Effect of Different Protein Levels on Nutrient Digestion Metabolism and Serum Biochemical Parameters in Calves 被引量:10

The current study has been performed to examine the effects of different dietary protein levels on the growth, nutrient digestion and absorption, as well as biochemical parameters in calves. Nine healthy newborn calves were selected, randomly divided into 3 groups and fed 3 milk replacers with different protein levels (18, 22, and 26%), respectively. Five period-digestion-metabolism trials were taken between 12-20, 22-30, 32-40, 42-50, and 52-60 days after birth. All 3 groups showed a similar growth curve during 11 to 61 experimental days, however, the growth rate of the 22% crude protein (CP) group was 8.89% higher than that of the 18% CP group and 19.48% higher than that of the 26% CP group, respectively. The apparent digestibility of dry material (DM) declined gradually with age, whereas, the apparent digestibility of N, extract ether (EE) rose slightly. Compared to the 22% CP and 26% CP group, calves fed with 18% CP apparently had a lower digestibility than DM, EE, and nitrogen (N). The average apparent digestibilities of N were 69.39, 75.36, and 74.55%, respectively. Both the apparent digestibility and retention of calcium (Ca) and phosphorus (P) were steady throughout the experiment, but the average apparent digestibility of P in the 26% CP group was only 63.83%, which was markedly lower than that of the 18% CP group (70.40%) and 22% CP group (69.73%). In addition, the serum concentrations of total protein (TP), albumin (ALB), and globulin (GLOB) of the 22% CP group were higher than those in the 18% CP and 26% CP groups. The urea N (BUN) content in the 18% CP group, on the other hand, was significantly lower than that of the other two groups. The highest glucose (GLU) concentration was found in the 22% CP group (5.38 mmol L^-1), at the end of the trials. The protein levels in the milk replacer affected the digestion metabolism of nutrition and the serum biochemical parameters of calves at different physiological phases. Calves fed with 22% CP milk replacer had a better growth performance than the other groups. 展开更多

This experiment was conducted to evaluate the effects of dietary protein and fat levels on growth performance and serum biochemical parameters of male minks during the growing period. A total, of 90 healthy male minks at the age of 60 d were randomly assigned into six groups with 15 replicates per group and one mink per replicate. The minks were fed six experimental diets with two protein levels (32% and 36% ) and three fat levels ( 10% , 20% and 30% ) for a 7-day preliminary period and then for 60-day experimental period. The results showed that the final body weight and average daily weight gain (ADWG) were significantly higher in low-protein, middle-fat group and low-protein, high-fat group than those in low-protein, low-fat group and high-protein, low-fat group (P 〈 0. O1 ). The final weight and ADWG were increased with dietary fat level increasing, and the two indices of 20% and 30% fat groups were significantly higher than those of 10% fat groups ( P 〈 0. O1 ). The ratio of feed intake to weight gain (F/G) in low-protein, high-fat group was the lowest, which was significantly lower than that in low- protein, low-fat group and high-protein, high-fat group (P 〈 0.05 ). Serum urea nitrogen (UN) in low-protein, high-fat group was significantly lower than that in other groups (P 〈 0. 01 ). Serum alanine transaminase (ALT) activity in 32% protein groups was significantly higher than that in 36% protein groups (P 〈 O. 05). Serum triglyceride (TG) content in low-protein, high-fat group and high-protein, high-fat group was significantly higher than that in low-protein, low-fat group and high-protein, low-fat groups (P 〈 0.05 ). Serum TG, total cholesterol (TCHO) and high-density lipoprotein (HDL) contents increased with the increase in dietary fat level, and they were all the highest in low-protein, high-fat group. The growth performance and serum biochemical parameters of minks were not affected by the interaction between dietary protein and fat levels (P 〉 O. 05 ). Considering all the factors, when the dietary protein is 32% and the fat level is 20% to 30% under the condition of this experiment, the dietary protein and fat utilization rates are higher, and the male minks during the growing period can get a better growth performance. 展开更多

Changes of hepatic biochemical parameters and proteomics in broilers with cold-induced ascites

