For this reason, the objective of this work was to describe the effect of age and dissociative anesthetics (ketamine and tiletamine), and their combinations with acepromazine, xylazine and zolazepam, on the physiological and blood biochemical parameters in Macaca mulatta. Methods: Eighty male Macaca mulatta were divided into four experimental groups depending on the anesthetic mixture applied. Each group of 20 males was divided into five sub‐groups according to age. Physiological parameters were recorded every five minutes during a 30‐minute period. A blood sample was drawn to analyze blood biochemistry. Results: Statistical analyses revealed significant differences in the physiological parameters between the ketamine‐acepromazine and ketamine‐xylazine groups compared to the control group. The analysis of blood biochemistry found significant differences by age and by anesthetic mixture among all groups. Conclusion: These findings contribute to standardizing this animal model in biological research.展开更多

In this study we conclude that among all drug analytical methods the cheapest and easiest test to screening opioids and other abuse drugs in urine and blood samples is strip test for rapid diagnosis and TLC (thin-layer chromatography) is appropriate confirmation method to drug abuse distinguishing. The all of samples were tested through as a view of clinical laboratory methods. Age ranges of female patients were 35 ~ 15 and age range of males patients were 45 ~ 15. All patients filled questionnaire and satisfy forms too. First, all fresh urine and blood samples were examined to confirm presence drugs abuses, depend on their addiction and treatment, so all samples were confirmed by two tests. Then they were examined to other clinical laboratory tests. All data were analyzed by t-test and were Anova one way and two ways of Anova Turkey, and p 〈 0.05 was considered significant. The p-value of this study was p = 0.0001. The results of this study were showed that 4% of abusers had mild increase in hematocrite level and 2% of narcotic drugs abusers had mild lower level of blood sugars than normal range and 4% of participants had increase liver enzymes such ALT (alanine transferase), AST (aspartat transferase), ALP (alkaline phosphatease) and 1% of them had renal failure. Although blood level BUN (blood urea nitrogen) and creatinin were examined to evaluation of their renal failure .The results in Tabriz/Iran undrevision of welfare organization clinics were approximately showed that positive results of addiction are in each of urine and blood samples. Because some of abusers directly consumed full long time agonist or partial agonists' drugs such as methadone and buprenorphine for their maintenance therapy in clinics. Also doing test on blood samples has high importance in distinguishing and confirmation of drugs abuse in samples. After breeding,Black-necked Grebes(Podiceps nigricollis)perform a moult-migration to autumn hypersaline staging sites,where they moult the flight feathers and forage on superabundant brine shrimp(Artemia spp.)before l...After breeding,Black-necked Grebes(Podiceps nigricollis)perform a moult-migration to autumn hypersaline staging sites,where they moult the flight feathers and forage on superabundant brine shrimp(Artemia spp.)before leaving for wintering areas.During the stay in moulting sites,the grebes experience changes in organs and muscle size(atrophy,hypertrophy),and almost double their body mass,which has been suggested to act as an insurance against a collapse in prey availability in late autumn.During two years we collected blood samples from hundreds of individuals at one of the most important European moulting sites(the Odiel marshes,SW Spain),which is a highly polluted area due to mining drainage and chemical industry.We assessed the potential effect of moulting stage,day of the year and body condition on 16 blood biochemical parameters.Because of the changes in prey availability and body composition of grebes,we expected some physiological adjustments during moult.Elevated levels of cholesterol suggested that birds in active moult increased foraging effort to face the costs of moulting.There was increased amount of lactate dehydrogenase,corresponding to periods of breast muscle atrophy.Birds in active moult augmented protein ingestion,likely to account for the requirements of feather growth.We also show that the probability of fasting due to low prey availability increased late in the moulting season,as demonstrated by an increase in plasmaβ-hydroxy-butyrate.展开更多
