的关键。g-C_(3)N_(4)半导体材料因优良的热稳定性和化学稳定性、易于合成以及能... 光催化降解技术高效、环保、节能的优点使其在环保领域具有广泛应用前景,为解决环境污染问题提供了新的思路。其中高效催化剂是实现污染物高效光催化降解的关键。g-C_(3)N_(4)半导体材料因优良的热稳定性和化学稳定性、易于合成以及能带结构易调控等优点在催化领域具有广阔的应用前景,但由于纯g-C_(3)N_(4)可见光利用率低、表面积小、导带位置较低,限制了其实际应用中的催化活性。本文通过光诱导还原方法实现了g-C_(3)N_(4)材料表面Ag/Ag_(2)O的原位负载,系统研究了Ag/Ag_(2)O负载量对材料催化性能的影响,实现低功率紫外光催化下水中罗丹明B的快速高效去除,研究发现50 mg的g-C_(3)N_(4)中在0.24 g/L的AgNO3溶液还原得到的g-C_(3)N_(4)/Ag/Ag_(2)O复合材料具有最佳的催化性能。同时将复合材料应用于实际水体中罗丹明B染料的催化降解,取得了良好催化效果。本研究为去除水环境有机物污染提供了一种高效、绿色的解决方法。 展开更多

弥漫大B细胞淋巴瘤合并淋巴浆细胞淋巴瘤/华氏巨球蛋白血症伴IgM及IgG共存的临床研究

目的 探讨弥漫大B细胞淋巴瘤合并淋巴浆细胞淋巴瘤/华氏巨球蛋白血症(LPL/WM)伴免疫球蛋白M(IgM)及免疫球蛋白G(IgG)共存的发病机制、诊断标准、治疗方案及预后。方法 分析1例弥漫大B细胞淋巴瘤合并LPL/WM伴IgM及IgG共存患者的临床资料... 目的 探讨弥漫大B细胞淋巴瘤合并淋巴浆细胞淋巴瘤/华氏巨球蛋白血症(LPL/WM)伴免疫球蛋白M(IgM)及免疫球蛋白G(IgG)共存的发病机制、诊断标准、治疗方案及预后。方法 分析1例弥漫大B细胞淋巴瘤合并LPL/WM伴IgM及IgG共存患者的临床资料,以“弥漫大B细胞淋巴瘤”“免疫球蛋白”“单克隆”“IgM”“IgG”“淋巴浆细胞淋巴瘤”“华氏巨球蛋白血症”“diffuse large B-cell lymphoma”“immune globulin”“monoclonal”“lymphoplasmacytic lymphoma”“Waldenstr9m macroglobulinemia”为检索词,对中国知网数据库、万方知识服务平台及PubMed数据库中2017年1月至2023年6月发表的文献进行检索。结果 该例患者明确诊断为弥漫大B细胞淋巴瘤合并LPL/WM伴IgM及IgG共存,确诊后经环磷酰胺+多柔比星+长春新碱+泼尼松(CHOP)方案、环磷酰胺+长春新碱+泼尼松(COP)方案治疗有效,但治疗期间病情进展,从发病到死亡存活10个月。依照上述检索条件并经过阅读文献题目与摘要筛选出7篇文献。阅读全文共筛出11例双克隆免疫球蛋白共存LPL/WM患者,其中IgM伴IgG双克隆6例,IgM伴免疫球蛋白A(IgA)双克隆2例,IgG伴IgA双克隆2例及IgMκ伴IgMλ双克隆1例。文献中提到的治疗方案主要包括硼替佐米+利妥昔单抗方案、COP方案、CHOP方案等,上述方案联合化疗均有效,但该病的发病机制尚不明确,尚无标准的治疗方案及确切的生存期。结论 弥漫大B细胞淋巴瘤合并LPL/WM伴IgM及IgG共存系首次报告,因该病发病率极低,发病机制尚不明确,因此目前尚无统一的治疗方案,预后尚不明确,临床需进一步提高对该病的认识,未来对于其发病机制及新型分子靶向药物的应用进行深入研究,将会大大改善患者的预后。 展开更多

