黑逍遥散调控p38MAPK/ATF2/COX-2信号通路对阿尔茨海默病大鼠神经炎症的影响

目的 观察黑逍遥散对Aβ_(1-42)所致阿尔茨海默病(AD)大鼠学习记忆能力的影响,并从p38MAPK/ATF2/COX-2信号通路介导的炎症级联反应探讨其作用机制。方法 双侧海马注射1μL Aβ_(1-42)溶液复刻AD大鼠模型。筛选造模成功的大鼠50只,随机... 目的 观察黑逍遥散对Aβ_(1-42)所致阿尔茨海默病(AD)大鼠学习记忆能力的影响,并从p38MAPK/ATF2/COX-2信号通路介导的炎症级联反应探讨其作用机制。方法 双侧海马注射1μL Aβ_(1-42)溶液复刻AD大鼠模型。筛选造模成功的大鼠50只,随机分为模型组、盐酸多奈哌齐组(0.5 mg/kg)和黑逍遥散低、中、高剂量组(4.25、8.5、17 g/kg),连续给药42 d。Morris水迷宫实验检测定位航行与空间探索能力,HE染色观察海马神经元病理结构改变,ELISA法检测海马组织Aβ_(1-42)、iNOS、PGE_(2)表达,RT-qPCR法检测海马组织p38、ATF2、COX-2 mRNA表达,Western blot法检测海马组织p-p38、p-ATF2、COX-2蛋白表达。结果 与模型组比较,黑逍遥散中、高剂量组和盐酸多奈哌齐组逃避潜伏期缩短(P<0.01),有效区域运动距离、目标象限滞留时间百分比增加(P<0.01),海马CA1区神经元排列有序,胞体清晰,凋亡细胞减少,海马组织Aβ_(1-42)、iNOS、PGE_(2)水平降低(P<0.05,P<0.01),p38、COX-2 mRNA表达降低(P<0.05,P<0.01),p38、p-p38、p-ATF2、COX-2蛋白表达降低(P<0.01)。结论 黑逍遥散可改善Aβ_(1-42)所致AD大鼠的认知能力,其机制可能与阻断p38MAPK/ATF2/COX-2信号通路传导,进而减轻炎症反应有关。 展开更多

目的:探讨前列腺素E2(prostaglandin E2,PGE2)、环氧合酶2(cyclooxygenase-2,COX-2)表达与牙槽骨成骨活性、内氧水平的关系。方法:选择2021年3月—2023年3月收治的56例慢性牙周炎且牙槽骨感染的患者为试验(牙周炎)组,同一时段53例健康... 目的:探讨前列腺素E2(prostaglandin E2,PGE2)、环氧合酶2(cyclooxygenase-2,COX-2)表达与牙槽骨成骨活性、内氧水平的关系。方法:选择2021年3月—2023年3月收治的56例慢性牙周炎且牙槽骨感染的患者为试验(牙周炎)组,同一时段53例健康牙槽骨为对照组。采用组织块培养法进行成骨细胞培养,采用改良Kaplow碱性磷酸酶(alkaline phosphatase,ALP)染色鉴定细胞,利用ELISA试剂盒检测COX-2、PGE2、破骨细胞抑制因子(osteoclastogenesis inhibitory factor,OPG)和核因子κB受体活化因子配体(receptor activator of nuclear factor-κb ligand,RANKL)等的表达,比较2组患者PGE2、COX-2、OPG、内氧水平、ALP和RANKL的表达水平,并分析其相关性。采用SPSS 27.0软件包对数据进行统计学分析。结果:牙周炎组的PGE2、COX-2、RANKL显著高于对照组,而OPG、内氧水平、ALP显著低于对照组(P<0.05)。PGE2、COX2呈高度正相关,与OPG、内氧水平和ALP呈高度负相关,但与RANKL呈高度正相关(P<0.05)。结论:PGE2、COX-2表达与ALP、内氧水平呈高度负相关,可考虑通过增加内氧水平,加大氧分压,并通过药物调节ALP水平,改变牙周炎或其他类似疾病的炎症状况。 展开更多

