<i>In-Vitro</i>Regeneration of <i>Citrus sinensis</i>(L.) Osbeck from Mature Seed Derived Embryogenic Callus on Different Solid Basal Media

In-vitro callus induction and regeneration method was developed using different plant growth regulators (PGRs), and basal media (Murashige and Skoog (MS), CHU (N6) and Gamborg (B5) media) of Citrus sinensis (L.) Osbeck. Observations of the effect of PGRs were carried out using different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D),1-naphthalene acetic acid (NAA) and combinations of 2,4-D and NAA using different basal media. This study found Citrus sinensis (L.) Osbeck exhibited a high frequency of callus induction on MS medium supplemented with 3 mg/L 2,4-D and callus induction frequency was 86.7% ± 3.4% whereas N6 and B5 showed lower callus induction frequency of 83.3% ± 8.8% and 82.2% ± 1.9% respectively compared to that of MS media with supplementation of the same hormone. Among the induced calli, the morphological analysis showed only 40% - 50% was embryogenic calli. Regeneration of plantlets from calli was done using different concentrations and combinations of auxin and cytokinin. The study showed that 3 mg/L 6-benzylaminopurine (BAP) supplemented medium has the maximum potential to promote regeneration of Citrus sinensis (L.) Osbeck from embryogenic calli with the frequency of 89.3% ± 8.8% but no regeneration occurred from the non-embryogenic calli. The regenerated plantlets were rooted on MS medium with supplementation of 5 mg/l NAA. These observations in Citrus sinensis (L.) Osbeck regeneration will be helpful for genetic improvement with desired traits. 展开更多

The results showed that B5 and WPM media were the optimal basal media and the optimal phyotohormone was BA of 1.0 mg/L or TDZ of 0.1mg/L; The concentrations of sucrose of 30g/L and agar of 6g/L were most suitable for the shoot regeneration; pH value from 5.8 to 6.6 were broadly effective, but the best at pH 5.8. 展开更多

以昆明小鼠的囊胚及胚胎干细胞为材料,用组成成份清楚的培养体系,研究不同的基础培养液(h-DMEM,DMEM/F12,Knockout DMEM)对昆明小鼠胚胎干细胞生长的影响。分别将小鼠囊胚置于以上3种培养液为基础的培养液(M1、M2、M3)中,4 d后观察原代... 以昆明小鼠的囊胚及胚胎干细胞为材料,用组成成份清楚的培养体系,研究不同的基础培养液(h-DMEM,DMEM/F12,Knockout DMEM)对昆明小鼠胚胎干细胞生长的影响。分别将小鼠囊胚置于以上3种培养液为基础的培养液(M1、M2、M3)中,4 d后观察原代集落形成率;取正常培养较小的胚胎干细胞集落,分别接种在上述培养液中培养观察;单细胞传代并观察结果;以碱性磷酸酶染色及OCT-4免疫荧光染色鉴定。3种基础培养基中原代集落形成率分别为100%,100%,96.3%;Knockout DMEM与DMEM/F12中的胚胎干细胞增殖的速度远快于h-DMEM中的增殖速度;单细胞传代的昆明小鼠胚胎干细胞都能形成集落;经AKP染色和OCT-4免疫荧光染色,证明3种培养体系中的胚胎干细胞均符合胚胎干细胞的特性。Knockout DMEM与DMEM/F12较DMEM更有利于胚胎干细胞的生长。 展开更多

Oberpr. and Vogt were studied in this report. A reliable protocol was developed for in vitro seed germination, multiplication and plantlet regeneration of Cladanthus mixtus. The seeds were sterilized and cultured in different media. Among the two basal media evaluated for symbiotic seed germination, gelled distilled water was found to be the best with a high percentage of seeds germination (100%) after 33 hours of culture. Seedlings were further transferred to different types of media. Result observed after 4 weeks showed that MS medium promotes the highest growth with an average of 2.75 ± 0.12 cm shoot length and 2.60 ± 0.29 shoots per explants, and the mean number of roots achieved 3.33 ± 0.17 root per explants with a length of 2.42 ± 0.16 cm. This study showed that macroelements of MS (1962) medium is essential for in vitro shoot multiplication, growth and rooting of shoots of Cladanthus mixtus L. 展开更多