Studies have shown that per-and polyfluoroalkyl substances(PFASs)may be hepatotoxic in animals or humans.However,data on clinical epidemiology are very limited.In this study,21PFASs were determined in patients with li...Studies have shown that per-and polyfluoroalkyl substances(PFASs)may be hepatotoxic in animals or humans.However,data on clinical epidemiology are very limited.In this study,21PFASs were determined in patients with liver diseases,with the highest median concentrations detected in the serum sample(26.7 ng/mL),followed by blood(10.7 ng/mL)and urine(5.02 ng/mL).Higher total PFAS concentrations were found in hepatocellular carcinoma(HCC)patients compared to non-HCC patients,with significant discrepancies in serum and blood samples.Besides,significant correlations were also found among PFAS concentrations and age,gender,body mass index(BMI),and liver function biomarkers levels.For example,PFAS concentrations are significantly higher in males than in females;Several serum PFASs concentrations increase with age and BMI,while the serum perfluorohexane sulfonic acid(PFHxS)concentrations are negatively correlated with age.In addition,multiple regression models adjusted for age,gender and BMI found that increased serum perfluorobutane sulfonic acid(PFBS),perfluoroheptane sulfonic acid(PFHpS)and perfluorohexylphosphonic acid(PFHxPA)conentrations are correlated with elevated alkaline phosphatase(ALP),aspartate aminotransferase(AST),and alpha-fetoprotein(AFP)(p<0.05).Our results provide epidemiological support for the future study on the potential clinical hepatotoxicity of PFAS.展开更多
An experiment arranged in 2×3 (2 concentrate supplement levels (CSL) and 3 breeds) factorial was carried out to evaluate the effect of CSL, breeds, and their interaction on feed intake, digestibility, feed utiliz...An experiment arranged in 2×3 (2 concentrate supplement levels (CSL) and 3 breeds) factorial was carried out to evaluate the effect of CSL, breeds, and their interaction on feed intake, digestibility, feed utilization efficiency, and blood profiles of Ethiopian fat-tail hair sheep. The CSL were 1% and 1.75% body weight, designated as L1 and L2, respectively. The breeds used were yearling intact male Blackhead Ogaden (BHO), Horro and Washera. Sixteen sheep from each breed were divided into two based on initial body weight and randomly allocated to two dietary treatments for digestibility and growth trials of 10 and 90 days, respectively. Grass hay as a basal diet was offered ad libitum to each sheep. Feed intake every day and live body weight at interval of ten days were recorded for the duration of the experiment. Blood samples and linear body measurements were taken a week before the end and on the last day of the experiment from all sheep, respectively. Dry matter and nutrient intakes, except concentrate supplement and CP, were influenced by the interaction. The higher (p<0.0001) DM digestibility was recorded from sheep supplemented with L2 than L1. Consequently, sheep supplemented with L2 had significantly higher (P<0.05) feed conversion efficiency (FCE), average daily gain (ADG) (57.3 vs 44.2 kg), total gain (TG) (5.3 vs 3.9 kg) and final body weight (FBW) than those supplemented with L1. Among breeds, Horro sheep recorded higher (P<0.05) DM intake, digestibility, and FCE than the rest of the breeds. Consequently, this breed has significantly (p<0.05) higher TG, ADG, and FBW. Some of the blood hematology (PCV, MCV, neutrophils, and monocytes) and sera metabolites (total protein, urea, and cholesterol) values were influenced by the interaction in favor of L2. Hemoglobin concentration was higher for Washera than the other breeds (p0.05). In conclusion, the study indicate the presence of variation in nutrient intake and utilization, performance traits, and blood profiles among breeds and between CSL, most of which were in favor of Horro breed and high level of concentrate.展开更多

The results revealed a significant positive association of mean daily intake of fat and urine sodium cxcretion with systolic, diastolic and mean arterial blood pressure. [Objectives]To investigate the clinical effect of different doses of Xuesaitong combined with autologous platelet-rich gel(APG)on patients with diabetic foot(DF).[Methods]90 patients with diabetic foot admitted to our...[Objectives]To investigate the clinical effect of different doses of Xuesaitong combined with autologous platelet-rich gel(APG)on patients with diabetic foot(DF).[Methods]90 patients with diabetic foot admitted to our hospital from February 2017 to February 2019 were enrolled in the study.According to the random number table method,the subjects were divided into study group A and B and control group C.Group A was given a low dose of Xuesaitong combined with APG,while group B was given high-dose Xuesaitong combined with APG and group C was treated only with APG.Patients in the three groups were observed and the changes of related indexes were detected.[Results]After treatment,with regard to the three groups,the fasting blood glucose,2-h postprandial blood glucose level,HbA1c,TNF-α,Hcy,blood urea nitrogen(BUN),creatinine(Cr)and 24-h urine protein levels were all decreased,yet AT-III level was increased,and granulation tissue coverage and thickness,wound clearance rate were increased,while the repair time of ulcer surface was significantly reduced.The above indexes were all significantly different(P<0.05).Compared with the control group C,the changes of the indexes in the study group A and B were the same as above,and the difference was significant(P<0.05).Compared with the study group A,the level of AT-III in group B was significantly increased(P<0.05),while TNF-α,Hcy,BUN,Cr and 24-h urine protein levels were significantly decreased(P<0.05).There was no significant difference in the other indexes between groups A and B(P>0.05).[Conclusions]Xuesaitong combined with APG could effectively reduce the blood sugar level of DF patients,improve the clinical indexes,promote wound healing,and the high-dose group had more significant advantages and was worthy of promotion.展开更多
