Pachyloen leaves. [Method] Using the chloro- phyll fluorescence system and fluorescence imaging system in the Mini-IMAGING- PAM, the chlorophyll fluorescence of Phyllostachys edulis cv. Pachyloen leaves were measured and the fluorescence parameters were calculated. [Result] The homo- geneities of the maximal quantum yield of PS II (Fv/Fm) and the leaf absorptivity (Abs) were higher, with CVs (Coefficient of Variation) of 1.58%-1.68% and 1.75%- 2.12% respectively, while the heterogeneities of the actual quantum yield (Y), non- photochemical quenching (NPQ/4), photochemical quenching(qP) and relative photo- synthetic rate (PS/50) were higher, with CVs of 9.60%-14.23%, 10.23%-13.02%, 11.92%-13.02% and 11.15%-17.74% respectively. The trends of heterogeneity change in Y, qP and PSI50 were basically the same with transversely higher het- erogeneity at two sides and lower heterogeneity in the middle, namely with larger CVs at the edges of leaves and smaller ones around the midrib. Longitudinally, the coefficients of variation of Y, qP and PS/50 decreased gradually (from top to bot- tom), which indicated that the heterogeneity declined from the leaf tip to the base. The trends of heterogeneity change in PS/50 and NPQ/4 were opposite. [Conclusion] The fluorescence parameters of Phyllostachys edulis cv. Pachyloen leaves revealed different heterogeneity.展开更多

[Objective]The aim was to study the effective factors on culturing Prunus salicina cv.Zaoshi embryos in vitro.[Method]Different age,culture medium and GA3 which affected culturing of prumssalicina embryos were discuss...[Objective]The aim was to study the effective factors on culturing Prunus salicina cv.Zaoshi embryos in vitro.[Method]Different age,culture medium and GA3 which affected culturing of prumssalicina embryos were discussed.[Result]The result showed that on the part of 26-41 d,after the blooming period,the embryos remained tiny and retained endosperms and showed no signs of change after having cultured for three generations.On the part of 48 d,after the blooming period,the endosperms had disappeared,the embryos kept growing until they filled the seed cavity;when they were planted on the MS culture medium,their survival rate reached 77%,in its first generation,the response of embryos was discernible.On the part of 65 d,after the blooming period,4.5% of their embryos grew into shoots on the MS culture medium;with the age of embryos growing,the survival rate of shoots increased until it reached 26% when the fruits went into ripeness;the embryos produced calli in their first generation of culturing.On the part of 65-83 d,after the blooming period,the embryos produced calli through more than 2-3 generations.On the part of 88 d,after the blooming period,the survival rate of shoots on the WPM culturing base doubled compared with that on the MS culturing base;on the same culture medium,the embryos were inhibited from growing into shoots when BA,KT or 2,4-D was added on to the culture medium.The survival rate of shoots was increased remarkably when the seeds were treated in 1 000 mg/L GA3.[Conclusion] This study provided experimental basis for the establishment of Prunus salicina cv.Zaoshi,embryos rescue techniques and cross breading.展开更多

Jinye and A. fruticosa as the research objects, the relative conductivity, MDA content, soluble sugar content, solu- ble protein content and proline content in leaf were determined during natural drop in temperature, and the SOD, POD and COD activity in leaf were determined under artificial low temperature to explore the adaptability of A. fruticosa L. cv. Jinye to low temperature. The results showed that with the decrease in natural temperature, the content of MDA in A. fruticosa L. cv. Jinye was lower than that in the control, and the contents of osmotic adjustment substances (soluble sugar, soluble protein and proline) increased, indicating that A. fruticosa L. cv. Jinye had a stronger toler- ance to low temperature compared with A. fruticosa. The activity of SOD, POD and CAT in A. fruticosa L. cv. Jinye increased continuously, higher than that in the con- trol, indicating that the resisting ability of A. fruticosa L. cv. Jinye against low tem- perature was superior to that of the control. More than 430 new varieties have been derived as mutants of rice (Oryza sativa L.) via the application of different mutagenic agents. Chemical mutagens such as ethyl methane sulphonate (EMS), diepoxybutane-derived (DEB), sodium azide and irradiation (Gamma rays, X-rays and fast neutrons) have been widely used to induce a large number of functional variations in rice and others crops. Among chemical mutagens, the alkylating agent, ethyl methane sulfonate (EMS) is the most commonly used in plants as it causes a high frequency of nucleotide substitutions, as detected in different genomes. In this study, seeds of potential genotype of the popular variety, (Oryza sativa L. spp. Indica cv. MR219) were treated with EMS at concentrations of 0.25%, 0.50%, 0.75%, 1%, 1.25%, 1.5% and 2%. Sensitivity to EMS was determined by various measurements on the M1 generation. As concentration of applied EMS increased, will decrease in germination, seedling height, root length and emergence under field conditions was observed in M1 generation as compared to the non-treatment control. Plant height and root length also decreased with increases in EMS mutagenesis in an approximately linear fashion. The LD25 and LD50 values were observed based on growth reduction of seedlings after EMS treatment with 0.25% and 0.50% on the rice variety (Oryza sativa L. spp. Indica cv. MR219).展开更多
Torreya grandis cv.Merrillii(Taxace,Torreya)is mainly distributed across the hilly areas of subtropical China and is well known for its nutritional value.In this study,the ameliorative effects of T.grandis seed oil on...Torreya grandis cv.Merrillii(Taxace,Torreya)is mainly distributed across the hilly areas of subtropical China and is well known for its nutritional value.In this study,the ameliorative effects of T.grandis seed oil on lipid metabolism were investigated,and the underlying mechanism was explored from the perspective of gut microbiota.Mice experiments showed that the rate of body mass gain in the group where the mice were fed a high-fat diet(HFD)and supplemented with 550 mg/(kg·day)T.grandis seed oil(HFD+TO550 group)was 42.27%,while it was 62.25%in the HFD group.Compared with the HFD group,the liver and fat indices,total cholesterol,triglycerides,and low-density lipoprotein cholesterol were reduced in the oil-supplement groups.Moreover,the oil supplement significantly changed the fatty acid composition and alleviated pathological damage to the liver caused by the high-fat diet.Additionally,the distinct clustering of bacteria in the composition of gut microbiota was observed in the oil treatment group compared with that in the HFD group.T.grandis seed oil significantly increased the abundance of the beneficial bacteria and short-chain fatty acid producers,including Lactobacillus,Bifidobacterium,Faecalibaculum and Allobaculum.Our results suggest that the supplements of T.grandis seed oil could alleviate hyperlipidemia caused by HFD.These positive effects are considered to be related with sciadonic acid(SCA)and are partially mediated by alterations in gut microbiota composition and functionality.展开更多

