Liver fibrosis is a necessary stage in the progression of chronic liver disease to cirrhosis. So far,no satisfactory drugs have been found to intervene in liver fibrosis.Liver microcirculation disorders are one of the important pathogenesis of chronic liver disease, and hepatic sinusoidal endothelial cells(HSECs)are the main cells that constitute the liver microcirculation barrier. In clinical practice,W-P bodies have been detected in HSECs of most patients with liver fibrosis.W-P bodies serve as a site for the synthesis and storage of vW factors,ET-1 and other cytokines that promote liver fibrosis. They can disrupt the structure and function of HSECs,cause liver microcirculation disorders, and exacerbate the progression of liver fibrosis.Previous studies have found that the Guangxi specialty ethnic medicine,C.kwangsiensis S.G.Lee et C.F.Liang,has definite effects in promoting blood circulation, resolving blood stasis, and resisting liver fibrosis.Based on this,a further research idea has been derived, stating that the blood circulation-promoting,blood stasis-resolving,and anti-liver fibrosis effects of C.kwangsiensis are produced by affecting the formation of W-P bodies, the synthesis and storage of contents in W-P bodies, and intervening in their exocytosis capacity. 展开更多

Objective: To explore the clinical efficacy of the principle of activating blood circulation to break stasis (ABCBS) and its influence on platelet membranous protein particle (GMP-140) and D 2 dimer (D-dimer) before and after treatment. Methods: Eighty-eight patients with blood stasis syndrome (BSS) of acute cerebral infarction (ACI) were randomly divided into two groups, both of which were treated with conventional treatment, i.e. with western medicine (WM), with Salvia injection added through intravenously dripping. One of the two groups was used as the control and the other group as the treated group who had ABCBS herbs orally taken in addition. The duration of treatment course for both groups was 3 weeks. Results: There were changes in both groups over clinical symptoms, nerve function deficit scoring and GMP-140, D-dimer, but the treated group showed significantly better than that of the control group, ( P <0.05). Conclusion: ABCBS principle could serve as an important auxiliary treating method for BSS of ACI, as it can effectively alter the blood of ACI patients which was viscous, condense, coagulant and aggregating. 展开更多

Objective: to study the influence of Chinese medicines for promoting blood circulation to remove blood stasis on fibrinogen (FIB) and D-dimer (DD) in plasma of patients with cerebral thrombosis. Method: 73 inpatients with acute cerebral thrombosis were randomly divided into a control group of 34 cases and a treatment group of 39 cases. The content of FIB and DD in plasma was detected before treatment and on the 7th and 14th days after treatment. Result: FIB content in plasma after treatment was lower than that before treatment in the control group (P<0.01) and more remarkable in the treatment group (P<0.001). There was an obvious difference in DD content before and after treatment in both groups. DD content on the 7th and 14th days after treatment in the treatment group was obviously higher than that in the control group (P<0.01 and P<0.05 respectively). Conclusion: Chinese medicines for promoting blood circulation to remove blood stasis can reduce the FIB content in plasma of patients with cerebral thrombosis, raise the DD content in plasma, cause the peak of DD content appear earlier and obviously improve hypercoagulability of blood in patients with cerebral thrombosis. 展开更多

OBJECTIVE Our previous studies demonstrated that various ingredients from the traditional Chinese medicine(TCM)for promoting blood circulation and removing blood stasis,as exemplified by cryptotanshinone and salvianolic acid B,exerted striking effects on modulating angiogenesis and vascular permeability, which suggests that they may be effective in treating vascular leak-driven diseases(e.g.tumor,cerebral cavernous malformation and diabetic retinopathy). However, the lack of reliable and advanced technologies and models sets up difficult hurdles for better understanding the role of TCM for promoting blood circulation and removing blood stasis. To this end, this study is to outline numerous cutting-edge platforms that can be utilized for exploring the function of TCM for promoting blood circulation and removing blood stasis in vascular leak-driven diseases.METHODS Two-photon laser scanning fluorescence microscopy was used to observe the interactions between neutrophils and blood vessels in a real-time manner.Dynamic flow system was employed to mimic the in vivo behaviors of neutrophils.RIP1-Tag5 spontaneous pancreatic cancer model was used to study the function of tumor blood vessels.CCM2ECKO(deletion of CCM2 in endothelial cells)mice were employed to establish the cerebral cavernous malformation(CCM)animal model.Micro-computed tomography(micro-CT)was utilized to assess the CCM lesion.Müller cell-knockout mouse model was used to study the progression of diabetic retinopathy.Vascular permeability in this model was assessed by fluorescein angiography.RESULTS The interactions between neutrophils and endothelial cells involve a series of complicated processes, including rolling, adhesion, intraluminal crawling and transmigration, which were all monitored in vivo by two-photon laser scanning fluorescence microscopy in a real-time manner.Dynamic flow system was capable of recapitulating the biological behaviors of neutrophils in vitro.Tumor vascular function in particular vascular perfusion could be assessed in the RIP1-Tag5 spontaneous pancreatic cancer model. In terms of CCM studies, specific deletion of CCM2 in endothelial cells resulted in the initiation of CCM lesion. The size and number of CCM lesions could be visualized and quantified by micro-CT.Furthermore,the Müller cell-knockout mouse model was able to precisely reflect the clinical symptoms of diabetic retinopathy.Vascular leak could be monitored at different time points using fluorescein angiography.CONCLUSION An array of high technologies and animal models can be used in investigating the occurrence and progression of multiple vascular leak-driven diseases. The pre-clinical and clinical studies of TCM for promoting blood circulation and removing blood stasis provide fundamental support for the application of the above-mentioned platforms, with the purpose of uncovering the scientific basis of TCM for promoting blood circulation and removing blood stasis. 展开更多