Despite the intensive investigations of this syndrome for many years, its pathogenesis remains unclear. The objective of this study was to analyze the difference in hepatic proteomics between ascites and healthy broilers by two-dimensional electrophoresis (2-DE) and matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF-MS). Changes of biochemical parameters of liver and blood were also determined. The results indicated that red blood cell counts (RBC), hematocrit (HCT) and haemoglobin (HGB) of ascites broilers were significantly greater than healthy broilers. Hepatic malondialdehyde (MDA) level of ascites broilers was significantly increased, and the activity of total superoxide dismutase (T-SOD) was significantly decreased. Hepatic lactic acid (LD) level of ascitic broilers were significantly lower than healthy ones, Serum glucose and cholesterol level of ascites broilers were significantly increased, and serum globulin level was significantly decreased in ascites broilers. There was no significant difference in triglyceride (TG) and blood urea nitrogen (BUN) level. The activity of liver hexokinase (HK) and succinodehydrogenase (SDH) in ascites broilers was significantly decreased, and there was no significant difference in the activity of liver pyruvate kinase (PK) and Na+-K+-ATPase. The hepatic proteomics analysis showed that 18 proteins expression difference were identified between ascites and healthy broilers. These proteins were mainly involved in: 1) cytoskeleton; 2) glucose, lipids and amino acid metabolism; 3) cell secretion; 4) cell apoptosis; 5) signal transduction; 6) immune and inflammatory response; and 7) cellular redox homeostasis. Mitochondrial isoform phosphoenolpyruvate carboxykinase (M-PEPCK) mainly participates in gluconeogenesis of chicken liver. In conclusion, liver oxidative damage was significantly aggravated, but antioxidant capacity was decreased in cold-induced ascites broilers. Serum glucose level was significantly increased, with liver M-PEPCK expression higher in ascites broilers, which implied that some potential regulatory reagents may reduce ascites susceptibility and mortality under cold temperature by increasing liver gluconeogenesis level. 展开更多

[ Objective] The study aimed to discuss the effects of ferrous fumarate on growth performance, blood biochemical parameters and content of trace element of Oncorhynchus mykiss. [ Method ] Juvenile rainbow trout with an initial weight of (89.2 ±0.2) g were fed with the basal fodder supplemented with different levels of ferrous fumarate (0, 20, 40, 80,160 and 480 mg/kg iron) for 60 d, and the six groups were named DO, D20, D40, D80, D160 and D480, wherein DO was as the control group, actually containing 62.60, 79.50, 99.60, 139.30, 215.20 and 538.40 mg/kg iron respectively. [ Result] The growth performance of juvenile rainbow trout was not affected by different dietary iron levels obviously ( P 〉 0.05). With the increase of dietary iron level, hemoglobin (Hb) content and number of red blood cells(RBC) rose firstly and then leveled off. No significance was found in hematocrit (Hct) among the six groups (P 〉0.05). Iron content in the whole body, vertebrae and muscle increased significantly with the improvement of dietary iron level( P 〈0.05), and iron concentration in the liver increased firstly and then leveled off in groups from D40 to D480. No significant difference in zinc content of the whole body was found among the six groups ( P〉0.05), while zinc content in the vertebrae and muscle in control group was significantly higher than that of other groups ( P〈0.05), and zinc concentration in the liver in groups DO and 1320 was significantly higher than that of other groups ( P 〈0.05). Copper content in the whole body increased significantly with the increase of dietary iron level (P 〈 0.05 ), while no significant difference was observed in the vertebrae ( P 〉0.05), and copper concentration in the muscle in control group was significantly lower than that of other groups ( P 〈0.05). Serum lysozyme (LZM) activity of group DO was significantly lower than that of other groups ( P 〈 0.05) except for group D480 ( P 〉 0.05). Catalase (CAT) activity of serum enhanced significantly ( P 〈 0.05) from DO to EH0 and then decreased obviously (P 〈0.05). [ Conclusion] Based on hemoglobin and iron content in the liver, the broken-line model analysis showed that the dietary iron level provided by ferrous fumarate for juvenile rainbow trout was estimated to be 99.8 and 100.4 mg/kg respectively in the experiment. 展开更多

This study was conducted to compare physiological characteristics between Meishan and Yorkshire piglets in their early lives. Six healthy purebred Meishan sows and Yorkshire sows with close farrowing dates were used in this research. The piglets sucked their respective sow’s milk for 14 days, then they were slaughtered to collect samples of blood, pancreas, contents of stomach, jejunum, cecum, colon as well as feces for analysis of blood biochemical parameters, digestive enzymes, and volatile fatty acid(VFA). The results showed that Yorkshire piglets had higher concentrations of high-density lipoprotein cholesterol(HDL-C) and total cholesterol(TC)(P < 0.05). Gastric lipase activity was higher in Meishan piglets but Yorkshire piglets had higher lactase activity(P < 0.05). The total VFA together with acetate and propionate in cecum and colon were higher in Meishan piglets than in Yorkshire piglets(P < 0.05),but acetate in jejunum and ratio of acetate to propionate in colon were lower in Meishan piglets than in Yorkshire piglets(P < 0.05). In conclusion, in early suckling period, significant differences exist in host metabolism and intestinal microbial metabolism between Meishan and Yorkshire piglets. 展开更多

