[Objective] This study aimed to compare the contents of total alkaloids in different solvent extracts from climbing groundsel herb produced in Guizhou Province and their antibacterial activity. [Method] The content of...[Objective] This study aimed to compare the contents of total alkaloids in different solvent extracts from climbing groundsel herb produced in Guizhou Province and their antibacterial activity. [Method] The content of total alkaloids was measured by UV spectrophotometry, and Oxford cups were used to investigate the antibacterial activity of each solvent extract. [Result] There was a good linear correlation between the absorbance measured by the UV spectrophotometer and the content of total al- kaloids within the concentration of 0.011 0-0.054 8 mg/ml at 207 nm, and the re- gression equation was Y=23.654X+0.021, R=0. 999 7 and the average recovery rate was 99.2%. The contents of total alkaloids in 60% ethanol extract, 95% ethanol and water ethanol were 38.71, 52.25 and 60.50 mg/g, respectively. The 60% ethanol ex- tract had strong antibacterial activity against Streptococcus pneumoniae, Staphylo- coccus aureus and Escherichia coil; the water extractive had stronger antibacterial activity against Escherichia colr, 95% ethanol extract had weak antibacterial activity, with no inhibition of Pseudomonas aeruginosa. [Conclusion] This method is accurate, simple, with good repeatability, which can be used for the determination of alkaloids content of the climbing groundsel herb; there is no positive correlation between the content of total alkaloids in different solvent extracts of climbing groundsel herb and their antibacterial activity.展开更多
There is no effective treatment to face Alzheimer’s disease complexity.Multitarget molecules are a good approach against the multiple physiopathological events associated with its development and progression.In this ...There is no effective treatment to face Alzheimer’s disease complexity.Multitarget molecules are a good approach against the multiple physiopathological events associated with its development and progression.In this context,N-((5-(3-(1-benzylpiperidin-4-yl)propoxy)-1-methyl-1H-indol-2-yl)methyl)-N-methylprop-2-yn-1-amine(ASS234)has been tested achieving promising results.ASS234 has demonstrated to cross the blood-brain barrier in vivo,and a good in silico safety profile being less toxic than donepezil.Besides,ASS234 reversibly inhibits human acetyl-and butyryl-cholinesterase,and irreversibly inhibits human monoamine oxidase A and B.Moreover,this multitarget molecule has antioxidant and neuroprotective properties,and inhibitsΑβ1–42 andΑβ1–40 self-aggregation.Inquiring about the mechanism of action,several signaling pathways related to Alzheimer’s disease had been explored showing that ASS234 induces the wingless-type MMTV integration site(Wnt)family and several members of the heat shock proteins family and moreover counteracts neuroinflammatory and oxidative stress-related genes promoting the induction of several key antioxidant genes.Finally,in vivo experiments with ASS234 in C57BL/6J mice displayed its ability to reduce amyloid plaque burden and gliosis in the cortex and hippocampus,ameliorating scopolamine-induced learning deficits.Here we gather the information regarding ASS234 evaluated so far,showing its ability to face different targets,necessary to counteract a neurodegenerative disease as complex as the Alzheimer’s disease.展开更多

The initiation of the floral elements is a centripetal spiral and the direction of the spiral is clockwise or anti-clockwise. However, the development of the androecium is highly unusual: in a longitudinal series of four stamens, the second stamen develops first from the inner to outer, then the third one, the fourth one and the first one in turn. The microsporogenesis and anther maturation follows the same developmental sequence. The tepals are different from the bracts and the stamens in both shape and size in the early developmental stage, but there is no difference between the stamens and carpels in the early developmental stage. Therefore, we established a spatio-temporal process of the floral morphogenesis of A. rivularis var. flore-minore and offer another meaning of the floral diversity patterns attributed to the level of the genus.展开更多
Objective:To investigate therapeutic mechanism in Jasminum amplexicaule(Oleaceae)and verify its main active component as quality control markers Methods:Established mouse models of diarrhea,intestinal angina,and infla...Objective:To investigate therapeutic mechanism in Jasminum amplexicaule(Oleaceae)and verify its main active component as quality control markers Methods:Established mouse models of diarrhea,intestinal angina,and inflammation were firstly used to select herb fractions with optimum efficacy,followed by an in vitro experiment to determine key targets associated with effects of J.amplexicaule extract.The selected fractions were isolated and purified,its components were identified,and the obtained compounds were verified for their effects on NF-κB and i NOS.Finally,effective compounds were administered to rats,their plasma pharmacokinetic parameters were calculated,and quality markers(QMs)reflecting therapeutic activities of J.amplexicaule were confirmed.Results:Trichloromethane and ethyl acetate fractions had significant anti-diarrheal,anti-inflammatory,and analgesic effects.The trichloromethane fraction also reduced BDNF,p38 MAPK,p-p38 MAPK,NF-κB p65,and p-NF-κB p65 levels in the ileum in a rhubarb-induced diarrhea mouse model.Additionally,it inhibited LPS-induced NF-κB transcription and nitric oxide(NO)production in RAW264.7 macrophages,which suppressed i NOS expression.Therefore,the trichloromethane fraction was further investigated.QMs candidate selection identified 17 compounds,and results of in-vitro therapeutic validation indicated that methyl caffeate and isochlorogenic acid B had the strongest anti-diarrheal,anti-inflammatory,and analgesic activities.After being validated by a UHPLC–MS-MS method,concentrations of these target compounds were accurately determined in the rat plasma and pharmacokinetic parameters were calculated.Cmax,tmax,and t1/2 were respectively 575.35 ng/mL(2.963 nmol/mL),0.5 h,and 0.45 h for methyl caffeate and 262.03 ng/m L(0.5034 nmol/mL),0.25 h,and 2.03 h for isochlorogenic acid B.Because these candidate compounds exhibited favorable pharmacokinetics,they were considered as QMs of J.amplexicaule.Conclusions:The present study accurately and effectively identified QMs of J.amplexicaule that act as indicators of efficacy and quality.展开更多
