The collision cross-sections(CCS)measurement using ion mobility spectrometry(IMS)in combination with mass spectrometry(MS)offers a great opportunity to increase confidence in metabolite identification.However,owing to...The collision cross-sections(CCS)measurement using ion mobility spectrometry(IMS)in combination with mass spectrometry(MS)offers a great opportunity to increase confidence in metabolite identification.However,owing to the lack of sensitivity and resolution,IMS has an analytical challenge in studying the CCS values of very low-molecular-weight metabolites(VLMs250 Da).Here,we describe an analytical method using ultrahigh-performance liquid chromatography(UPLC)coupled to a traveling wave ion mobility-quadrupole-time-of-flight mass spectrometer optimized for the measurement of VLMs in human urine samples.The experimental CCS values,along with mass spectral properties,were reported for the 174 metabolites.The experimental data included the mass-to-charge ratio(m/z),retention time(RT),tandem MS(MS/MS)spectra,and CCS values.Among the studied metabolites,263 traveling wave ion mobility spectrometry(TWIMS)-derived CCS values(TWCCSN2)were reported for the first time,and more than 70%of these were CCS values of VLMs.The TWCCSN2 values were highly repeatable,with inter-day variations of<1%relative standard deviation(RSD).The developed method revealed excellent TWCCSN2 accuracy with a CCS difference(DCCS)within±2%of the reported drift tube IMS(DTIMS)and TWIMS CCS values.The complexity of the urine matrix did not affect the precision of the method,as evidenced by DCCS within±1.92%.According to the Metabolomics Standards Initiative,55 urinary metabolites were identified with a confidence level of 1.Among these 55 metabolites,53(96%)were VLMs.The larger number of confirmed compounds found in this study was a result of the addition of TWCCSN2 values,which clearly increased metabolite identification confidence.展开更多
Out-of-plane microneedle structures are widely used in various applications such as transcutaneous drug delivery and neural signal recording for brain machine interface.This work presents a novel but simple method to ...Out-of-plane microneedle structures are widely used in various applications such as transcutaneous drug delivery and neural signal recording for brain machine interface.This work presents a novel but simple method to fabricate high-density silicon(Si)microneedle arrays with various heights and diverse cross-sectional shapes depending on photomask pattern designs.The proposed fabrication method is composed of a single photolithography and two subsequent deep reactive ion etching(DRIE)steps.First,a photoresist layer was patterned on a Si substrate to define areas to be etched,which will eventually determine the final location and shape of each individual microneedle.Then,the 1st DRIE step created deep trenches with a highly anisotropic etching of the Si substrate.Subsequently,the photoresist was removed for more isotropic etching;the 2nd DRIE isolated and sharpened microneedles from the predefined trench structures.Depending on diverse photomask designs,the 2nd DRIE formed arrays of microneedles that have various height distributions,as well as diverse cross-sectional shapes across the substrate.With these simple steps,high-aspect ratio microneedles were created in the high density of up to 625 microneedles mm^(-2)on a Si wafer.Insertion tests showed a small force as low as~172μN/microneedle is required for microneedle arrays to penetrate the dura mater of a mouse brain.To demonstrate a feasibility of drug delivery application,we also implemented silk microneedle arrays using molding processes.The fabrication method of the present study is expected to be broadly applicable to create microneedle structures for drug delivery,neuroprosthetic devices,and so on.展开更多

Double-differential cross sections for 1s and 2s sub...The ionization process in the collisions of He^2+ with C^q+ (q = 0-5) is investigated by using the continuum-distorted-wave eikonal-initial-state approximation. Double-differential cross sections for 1s and 2s sub-shells are obtained at the electron-ejected angle θ = 0° with the projectile energy ranging from 30keV/u to 10MeV/u. Variation of ionization mechanisms with q in C^q+ is studied, and the dependences on the projectile energies and target sub-shells are also discussed. It is found that in the whole energy range, the absolute values of soft collision (SC) and binary encounter (BE) peaks decrease with increasing q. For the lower incident energies, the electron capture to the projectile continuum (ECC) peak decrease with increasing q as well as SC and BE peaks. For the higher incident energies (〉 1 MeV/u), the absolute value of ECC peak increases with increasing q, so that the crossings of cross sections appear for C^q+ with different q. This can be explained by the matching of velocities between the projectile and the electron initially bound to the target.展开更多