目的:探究宣痹汤抑制环氧合酶-2(COX-2)通路对急性痛风性关节炎(GA)大鼠炎症的影响。方法:SD大鼠60只,随机分为对照组、模型组、塞来昔布组(20 mg/kg)、宣痹汤低剂量组(5 g/kg)、宣痹汤中剂量组(10 g/kg)、宣痹汤高剂量组(20 g/kg),每... 目的:探究宣痹汤抑制环氧合酶-2(COX-2)通路对急性痛风性关节炎(GA)大鼠炎症的影响。方法:SD大鼠60只,随机分为对照组、模型组、塞来昔布组(20 mg/kg)、宣痹汤低剂量组(5 g/kg)、宣痹汤中剂量组(10 g/kg)、宣痹汤高剂量组(20 g/kg),每组10只。采用Coderre经典方法建立大鼠痛风模型,对各组大鼠一般情况进行观察,步态分析,评估关节肿胀程度;利用HE染色技术对大鼠踝关节滑膜组织进行观察;ELISA测定大鼠关节液中炎症因子IL-1β、TNF-α、PGE_(2)、LTB_(4)水平;免疫印迹法检测大鼠踝关节滑膜组织PGE受体2(EP2)和LTB受体1(BLT1)的表达情况;qRT-PCR测定COX-2、5-脂氧合酶(5-LOX)mRNA表达的影响。结果:模型组大鼠关节肿胀、跛足、毛发暗淡、精神萎靡;塞来昔布组、宣痹汤低、中、高剂量组的大鼠症状明显改善;相比于对照组,模型组大鼠步态评价、关节肿胀程度、组织病理学改变、IL-1β、TNF-α、PGE_(2)、LTB_(4)、COX-2、5-LOX、EP2、BLT1水平显著升高(P<0.05);与COX-2抑制剂塞来昔布组相比,宣痹汤(20 g/kg)在接受MSU晶体处理的大鼠中表现出更好的抗关节炎特性,同时伴有IL-1β、TNF-α、PGE_(2)、LTB_(4)、COX-2、5-LOX、EP2、BLT1表达的减少。结论:宣痹汤抑制GA大鼠中COX-2、5-LOX来改善MSU晶体诱导的炎症,并可能作为一种治疗GA新策略。 展开更多

Low-density lipoprotein receptor-related protein 2(LRP2)is a multifunctional endocytic receptor expressed in epithelial cells.In mammals,it acts as an endocytic receptor that mediates the cellular uptake of cholesterol-containing apolipoproteins to maintain lipid homeostasis.However,little is known about the role of LRP2 in lipid homeostasis in insects.In the present study,we investigated the function of LRP2 in the migratory locust Locusta migratoria(LmLRP2).The mRNA of LmLRP2 is widely distributed in various tissues,including integument,wing pads,foregut,midgut,hindgut,Malpighian tubules and fat body,and the amounts of LmLRP2 transcripts decreased gradually in the early stages and then increased in the late stages before ecdysis during the nymphal developmental stage.Fluorescence immunohistochemistry revealed that the LmLRP2 protein is mainly located in cellular membranes of the midgut and hindgut.Using RNAi to silence LmLRP2 caused molting defects in nymphs(more than 60%),and the neutral lipid was found to accumulate in the midgut and surface of the integument,but not in the fat body,of dsLmLRP2-treated nymphs.The results of a lipidomics analysis showed that the main components of lipids(diglyceride and triglyceride)were significantly increased in the midgut,but decreased in the fat body and hemolymph.Furthermore,the content of total triglyceride was significantly increased in the midgut,but markedly decreased in the fat body and hemolymph in dsLmLRP2-injected nymphs.Our results indicate that LmLRP2 is located in the cellular membranes of midgut cells,and is required for lipid export from the midgut to the hemolymphand fat body in locusts. 展开更多

