Here, we further examined the combined effects of SM and DO on rat myocardial ischemia/reperfusion injury. The possible mechanism of SM and DO also were elucidated. Methods DO was divided into aqueous extract of lignum dalbergiae odoriferae (DOW) and lignum dalbergiae odoriferae oil (DOO). Sprague-Dawley rats were randomized to seven groups: sham group, model group, treatment groups inclu- ding SM (10 g · kg^-1), DOW (5 g · kg^-1), DOO (0.5 ml · kg^-1), SM + DOW (10 g · kg^-1 + 5 g · kg^-1), SM + DOO ( 10 g · kg^-1 + 0. 5 ml · kg^-1). Rats were pretreated with homologous drug for 7 days and then subjec- ted to 30 rain of ischemia followed by 180 rain of reperfusion. Electrocardiogram (ECG) and heart rate were moni- tored and recorded continuously. At the end of reperfusion, blood samples were collected to determine the serum levels of creatine kinase-MB (CK-MB) and lactate dehydrogenase (LDH). Hearts were harvested to assess heart- body rate, infarct size and histopathological changes as well. Maximum and minimum effective points were deter- mined by measuring indicators associate with myocardial injury at different time-points of reperfusion (Smin, 15min, 30min, 45rain, 60min, 120min, 180min). The potential therapeutic mechanism of SM and SM + DOO were carried out by detecting superoxide dismutase (SOD), malondialdehyde (MDA), tumor necrosis factor-alpha (TNF-alpha) and interleukin 6 (IL-6). Results The results showed SM and DO can ameliorate cardiac function respectively, and this cardioprotective effect was further strengthened by their combinations. Among all the combi- nations, SM + DOO showed predominant potential to improve ECG and heart rate, reduce heart-body rate (28.5% + 1.4% , P 〈 0.01 vs model) and myocardial infarct size ( 20.96% + 1.61% , P 〈 0.01 vs model, P 〈 0.05 vs SM) , attenuate histopathological damage, decrease the levels of CK-MB and LDH (P 〈 0.01 vs model, P 〈 0.05 vs SM). The maximum effective points of SM and SM + DOO were 15min and 30rain respectively, and the minimum effective points of them were 180rain. In reducing serum level of MDA, TNF-alpha, IL-6 and increasing SOD activ- ity, SM + DOO was similar to SM. Conclusion The results of this study indicated that SM + DOO have combined effects that are highly effective than single pretreatment against myocardial ischemie reperfusion injury in rats. The possible mechanism of SM and DO were likely through its anti-oxidant and anti-inflammatory properties, and thus may be an effective and promising medicine for both prophylaxis and treatment of ischemic heart disease.展开更多

Methods The compounds of Astragalus mongholicus Bunge were isolated by chromatography and the structures were elucidated on the basis of spectral data interpretation. Their antioxidant activities were detected by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities in a cell-free system. Meanwhile, the effects against XA/XO-induced toxicity were assessed using MTT assay in PC12 cells. Results Ten principal constituents were isolated and identified as formononetin (Ⅰ), ononin (Ⅱ), calycosin (Ⅲ), calycosin-7-O-β-D-glucoside (Ⅳ), 9,10-dimethoxypterocarpan-3-O-β-D-glucoside (Ⅴ), adenosine (Ⅵ), pinitol (Ⅶ), daucosterol (Ⅷ), β-sitoster (Ⅸ) and saccharose (Ⅹ) from Astragalus mongholicus Bunge. The compounds Ⅰ, Ⅲ, and Ⅳ scavenged DPPH free radicals in vitro. Formononetin and calycosin were found to inhibit XA/XO-induced cell injury significantly, with an estimated EC50 of 50 ng/mL. Conclusion Compound Ⅲ, Ⅵ, and Ⅶ are first reported in this plant. Calycosin exhibits the most potent antioxidant activity both in the cell-free system and in the cell system.展开更多

The seeds had a high kernel percentage (53.67%± 7.51), oil content (52.21%± 4.01), and biodiesel yield (99.77%± 0.21). Among the fatty acids in the oil were high percentages of linoleic acid (41.66 ± 2.26)% and oleic acid (28.44%± 2.03). Most of the fuel properties complied well with the ASTM D6751-10, EN 14214-08, and GB/T 20828-2014 standards. The 13 sampling areas were grouped into four clusters based on different kernel percentage, oil content, biodiesel yield, and fatty acid composition. The results showed that the quality of kernel oils from seeds from Ar Horqin Banner was the best, although kernel oils from seeds in all 13 areas were suitable for biodiesel production. This study provides a basis for selecting optimal sites to harvest seeds from X. sorbifolia.展开更多

Methods Cell proliferation was measured by a newly developed cell proliferation reagent, WST-1. Cell apoptosis was assayed by flow cytometry through detecting annexin V. Nitric oxide production was evaluated using confocal laser scanning microscopy with diaminofluorescein diacetate (DAF-2, DA). Cell aldose reductase (AR) activity, as well as the effect of Epalrestat and interleukin-1β were also explored. Results WST assay showed that cell proliferation induced by serum was significantly inhibited by SAaB (P〈0.01). Flow cytometry analysis revealed that SAaB could enhance apoptotic rate of VSMCs (P〈0.01). Nitric oxide production was significantly enhanced after administration of SAaB and interleukin-Iβ Moreover, AR activity of VSMCs was also remarkably inhibited by both SAaB and Epalrestat (P〈 0.01). Conclusion SAaB can inhibit proliferation and enhance apoptosis of VSMCs. It may protect vascular cells by inhibiting VSMC proliferation and augmenting apoptotic rate of VSMCs via NO-dependent pathway.展开更多