Wild rodents were collected using live snap traps in pistachio gardens of Kerman Province, Southeast Iran from 2007 to 2009, then some physiological parameters of them were measured. The samples were identified as follow: Nesokia indica, Meriones persicus, Meriones lybicus and Tatera indica. Blood samples were obtained from the heart, then the blood parameters (glucose, cholesterol, triglyceride, total protein, HDL, red and white blood cell number) in wild species of rodents and laboratory rat were compared. The results showed that there were no significant differences in serum glucose, triglyceride, HDL and total protein levels among different experimental groups. The concentration of cholesterol in T. indica was more than that in N. indica (P < 0.01). The total numbers of red blood cells also showed significant difference between wild garden rodent species and laboratory rat (P < 0.01), while the numbers of white blood cells showed no significant difference. 展开更多

Objective:To determine the beneficial effects of the methanol extract of Synsepalum dulcificum on some biochemical parameters.Methods:In this study,rats were orally administered(gavage)with methanol extracts at doses of 0 mg/kg(Group 1,as normal group),100 mg/kg(Group 2),200 mg/kg(Group 3)and 500 mg/kg(Group 4)body weight per day for 28 d.Results:Acute toxicity study showed that the methanol extract was not toxic to rats up to 5000 mg/kg.From the results,the 100 mg/kg doses of the extract significantly(P<0.05)reduced serum levels of bilirubin,low density lipoprotein,alanine aminotransferase and glucose after 14 d compared with those after 28 d.A significant difference(P<0.05)was observed in the malondialdehyde and serum protein concentration in Group 4 while glucose concentration decreased significantly(P<0.05)in Group 1 and Group 4 after 14 d compared with 28 d.The high density lipoprotein significantly increased(P<0.05)in Group 3.Conclusions:The fruit has no negative effect on some biochemical parameters in albino rats. 展开更多

[ Objective ] This experiment was conducted to study the effects of dietary betaine on peiformance, egg quality and serum biochemical parameters of lay- ing hens under heat stress condition. [ Method] A total of 600 22-week-old commercial Roman laying hens were randomly divided into five groups, with eight repli- cates in each group and 15 hens in each replicate. Group I was taken as the positive control group, and laying hens in group ] were fed with basal diets in anrmal thermal environment. Temperature humidity index of group 1 was between 64.9 and 68.9. Group II was taken as the negative control group, and laying hens in group II were fed with basal diets under heal stress condition. Temperature humidity index of group II was higher than 72. laying hens in groups llI-V were fed with basal diets supplemented with 200,400 and 600 mg/kg betaine under heat stress condition respectively, and temperature humidity indexes in groups III-V were all higher than 72. The experiment lasted for 14 weeks. [ Results ] Results showed that there were no significant differences in average daily feed intake, feed egg ratio and broken egg rate among different test groups (P 〉 0.05 ). Hen-housed laying rate, hen-housed egg yield, serum total protein (TP) content and serum alkaline phosphatase (AKP) activity in group II were significantly lower than those in group I( P 〈0.05 ). However, activities of glutamic-oxalacetic transaminase (GOT), creatine kinase (CK) and glutamic-pyruvic transaminase (GPT) in group II were significantly higher than those in group i (P 〈0.05 ). Hen-housed laying rate, hen-housed egg yield and serum TP content in group IV were significantly higher than those in group II( P 〈 0.05 ). Hen-housed egg yield, serum TP eontent and serum albumin (ALB) content in group V were significantly higher than those in group II(P 〈0.05). Activities of serum CK and Girl in groups IV and V were significantly lower than those in group II ( P 〈 0.05 ), serum triglyceride (TG) content in group V was significantly lower than that in group 11 ( P 〈 0.05 ). [ Conclusion] Metabolism and physiological function of laying hens could be affected by heat stress, and which caused a decline in performance. Adding of dietary betaine in diets could improve hen-housed egg production and hen-housed laying rate, which could also improve the health of laying hens under heat stress condi- tion. A total of 144 mixed sex matured chickens of Cosmopolitan (C), Improved Horro (H), Cosmopolitan♂*Improved Horro♀(CH), Improved Horro♂*Cosmopolitan♀ (HC), Indigenous (L) and Koekkoek (KK) were used to determine serum biochemical, of which 36 chickens were also used for fatty acid profile study. Completely randomized design in 6 × 2 factorial arrangements was set up. Serum biochemical and fatty acid profiles were determined by Roche/Hitachi cobas c 501 and gas liquid chromatography (GC) procedures, respectively. There were significant differences (P ≤ 0.001) in Total cholesterol (TC), Triglyceride (TG), Low-density lipoprotein (LDL) and High-density lipoprotein (HDL) among genotypes and between sexes. HDL was inversely related with TC, TG and LDL. Male had significantly higher (P ≤ 0.001) TC, TG, LDL but lower HDL than female. SFA, (Myristic, Pentadecanoic and Palmitic acids) had significantly (P < 0.05) varied among genotypes. However, Margaric (P ≤ 0.01) and stearic (P ≤ 0.001) acids were significantly different between sexes. Myristoleic and Palmitoleic acids significantly vary (P ≤ 0.05) among genotypes and between sexes. Oleic (P ≤ 0.01) and Eicosenoic (P ≤ 0.001) significantly influenced by sex. Moreover, Linoleic had significantly (P ≤ 0.01) affected by genotypes. Nevertheless, α-linolenic acid significantly (P ≤ 0.01) varied between sexes. The serum biochemical differed across genotypes and between sexes. The difference in the number of carbons, double bond and position of the double bond could affect fatty acid profile among genotypes and between sexes. Chicken products with higher level TC, TG, LDL and SFA might affect human health problems. It could also be interesting topic for future studies. 展开更多

