The specific antisera for the two strains of vibrio were prepared with New Zealand rabbit and the antiserum and cross-reactive efficacy was tested by coagulation in tube. It showed that the goat anti-rabbit IgG had been labeled by fluorescence isothiocyanate (FITC). The results showed that positive reactions were 100% for the large yellow croaker Pseudosciaena crocea with typical symptom of vibrio infection, while the positive reaction to the pathogen in healthy yellow croakers reached 40%, but seemed negative for aquaculture water. The results demonstrated that this fast and indirect fluorescent antibody assay can be used not only to test the vibrio pathogen in diseased yellow croaker but also in infected animals with no symptom.展开更多
BACKGROUND Although ABO-nonidentical and ABO-incompatible liver transplantation(LT)are other options for end-stage liver disease treatment,the development of antibodies against blood group antigens(anti-A/B antibodies...BACKGROUND Although ABO-nonidentical and ABO-incompatible liver transplantation(LT)are other options for end-stage liver disease treatment,the development of antibodies against blood group antigens(anti-A/B antibodies)is still a challenge in managing and follow-up of the recipients.CASE SUMMARY A 56-year-old male with end-stage liver disease with rapid deterioration and poor prognosis was considered to receive a deceased ABO-nonidentical liver graft.All required tests were performed according to our pre-LT diagnostic protocol.The orthotopic LT procedure involving O+donor and A1B+recipient was performed.Our treatment strategy to overcome the antibodymediated rejection included a systemic triple immunosuppressive regimen:methylprednisolone,mycophenolate mofetil,and tacrolimus.The immunological desensitization consisted of the chimeric anti-CD20 monoclonal antibody rituximab and intravenous immunoglobulins.The patient was also on antibiotic treatment with amoxicillin/clavulanate,cefotaxime,and metronidazole.On the 10th postoperative day,high titers of IgG anti-A and anti-B antibodies were found in the patient’s plasma.We performed a liver biopsy,which revealed histological evidence of antibody-mediated rejection,but the rejection was excluded according to the Banff classification.The therapy was continued until the titer decreased significantly on the 18th postoperative day.Despite the antibiotic,antifungal,and antiviral treatment,the patient deteriorated and developed septic shock with anuria and pancytopenia.The conservative treatment was unsuccessful,which lead to the patient’s fatal outcome on the 42nd postoperative day.CONCLUSION We present a patient who underwent ABO-nonidentical LT from a deceased donor.Even though we implemented the latest technological advancements and therapeutic approaches in the management of the patient and the initial results were promising,due to severe infectious complications,the outcome was fatal.展开更多
AIM: To explore a specific diagnostic method for local recurrent rectal cancer.METHODS: Immunoscintigraphy with 99mTc-labeled anti-CEA monoclonal antibody (MoAb) CL-58 was performed for patients suspected of having a ...AIM: To explore a specific diagnostic method for local recurrent rectal cancer.METHODS: Immunoscintigraphy with 99mTc-labeled anti-CEA monoclonal antibody (MoAb) CL-58 was performed for patients suspected of having a postoperative local recurrent rectal cancer and the findings were compared with the results of conventional imaging and pathology.RESULTS: A total of 36 patients with a suspected local recurrent rectal cancer underwent immunoscintigraphy with 99mTc-conjugated CL58. Local recurrence of rectal cancer was identified in 31 patients and established in 30 during operation, endoscopy and pathological examination. No local recurrence was found in 5 patients without specific accumulation of 99mTc during the follow-up. Immunoscintigraphy had a positive rate of 86.11%, a specificity of 83.33%, and a sensitivity of 100%.CONCLUSION: Immunoscintigraphy has a highly specific and predictive value for detecting local recurrent rectal cancer, especially after abdominal perineal resection (APR).展开更多

Methods: CEA antigen was extracted from metastasized liver of patients with colorectal can...Objective: To study the preparation and characterization of monoclonal antibody (McAb) against carcinoembryonic antigen (CEA). Methods: CEA antigen was extracted from metastasized liver of patients with colorectal cancer and used for the preparation of McAb against CEA by hybridoma technique. Immunoreactivity of McAb to CEA antigen was evaluated using ELISA. Mouse ascites was purified by two steps, high performance liquid chromatography (HPLC) using protein A and high performance hydroxylapatite (HPHT). Normal adult tissues and tumor specimen were used for immunohistochemical evaluation of the McAb. Isotope 99mTc labeled CEA McAb was used for biodistribution in tumor-bearing mouse. Results: Purified CEA antigen was a glycoprotein of 180 kD. Anti-CEA McAb affinity constant was 7.4×109/M. The McAb showed positive staining in 54–88% of colorectal cancer, gastric cancer and lung cancer, while negative for normal tissues. 24 hours after injection of 99mTc labeled McAb, tumor ID%/g was higher than 15% and tumor/blood, tumor/kidney and tumor/liver were 1.82, 1.51 and 2.92 respectively. T/NT ratios of other viscera were over 3.0. Conclusion: Purified CEA antigen had very good immunogenicity. The anti-CEA McAb was highly specific. 99mTc labeled McAb was stabled both in vivo and in vitro. In vivo distribution result was satisfactory. McAb CL58 may be useful for RII and RIGS.展开更多