Krill oil(KO)exhibits various biological activities,such as anti-inflammatory and antitumor effects.However,the inhibitory effects of benign prostatic hyperplasia(BPH)in vitro and in vivo have not yet been studied.This study investigated the anti-BPH effects of KO extracted by an enzymatic hydrolysis method.KO treatment inhibited the proliferation of WMPY-1 and BPH-1 cells by induction of G0/G1 phase arrest through the modulation of positive and negative regulators in both prostate cell types.KO treatment stimulated phosphorylation of c-Jun N-terminal kinase(JNK)and p38 signaling.In addition,KO changed the expression of BPH-related markers(5α-reductase,androgen receptor,FGF,Bcl-2,and Bax)and the activity of the proliferation-mediated NF-κB binding motif.KO-induced levels of proliferation-mediated molecules of prostate cells were attenuated in the presence of siRNA-specific p-38(si-p38)and JNK(si-JNK).Furthermore,the administration of KO alleviated prostate size and weight and the cell layer thickness of prostate glands in a testosterone enanthate-induced BPH rat model.KO treatment altered the level of dihydrotestosterone in serum and the expression levels of BPH-related markers in prostate tissues.Finally,KO-mediated inhibition of prostatic growth was validated by histological analysis.These results suggest that KO has an inhibitory effect on BPH in prostate cells in vitro and in vivo.Thus,KO might be a potential prophylactic or therapeutic agent for patients with BPH. 展开更多

Students'demand for online learning has exploded during the post-COvID-19 pandemic era.However,due to their poor learning experience,students'dropout rate and learning performance of online learning are not always satisfactory.The technical advantages of Beyond Fifth Generation(B5G)can guarantee a good multimedia Quality of Experience(QoE).As a special case of multimedia services,online learning takes into account both the usability of the service and the cognitive development of the users.Factors that affect the Quality of Online Learning Experience(OL-QoE)become more complicated.To get over this dilemma,we propose a systematic scheme by integrating big data,Machine Learning(ML)technologies,and educational psychology theory.Specifically,we first formulate a general definition of OL-QoE by data analysis and experimental verification.This formula considers both the subjective and objective factors(i.e.,video watching ratio and test scores)that most affect OLQoE.Then,we induce an extended layer to the classic Broad Learning System(BLS)to construct an Extended Broad Learning System(EBLS)for the students'OL-QoE prediction.Since the extended layer can increase the width of the BLS model and reduce the redundant nodes of BLS,the proposed EBLS can achieve a trade-off between the prediction accuracy and computation complexity.Finally,we provide a series of early intervention suggestions for different types of students according to their predicted OL-QoE values.Through timely interventions,their OL-QoE and learning performance can be improved.Experimental results verify the effectiveness oftheproposed scheme. 展开更多

In Beyond the Fifth Generation(B5G)heterogeneous edge networks,numerous users are multiplexed on a channel or served on the same frequency resource block,in which case the transmitter applies coding and the receiver uses interference cancellation.Unfortunately,uncoordinated radio resource allocation can reduce system throughput and lead to user inequity,for this reason,in this paper,channel allocation and power allocation problems are formulated to maximize the system sum rate and minimum user achievable rate.Since the construction model is non-convex and the response variables are high-dimensional,a distributed Deep Reinforcement Learning(DRL)framework called distributed Proximal Policy Optimization(PPO)is proposed to allocate or assign resources.Specifically,several simulated agents are trained in a heterogeneous environment to find robust behaviors that perform well in channel assignment and power allocation.Moreover,agents in the collection stage slow down,which hinders the learning of other agents.Therefore,a preemption strategy is further proposed in this paper to optimize the distributed PPO,form DP-PPO and successfully mitigate the straggler problem.The experimental results show that our mechanism named DP-PPO improves the performance over other DRL methods. 展开更多

BACKGROUND Phospholipase A2(PLA2)enzymes are pivotal in various biological processes,such as lipid mediator production,membrane remodeling,bioenergetics,and maintaining the body surface barrier.Notably,these enzymes play a significant role in the development of diverse tumors.AIM To systematically and comprehensively explore the expression of the PLA2 family genes and their potential implications in cholangiocarcinoma(CCA).METHODS We conducted an analysis of five CCA datasets from The Cancer Genome Atlas and the Gene Expression Omnibus.The study identified differentially expressed genes between tumor tissues and adjacent normal tissues,with a focus on PLA2G2A and PLA2G12B.Gene Set Enrichment Analysis was utilized to pinpoint associated pathways.Moreover,relevant hub genes and microRNAs for PLA2G2A and PLA2G12B were predicted,and their correlation with the prognosis of CCA was evaluated.RESULTS PLA2G2A and PLA2G12B were discerned as differentially expressed in CCA,manifesting significant variations in expression levels in urine and serum between CCA patients and healthy individuals.Elevated expression of PLA2G2A was correlated with poorer overall survival in CCA patients.Additionally,the study delineated pathways and miRNAs associated with these genes.CONCLUSION Our findings suggest that PLA2G2A and PLA2G12B may serve as novel potential diagnostic and prognostic markers for CCA.The increased levels of these genes in biological fluids could be employed as non-invasive markers for CCA,and their expression levels are indicative of prognosis,underscoring their potential utility in clinical settings. 展开更多

