After gene mutation, the pcDNA3.1/APP595/596 plasmid was transfected into HEK293 cells to establish a cell model of Alzheimer's disease. The cell model was treated with donepezil or compound Danshen tablets after culture for 72 hours. Reverse transcription-PCR showed that the mRNA expression of amyloid protein precursor decreased in all groups following culture for 24 hours, and that there was no significant difference in the amount of decrease between donepezil and compound Danshen tablets. Our results suggest that compound Danshen tablets can reduce expression of the mRNA for amyloid protein precursor in a transgenic cell model of Alzheimer's disease, with similar effects to donepezil. 展开更多

Recovery following stroke involves neurogenesis and axonal remodeling within the ischemic brain. Gualou Guizhi decoction (GLGZD) is a Chinese traditional medicine used for the treatment of post-stroke limb spasm. GLGZD has been reported to have neuroprotective effects in cerebral ischemic injury. However, the effects of GLGZD on neurogenesis and axonal remodeling following cerebral ischemia remain unknown. In this study, a rat model of focal cerebral ischemia/reperfusion was established by middle cerebral artery occlusion. Neurologi- cal function was assessed immediately after reperfusion using Longa's 5-point scoring system. The rats were randomly divided into vehicle and GLGZD groups. Rats in the sham group were given sham operation. The rats in the GLGZD group were intragastrically administered GLGZD, once daily, for 14 consecutive days. The rats in the vehicle and sham groups were intragastrically administered distilled water. Modified neurological severity score test, balance beam test and foot fault test were used to assess motor functional changes. Nissl staining was performed to evaluate histopathological changes in the brain. Immunofluorescence staining was used to examine cell proliferation using the marker 5-bromo-2'-deoxyuridine (BrdU) as well as expression of the neural precursor marker doublecortin (DCX), the astrocyte marker glial fibrillary acidic protein (GFAP) and the axon regeneration marker growth associated protein-43 (GAP-43). GLGZD substan- tially mitigated pathological injury, increased the number of BrdU, DCX and GFAP-immunoreactive cells in the subventricular zone of the ischemic hemisphere, increased GAP-43 expression in the cortical peri-infarct region, and improved motor function. These findings suggest that GLGZD promotes neurological functional recovery by increasing cell proliferation, enhancing axonal regeneration, and in- creasing the numbers of neuronal precursors and astrocytes in the peri-infarct area. 展开更多

Objective: To observe the clinical effect of Rebixiao granule (热痹消颗粒剂, RBXG) in treating repeatedly attacking acute gouty arthritis and through experimental study on blood uric acid to explore RBXG's therapeutic mechanism. Methods: Ninety repeatedly attacking acute gouty arthritis patients were divided into the treated group ( n =60) and control group ( n =30). The treated group was treated with RBXG, and the control group was treated with Futalin tablets (diclofenac sodium). The baseline treatment including good rest, low purine diet, sufficient water drinking and urine alkalization, etc. was then given to both groups. Hypoxanthine 600 mg/kg and niacin 100 mg/kg was applied to hyperuricemic mice by gastrogavage to establish the animal models. Results: The clinical effective rate of the treated group was 95.0% and that of the control 90.0%. Good therapeutic effects were won, insignificant difference ( P >0.05)was shown between the two groups. However, the cure rate of the treated group was 26.7% while that of the control group was 10.0%, with significant difference ( P <0.01) shown between them. The treated group had its blood uric acid lowered, which was significantly different ( P <0.05) from that of the control group. The animal experiment indicated that all the three groups treated with different dosages of RBXG, as well as the Ash bark and Smilax glabra rhizome groups had their blood uric acid content reduced in the hyperuricemic mice. Conclusion: RBXG has a quicker initiation and better treatment effects than sole anti-inflammatory and analgesic agents on the treatment of repeatedly attacking acute gouty arthritis, showing no obvious toxic or adverse reactions and therefore good for long-term administration and likely to be a safe TCM preparation to control the symptoms and reduce the onsets of repeatedly attacking of acute gouty arthritis. The animal experiment shows that both the compound preparation and part of the single ingredients in the recipe have the function of reducing blood uric acid. However, the compound recipe has better therapeutic effects, proving to be superior to single drugs. 展开更多

