The objective of this study was to analyze the relationship of somatic cell count (SCC) with milk yield, fat and protein percentage, fat and protein yield using analysis of variance and correlation analysis in Chine...The objective of this study was to analyze the relationship of somatic cell count (SCC) with milk yield, fat and protein percentage, fat and protein yield using analysis of variance and correlation analysis in Chinese Holstein population. The 10 524 test-day records of 568 Chinese Holstein Cattle were obtained from 2 commercial herds in Xi'an region of China during February 2002 to March 2009. Milk yield, fat percentage, fat and protein yield initially increased and then dropped down with parity, whereas protein percentage decreased and SCC increased. Analysis of variance showed highly significant effects of different subclasses SCC on milk yield and composition (P〈 0.01). Compared with milk yield with SCC ≤ 200 000 cells mL-1, milk yield losses with SCC of 200 000-500 000 cells mL-1, 501000-1 000 000 cells mL-1, ≥ 1 000 000 cells mL-1 were 0.387, 0.961 and 2.351 kg, respectively. The highly significant negative correlation coefficient between somatic cell score (SCS) and milk and protein yield, milk yield and fat and protein percentage, protein percentage and fat yield were -0.084, -0.037, -0.061, -0.168, and -0.088, respectively (P〈 0.01). The highly significant positive correlation coefficients between SCS and fat yield and fat and protein percentage, milk yield and fat and protein yield, fat percentage and protein percentage and fat yield, protein yield and protein percentage and fat yield were 0.041, 0.177, 0.105, 0.771, 0.865, 0.122, 0.568, 0.318, and 0.695, respectively (P〈 0.01). There was no significant relationship between fat percentage and protein yield (P 〉 0.05). The results of the present study first time provide the relevant base-line data for assessing milk production at Xi'an region of China.展开更多
Although Coxiella burnetii is considered to be an obligate intracellular bacterium and grows in embryonated eggs, laboratory animals and cell culture, recently it has been grown in cell-free media and on agar plates. ...Although Coxiella burnetii is considered to be an obligate intracellular bacterium and grows in embryonated eggs, laboratory animals and cell culture, recently it has been grown in cell-free media and on agar plates. This current study was conducted to compare four cell lines for their yield of C. burnetii. Four different isolates of C. burnetii (Henzerling, Arandale, Cumberland and Timony) were grown in DH82, L929, Vero and XTC-2 cell lines. The DH82 and XTC-2 cells lines produced the highest C. burnetii yield which was slightly less than the yields achieved in recently published studies using cell free media. The Arandale isolate of C. burnetii produced a significantly higher yield in DH82 cells compared to XTC-2 cells (P 0.03).展开更多

Four clinical studies are on going in our institution and enrol patients with cardiac diseases, coronary disease and type 2 diabetes, patients with osteoarthritis. Density gradient is used to separate bone marrow mononuclear cells. Cell processing looses are significant. To find out critical control points we analysed processing process and differences in cell yields between operators performing cell extraction. Bone marrow mononuclear cells were isolated using Ficoll density gradient centrifugation. Cells were counted using flow cytometry for mononuclear cell total counts, CD34+ population count and viability analysis. The patients who underwent bone marrow aspiration followed by cell isolation received cell suspension for transplantation. Two cells processing for separate patients were performed at once. Same standard operation procedures were applied. Processing looses between operators performing cell extraction were analysed. Bone marrow samples from eight patients were processed. Mononuclear cells were extracted. Operator performances were compared. Operator A average bone marrow mononuclear cell yield in starting material was 9,97 ± 9,98 %, CD34+ population yield 75,46±79,67%. Operator B average bone marrow mononuclear cell yield in starting material was 24,68 ± 14,8 %, CD34+ population yield 70,42 ±44.84%. Operator A average cell viability in starting material was 45,24 ± 9,55%, after cell processing 42,96 ± 23,66 %. Operator B average cell viability in starting material was 49,85 ± 5,48%, after cell processing 69,52 ± 6,65 %.展开更多
Rice(Oryza sativa L.)is a major food crop worldwide.Plant height and yield are important agronomic traits of rice.Several genes regulating plant height and/or yield have been cloned.However,the molecular mechanisms co...Rice(Oryza sativa L.)is a major food crop worldwide.Plant height and yield are important agronomic traits of rice.Several genes regulating plant height and/or yield have been cloned.However,the molecular mechanisms coordinating plant height and yield are not fully characterized.Here,we report a novel gene,OstMAPKKK5 that encodes a truncated variant of a mitogen-activated protein kinase kinase kinase 5(OsMAPKKK5)lacking an intact kinase domain.Transgenic plants overexpressing OstMAPKKK5 in indica cultivar 9311 showed increased plant height,grain length,grain width,1000-grain weight,grain number per main panicle,and yield.Real-time quantitative PCR showed that OstMAPKKK5 was widely expressed in various tissues and developmental stages.The increased plant height and yield were attributed to enlarged cell size.Overexpression of OstMAPKKK5 led to higher contents of various forms of endogenous gibberellin(GA),especially the most common active forms,GA1,GA3,GA4.We concluded that OstMAPKKK5 positively regulates plant height and yield in rice by affecting cell size,and that its underlying mechanism is based on increased endogenous GA content.展开更多

From the full width at h...In situ synchrotron angle-dispersive x-ray diffraction experiments on niobium powders have been conducted at pressures up to 61 GPa and room temperature using the diamond anvil cell technique. From the full width at half maximum of the measured diffraction lines, the yield strength was derived with the line-width analysis theory. The niobium powder sample was found to be compressed more packed firstly and then yielded at~14 GPa–18 GPa. Following an initial increase in the yield strength with pressure, an obvious decrease was observed occurring at ~42 GPa–47 GPa accompanying with a typical pressure dependence above 47 GPa. The experimentally observed anomalous softening of the yield strength in niobium surprisingly follows the trend of the predicted unusual softening in the shear modulus by the recent theoretical investigations. The possible mechanisms, applicable to interpret the yield strength softening of materials at high pressure,were also discussed in detail.展开更多

This work investigates an accident during the pyrochemical extraction of Uranium and Plutonium from PWR spent fuel in an argon atmosphere hot cell. In the accident, the heavy metals (U and Pu) being extracted are acci...This work investigates an accident during the pyrochemical extraction of Uranium and Plutonium from PWR spent fuel in an argon atmosphere hot cell. In the accident, the heavy metals (U and Pu) being extracted are accidently exposed to air from a leaky instrument penetration which goes through the cell walls. The extracted pin size pieces of U and Pu metal readily burn when exposed to air. Technicians perform the electrochemical extraction using manipulators through a 4 foot thick hot cell concrete wall which protects them from the radioactivity of the spent fuel. Four foot thick windows placed in the wall allow the technicians to visually control the manipulators. These windows would be exposed to the heat of the metal fire. This analysis determines if the thermal stress caused by the fire would crack the windows and if the heat would degrade the window seals allowing radioactivity to escape from the cell.展开更多