Walnut dreg protein hydrolysates(WDPHs)exhibit a variety of biological activities,however,the cyclooxygenase-2(COX-2)inhibitory peptide of WDPHs remain unclear.The aim of this study was to rapidly screen for such peptides in WDPHs through a combination of in silico and in vitro analysis.In total,1262 peptide sequences were observed by nano liquid chromatography/tandem mass spectrometry(nano LC-MS/MS)and 4 novel COX-2 inhibitory peptides(AGFP,FPGA,LFPD,and VGFP)were identified.Enzyme kinetic data indicated that AGFP,FPGA,and LFPD displayed mixed-type COX-2 inhibition,whereas VGFP was a non-competitive inhibitor.This is mainly because the peptides form hydrogen bonds and hydrophobic interactions with residues in the COX-2 active site.These results demonstrate that computer analysis combined with in vitro evaluation allows for rapid screening of COX-2 inhibitory peptides in walnut protein dregs. 展开更多

BACKGROUND Thrombocytopenia 2,an autosomal dominant inherited disease characterized by moderate thrombocytopenia,predisposition to myeloid malignancies and normal platelet size and function,can be caused by 5’-untranslated region(UTR)point mutations in ankyrin repeat domain containing 26(ANKRD26).Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1)have been identified as negative regulators of ANKRD26.However,the positive regulators of ANKRD26 are still unknown.AIM To prove the positive regulatory effect of GATA binding protein 2(GATA2)on ANKRD26 transcription.METHODS Human induced pluripotent stem cells derived from bone marrow(hiPSC-BM)INTRODUCTION Ankyrin repeat domain containing protein 26(ANKRD26)acts as a regulator of adipogenesis and is involved in the regulation of feeding behavior[1-3].The ANKRD26 gene is located on chromosome 10 and shares regions of homology with the primate-specific gene family POTE.According to the Human Protein Atlas database,the ANKRD26 protein is localized to the Golgi apparatus and vesicles,and its expression can be detected in nearly all human tissues[4].Moreover,UniProt annotation revealed that ANKRD26 is localized in the centrosome and contains coiled-coil domains formed by spectrin helices and ankyrin repeats[5,6].The most common disease related to ANKRD26 is thrombocytopenia 2(THC2),which is a rare autosomal dominant inherited disease characterized by lifelong mild-to-moderate thrombocytopenia and mild bleeding[7-9].Caused by the variants in the 5’-untranslated region(UTR)of ANKRD26,THC2 is defined by a decrease in the number of platelets in circulating blood and results in increased bleeding and decreased clotting ability[8,10].Due to the point mutations that occur in the 5’-UTR of ANKRD26,its negative transcription factors(TFs),Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1),lose their repression effect[11].The persistent expression of ANKRD26 increases the activity of the mitogen activated protein kinase and extracellular signal regulated kinase 1/2 signaling pathways,which are potentially involved in the regulation of thrombopoietin-dependent signaling and further impair proplatelet formation by megakaryocytes(MKs)[11].However,the positive regulators of ANKRD26,which might be associated with THC2 pathology,are still unknown. 展开更多

Pancreatic ductal adenocarcinoma(PDAC)is one of the most aggressive solid malignancies.A specific mechanism of its metastasis has not been established.In this study,we investigated whether Neural Wiskott-Aldrich syndrome protein(N-WASP)plays a role in distant metastasis of PDAC.We found that N-WASP is markedly expressed in clinical patients with PDAC.Clinical analysis showed a notably more distant metastatic pattern in the N-WASP-high group compared to the N-WASP-low group.N-WASP was noted to be a novel mediator of epithelialmesenchymal transition(EMT)via gene expression profile studies.Knockdown of N-WASP in pancreatic cancer cells significantly inhibited cell invasion,migration,and EMT.We also observed positive association of lysyl oxidase-like 2(LOXL2)and focal adhesion kinase(FAK)with the N-WASP-mediated response,wherein EMT and invadopodia function were modulated.Both N-WASP and LOXL2 depletion significantly reduced the incidence of liver and lung metastatic lesions in orthotopic mouse models of pancreatic cancer.These results elucidate a novel role for N-WASP signaling associated with LOXL2 in EMT and invadopodia function,with respect to regulation of intercellular communication in tumor cells for promoting pancreatic cancer metastasis.These findings may aid in the development of therapeutic strategies against pancreatic cancer. 展开更多