Objective:Based on data mining technology, we attempted to explore the medication rules of modern traditional Chinese medicine (TCM)compounds in non-small cell lung cancer (NSCLC) treatment, to provide a reference for clinical drug use. Methods:From 2010 to 2017, TCM compounds used for NSCLC treatment were collected from the Beijing 301 Hospital. The modern TCM compounds utilized in the treatment of NSCLC were established in the prescription database. Excel, SPSS 22, and SPSS Modeler14.2 software were utilized for the frequency analysis, factor analysis, cluster analysis, and association analysis. Then, the quantitative and qualitative analyses of the regularity of TCM compound medications were performed, and the possible mechanism was discussed. Results:The treatment of NSCLC using Chinese herbal compounds involved 231 prescriptions, 389 types of Chinese herbs, and 135 types of high-frequency Chinese herbs. Of these, Fritillaria cirrhosa, stir-baked fried Scutellariae, raw Os Draconis, Poria cocos (Schw.) Wolf, and Scutellaria barbata were the top five frequently prescribed Chinese herbs. Among the 39 types of drugs, heat-clearing and detoxifying drugs and qi-tonifying drugs were the leading. Cold, warm, flat, slightly cold, sweet, bitter, and pungent of four properties and five tastes and the meridians of lung, spleen, and stomach were most commonly selected. Factor analysis extracted 12 common factors, and the cumulative contribution rate was 65.595%, which mainly contained tonifying qi and blood;tonifying yin, clearing away heat, and eliminating stagnation;tonifying the spleen, regulating qi, and eliminating phlegm.Forty drug groups were obtained by cluster analysis;a total of 63 association rules were obtained by association analysis. The pairs of Poria cocos→dried tangerine peel and fried Atractylodes macrocephala→dried tangerine peel were commonly used in NSCLC, while the three most frequent herb groups were raw Astragalus→fried A. macrocephala and Poria cocos;raw-medicated leaven→fried A. macrocephala and Poria cocos;and dried tangerine peel→fried A. macrocephala and Poria cocos. Conclusion:Lung cancer is mainly caused by qi stagnation, phlegm obstruction, phlegm, and blood stasis. Based on the principle of strengthening the body and dispelling pathogens, clinical treatment of NSCLC involves clearing heat and detoxifying, tonifying the spleen, regulating qi, eliminating phlegm to dispel pathogens, and tonifying qi and blood to strengthen the body. 展开更多

Cerebral ischemia-reperfusion injury(CIRI) refers to a pathological phenomenon that aggravates the injury after the restoration of blood perfusion and oxygen supply to the cerebral ischemia-induced tissues and organs, with a relatively high incidence. The traditional Chinese medicine(TCM) believes that Qi deficiency and blood stasis are the cause of CIRI. Therefore, Chinese medicine for tonifying Qi and activating blood is regarded as an important choice for the treatment of CIRI. In recent years, it has been found that many Chinese herbal medical ingredients and compound Chinese medicine(CCM) have significant anti-CIRI effects, and their mechanisms of action mainly include improving brain blood supply, neuroprotection, regulating signal pathways such as TLR4/HO-1/Bcl-2, protecting mitochondrial function, regulating related protein levels, and regulating oxidative molecule levels. This article summarizes and introduces the pharmacological mechanisms of Tonifying-Qi and activating-blood Chinese medicine and CCM which have the function of anti-CIRI. Our goal is to provide effective reference for further researches on the cerebral protection of related TCMs or compounds and their clinical application. 展开更多