Fully expanded leaf explants were cultured on Murashige and Skoog’s (MS) medium supplemented with thidiazuron (TDZ) (0.5-4.0 mg/l), 6-benzylaminopurine (BAP) (0.5-2.0 mg/l), indole acetic acid (IAA) (2.0 mg/l), gibberlic acid (GA3) (1.0-2.0 mg/l) silver nitrate (AgNO3) (2.0 mg/l) and different carbon sources such as sucrose, fructose and glucose (10%-30%) either individually or in combination to induce adventitious buds and regeneration. The highest percentage (63%) of adventitious bud formation and regeneration (68%) was achieved in the medium containing MS with TDZ (1.0 mg/l), IAA (2.0 mg/l) and AgNO3 (2.0 mg/l). For subsequent regeneration and shoot elongation the MS medium having BAP (1.0 mg/l), GA3 (2.0 mg/l) and AgNO3 (2.0 mg/l) was found to be suitable. Amongst the carbon sources tested, the most suitable carbon source was found to be sucrose (3%) followed by fructose (2%) for adventitious bud formation. Excised in vitro shoots were rooted (60%-80%) in half strength MS medium supplemented with indole-3-butyric acid (1.0 mg/l). The well rooted plantlets were hardened in soil + sand + farm yard manure (FYM) mixture with a success rate of 70%-90%. Since in vitro regeneration is highly genotype-dependent in mulberry, the standardized protocol can be effectively used for further improvement of this leading genotype using biotechnological approaches.展开更多

Purple to provide reference for the certification and development of new varieties.[Method] With P. purpureum Schumab cv. Purple as experimental material, the variety comparison tests were conducted in Nanning, Lingyun, and Tiandeng in Guangxi from 2007 to 2009, and variety regional trials were conducted in Danzhou of Hainan, Fuzhou of Fujian and Zhanjiang of Guangdong from 2011 to 2013. Its comprehensive characters of plant height, hay yield, stem-leaf ratio and nutritional components were determined, and were compared with those of P. purpureum × P.americanum cv. Reyan No.4 and P. purpureum Schum cv. Guiminyin. [Result] The average height of P. purpureum Schumach cv. Purple was 706.54 cm, which was5.92% and 8.44% lower than that of P. purpureum × P. americanum cv. Reyan No.4 and P. purpureum Schum cv. Guiminyin, respectively. The average hay yield of P. purpureum Schumach cv. Purple was 32.74 t/ha in Nanning, Lingyun, and Tiandeng of Guangxi from 2007 to 2009, and 22.41 t/ha in Danzhou of Hainan, Fuzhou of Fujian and Zhanjiang of Guangdong from 2011 to 2013. Although its hay yield was lower than two control varieties, the difference was not significant(P 0.05).The stem-leaf ratio of P. purpureum Schumach cv. Purple was 0.98:1. Moreover,the analysis on the nutritional components of dry matter showed that compared with P. purpureum × P. americanum cv. Reyan No.4 and P. purpureum Schum cv.Guiminvin, the crude fiber of P. purpureum Schumab cv. Purple was decreased by4.76% and 2.78%, respectively; the neutral detergent fiber decreased by 5.07% and2.22%, respectively; the acid detergent fiber was decreased by 2.54% and 1.99%,respectively; the crude fat was increased by 64.29% and 43.75%, respectively; the calcium content was increased by 28.00% and 33.33%, respectively; the phosphorus content was increased by 9.68% and 22.73%, respectively; the total content of amino acid was increased by 43.23% and 68.41%, respectively; the anthocyanin content of leaf was increased by 400.00% and 216.90%, respectively(P 0.05).[Conclusion] P. purpureum Schumach cv. Purple is a new high-quality forage variety with wide adaptability, high yield, rich nutrient and good palatability, which is appropriate for comprehensive exploitation and application in the southern region of China.展开更多

