P4 medicine (personalized, predictive, preventive, and participatory/precision) similar to Chinese medicine theory is beginning to representing the pioneer in the Western world. In this review, we summarized different domains of Chinese medicine. Based on the basic of promoting health wellness, we compared the fundamental theory of Chinese medicine to the new merging P4 medicine idea of Western medicine. We also discussed the potential for using modern computational medicine technique to integrate Chinese medicine theory and Western medicine theory. 展开更多

利用电喷雾-四极杆-飞行时间串联质谱(ESI-Q-TOF MS/MS)技术,在负离子模式下,探讨巴戟天中4种环烯醚萜苷和2种蒽醌成分的质谱裂解途径。通过[M-H]-获得化合物的相对分子质量信息,进一步对[M-H]-进行碰撞诱导解离,获得相应化合物的裂解... 利用电喷雾-四极杆-飞行时间串联质谱(ESI-Q-TOF MS/MS)技术,在负离子模式下,探讨巴戟天中4种环烯醚萜苷和2种蒽醌成分的质谱裂解途径。通过[M-H]-获得化合物的相对分子质量信息,进一步对[M-H]-进行碰撞诱导解离,获得相应化合物的裂解途径。结果表明,环烯醚萜苷主要的裂解途径是首先脱去母环上的功能基团,如中性丢失H_2O、CO_2、CH_3COOH和糖单元等部分;其次是二氢吡喃环和糖环的断裂,m/z113、101为环烯醚萜苷母环断裂的特征碎片离子。蒽醌类化合物的裂解行为是连续失去CO,也可以失去CO_2。这些质谱裂解行为的研究有助于环烯醚萜苷和蒽醌类化合物的结构解析,也可为其他同类化合物的鉴定提供依据。 展开更多

Brazilin Actuates Ferroptosis in Breast Cancer Cells via p53/SLC7A11/GPX4 Signaling Pathway

Objective:To investigate the mechanism of induction of ferroptosis by brazilin in breast cancer cells.Methods:Breast cancer 4T1 cells were divided into 6 groups:control,brazilin 1/2 half maximal inhibitory concentration(ICso),ICso,2×ICso,erastin(10μg/mL)and capecitabine(10μg/mL)groups.The effect of brazilin on the proliferation of 4T1 cells was detected by cell counting kit-8 assay,and the treatment dose of brazilin was screened.The effect of brazilin on the mitochondrial morphology of 4T1 cells,and the mitochondrial damage was evaluated under electron microscopy.The levels of Fe+,reactive oxygen species(ROS),malondialdehyde(MDA),glutathione(GSH)and glutathione peroxidase 4(GPX4)were estimated using various detection kits.The invasion and migration abilities of 4T1 cells were detected by scratch assay and transwell assay.The expressions levels of tumor protein p53,solute carrier family 7 member 11(SLC7A11),GPX4 and acyl-CoA synthetase long-chain family member 4(ACSL4)proteins were quantified by Western blot assay.Results:Compared to the control group,the 10(1/2 IC_(50)),20(IC_(50))and 40(2×IC_(50))μg/mL brazilin,erastin,and capecitabine groups showed a significant decrease in the cell survival rate,invasion and migration abilities,GSH,SLC7A11 and GPX4 protein expression levels,and mitochondrial volume and ridge(P<0.05),and a significant increase in the mitochondria membrane density,Fe2+,ROS and MDA levels,and p53 and ACSL4 protein expression levels(P<0.05).Conclusions:Brazilin actuated ferroptosis in breast cancer cells,and the underlying mechanism is mainly associated with the p53/SLC7A11/GPX4 signaling pathway. 展开更多

Objective To evaluate the effect of Guilu Erxian Glue(龟鹿二仙胶,GEG)on cyclophosphamide(CTX)-induced bone marrow hematopoietic stem cells(HSCs)senescence in mice and explore the underlying mechanism.Methods The H22 liver cancer ascites lump model was established in male Kunming mice by injecting intraperitoneally(i.p.)with 5×10^6/mL H22 cells per mouse.Fifty tumor-bearing mice were divided into the control,model,pifithrin-α,GEG,and GEG+pifithrin-αgroups using a random number table,10 mice in each group.CTX(100 mg/kg i.p.)was administrated to mice from day 1 to day 3(d1–d3)continuously except for the control group.The mice in the pifithrin-α,GEG and GEG+pifithrin-αgroups were treated with pifithrin-α(2.2 mg/(kg·d)i.p.)for 6 consecutive days(d4–d9),GEG(9.5 g/(kg·d)i.p.)for 9 consecutive days(d1–d9),and GEG plus pifithrin-α,respectively.HSCs were collected after 9-d drug treatment.The anti-aging effect of GEG was studied by cell viability,cell cycle,andβ-galactosidase(β-gal)assays.The mRNA and protein expressions of cyclin-dependent kinase 2(CDK2),CDK4,inhibitor of cyclin-dependent kinase 4a encoding the tumor suppressor protein p16^(p16^INK4a),p21^Cip1/Waf1,p53,and phosphorylated retinoblastoma(pRb)were evaluated by quantitative real-time reverse transcription-polymerase chain reaction and semi-quantitative Western blot,respectively.Results Compared with the model group,GEG increased cell viability as well as proliferation(P<0.05 or P<0.01)and reducedβ-gal expression.Furthermore,GEG significantly decreased the expressions of p16^INK4a,p53 and p21^Cip1/Waf1 proteins,and increased the expressions of CDK2,CDK4 and pRb proteins compared with the model group(P<0.05 or P<0.01).Conclusion GEG can alleviate CTX-induced HSCs senescence in mice,and the p16^INK4a-Rb signaling pathway might be the underlying mechanism. 展开更多

目的:建立保健食品及中成药中非法添加8种非甾体抗炎药物的检测方法。方法:采用溶剂超声或液-液萃取方式对样品进行提取富集,气相色谱-质谱联用(GC-MS),Scan模式进行定性检测,选择离子扫描模式(SIM)进行定量。采用DB-5 ms毛细管色谱柱(3... 目的:建立保健食品及中成药中非法添加8种非甾体抗炎药物的检测方法。方法:采用溶剂超声或液-液萃取方式对样品进行提取富集,气相色谱-质谱联用(GC-MS),Scan模式进行定性检测,选择离子扫描模式(SIM)进行定量。采用DB-5 ms毛细管色谱柱(30 m×0.25 mm×0.25μm),程序升温:初始温度120℃(1 min),10℃.min-1升至230℃,15℃.min-1升至270℃(20 min),选择EI离子源,溶剂延迟2.5 min,质谱扫描40~350 amu。结果:在选定的色谱条件下,在30 min内快速检测8种非甾体抗炎药物,得到较好的分离;样品筛查结果发现2批样品中分别添加了对乙酰氨基酚和双氯芬酸钠。结论:该法快速﹑准确,专属性强,可用于保健食品及中成药中添加非甾体抗炎药的快速检测。 展开更多