Hepatic fibrosis is a reversible pathological phenomenon in the early and middle stages, but but no satisfactory intervention drugs have been available so far.Recent studies have suggested that microcirculation disturbance of liver is one of the important pathogenesis of chronic liver disease, the improvement of microcirculation is beneficial to the recovery of liver function and the delay of liver fibrosis.Hepatic stellate cells are the core cells of hepatic fibrosis, and also the most critical cells that affect the microcirculation of the liver. While TLR4/MyD88/NF-κB and TLR4/MyD88/MAPKs which are based on the action of hepatic stellate cells are two pathways that have very important influence on the inflammatory response of liver, the proliferation and apoptosis of hepatic stellate cells, and the secretion of fibrogenic cytokines. It was found that Plumbapin, the active ingredient of Guangxi specialty ethnic medicine, has the definite effect of promoting blood circulation and removing blood stasis and anti-hepatic fibrosis, but its mechanism is not clear. In this study, the research progress of the above problems was reviewed, and further research ideas were derived as follows:the pharmacological effect of Plumbapin on anti-hepatic fibrosis, promoting blood circulation and removing stasis was based on the influence of TLR4/MyD88/NF-κB and MAPKs signal pathway. 展开更多

To reveal the resonance suppression mechanism of the blood circulation in dragonfly wings,a numerical modeling method of dragonfly wings based on Voronoi diagrams is proposed, and the changes in mass, aerodynamic damping, and natural frequencies caused by blood circulation in veins are investigated. The equivalent mass of blood, boundary conditions, and aerodynamic damping are calculated theoretically.Modal analysis and harmonic response analysis of wing models with different blood circulation paths are performed using the finite-element method(FEM).The vibration reduction ratioδis introduced to compare the damping efficiency of different mass regions. Finally,a natural frequency testing device is constructed to measure the natural frequencies of dragonfly wings. The results indicate that the shape, mass, and natural frequencies of the dragonfly wing model constructed by numerical method agree well with reality. The mass distribution on the wing can be altered by blood circulation, thereby adjusting the natural frequencies and achieving resonance suppression. The highestδof 1.013 is observed in the C region when blood circulates solely in secondary veins, but it is still lower than theδof 1.017 when blood circulates in complete veins. The aerodynamic damping ratio(1.19–1.79%)should not be neglected in the vibration analysis of the beating wing. 展开更多

Vertebrobasilar dolichoectasia(VBD),a rare posterior circulation vascular variant disease,is an important risk factor for many acute cerebrovascular diseases. An insufficient understanding of VBD often leads to misdiagnose. Two cases of VBD that were initially diagnosed as posterior circulation watershed infarction are reported here.Absence of common causes of stroke including hypoperfusion, blood system diseases, carotid and aortic dissection, and eosinophil elevation, the symptoms of the 2 patients met the diagnostic criteria of VBD.Both patients displayed symptoms that were in line with the Traditional Chinese Medicine(TCM)syndrome pattern of"Qi deficiency and blood stasis".Accordingly, they were comprehensively treated with Supplementing Qi and activating blood circulation method. The clinical manifestations of the 2 patients were remarkably improved and no recurrence of watershed infarction was found in a 1-year follow-up.A detailed medical history and laboratory examination are capable of improving diagnostic accuracy of VBD.TCM treatment based on syndrome identification might be a promising candidate for VBD management. 展开更多