Methods: In a blinded, randomized, Kangfuxin Solution(康复新液, KFX)-controlled, single-centre clinical trial, 120 patients with lung, esophagus or mediastinal cancer were prospectively enrolled and assigned to the treatment group(60 cases) and control group(60 cases) by the random number table method. All patients received concurrent or sequential radiotherapy(2 Gy per day, 5 times per week, for 4 weeks) and were treated for 4 weeks since the radiation therapy. Patients in the treatment group were given 12 mg CZSG orally, thrice daily, while patients in the control group were given 10 m L KFX orally, thrice daily. The major indicators were observed, including the incidence and grade of esophagitis, time of occurrence and duration. Minor indicators were changes of CM symptoms, weight and Karnofsky Performance Status(KPS) Scale during 4 weeks from the beginning, recorded once a week. Blood routine examination and hepatorenal function were detected at the 2nd and 4th weeks. Results: The incidence and grade of ARIE were significantly decreased in the treatment group compared with the control group(P〈0.05). CZSG appeared to significantly delay the time of ARIE occurrence and reduce the duration compared with KFX(P〈0.05). The scores of CM symptoms, KPS and weight were improved significantly in the treatment group compared with the control group(P〈0.05). There were no blood routine and hepatorenal function abnormal or obvious side-effects in both groups. Hemoglobin was improved and neutrophil and interleukin 6 were decreased in both groups after 4-week treatment compared with before treatment(P〈0.05), and there was no significant difference between the two groups(P〉0.05). Conclusions: CZSG can decrease the incidence and grade of ARIE, delay the time of occurrence, reduce duration and alleviate the damage of ARIE. It is safe and effective in the prevention and cure of ARIE. 展开更多

Background:Non-alcoholic fatty liver disease(NAFLD)is a multifactorial liver metabolic disease, which af-fects nearly a quarter of the world’s population.Yunpi Qushi Jiangzhuo Recipe(QSJZR)is a traditional Chi-nese medicine compound, which is composed of Amomum kravanh Pierre ex Gagnep,Coix lacryma-jobi L.var.ma-yuen(Roman.)Stapf,Prunus armeniac a L.var.ansu Maxim,Salvia miltiorrhiza Bge,Nelumbo nucifera Gaertn,Alisma plantago-aquatica Linn,Polyporus umbellatus(Pers.)Fries,Poria cocos(Schw.)Wolf, and Artemisia capillaris Thunb.QSJZR has a certain therapeutic effect on NAFLD patients, but its specific mechanism is not clear. Objective:To investigate the effect of QSJZR on high-fat diet(HFD)-fed NAFLD mice and its mechanism.Methods:Twenty-four SPF female C57BL/6 J mice(21.0±0.5 g)were randomly divided into normal diet(ND)group,HFD group and QSJZR group.ND group was given basal diet, while the other two groups were given HFD.Meanwhile, each mouse in QSJZR group was given 0.2 mL/day(containing 2.27 g crude drug per mL)QSJZR,ND group and HFD group were given the same amount of normal saline for 13 consecutive weeks. Then, the serum was collected for biochemical assay, and the liver was removed for pathological examination and biochemical analysis. Results:Body weight and white fat weight of the HFD-induced NAFLD mice significantly decreased after min-istration with QSJZR,while liver weight had no significant change.QSJZR also significantly reduced liver and serum triglyceride levels, and alleviated hepatocyte lipid deposition by regulating genes and proteins expression related to lipid metabolism, including AMPK,SREBP1C,CPT1A and ACC.In addition, compared with HFD group, liver malondialdehyde(MDA)content was lower in QSJZR group, while glutathione peroxidase(GPx)content was higher.Serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)levels were lower in QSJZR group than those in HFD group.Treatment with QSJZR significantly alleviated liver injury by increasing BCL2/BAX protein ratio and down-regulating ASK1/JNK pathway. Conclusion:Administration of QSJZR to NAFLD mice once daily for 13 weeks can decrease lipid levels, and alleviate liver damage. These results suggest that QSJZR might be used to treat NAFLD,although more studies need to be conducted for further verification. 展开更多