Beyond-5G(B5G)aims to meet the growing demands of mobile traffic and expand the communication space.Considering that intelligent applications to B5G wireless communications will involve security issues regarding user data and operational data,this paper analyzes the maximum capacity of the multi-watermarking method for multimedia signal hiding as a means of alleviating the information security problem of B5G.The multiwatermarking process employs spread transform dither modulation.During the watermarking procedure,Gram-Schmidt orthogonalization is used to obtain the multiple spreading vectors.Consequently,multiple watermarks can be simultaneously embedded into the same position of a multimedia signal.Moreover,the multiple watermarks can be extracted without affecting one another during the extraction process.We analyze the effect of the size of the spreading vector on the unit maximum capacity,and consequently derive the theoretical relationship between the size of the spreading vector and the unit maximum capacity.A number of experiments are conducted to determine the optimal parameter values for maximum robustness on the premise of high capacity and good imperceptibility. 展开更多

目的探讨G蛋白信号调节子(RGS)16及高迁移率族蛋白B1(HMGB1)蛋白表达在高级别宫颈鳞状上皮内病变(HSIL)冷刀锥切术(CKC)后复发中预测价值。方法选择2021年1月至2022年12月徐州医科大学附属连云港医院收治的患者均行CKC后且术后病理切缘... 目的探讨G蛋白信号调节子(RGS)16及高迁移率族蛋白B1(HMGB1)蛋白表达在高级别宫颈鳞状上皮内病变(HSIL)冷刀锥切术(CKC)后复发中预测价值。方法选择2021年1月至2022年12月徐州医科大学附属连云港医院收治的患者均行CKC后且术后病理切缘阴性的238例宫颈高级别鳞状上皮内病变(HSIL)患者,术中采集病灶组织,采用免疫组化法测定RGS16、HMGB1蛋白表达情况。术后随访1年,根据患者术后1年是否复发分为复发组与非复发组。对比2组RGS16、HMGB1蛋白表达情况,对两组临床资料进行单因素分析,将单因素分析差异有统计学意义的指标纳入多因素Logistic回归分析,筛选HSIL患者术后复发的影响因素。绘制受试者工作特征曲线(ROC),以曲线下面积(AUC)评估预测价值。结果随访1年,无失访,HSIL患者术后复发24例,复发率为10.08%,剩余214例均未复发。单因素分析结果显示,复发组产次、有腺体累及、术后人类乳头瘤病毒(HPV)持续感染、术后液基细胞学阳性例数占比、RGS16蛋白阳性表达率、HMGB1蛋白阳性表达率高于非复发组(P<0.05)。多因素Logistic回归分析结果显示,RGS16蛋白阳性表达、HMGB1蛋白阳性表达为HSIL患者术后复发的影响因素(P<0.05)。ROC曲线结果显示,RGS16蛋白、HMGB1蛋白阳性表达及二者联合预测HSIL患者术后复发的AUC值分别为0.740、0.790、0.818(P<0.05),且二者联合的AUC值更高(P<0.05)。结论RGS16蛋白、HMGB1蛋白阳性表达可用于预测HSIL患者术后复发风险,且二者联合具有更高的预测价值。 展开更多

Backscatter communications will play an important role in connecting everything for beyond 5G(B5G)and 6G systems.One open challenge for backscatter communications is that the signals suffer a round-trip path loss so that the communication distance is short.In this paper,we first calculate the communication distance upper bounds for both uplink and downlink by measuring the tag sensitivity and reflection coefficient.It is found that the activation voltage of the envelope detection diode of the downlink tag is the main factor limiting the back-scatter communication distance.Based on this analysis,we then propose to implement a low-noise amplifier(LNA)module before the envelope detection at the tag to enhance the incident signal strength.Our experimental results on the hardware platform show that our method can increase the downlink communication range by nearly 20 m. 展开更多
