The...A 120-day feeding experiment was conducted to investigate the effects of partial replacement of brown fish meal (BFM) by fermented soybean meal (FSBM) in diets of Chinese soft-shelled turtle (Pelodiscus sinensis). The turtles (initial mean body weight, (115.52 ± 1.05) g) were fed with three experimental diets, in which 0%, 4.72% and 9.44% BFM protein was replaced by 0%, 3% and 6% FSBM, respectively. Results showed that the feeding rate (FR), specific growth rate (SGR) and feed efficiency ratio (FER) of turtles fed with the diet containing 3% FSBM were not significantly different from the control group (0% FSBM) (P > 0.05). However, FR, SGR and FER of turtles fed with the diet containing 6% FSBM were significantly lower than those of the control group (P < 0.05). No significant differences were observed in the activities of serum glutamic-oxalacetic transaminase and glutamicpyruvic transaminase among dietary treatments (P > 0.05). However, the uric acid concentration in turtles fed with the diet containing 3% or 6% FSBM was significantly lower than that in the control group (P < 0.05). There were no significant differences in the activities of lysozyme, alkaline phosphatase and total superoxide dismutase among dietary treatments (P > 0.05). The results suggested that FSBM could replace 4.72% BFM protein in turtle diets without exerting adverse effects on turtle growth, feed utilization and measured immune parameters.展开更多

Apparent digestibility (AD) of dry matter (DMAD), protein (PAD) and protein real digestibility (PRD) were significantly affected by ration level, but not by feeding frequency when the ration level was similar. However, the feeding frequency affected the AD, DMAD, PAD and PRD significantly when the turtles were fed to satiation. The relationship between fecal protein content (Y) and protein intake (At) can be expressed as a quadric equation: Y=-0.1742+0.1476X-0.0003X^2 (r^2=0.876, n=27, F=93.92, P〈0.01).展开更多
目前,细菌性疾病是中华鳖养殖中的常见疾病。实验采用常规方法从呈白底板症状的中华鳖(Pelodiscus sinensis)肝脏、脾脏、肾脏、胃积液等中分离纯化病原菌,经形态观察、生理生化鉴定和16S r RNA序列分析确定其种属,通过回归感染实验判...目前,细菌性疾病是中华鳖养殖中的常见疾病。实验采用常规方法从呈白底板症状的中华鳖(Pelodiscus sinensis)肝脏、脾脏、肾脏、胃积液等中分离纯化病原菌,经形态观察、生理生化鉴定和16S r RNA序列分析确定其种属,通过回归感染实验判断分离菌株致病性,并以纸片扩散法测定其药物敏感性。结果显示:自病鳖中分离可得到一种优势菌落,其表面湿润光滑、微隆起、半透明,呈灰黄色;基于VITEK 2的64个生化反应的分析提示,分离菌理化特性与迟缓爱德华氏菌(Edwardsiella tarda)基本一致;16S rRNA序列分析显示,其与迟缓爱德华氏菌同源性达98%以上。腹腔注射回归感染实验引起稚鳖死亡,且细菌分离能再次得到相同分离株,表明分离到的迟缓爱德华氏菌具有致病性。药敏试验表明该菌对庆大霉素、阿米卡星、新霉素、头孢哌酮、氨曲南等5种抗生素较为敏感。本研究可为中华鳖病害的正确诊断和科学防治提供参考。展开更多
In this paper, RT PCR analysis on SOX gene of seven tissues from the Chinese soft shelled turtle (Pelodiscus sinensis) was studied, and SOX gene fragments of expression from the testicle, brain, heart, kidney and sple...In this paper, RT PCR analysis on SOX gene of seven tissues from the Chinese soft shelled turtle (Pelodiscus sinensis) was studied, and SOX gene fragments of expression from the testicle, brain, heart, kidney and spleen were cloned using RT PCR products.The results show that SOX genes has specific expression in the testicle, brain, spleen, cardiac muscle and kidney and isn’t expression in muscle, liver and ovary of femel.The results of sequence reveal that the SOX genes of expression in the testicle are TSSOX1、TSSOX4 TSSOX5 and TSSOX9, and those are TSSOX2 and TSSOX4 in the brain, and this is TSSOX4 in the spleen and heart tissue, and those are TSSOX2 and TSSOX3 in the kidney tissue.This suggests that the SOX gene act important role not only on the sex determination, but also on the development of neural system, immunocyte system and the differentiation of male germ cell.展开更多