The method used was the questionnaire for the food profile and blood samples were taken to measure the biochemical parameters of the athletes. A total of 64 athletes, i.e. 20 girls and 44 boys divided into two groups (group 1 or experimental group (EG), 32 endurance athletes and group 2 or control group (CG), 32 walkers who participated in a cross-sectional study in Brazzaville. The subjects were respectively 26.16 ± 2.79 years old for the EG and 27.44 ± 3.34 years old for the CG. The results indicated that quality foods were difficult to access. The main course was more consumed compared to the starter and dessert (60% of girls and 63.63 of boys). However, biochemical parameters showed a significantly lower serum creatinine concentration in EG subjects compared to CG subjects (0.86 ± 0.06 mg/dl vs 1.04 ± 0.16 mg/dl;t = ?5.95;p 0.05). Similarly, Total Cholesterol was significantly low (1.66 ± 0.34 g/l vs 2.09 ± 0.50 g/l;t = ?3.99;p < 0.000). Cholesterol concentrations of EG subjects (HDL-C) were significantly elevated (0.98 ± 0.49 g/l vs 0.48 ± 0.18 g/l;t = 5.34;p < 0.000). In conclusion, the biochemical data were normal but the food profile of the runners was unfavorable, inadequate and unbalanced. This is considered to be a performance limiting factor. 展开更多

The objective of this study is to describe the variations of hematological and biochemical parameters of hemolysis in sickle cell patients in critical and inter-critical periods. </span><b><span style="font-family:Verdana;">Methods:</span></b><span style="font-family:Verdana;"> This is a descriptive cross-sectional study including sickle cell patients followed regularly at the National Reference Center for Sickle Cell Disease (CNRDr) from November 2019 to March 2020. A sample of 167 patients (sickle cell subjects in crisis and in steady state as well as control subjects) was randomly selected during the study period. The blood count was performed using a Sysmex-XN 350 automated system and the biochemical parameters were determined using the Cobas e 311 automated system. Statistical analysis was performed with SPSS version 22 software. </span><b><span style="font-family:Verdana;">Results:</span></b><span style="font-family:Verdana;"> The study showed that the mean cholesterol level in controls was 4.16 ± 0.77 ul compared with 9.64 ± 4.34 ul in sickle cell crisis subjects. Hb and HCT levels were significantly higher in controls compared with sickle cell subjects in crisis. During crisis, total bilirubin, direct bilirubin, triglycerides, LDH, AST, and CRP were significantly elevated. Hematological parameters such as Hb and HCT were elevated in controls, while the mean WBC value and RET were higher in sickle cell patients in steady state. The mean values of the biochemical parameters were higher in sickle cell patients in steady state. </span><b><span style="font-family:Verdana;">Conclusion:</span></b><span style="font-family:Verdana;"> Evaluation of the influence of sickle cell trait on biochemical and hematological parameters showed significant differences between sickle cell and control subjects.</span></span> 展开更多