Changshou Shatian You infected with Agrobacterium rhizogenes were carried out in darkness or in light. The optimizing culture system of Ri T-DNA transformed roots for C. grandis cv. Changshou Shatian You was constructed as follows: After the ventral wounded striations on the single activation cotyledon were inoculated by A. rhizogenes A4 (logarithmic period), they were cocultured at (25 ±2)℃ in darkness for 25-30 days; some transformed roots were generated from wounded striations of most cotyledons. The genetically transformed ratio is (83 ± 11)%. Axenic Ri T-DNA transformed roots (hairy roots) were harvested after five subcultures. Explants were activated on MT medium. The MS medium was used for subculture of transformed roots. Mass Ri T-DNA transformed roots in which the hormone was produced independently were harvested from this optimizing culture system. White, fresh Ri T-DNA transformed roots were (1.14 ±0.07) cm long, (0.73 ±0.04) mm wide, and the growth direction of transformed roots was negative geotropism.展开更多

文摘[Objective] The present experiment was conducted to investigate the yield,adaptability and quality characteristics of Pennisetum purpureum Schumach cv. Purple to provide reference for the certification and development of new varieties.[Method] With P. purpureum Schumab cv. Purple as experimental material, the variety comparison tests were conducted in Nanning, Lingyun, and Tiandeng in Guangxi from 2007 to 2009, and variety regional trials were conducted in Danzhou of Hainan, Fuzhou of Fujian and Zhanjiang of Guangdong from 2011 to 2013. Its comprehensive characters of plant height, hay yield, stem-leaf ratio and nutritional components were determined, and were compared with those of P. purpureum × P.americanum cv. Reyan No.4 and P. purpureum Schum cv. Guiminyin. [Result] The average height of P. purpureum Schumach cv. Purple was 706.54 cm, which was5.92% and 8.44% lower than that of P. purpureum × P. americanum cv. Reyan No.4 and P. purpureum Schum cv. Guiminyin, respectively. The average hay yield of P. purpureum Schumach cv. Purple was 32.74 t/ha in Nanning, Lingyun, and Tiandeng of Guangxi from 2007 to 2009, and 22.41 t/ha in Danzhou of Hainan, Fuzhou of Fujian and Zhanjiang of Guangdong from 2011 to 2013. Although its hay yield was lower than two control varieties, the difference was not significant(P 0.05).The stem-leaf ratio of P. purpureum Schumach cv. Purple was 0.98:1. Moreover,the analysis on the nutritional components of dry matter showed that compared with P. purpureum × P. americanum cv. Reyan No.4 and P. purpureum Schum cv.Guiminvin, the crude fiber of P. purpureum Schumab cv. Purple was decreased by4.76% and 2.78%, respectively; the neutral detergent fiber decreased by 5.07% and2.22%, respectively; the acid detergent fiber was decreased by 2.54% and 1.99%,respectively; the crude fat was increased by 64.29% and 43.75%, respectively; the calcium content was increased by 28.00% and 33.33%, respectively; the phosphorus content was increased by 9.68% and 22.73%, respectively; the total content of amino acid was increased by 43.23% and 68.41%, respectively; the anthocyanin content of leaf was increased by 400.00% and 216.90%, respectively(P 0.05).[Conclusion] P. purpureum Schumach cv. Purple is a new high-quality forage variety with wide adaptability, high yield, rich nutrient and good palatability, which is appropriate for comprehensive exploitation and application in the southern region of China.

文摘Genetic transformation experiments of the different explants from Citrus grandis cv. Changshou Shatian You infected with Agrobacterium rhizogenes were carried out in darkness or in light. The optimizing culture system of Ri T-DNA transformed roots for C. grandis cv. Changshou Shatian You was constructed as follows: After the ventral wounded striations on the single activation cotyledon were inoculated by A. rhizogenes A4 (logarithmic period), they were cocultured at (25 ±2)℃ in darkness for 25-30 days; some transformed roots were generated from wounded striations of most cotyledons. The genetically transformed ratio is (83 ± 11)%. Axenic Ri T-DNA transformed roots (hairy roots) were harvested after five subcultures. Explants were activated on MT medium. The MS medium was used for subculture of transformed roots. Mass Ri T-DNA transformed roots in which the hormone was produced independently were harvested from this optimizing culture system. White, fresh Ri T-DNA transformed roots were (1.14 ±0.07) cm long, (0.73 ±0.04) mm wide, and the growth direction of transformed roots was negative geotropism.