3-Methoxyphencyclidine(3-MeO-PCP)is a new psychoactive substance that belongs to the phencyclidines family,first identified in Europe in 2012.This drug presents a stronger binding to N-methyl-D-aspartate(NMDA)receptor...3-Methoxyphencyclidine(3-MeO-PCP)is a new psychoactive substance that belongs to the phencyclidines family,first identified in Europe in 2012.This drug presents a stronger binding to N-methyl-D-aspartate(NMDA)receptors when compared to phencyclidine,which results in more potent effects,even at low concentrations.Very few articles have been published regarding 3-MeO-PCP in forensic toxicology.In this paper,the authors present a fatal 3-MeO-PCP intoxication case.In addition to the detection of the parent drug,metabolites were investigated in urine and,for the first time in the scientific literature,in blood.3-MeO-PCP and its metabolites were quantitated by liquid chromatography-tandem mass spectrometry system(LC-MS/MS).Identification was confirmed by liquid chromatography-high resolution mass spectrometry(LC-HRMS).3-MeO-PCP tested positive in femoral blood(3525 ng/mL)and urine(7384 ng/mL).The femoral blood concentration was higher than the fatal concentrations range already reported in the literature(from 50 to 3200 ng/mL).3-MeO-PCP metabolites,including O-demethyl-3-MeO-PCP,piperidine-OH-3-MeO-PCP,O-demethyl-piperidine-di-OH-3-MeO-PCP and piperidine-di-OH-3-MeO-PCP,were detected in blood.In addition,two new metabolites,O-demethyl-piperidine-OH-3-MeO-PCP and O-demethyl-cyclohexyl-OH,were identified in both blood and urine.Unfortunately,due to the lack of reference material on the market,it was not possible to measure the concentration of these metabolites.However,the ratios between the metabolites and the parent drug were useful to estimate their analytical response and prevalence.At this time,considering the low ratios(<1)between metabolites and parent drug,metabolites testing does not seem useful to increase the detection window of the drug.展开更多
A simple,rapid and sensitive liquid chromatography with tandem mass spectrometry method for the determination of periplocymarin in human blood and urine was developed.The digoxin‑d3 was used as an internal standard.Pe...A simple,rapid and sensitive liquid chromatography with tandem mass spectrometry method for the determination of periplocymarin in human blood and urine was developed.The digoxin‑d3 was used as an internal standard.Periplocymarin and digoxin‑d3(IS)were processed with ethyl acetate by liquid–liquid extraction.The chromatographic separation was performed on a Shim‑pack XR‑ODSIII C18 column with a 7 min gradient elution using methanol‑ammonium formate(5 mmol/L)as mobile phase at a flow rate of 0.3 mL/min(65:35,v/v).The detection was performed on a triple quadrupole tandem mass spectrometer using positive‑ion mode electrospray ionization in selected reaction monitoring mode.The periplocymarin was well separated from the internal standard.Two calibration curves were linear within the concentration range 0.01–1µg/mL.The limit of detection and quantification of blood and urine samples were both estimated at 0.005 and 0.01µg/mL.The interday and intraday precisions,accuracy,and recovery were assessed to verify this method.The results showed that the method was suitable for the determination of periplocymarin in forensic toxicological analysis and clinical diagnosis.展开更多

文摘Studies have shown that per-and polyfluoroalkyl substances(PFASs)may be hepatotoxic in animals or humans.However,data on clinical epidemiology are very limited.In this study,21PFASs were determined in patients with liver diseases,with the highest median concentrations detected in the serum sample(26.7 ng/mL),followed by blood(10.7 ng/mL)and urine(5.02 ng/mL).Higher total PFAS concentrations were found in hepatocellular carcinoma(HCC)patients compared to non-HCC patients,with significant discrepancies in serum and blood samples.Besides,significant correlations were also found among PFAS concentrations and age,gender,body mass index(BMI),and liver function biomarkers levels.For example,PFAS concentrations are significantly higher in males than in females;Several serum PFASs concentrations increase with age and BMI,while the serum perfluorohexane sulfonic acid(PFHxS)concentrations are negatively correlated with age.In addition,multiple regression models adjusted for age,gender and BMI found that increased serum perfluorobutane sulfonic acid(PFBS),perfluoroheptane sulfonic acid(PFHpS)and perfluorohexylphosphonic acid(PFHxPA)conentrations are correlated with elevated alkaline phosphatase(ALP),aspartate aminotransferase(AST),and alpha-fetoprotein(AFP)(p<0.05).Our results provide epidemiological support for the future study on the potential clinical hepatotoxicity of PFAS.