The gut is home to a large number of intestinal microbiota that play an important role in the metabolism and immune system of the host.A growing body of evidence suggests that a high-fat diet is closely associated with many metabolic disorders,including fatty liver and type 2 diabetes.According to reports,Tartary buckwheat extract has a positive effect on intestinal microbiota in animals.The effects of Tartary buckwheat on biochemical indexes and intestinal microflora in mice were studied.Tartary buckwheat protein(FGP),Tartary buckwheat resistant starch(FGS)and Tartary buckwheat flour(FGF)alleviated organ damage in mice and lowered the atherosclerotic index(AI)in plasma.Otherwise,principal coordinate analysis(PCoA)showed that intestinal bacterial structure of FGF were separated apparently from other groups.The Firmicutes/Bacteroidetes(F/B)value of the high-fat(HF)-FGF group was significantly lower than that of the HF-FGP and HF-FGS groups.FGF significantly increases the abundance of beneficial bacteria such as Bifidobacterium,while decreasing the abundance of lipopolysaccharide(LPS)-producing bacteria.Observation of blood lipid metabolism parameters and analysis of the intestinal microbiota suggested that FGF can be more effective than FGP and FGS to reduce the effects of a high-fat diet in mice,restoring the blood parameters to values similar of those in mice fed a low-fat diet.FGF may be used to prevent or treat blood lipid metabolism disorders and intestinal microbiota disorders in mice fed a high-fat diet. 展开更多

However, the security o... Momordica foetida is a plant widely used in tropical Africa to manage gastroenteric diseases. Previous studies demonstrated interesting antibacterial activity against human pathogenic bacteria. However, the security or toxicity of methanol leaf extract has not been determined yet. The aim of this study was to evaluate the acute and sub-acute toxicity of the leaf extract of Momordica foetida. In the acute toxicity study, a single oral dose of 5000 mg/kg body weight was administered to rats which were observed for 14 days in order to identify signs of toxicity or death. In the sub-acute toxicity, the animals were treated with 250, 500 and 1000 mg/kg of the extract for 28 consecutive days. Body weights and behavior were noted throughout the experiment. Upon treatment, blood and urine were collected for hematological and biochemical analysis. Liver, lungs, heart, kidneys, testes and ovaries were analyzed for relative weights and histopathology. The acute toxicity study of M. foetida leaf extract revealed no signs of toxicity related to the treatment, indicating that the median-lethal-dose (LD50) value is greater than 5000 mg/Kg of body weight. In the sub-acute toxicity assay, the extract did not affect the general behavior of animals, meanwhile, it led to a significant increase in the levels of red blood cells, platelets, hemoglobin, granulocytes and Mid-Cells (MIDs). Biochemical parameters showed an increase in total cholesterol, HDL cholesterol, serum urea, serum and urinary glucose and a decrease in urinary proteins, serum creatinine, urinary urea levels, serum activities of AST, ALT and proteins levels, as well as increases in lung, spleen and ovaries relative weight were noticed, all compared to control animals. Histological analysis revealed a normal architecture of kidneys, liver, heart, lung, ovaries and testes. This study provides valuable data on the safety of per os administration of Momordica foetida leaf methanol extract that could be very useful for future assays. 展开更多

Cassava (Manihot esculenta Crantz) is the third largest source of calories in tropical countries and the sixth most important food crop in the World. However, the short shelf life of its storage roots after harvest due to a rapid post-harvest physiological deterioration (PPD) makes the roots to be considered as a risky product to market. The objectives of this work were to investigate the influence of two harvest periods on cassava agronomic parameters and their physiological response to PPD. Three cassava cultivars 96/1414, I070593 and LMR were selected for the experiment and harvested at 10 and 12 months after planting (MAP). The response to PPD was assessed during storage at 0, 3, 8 and 15 days after harvest (DAH). Total proteins content, soluble sugars and starch, total polyphenols compounds, polyphenoloxidase and peroxidase activities were recorded during storage. Results showed large variation among the parameters at the two harvest periods across the cultivars. High number of tubers was recorded in all the cultivars at 12 MAP and a significant increase in storage roots length was observed in 96/1414 and LMR from 10 MAP to 12 MAP (25 ± 5.1 to 41.3 ± 5.9 and 22.6 ± 3.3 to 27.9 ± 4.8) respectively. A reduction of about 49% of tubers weight was observed in I070593 from 10 to 12 MAP while an increase of about 36% and 11% were recorded in LMR and 96/1414 respectively. Tubers from I070593 showed less susceptibility to PPD when harvested at 10 MAP compared to those from LMR and 96/1414 where less susceptibility to PPD were recorded at 12 MAP. An increase in soluble sugars content, total proteins content and peroxidase activity subsequently to a decrease in starch content were recorded during storage from 8 to 15 days after harvest especially at 10 MAP in I070593 and at 12 MAP in LMR and 96/1414. High content of total phenolic compounds and less activity of polyphenol oxidase were correlated to PPD susceptibility. This work opens a new insight issue of the consideration of the appropriate harvest time of the cultivars as a tool to better control the onset of postharvest physiological deterioration. 展开更多

