One kind of novel BLMs was fabricated by patch-clamp pipette technology characterized in considerably sensitive to changes of electrochemical parameters.Detectiye currents and voltage presented linear relationship when BLMs was formed and it could be confirmed by Gramicidin method.Ion current was increased by dihexyl (C_ (12)) modified ssDNA fixed on the BLMs and also indicated linear relationship to ssDNA's concentration due to the interaction of (C_ 12)-ssDNA and BLMs.Further more,the regression equations were different from BLMs fixed with ssDNA probe and a blank control BLM in the same experimental conditions.The ssDNA probe was successfully fixed on patch-clamp pipette supported-BLMs.Based on our studies,a biosensor with reactive element of patch-clamp pipette-supported BLMs has been established. 展开更多

One of the critical issues from the application viewpoint is the stability of the bio-nano hybrid under mechanical perturbations. In this work we explore mechanical responses of the interface between lipid bilayer and graphene under hydrostatic coating provides remarkable resistance to the pressure or indentation loads, We find that graphene loads, and the intercalated water layer offers additional protection. These findings are discussed based on molecular dynamics simulation results that elucidate the molecular level mechanisms, which provide a basis for the rational design of bionanotechnology- enabled aoolications such as biomedical devices and nanotheraoeutics. 展开更多

Not only do they serve as the protective boundary of cells and their internal organelles, they also organize and host major parts of the biochemical machinery for cellular communication and transmembrane transport. To study aspects of cellular membranes in a controlled manner, solid supported planar bilayers have served as reliable tools for many decades. They have been used in a large variety of studies ranging from fundamental investigations of membranes and their constituents to the dissection of cellular signaling mechanisms. However, there are limitations to these systems and recently a class of new systems in which the lipid bilayer is supported on a soft, polymer cushion has emerged. However, it is well established that p... Biological cell membranes are complex structures containing mainly lipids and proteins. Functional aspects of such membranes are usually attributed to membrane integral proteins. However, it is well established that parameters of the lipid matrix are modifying the function of proteins. Additionally, electrical capacity and conductance of the plain lipid matrix of membranes are contributing directly to cellular functions as there is, for example, the propagation of action potentials. Accordingly the dependence of these parameters on changes of gravity might be important in the field of life sciences under space conditions. In this study consequently we have performed experiments in parabolic flight campaigns utilizing the patch-clamp technology to investigate conductance and capacity of plain lipid vesicle membranes under conditions of changing gravity. Both capacity and conductance were found to be gravity dependent. The changes in capacity could be contributed to changes in membrane geometry. Significant permeability in plain lipid membranes could be only observed at high potentials, where spontaneous current fluctuations occurred. 细胞膜是保护细胞免受外源性污染物破坏的屏障,在细胞与外界进行物质运输、能量转换、信息传递等活动中具有不可替代的作用。当前针对环境污染物诱导细胞膜损伤的研究相对零散,未成体系。本文聚焦近5年国内外环境污染物诱导细胞膜损伤... 细胞膜是保护细胞免受外源性污染物破坏的屏障,在细胞与外界进行物质运输、能量转换、信息传递等活动中具有不可替代的作用。当前针对环境污染物诱导细胞膜损伤的研究相对零散,未成体系。本文聚焦近5年国内外环境污染物诱导细胞膜损伤的相关研究并对其进行归纳总结,主要从脂质双分子层、膜蛋白、细胞骨架以及膜微结构域4个方面分析了环境污染物对细胞膜的损伤影响。从脂质双分子层结构变化、脂质过氧化以及膜通透性和完整性等方面描述了环境污染物对脂质双分子层的损伤;从膜蛋白活性、蛋白表达等方面阐述了环境污染物对细胞膜蛋白的损伤;从微管、微丝和中间纤维的变化归纳了环境污染物对细胞骨架的损伤;从脂筏和小窝的变化概括了环境污染物对细胞膜微结构域的损伤。最后,对环境污染物诱导细胞膜损伤的研究现状进行了思考,并对未来研究趋势进行了展望。 展开更多

研究了以C60镶嵌物修饰的固体支撑双层类脂膜(s-BLM)体系对I-的电化学传感行为。结果表明,在s-BLM膜中镶嵌C60,体系对I-的检测灵敏度达到10-7mol L。通过对s-BLM膜中镶嵌物的选择,可获得更高稳定性和检出限的离子传感器。

In this work, the previously developed coarse-grained (CG) particle models for biomembranes and red blood cells (RBCs) are reviewed, and the advantages of the CG particle methods over the continuum and atomistic simulations for modeling biological phenomena are discussed. CG particle models can largely increase the length scale and time scale of atomistic simulations by eliminating the fast degrees of freedom while preserving the mesoscopic structures and properties of the simulated system. Moreover, CG particle models can be used to capture the microstructural alternations in diseased RBCs and simulate the topological changes of biomembranes and RBCs, which are the major challenges to the typical continuum representations of membranes and RBCs. The power and versatility of CG particle methods are demonstrated:through simulating the dynamical processes mvolving significant topological .changes e.g. lipid self-assembly vesicle fusion and membrane budding. 展开更多

Research had shown that lipids change the risk for dementia, especially some omega-3 lipids appear to lower Alz-heimer’s risk, yet only limited research exists on the modulation of NMDA receptor channels by lipids. Here we review recent literature concerning molecular determinants that influence the NMDA receptor channel gating via membrane lipids and fatty acids with profound significance for understanding how altered NMDA signalling leads to neuronal cell death linked to age-related dementia’s. Future discovery of lipid-like modulators of NMDA receptor function offer the potential for the development of new bioceu-ticals and affordable nutritional supplements to combat neuronal degeneration as well as to promote well being and healthy aging. 展开更多