In this study, an aged mouse model was established through intraperitoneal injection of D-galactose for 8 weeks, followed by treatment with 40, 20, or 10 mg/kg orientin, vitexin, or a positive control (vitamin E) via intragastric administration for an additional 8 weeks. Orientin, vitexin, and vitamin E improved the general medical status of the aging mice and significantly increased their brain weights. They also produced an obvious rise in total antioxidant capacity, superoxide dismutase, catalase, and glutathione peroxidase levels in the serum, and the levels of superoxide dismutase, catalase and glutathione peroxidase, Na+-K+-ATP enzyme, and Ca2+-Mg2+-ATP enzyme in the liver, brain and kidneys. In addition, they significantly reduced malondialdehyde levels in the liver, brain and kidney and lipofuscin levels in the brain. They also significantly improved the neuronal ultrastructure. The 40 mg/kg dose of orientin and vitexin had the same antioxidant capacity as vitamin E. These experimental findings indicate that orientin and vitexin engender anti-aging effects through their antioxidant capacities.展开更多

Methods The collagen synthesis of hepatocytes were stimulated by the addition of carbon tetrachloride (CCl 4 ) to the culture medium, the concentration of type procollagen (PC) in the culture medium and the hydroxyproline (Hyp) in hepatocytes were determined, as well as the activity of se dependent glutathione peroxidase (Se GSH Px) and the concentration of malondiadehyde (MDA) in the culture medium. Results A significant decrease in PC, Hyp and MDA production, and the significant increase in Se GSH Px activity were observed in the cultures pretreated with 1 g L -1 SMB for 4 hours compared with the untreated cultures. Analysis of the Se GSH Px/MDA ratio in SMB pretreated group showed more marked increase compared to that of the untreated group ( P <0.01). There was a significant negative correlation between the ratio of Se GSH Px/MDA and the concentration of PC in SMB pretreated group ( r=-0.9017, P <0.01). Conclusion Our results indicate that SMB may suppress the collagen synthesis of cultured human fetal hepatocytes stimulated by CCl 4 , and its mechanism may be related to the increase in Se GSH Px/MDA ratio and the enhancement of hepatocytes antioxidation capability.展开更多

The results showed that SMB had significant effect inhibiting lipid peroxidation of the hepatocytes when exposed to CCl4 as compared with the non-protective co4trol. SMB could decrease the production of malondialdehyde (MDA) and the release of ALT, and increase the synthesis of albumin (ALB) and the activity of Se-dependent glutothione peroxidase (Se-GSH-Px). These results suggest that the suppression of lipid peroxidation may be largely due to the increased activity of Se-GSH-Px.展开更多

The possible mechanism of SM and DO were likely through its anti-oxidant and anti-inflammatory properties, and thus may be an effective and promising medicine for both prophylaxis and treatment of ischemic heart disease.
文摘To evaluate the antioxidant activities of different chemical constituents from Astragalus mongholicus Bunge and their protection against xanthine (XA)/xanthine oxidase (XO)-induced toxicity in PC12 cells. Calycosin exhibits the most potent antioxidant activity both in the cell-free system and in the cell system.

文摘Oil content from seed kernels of Xanthoceras sorbifolia from 13 areas in China was analyzed by gas chromatography–mass spectrometry to determine oil characteristics and biodiesel properties. This study provides a basis for selecting optimal sites to harvest seeds from X. sorbifolia.

文摘Objective To investigate the effects of saponins from Anemarrhena asphodeloides Bunge (SAaB) (Botanical Name: Anemarrhena Asphodeloidis Rhizoma) on the growth of vascular smooth muscle cells (VSMCs). It may protect vascular cells by inhibiting VSMC proliferation and augmenting apoptotic rate of VSMCs via NO-dependent pathway.

文摘Total flavonoids are the main pharmaceutical components of Trollius chinensis Bunge, and orientin and vitexin are the monomer components of total flavonoids in Trollius chinensis Bunge. These experimental findings indicate that orientin and vitexin engender anti-aging effects through their antioxidant capacities.

Seeds with diameter φ 〉 2.0 mm were used as explants and achieved relatively high surviv- al rate of aseptic seedlings, which reached above 65%. [ Conclusion] This study established a surface sterilization method for tissue culture of H. ammodendron ( C. A. Mey. ) Bunge seeds.
文摘Objective The effects of salvia miltiorrhiza bunge (SMB) on collagen synthesis in the human fetal hepatocytes culture were studied. Conclusion Our results indicate that SMB may suppress the collagen synthesis of cultured human fetal hepatocytes stimulated by CCl 4 , and its mechanism may be related to the increase in Se GSH Px/MDA ratio and the enhancement of hepatocytes antioxidation capability.

文摘The role of salvia miltiorrhiza bunge (SMB) In protection of lipid peroxidation induced by CCl4 was studied in primary cultured human fetal hepatocytes. These results suggest that the suppression of lipid peroxidation may be largely due to the increased activity of Se-GSH-Px.