BACKGROUND Recently,type 2 diabetic osteoporosis(T2DOP)has become a research hotspot for the complications of diabetes,but the specific mechanism of its occurrence and development remains unknown.Ferroptosis caused by iron overload is con-sidered an important cause of T2DOP.Polycytosine RNA-binding protein 1(PCBP1),an iron ion chaperone,is considered a protector of ferroptosis.AIM To investigate the existence of ferroptosis and specific role of PCBP1 in the development of type 2 diabetes.METHODS A cell counting kit-8 assay was used to detect changes in osteoblast viability under high glucose(HG)and/or ferroptosis inhibitors at different concentrations and times.Transmission electron microscopy was used to examine the morpho-logical changes in the mitochondria of osteoblasts under HG,and western blotting was used to detect the expression levels of PCBP1,ferritin,and the ferroptosis-related protein glutathione peroxidase 4(GPX4).A lentivirus silenced and overex-pressed PCBP1.Western blotting was used to detect the expression levels of the osteoblast functional proteins osteoprotegerin(OPG)and osteocalcin(OCN),whereas flow cytometry was used to detect changes in reactive oxygen species(ROS)levels in each group.RESULTS Under HG,the viability of osteoblasts was considerably decreased,the number of mitochondria undergoing atrophy was considerably increased,PCBP1 and ferritin expression levels were increased,and GPX4 expression was decreased.Western blotting results demonstrated that infection with lentivirus overexpressing PCBP1,increased the expression levels of ferritin,GPX4,OPG,and OCN,compared with the HG group.Flow cytometry results showed a reduction in ROS,and an opposite result was obtained after silencing PCBP1.CONCLUSION PCBP1 may protect osteoblasts and reduce the harm caused by ferroptosis by promoting ferritin expression under a HG environment.Moreover,PCBP1 may be a potential therapeutic target for T2DOP. 展开更多

In this editorial,we comment on the article by Liu et al published in the recent issue of the World Journal of Diabetes(Relationship between GCKR gene rs780094 polymorphism and type 2 diabetes with albuminuria).Type 2 diabetes mellitus(T2DM)is a chronic disorder characterized by dysregulated glucose homeostasis.The persistent elevated blood glucose level in T2DM significantly increases the risk of developing severe complications,including cardiovascular disease,re-tinopathy,neuropathy,and nephropathy.T2DM arises from a complex interplay between genetic,epigenetic,and environmental factors.Global genomic studies have identified numerous genetic variations associated with an increased risk of T2DM.Specifically,variations within the glucokinase regulatory protein(GCKR)gene have been linked to heightened susceptibility to T2DM and its associated complications.The clinical trial by Liu et al further elucidates the role of the GCKR rs780094 polymorphism in T2DM and nephropathy development.Their findings demonstrate that individuals carrying the CT or TT genotype at the GCKR rs780094 locus are at a higher risk of developing T2DM with albuminuria compared to those with the CC genotype.These findings highlight the importance of genetic testing and risk assessment in T2DM to develop effective preventive strategies and personalized treatment plans. 展开更多