Objective: To observe the regulatory effect of Chinese drugs for activating blood circulation (ABC) and for activating blood circulation and detoxifying (ABCD) on indices of thrombosis, inflammatory reaction, and tissue damage in a rabbit model of toxin-heat and blood stasis syndrome. Methods: Fifty-four rabbits were randomized into the normal control group, model group, simvastatin group (simvastatin, 0.93 mg/kg per day), ABC group [Xiongshao Capsule (~ff_), 0.07 g/kg per day], and ABCD group [Xiongshao Capsule, 0.07 g/kg per day, and Huanglian Capsule (-~L^t), 0.14 g/kg per day]. All except the normal control group received a single injection of bovine serum albumin and were fed with high-fat diets for 6 weeks. At the end of week 4 of giving high- fat diets, a dose of endoxitin was given by ear vein injection, and a randomized 2-week treatment was initiated. At the end of treatment, blood lipids, circulating endothelial cells, and the pathological changes of the aortic arch were assessed. The serum levels of matrix metalloproteinases (MMP-9), tissue inhibitors to metalloproteinase (TIMP-1), granule membrane protein-140 (GMP-140), plasminogen activator inhibitor-1 (PAl-l), high-sensitivity C-reactive protein (hs-CRP), interleukin-6 (IL-6), and tumor necrosis factor- ~ (TNF- oL ) were determined. Results: Compared with the model group, ABCD group showed decreased serum triglycaride (TG) level, improvement in the pathological change in the aortic arch, and reduction in the number of circulating endothelial cells (4.00_+ 1.41 per 0.9 p~ L for ABCD group vs 7.83_+ 1.72 per 0.9 μL for the model group). In addition, the levels of serum GMP-140, PAl-l, and IL-6 in ABCD group were also significantly reduced [0.79+0.20 ng/mL, 5.23± 1.39 ng/mL, 40.64 ± 10.11 pg/mL for ABCD group vs 1.08 ± 0.31 ng/mL, 7.28± 2.01 ng/mL, 54.44 ± 13.56 pg/mL for the model group, respectively, P〈0.05]. A trend showing improvement in the indices of thrombosis, inflammatory reaction, and tissue damage was observed in the ABC group when compared to the model group, but the changes were not statistically significant (P〉0.05). Conclusions: Chinese drugs for activating blood circulation and detoxifying have beneficial effects on regulating indices of thrombosis (GMP-140 and PAl-l) and inflammatory reaction (IL-6) in rabbit model with toxic-heat and blood stasis. The effect of the activating blood circulation and detoxifying drugs in regulating the levels of serum GMP-140, PAl-l, and IL-6 was superior to that of the activating blood circulation drugs. 展开更多

Objective:To observe the pharmaceutical effect of Chinese drugs for activating blood circulation (Xiongshao Capsule,XSC,芎芍胶囊)and for activating blood circulation and detoxification(Xiongshao Capsule and Huanglian Capsule,XSHLC,黄连胶囊)in terms of the indices of thrombosis, inflammatory reaction and tissue damage related factors in experimental carotid artery thrombosis rats. Methods:Fifty Wistar rats were randomly divided into the sham operation group, the model group, the Simvastatin group(SG),the activating blood circulation(ABC)group, and the activating blood circulation and detoxifying(ABCD)group, with 10 rats in each group.Simvastatin(1.8 mg/kg),XSC(0.135 g/kg)and XSHLC(0.135 g/kg)were administered to Simvastatin,ABC and ABCD group by gastrogavage, and an equal volume of normal saline was given to the sham operation group and the model group. After 2 weeks of successive medication, the rats in the model and all drug therapy groups were made into experimental carotid artery thrombosis model. The serum levels of matrix metalloproteinases(MMP-9),tissue inhibitors to metalloproteinase(TIMP-1),granule membrane protein-140(GMP-140),tissue-type plasminogen activator(t-PA),high-sensitivity C-reactive protein(hs-CRP) and interleukin-6(IL-6)were detected with enzyme-linked immunoassay 24 h later. Results:Compared with the model group, the levels of serum GMP-140,hs-CRP,IL-6 and MMP-9 were significantly decreased, and the level of t-PA was significantly increased in the ABC and ABCD group(P〈0.05),while the level of serum hs-CRP in ABCD group decreased significantly compared with that in the ABC group(P〈0.05).Conclusions:Chinese drugs both for activating blood circulation and for activating blood circulation and detoxifying have good effects on regulating indices of thrombosis, inflammatory reaction and tissue damage in experimental carotid artery thrombosis rats. The effect of activating blood circulation and detoxifying drugs on regulating the level of serum hs-CRP is superior to that of activating blood circulation drug alone. 展开更多