文摘An experiment arranged in 2×3 (2 concentrate supplement levels (CSL) and 3 breeds) factorial was carried out to evaluate the effect of CSL, breeds, and their interaction on feed intake, digestibility, feed utilization efficiency, and blood profiles of Ethiopian fat-tail hair sheep. The CSL were 1% and 1.75% body weight, designated as L1 and L2, respectively. The breeds used were yearling intact male Blackhead Ogaden (BHO), Horro and Washera. Sixteen sheep from each breed were divided into two based on initial body weight and randomly allocated to two dietary treatments for digestibility and growth trials of 10 and 90 days, respectively. Grass hay as a basal diet was offered ad libitum to each sheep. Feed intake every day and live body weight at interval of ten days were recorded for the duration of the experiment. Blood samples and linear body measurements were taken a week before the end and on the last day of the experiment from all sheep, respectively. Dry matter and nutrient intakes, except concentrate supplement and CP, were influenced by the interaction. The higher (p<0.0001) DM digestibility was recorded from sheep supplemented with L2 than L1. Consequently, sheep supplemented with L2 had significantly higher (P<0.05) feed conversion efficiency (FCE), average daily gain (ADG) (57.3 vs 44.2 kg), total gain (TG) (5.3 vs 3.9 kg) and final body weight (FBW) than those supplemented with L1. Among breeds, Horro sheep recorded higher (P<0.05) DM intake, digestibility, and FCE than the rest of the breeds. Consequently, this breed has significantly (p<0.05) higher TG, ADG, and FBW. Some of the blood hematology (PCV, MCV, neutrophils, and monocytes) and sera metabolites (total protein, urea, and cholesterol) values were influenced by the interaction in favor of L2. Hemoglobin concentration was higher for Washera than the other breeds (p0.05). In conclusion, the study indicate the presence of variation in nutrient intake and utilization, performance traits, and blood profiles among breeds and between CSL, most of which were in favor of Horro breed and high level of concentrate.

文摘Extracellular vesicles(EVs)are heterogeneous nano-sized vesicles of endocytic origin shed into blood and other body fluids such as urine,saliva,seminal fluid,ascites,amniotic liquid,synovial fluid,breast milk and cerebrospinal fluid(CSF)by quite all cell types.EVs actively contribute to intercellular communication as they carry bioactive molecules that are selectively incorporated by the originating cell and are delivered to recipient cells over long and short distances(Simons and Raposo,2009).EVs can be divided into three main groups according to their size and cellular origin:1)exosomes(40–120 nm),that have an endocytic origin and are formed by inward budding of the limiting membrane of multivescicular bodies,which fuse with the plasma membrane and release exosomes into the extracellular space;2)microvesicles(50–1000 nm),budding directly off the plasma membrane;3)apoptotic bodies(>1000 nm),which are released during apoptosis.Besides originating via distinct processes,different subtypes of EVs carry different proteins within their membrane and luminal compartments that reflect the phenotype of the tissue of origin.
文摘Dietacy surveys were done using a standardized 24-hour recall method and the timed overnight urine sodium,potassium and creatinine (standardized to 8 hours)were determined in a total of 178 farmers, aged 25 to 59 years,of Jiangsu province from Autumn 1984 to Spring 1985.A common food table was used to estimate nutrients intake.For each subject of the test group the relationship of nutritional factors and dietary electrolytes to blood pressure was studied by multiple regression analyses adjusted for age,sex, body mass index,hypertensive family history and alcohol intake habit. The results revealed a significant positive association of mean daily intake of fat and urine sodium cxcretion with systolic, diastolic and mean arterial blood pressure. Whereas the total energy(Kcal)intake is negatively associated with the systolic blood pressure.