目的探究血清红细胞沉降率(erythrocyte sedimentation rate,ESR)、C反应蛋白(C-reactive protein,CRP)、环氧合酶-2(cycloxygenase-2,COX-2)指标水平对骨科开放性损伤患者术后感染诊断价值与全身炎症反应综合征(systemic inflammatory ... 目的探究血清红细胞沉降率(erythrocyte sedimentation rate,ESR)、C反应蛋白(C-reactive protein,CRP)、环氧合酶-2(cycloxygenase-2,COX-2)指标水平对骨科开放性损伤患者术后感染诊断价值与全身炎症反应综合征(systemic inflammatory response syndrome,SIRS)评分的相关性。方法选取中国人民武装警察部队重庆市总队医院骨科2019年7月至2022年7月诊治的98例骨科开放性损伤患者作为研究对象,根据患者术后是否感染分为感染组和未感染组。感染组43例,男25例,女18例;年龄21~65岁,平均(56.19±4.33)岁。未感染组55例,男29例,女26例;年龄21~66岁,平均(56.37±4.49)岁。采用双抗体酶联免疫吸附剂测定(enzyme linked immuno sorbent assay,ELISA)检测患者血清ESR、CRP和COX-2水平;对患者行常规血液检查和生命体征的监测,计算SIRS评分;对比两组ESR、CRP和COX-2水平;分析血清ESR、CRP和COX-2对骨科创伤患者术后感染诊断价值;对比骨科创伤感染组和未感染组手术前后SIRS评分差异;Pearson分析ESR、CRP和COX-2与SIRS评分相关性。结果术后两组ESR、CRP和COX-2水平均升高,且感染组表达明显高于未感染组(P<0.05);ESR、CRP和COX-2联合检测在骨科术后感染患者中诊断评估中的受试者工作特征(receiver operating characteristic,ROC)曲线下面积(area under curve,AUC)为0.899,具有较高的特异性以及敏感度,显著高于单独ESR、CRP检测,且联合诊断对骨科术后感染患者具有高度一致性(P<0.05);术后3 d两组SIRS评分均升高,且感染组评分明显高于未感染组(P<0.05);Pearson分析骨折术后感染患者ESR、CRP和COX-2与SIRS评分相关性,发现均与SIRS评分存在显著正相关(P<0.001)。结论血清ESR、CRP和COX-2在骨科术后感染患者中呈现高表达,联合检测对患者感染发生诊断价值较高;且血清ESR、CRP和COX-2与SIRS评分呈现显著正相关,临床可对患者实行指标及时检测,以降低术后感染的发生率。 展开更多

Fibroids, also called leiomyomas or myomas, are communal tumors of the muscle or uterine wall that affect about 20% of females who are of reproductive age. They can look as if singly or in clusters, and they often cease to grow after menopause. Fibroids can be classified as intramural, sub serosal, pedunculated, or submucosal based on where they are positioned in the uterus. Although fibroids are benign, they can grow quickly and cause a range of symptoms, such as pelvic pressure, heavy menstrual flow, and infertility. As a result, fibroids are a main reason behind hysterectomy surgeries. The majority of cases of breast cancer are ductal and lobular cancers, making it the second utmost common cancer in women international. Gene mutations like those in BRCA1 or BRCA2 knowingly raise the risk of breast and other cancers, typically with an earlier cancer onset. Cancer risk is influenced by a complex interplay of genetic abnormalities, environmental factors, and lifestyle selections. Further research into these relations is domineering. Although they are common in uterine leiomyomas, especially multiple leiomyomas, MED12 mutations do not significantly correlate with tumor size. These mutations have also been noticed in smooth muscle tumors and leiomyosarcomas, two other types of uterine cancer. The identification of MED12 mutations as the sole genetic abnormality originates in leiomyomas raises the opportunity of a role in the genesis of cancer. 10% - 15% of women who are of reproductive age have endometriosis, which grants serious difficulties because of its chronic nature and range of clinical symptoms. Even after effective surgeries, issues reoccur often, adding to the enormous financial burden. The effects of MED12 mutations have been experiential in recent studies examining the molecular causes of endometriosis-associated infertility, which have shown anomalies in cellular connections and signaling cascades. Computational techniques were used in this study to investigate LifeGreenTM’s potential to prevent uterine fibroids and breast cancer. The efficacy of LifeGreenTM as a preventive measure or a treatment for common gynecological matters was examined and modeled. We investigated the mechanisms underlying LifeGreenTM’s benefits in the treatment of uterine fibroids and breast cancer using computational techniques. Our research contributes to our understanding of its potential therapeutic benefits for women’s health. 展开更多

