Objective:To investigate the effects of ivabradine on Notch and NF-kappa B signaling pathway in myocardial cells of rats with myocardial infarction.Methods: The model of myocardial infarction was established by ligati...Objective:To investigate the effects of ivabradine on Notch and NF-kappa B signaling pathway in myocardial cells of rats with myocardial infarction.Methods: The model of myocardial infarction was established by ligating the left anterior descending coronary artery. The surviving rats were randomly divided into model group (MI group,n=8) and treatment group (IVA group,n=8). Rats with the same location but without ligation of the left anterior descending coronary artery were used as control group (CON group,n = 8). IVA was administered for 28 d. Hemodynamic and cardiac function indexes of all rats were measured: heart rate (HR), systolic pressure (SBP), diastolic pressure (DBP), mean arterial pressure (MAP), left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure (LVEDP) and maximum rate of increase and decrease of left ventricular internal pressure (+dp/dt);left ventricular mass index, left ventricular cross-sectional diameter and infarct area;The expression of Notch signaling pathway components mRNA (Notch-1, Dll-4, Hes-1) in rat cardiomyocytes was detected by PT-PCR, and the expression of DICD-1 and P65 protein was detected by western-blot. One-way ANOVA was used for comparison between groups, and SNK was used for comparison between groups.Results: SBP, DBP, MAP, LASP, LVEDP and (+dp/dt) in MI group were lower than those in control group (P<0.05), while IVA was higher than those in MI group (P<0.05). Left ventricular mass index and left ventricular sectional diameter in MI group were significantly higher than those in control group (P<0.05), but lower than those in IVA group (P<0.05). The expression of Notch-1 in MI group was significantly higher than that in control group (P<0.05), but lower than that in IVA group (P<0.05). There was no significant difference in the expression of Dll-4 and Hes-1 mRNA between the three groups (P>0.05). The expression levels of NICD-1 and P 65 in MI group were significantly higher than those in CON group (P<0.05), but lower than those in IVA group (P<0.05). Conclusion: IVA may improve cardiac function and inhibit ventricular remodeling in rats with myocardial infarction through Notch and NF-kappa B signaling pathways.展开更多

Bu...Fracture-surface topography analysis (FRASTA) was successfully used to simulate the process of crack and calculate parameters of crack such as the crack opening angle (COA) and crack-tip opening angle (CTOA). But because the amount of calculation is large, errors are often made during the course of calculation. FRASTA simulation software, namely fracture surface analyst (FSA ) is developed and a series of center-cracked tension (CCT) and double edge notched (DEN) specimens of different materials with different sizes are tested, their fracture surfaces are scanned by non-contact 319 scanning system and the elevation data of the fracture surfaces is recorded. FSA used the recorded elevation data of fracture surfaces to simulate the process of crack based on the principle of FRASTA. Results show that FSA can accurately simulate the process and calculate the parameters of crack.展开更多
BACKGROUND Malignant peritoneal mesothelioma is an unusual disease process characterized radiologically by ascites and infiltration of the peritoneum by multiple small tumor nodules.Both parietal and visceral peritone...BACKGROUND Malignant peritoneal mesothelioma is an unusual disease process characterized radiologically by ascites and infiltration of the peritoneum by multiple small tumor nodules.Both parietal and visceral peritoneum are involved by the multiple malignant tumor nodules.Computed tomography(CT)has been used to identify the anatomic pathology induced by the progression of this malignant process.AIM To identify and then describe unusual CT images in patients with malignant peritoneal mesothelioma.Recognition of these unusual radiologic findings can cause the radiologist to be suspicious of this rare malignant process.METHODS In 100 patients who were to undergo definitive treatment of malignant peritoneal mesothelioma,the findings on preoperative CT scans were catalogued.Many of these changes were repeatedly noted on the CT scans.Other pathologic CT images were less common.These unusual radiologic presentations were specially studied in this manuscript.RESULTS Eight unusual radiologic presentations of malignant peritoneal mesothelioma were selected for study.These unusual findings included a mass occurring within a Spigelian hernia,infiltration of the splenic parenchyma by spherical mesothelioma masses,infiltration of the lower mediastinum by tumor,a mesothelioma mass within a left inguinal canal,enlarged cardiophrenic angle lymph nodes,pleural plaques associated with the progression of malignant peritoneal mesothelioma,splenic notches caused by disease surrounding the spleen,and a mass greater than 5 cm associated with the proximal jejunum and directly adjacent to the anatomic location of the Treitz ligament.CONCLUSION There are unusual radiologic presentations of malignant peritoneal mesothelioma that are important to recognize in order to accurately diagnose this disease by CT.展开更多

Here, we report the impact of hypoxia-induced Jagged1 expression in cardiomyocytes(CMs) for driving the differentiation of cardiac stem cells(CSCs).Forced hypoxia-inducible factor 1α(HIF-1α) expression and physical hypoxia(5% O_2) treatment could induce Jagged1 expression in neonatal rat CMs. Pharmacological inhibition of HIF-1α by YC-1 attenuated hypoxia-promoted Jagged1 expression in CMs. An ERK inhibitor(PD98059), but not inhibitors of JNK(SP600125), Notch(DAPT), NF-κB(PTDC), JAK(AG490), or STAT3(Stattic) suppressed hypoxiainduced Jagged1 protein expression in CMs. c-Kit^+ CSCs isolated from neonatal rat hearts using a magnetic-activated cell sorting method expressed GATA4, SM22α or vWF, but not Nkx2.5 and cTnI.Moreover, 87.3% of freshly isolated CSCs displayed Notch1 receptor expression. Direct co-culture of CMs with BrdU-labeled CSCs enhanced CSCs differentiation, as evidenced by an increased number of BrdU^+/Nkx2.5^+ cells, while intermittent hypoxia for 21 days promoted co-culture-triggered differentiation of CSCs into CM-like cells. Notably, YC-1 and DAPT attenuated hypoxia-induced differentiation.Our results suggest that hypoxia induces Jagged1 expression in CMs primarily through ERK signaling,and facilitates early cardiac lineage differentiation of CSCs in CM/CSC co-cultures via HIF-1α/Jagged1/Notch signaling.展开更多
