Monocytes (MC), lymphocytes (LC) and Kupffer cells (KC) were isolated respectively from blood and surgical liver samples of patients suffering from he-patocellular carcinoma (HCC). 13 patients were given BCG, mixed ba...Monocytes (MC), lymphocytes (LC) and Kupffer cells (KC) were isolated respectively from blood and surgical liver samples of patients suffering from he-patocellular carcinoma (HCC). 13 patients were given BCG, mixed bacterium vaccine (MBV) and human white blood cell interferon (IFN), the other 3 patients were not treated with any biological immune stimulants (BIS) and served as controls. The cytosta-tic and cytotoxic effects of MC and KC on human hepatoma SMMC-7721 (TC) were assayed in vitro and the numbers of T total (Tt), T helper (Th) and T suppressor (Ts) cells were counted using CD monoclonal antibody immunofluorescence. The results were as follows: (1) On the 7th day after the first administration of BIS, the cytostatic and cytotoxic effects of MC on TC showed obvious increase over pre-administration. The activity of BIS was 1 ?5 times as high as that in the controls. (2) After 3 administrations, the cytostatic effect of MC on TC increased to the normal level (84%), while the controls remained as before (45%). (3) On the 7th day after first administration, cytostatic and cytotoxic effects of KC on TC were 0.5 and 1 times higher respectively than those of the controls. (4) The numbers of Tt and Th of patients given BIS increased continuously; on the contrary Ts decreased in number. These results indicate that combined use of BCG, MBV and IFN can actively enhance the immune anti-hepatoma function of patients suffering from HCC.展开更多

Methods: Convenience sampling method was used to select inpatients receiving biologics treatment in Rheumatology and Immunology Department of a grade A hospital in Jingzhou city from May 2020 to April 2022. 75 patients from May 2020 to April 2021 were selected as the control group, and 75 patients from May 2021 to April 2022 were selected as the observation group. The control group was given routine care. The observation group was treated with CNP combined with cluster nursing mode on the basis of the control group, and the incidences of adverse infusion reactions, total treatment time, patient satisfaction and nurse satisfaction were compared between the two groups. This study evaluated the prognostic potential of an immune score system to supplement the TNM staging system. Mothodsg An immunohistochemical analysis was conducted to assess the density of T cells, B cells, and myeloid-derived suppressor cells (MDSCs) in cancer tissues from 100 stage IIIA gastric cancer patients; the expression of the high-mobility group protein B1 (HMGB1) was also evaluated in cancer cells. The relationship between the overall survival (OS), disease-free survival (DFS), and immunological parameters was analyzed.Results: An immune score system was compiled based on the prognostic role of the density ofT cells, B cells, MDSCs, and the expression of HMGB1 in cancer tissues. The median 5-year survival of this group of patient was 32%. However, the 5-year survival rates of 80.0%, 51.7%, 0%, 5.8%, and 0% varied among the patients with an immune score of 4 to those with an immune score of 0 based on the immune score system, respectively. Similarly, differences in DFS rates were observed among the immune score subgroups. Concluslons: An immune score system could effectively identify the prognostic heterogeneity within stage IliA gastric cancer patients, implying that this immune score system may potentially supplement the TNM staging system, and help in identifying a more homogeneous group of patients who on the basis of prognosis can undergo adjuvant therapy.展开更多

Using the heuristic antibody generation method based on the physiology immune mechanism, the validity of the scheduling optimization is improved, and based on the immune and expert system under the event-driven constraints, not only Job-shop scheduling problem with multi-objective can be solved, but also the disturbance of the production be handled rapidly. A case of the job-shop scheduling is studied and dynamic optimal solutions with multi-objective function for agile manufacturing are obtained in this paper. And the event-driven dynamic rescheduling result is compared with right-shift rescheduling and total rescheduling.展开更多

文摘目的基于生物信息学方法分析创伤后脓毒症的关键基因及其与免疫基因相关性。方法从基因表达数据库(geneexpressionomnibus,GEO)中下载GSE12624数据集,筛选创伤后脓毒症患者外周血中差异基因,对差异基因进行基因本体(gene ontology,GO)和京都基因与基因组百科全书(Kyotoencyclopediaofgenesandgenomes,KEGG)富集分析,使用String、Cytoscape构建蛋白质-蛋白质互作网络(Protein-Protein Interaction Networks,PPI)及基于Cytohubba插件选出关键基因,通过受试者操作特征曲线的曲线下面积选出枢纽基因,验证其在创伤后脓毒症中的表达情况,通过基因集富集分析(gene set enrichment analysis,GSEA)、基因集变异分析(gene set variation analysis,GSVA)、免疫浸润分析以及分析其与免疫基因、免疫细胞相关性。结果在GSE12624数据集中获得362个差异基因,GO富集分析示差异基因参与信号转导、细胞凋亡、炎症反应相关的生物学过程,KEGG富集分析示,差异基因主要富集在补体和凝血级联通路、细胞周期等信号通路;选出曲线下面积最大值(0.815)的基因AURKB作为创伤后脓毒症的枢纽基因,其在创伤后脓毒症中高表达;通过GSEA富集分析显示AURKB高表达组主要富集到代谢、细胞周期、P53信号通路等相关的基因集,GSVA富集分析显示AURKB高表达组主要集到糖胺聚糖降解、花生四烯酸代谢通路、焦点黏附通路等相关途径;AURKB的表达与免疫基因(IL21R、CXCL10)的表达、免疫细胞(初始CD4+T细胞、滤泡辅助性T细胞)浸润呈正相关。结论AURKB在创伤后脓毒症中表达上调,参与创伤后脓毒症的免疫调节通路,可作为创伤后脓毒症早期预警的分子标志物及干预潜在靶标。