Background:Radix Aconiti Lateralis Preparata(Fu-zi)is a traditional Chinese medicinal herb,which has been widely used in the clinic and has potent anti-inflammatory activities.we aimed to explore the mechanisms of extract containing alkaloids from different Fu-zi Processed Products(FPP)in treating inflammation,especially rheumatoid arthritis(RA).Methods:Firstly,using network pharmacology technology,the ingredients,and targets of Fu-zi were obtained by searching and screening,the targets involving RA were acquired,the intersection targets were constructed a"component-target-pathway"network.A comprehensive investigation was conducted on the anti-rheumatoid arthritis mechanisms of 5 FPPs in lipopolysaccharide(LPS)induced RAW264.7 cells,which serve as a model for RA.The production of NO and inflammatory cytokines were measured by ELISA kit.Quantitative Real-time PCR(qRT-PCR)was utilized to measure the mRNA levels.COX-2/PGE2 signaling pathway-associated proteins were determined by western blot.Results:According to a network pharmacological study,16 chemical components and 43 common targets were found in Fu-zi and 6 key targets including PTGS2 were closely related to the mechanism of Fu-zi in treating RA.The in vitro study revealed that the levels of NO,TNF-α,and IL-1βwere substantially decreased by the 5 FPPs.The 5 FPPs significantly suppressed the expression of proteins COX-2,iNOS,and NF-κB,with particularly notable effects observed for PFZ and XFZ.Conclusion:Altogether,these results demonstrated that the 5 PPS containing alkaloids have a good anti-RA-related inflammatory effect,and the mechanism may be related to COX-2/PGE2 signaling pathway,particularly,Fu-zi prepared utilizing a traditional Chinese technique. 展开更多

多层螺旋CT联合血清COX-2、SCC-Ag在宫颈癌患者淋巴结转移诊断中的价值

目的:本研究将多层螺旋CT与血清环氧合酶(COX-2)、鳞状细胞癌抗原(SCC-Ag)联合应用于宫颈癌患者淋巴结转移诊断中,并对该方式的诊断效能进行分析。方法:选取2021年1月—2023年1月在南京大学医学院附属盐城第一医院收治的80例经手术治疗... 目的:本研究将多层螺旋CT与血清环氧合酶(COX-2)、鳞状细胞癌抗原(SCC-Ag)联合应用于宫颈癌患者淋巴结转移诊断中,并对该方式的诊断效能进行分析。方法:选取2021年1月—2023年1月在南京大学医学院附属盐城第一医院收治的80例经手术治疗的宫颈癌患者,以病理检查结果作为判定是否发生淋巴结转移的金标准,其中转移32例设为研究组,未转移48例设为对照组。对患者进行多层螺旋CT及血清COX-2、SCC-Ag检测,对不同检测方式的诊断效能进行分析。结果:研究组的血清COX-2、SCC-Ag水平以及多层螺旋CT参数中的血量(BV)、峰值强化(PEI)、灌注值(PF)均高于对照组,而研究组多层螺旋CT参数中的达峰时间(TTP)低于对照组,差异有统计学意义(均P<0.05);多层螺旋CT检查后可见27例转移,53例未转移;血清COX-2、SCC-Ag检查后可见29例转移,51例未转移;联合检测可见31例转移,49例未转移。其中联合检测的阳性预测值为96.77%,阴性预测值为95.92%;对比曲线下面积后可见多层螺旋CT<血清COX-2、SCC-Ag<联合检测,其中联合检测的曲线下面积为0.957(P=0.027,95%CI:0.904~1.000),灵敏度为90.60%,特异度为95.80%。结论:多层螺旋CT联合血清COX-2和SCC-Ag诊断宫颈癌淋巴结转移具有显著的诊断效能,有助于临床医生判断淋巴结转移的危险程度并尽早制定治疗计划,以改善患者的预后。 展开更多