文摘[Objectives]To investigate the clinical effect of different doses of Xuesaitong combined with autologous platelet-rich gel(APG)on patients with diabetic foot(DF).[Methods]90 patients with diabetic foot admitted to our hospital from February 2017 to February 2019 were enrolled in the study.According to the random number table method,the subjects were divided into study group A and B and control group C.Group A was given a low dose of Xuesaitong combined with APG,while group B was given high-dose Xuesaitong combined with APG and group C was treated only with APG.Patients in the three groups were observed and the changes of related indexes were detected.[Results]After treatment,with regard to the three groups,the fasting blood glucose,2-h postprandial blood glucose level,HbA1c,TNF-α,Hcy,blood urea nitrogen(BUN),creatinine(Cr)and 24-h urine protein levels were all decreased,yet AT-III level was increased,and granulation tissue coverage and thickness,wound clearance rate were increased,while the repair time of ulcer surface was significantly reduced.The above indexes were all significantly different(P<0.05).Compared with the control group C,the changes of the indexes in the study group A and B were the same as above,and the difference was significant(P<0.05).Compared with the study group A,the level of AT-III in group B was significantly increased(P<0.05),while TNF-α,Hcy,BUN,Cr and 24-h urine protein levels were significantly decreased(P<0.05).There was no significant difference in the other indexes between groups A and B(P>0.05).[Conclusions]Xuesaitong combined with APG could effectively reduce the blood sugar level of DF patients,improve the clinical indexes,promote wound healing,and the high-dose group had more significant advantages and was worthy of promotion.
文摘3-Methoxyphencyclidine(3-MeO-PCP)is a new psychoactive substance that belongs to the phencyclidines family,first identified in Europe in 2012.This drug presents a stronger binding to N-methyl-D-aspartate(NMDA)receptors when compared to phencyclidine,which results in more potent effects,even at low concentrations.Very few articles have been published regarding 3-MeO-PCP in forensic toxicology.In this paper,the authors present a fatal 3-MeO-PCP intoxication case.In addition to the detection of the parent drug,metabolites were investigated in urine and,for the first time in the scientific literature,in blood.3-MeO-PCP and its metabolites were quantitated by liquid chromatography-tandem mass spectrometry system(LC-MS/MS).Identification was confirmed by liquid chromatography-high resolution mass spectrometry(LC-HRMS).3-MeO-PCP tested positive in femoral blood(3525 ng/mL)and urine(7384 ng/mL).The femoral blood concentration was higher than the fatal concentrations range already reported in the literature(from 50 to 3200 ng/mL).3-MeO-PCP metabolites,including O-demethyl-3-MeO-PCP,piperidine-OH-3-MeO-PCP,O-demethyl-piperidine-di-OH-3-MeO-PCP and piperidine-di-OH-3-MeO-PCP,were detected in blood.In addition,two new metabolites,O-demethyl-piperidine-OH-3-MeO-PCP and O-demethyl-cyclohexyl-OH,were identified in both blood and urine.Unfortunately,due to the lack of reference material on the market,it was not possible to measure the concentration of these metabolites.However,the ratios between the metabolites and the parent drug were useful to estimate their analytical response and prevalence.At this time,considering the low ratios(<1)between metabolites and parent drug,metabolites testing does not seem useful to increase the detection window of the drug.

文摘A simple,rapid and sensitive liquid chromatography with tandem mass spectrometry method for the determination of periplocymarin in human blood and urine was developed.The digoxin‑d3 was used as an internal standard.Periplocymarin and digoxin‑d3(IS)were processed with ethyl acetate by liquid–liquid extraction.The chromatographic separation was performed on a Shim‑pack XR‑ODSIII C18 column with a 7 min gradient elution using methanol‑ammonium formate(5 mmol/L)as mobile phase at a flow rate of 0.3 mL/min(65:35,v/v).The detection was performed on a triple quadrupole tandem mass spectrometer using positive‑ion mode electrospray ionization in selected reaction monitoring mode.The periplocymarin was well separated from the internal standard.Two calibration curves were linear within the concentration range 0.01–1µg/mL.The limit of detection and quantification of blood and urine samples were both estimated at 0.005 and 0.01µg/mL.The interday and intraday precisions,accuracy,and recovery were assessed to verify this method.The results showed that the method was suitable for the determination of periplocymarin in forensic toxicological analysis and clinical diagnosis.
文摘Metabonomics analysis of the urine of rats with Qi deficiency and blood stasis syndrome has been performed by comparison with those of normal rats based on NMR techniques. The relative contents of formate,creatinine,2-oxoglutarate(2-OG) ,citrate,taurine,trimethylamine-N-oxide(TMAO) ,succinate and hippurate in the urine of the rats with Qi deficiency and blood stasis syndrome have been changed. These results have provided evidence for understanding the mechanism and the therapy of Qi defi-ciency and blood stasis syndrome.