Bridelia micrantha, commonly known as coastal golden leaf, is a member of the family Phyllanthaceae. In preliminary studies nine fractions, named F1 - F9, were obtained by fractionating the crude methanol extract of the stem bark of Bridelia micrantha using column chromatographic techniques. The fraction F6 was the most active when tested for antibacterial activity. Thus, toxicity of this fraction was investigated for further use. The present study evaluated the acute and sub-chronic toxicity of the crude methanolic bark extract of Bridelia micrantha and its fraction. The acute toxicity was carried out according to the experimental protocol of Organization for Economic Co-operation and Development (OECD). The plant extract or the fraction F<sub>6</sub> was administered orally to female mice at a single dose of 2000 mg/kg and the animals were observed for any behavioral changes or mortality for 14 days. In the sub-chronic toxicity study, the extract and fraction were administered orally at 200, 400 and 800 mg/kg bw/day for 28 days to healthy Wistar rats. The general behavior and body weight of the rats were recorded daily. At the end of the experimental period, hematological and biochemical analyses, changes in vital organ weight (liver, lung, heart, spleen and kidney), and histopathological examination of the liver and kidney were performed. No mortality or adverse effects were noted at the 2000 mg/kg dose during the oral acute toxicity test. In the sub-chronic study, the crude methanolic bark extract of Bridelia micrantha and the fraction F<sub>6</sub> induced no mortality or treatment-related adverse effects on body weight, general behavior, relative organ weights, hematological and biochemical parameters. Histopathological examination of the liver and kidney showed normal architecture suggesting no morphological alterations. In conclusion, the oral administration of the crude methanolic bark extract of Bridelia micrantha and the fraction F<sub>6</sub> for 28 days at a dosage of up to 800 mg/kg did not induce toxicological damage in rats. From acute toxicity study, the median lethal dose (LD<sub>50</sub>) of the crude methanolic bark extract of Bridelia micrantha and the fraction F<sub>6</sub> was estimated to be more than 2000 mg/kg. 展开更多

Objective To test possible antioxidant activity of n-hexane extract of Podophyllum hexandrum under in vitro and in vivo conditions. Methods The in vitro antioxidant activity was evaluated by the ability of the extract to interact with the stable free radical DPPH, Superoxide (02), Hydroxyl (OH), Hydrogen peroxide (H202) radicals, and reducing power ability of the extract was also evaluated. Under in vivo conditions the extract was evaluated for its hepatoprotective activity by measuring different biochemical parameters, such as serum alanine aminotransaminase, serum aspartate aminotransaminase and serum lactate dehydrogenase and antioxidant enzymes. Antioxidant status was estimated by determining the activities of antioxidative enzymes, glutathione reductase (GR), glutathione peroxidase (GPx), glutathione-S-transferase (GST) and superoxide dismutase (SOD), and by determining the levels of reduced glutathione (GSH) and thiobarbituric acid reactive substances (TBARS). Results Hexane extract of P. hexondrum exhibited good radical scavenging capacity in neutralization of DPPH, O2-, OH, and H202 radicals in a dose dependent manner, n-hexane extract of Podophyllum hexandrum at the doses of 20, 30, and 50 mg/kg-day produced hepatoprotective effect by decreasing the activity of serum marker enzymes, while it significantly increased the levels of glutathione (GSH), glutathione peroxidase (GPx), glutathione reductase (GR), super oxide dismutase (SOD), and glutathione-S-transferase (GST) in a dose dependant manner. The effect of n-hexane extract was comparable to that of standard antioxidant vitamin E. Conclusion The extract of Podophyllum hexandrum possess free radical scavenging activity under in vitro conditions and could protect the liver tissue against CCI4 induced oxidative stress probably by increasing antioxidant defense activities. 展开更多