The fully relativistic distorted-wave method is used in cross-section calculations. The programme is applied to He- and Li-like ions. The calculated values of line polarization are compared with other theoretical results and experimental values. For He-like U, at lower incident energy, the present polarization agrees with the other theoretical ones within 1%, while at higher energy, the differences increase up to about 10%. For He-like Fe and Ti, the present results of polarization degree for most of the lines agree with the experimental data within the experimental error bars. For the Li-like Ti line q (ls2s2p^2p3/2 to ls^22s), the present value of the polarization agree excellently with another theoretical one, and both the values are consistent with the measured data within the experimental error bar.展开更多

A 75 012701) the effective charge of the projectile is introduced to theoretically estimate the value of R for the partially stripped ions impacting on helium atoms. The results calculated from our "effective charge" model are in good agreement with the experimental data, and the dependence of the effective charge on the ionization energy of the projectile is also discussed qualitatively.展开更多

The cross-section ratios σ^2E/σ^SC, σ^3E/σ^SC, σ^4E/σ^SC and σ^SE/σ^SC are approximately the c...The relative partial cross sections for ^13C^6+ -Ar collisions at 4.15-11.08 ke V/u incident energy are measured. The cross-section ratios σ^2E/σ^SC, σ^3E/σ^SC, σ^4E/σ^SC and σ^SE/σ^SC are approximately the constants of 0.514-0.05, 0.204-0.03, 0.064-0.03 and 0.024-0.01 in this region. The significance of the multi-electron process in highly charged ions (HCIs) with argon collisions is demonstrated (σ^ME/σ^SC as high as 0.794-0.06). In multi-electron processes, it is shown that transfer ionization is dominant while pure electron capture is weak and negligible. For all reaction channels, the cross-sections are independent of the incident energy in the present energy region, which is in agreement with the static characteristic of classic models, i.e. the molecular Coulomb over-the-barrier model (MCBM), the extended classical over-the-barrier model (ECBM) and the semiempirical scaling laws (SL). The result is compared with these classical models and with our previous work of ^13C^6+ -Ne collisions [Chin. Phys. Lett. 23 (2006) 95].展开更多
Objective: Our group has previously observed that in patients with small-cell lung cancers (SCLCs), the expression of a tumor antigen, glioma big potassium (gBK) ion channel, is higher at the time of death than w...Objective: Our group has previously observed that in patients with small-cell lung cancers (SCLCs), the expression of a tumor antigen, glioma big potassium (gBK) ion channel, is higher at the time of death than when the cancer is first treated by surgical resection. This study aimed to determine whether this dichotomy was common in other potential lung tumor antigens by examining the same patient samples using our more extensive profile analysis of tumor-antigen precursor protein (TAPP). We then tested the hypothesis that therapeutic intervention may inadvertently cause this increased gBK production. Methods: SCLC samples (eight surgical resections and three autopsy samples) and three control lungs were examined by quantitative real-time polymerase chain reaction for 42 potential TAPPs that represent potential T-cell-mediated immunological targets. Results: Twenty-two TAPP mRNAs displayed the same profile as gBK, i.e., more mRNAs were expressed at autopsy than in their surgical counterparts. B-cyclin and mouse double minute 2, human homolog of PS3-binding protein were elevated in both autopsy and surgical specimens above the normal-lung controls. When HTB119 cells were incubated with doxorubicin, gBK was strongly induced, as confirmed by intracellular flow cytometry with a gBK-specific antibody. Conclusion: Our findings suggested that more immunological targets became available as the tumor responded to chemotherapy and proceeded toward its terminal stages.展开更多

In the present study polystyrene (PS) was irradiated with 107A...The study of radiation damage of high- molecular weight substances due to MeV ion interactions is of interest for engineering and scientific applications. In the present study polystyrene (PS) was irradiated with 107Ag ions of three different charge states (q) 11+, 14+ and 25+ and of 130 MeV energy. The emission of hydrogen from PS was monitored as a function of the incident ion fluence. The experimental results showed that the hydrogen depletion per incident ion from PS varies as qn, where n was found to be 2.1 as compared to the value 2.7 to 3.0 reported in the literature. Radii of the nanometric damaged zones or ion tracks formed were analyzed from the slope of the hydrogen depletion versus ion fluence curves as a function of charge state of incident ion. These have values between 3.2 - 6.8 nm. These radii were found to depend upon the charge state of the incident ion and vary as qm, where m has the value 0.9.展开更多

The agreement between the measured data and the attempted fits are, on the whole, modestly satisfactory excepting a very narrow central region in the vicinity of y = η = 0. At last, hints to how the steps suggested in the main body of the text to proceed with the description of the measured data given in the plots could lead finally to a somewhat systematic methodology have also been made.展开更多
