Background:The important roles of liver and kidney in the elimination of injurious chemicals make them highly susceptible to the noxious activities of various toxicants including cobalt chloride(CoCl_(2)).This study w...Background:The important roles of liver and kidney in the elimination of injurious chemicals make them highly susceptible to the noxious activities of various toxicants including cobalt chloride(CoCl_(2)).This study was designed to investigate the role of glycine in the mitigation of hepato-renal toxicities associated with CoCl_(2)exposure.Methods:Forty-two(42)male rats were grouped as Control;(CoCl_(2);300 ppm);CoCl_(2)+Glycine(50 mg/kg);CoCl_(2)+Glycine(100 mg/kg);Glycine(50 mg/kg);and Glycine(100 mg/kg).The markers of hepatic and renal damage,oxidative stress,the antioxidant defense system,histopathology,and immunohistochemical localization of neutrophil gelatinase associated lipocalin(NGAL)and renal podocin were evaluated.Results:Glycine significantly reduced the markers of oxidative stress(malondialdehyde content and H_(2)O_(2) generation),liver function tests(ALT,AST,and ALP),markers of renal function(creatinine and BUN),and decreased the expression of neutrophil gelatinase-associated lipocalin(NGAL)and podocin compared with rats exposed to CoCl_(2)toxicity without glycine treatment.Histopathology lesions including patchy tubular epithelial necrosis,tubular epithelial degeneration and periglomerular inflammation in renal tissues,and severe portal hepatocellular necrosis,inflammation,and duct hyperplasia were observed in hepatic tissues of rats exposed to CoCl_(2)toxicity,but were mild to absent in glycine-treated rats.Conclusion:The results of this study clearly demonstrate protective effects of glycine against CoCl_(2)-induced tissue injuries and derangement of physiological activities of the hepatic and renal systems in rats.The protective effects are mediated via augmentation of total antioxidant capacity and upregulation of NGAL and podocin expression.展开更多

The results show that the precipitation reaction is diffusion-controlled. The main factors affecting the oxidation rate such as the stirring speed, solution temperature, ozone partial pressure, initial concentration and flow rate were investigated. The kinetics equation of each condition was established. The results indicate that the oxidation rate is independent of the initial concentration or solution temperature. The oxidation rate increases obviously with increasing the stirring speed. The linear relationship between ozone partial pressure or flow rate and oxidation rate is found.展开更多

Through the comparative analysis of the four spectra we have categorically assigned the OH-related vibration modes of hydroxyl groups in the trimeric hydrogen bond environment (Co3 =OH)3 … Cl/Br, and tentatively suggested vibration modes of O-Co-O, Co O and Cl/Br-Co-Cl/Br units. The accumulation of excessive reactive oxygen species can exacerbate any injury of retinal tissue because free radicals can trigger lipid peroxidation,protein damage and DNA fragmentation.Increased oxidative stress is...The accumulation of excessive reactive oxygen species can exacerbate any injury of retinal tissue because free radicals can trigger lipid peroxidation,protein damage and DNA fragmentation.Increased oxidative stress is associated with the common pathological process of many eye diseases,such as glaucoma,diabetic retinopathy and ischemic optic neuropathy.Many studies have demonstrated that Lycium barbarum polysaccharides(LBP)protects against oxidative injury in numerous cells and tissues.For the model of hypoxia we used cultured retinal ganglion cells and induced hypoxia by incubating with 200μM cobalt chloride(CoCl2)for 24 hours.To investigate the protective effect of LBP and its mechanism of action against oxidative stress injury,the retinal tissue was pretreated with 0.5 mg/mL LBP for 24 hours.The results of flow cytometric analysis showed LBP could effectively reduce the CoCl2-induced retinal ganglion cell apoptosis,inhibited the generation of reactive oxygen species and the reduction of mitochondrial membrane potential.These findings suggested that LBP could protect retinal ganglion cells from CoCl2-induced apoptosis by reducing mitochondrial membrane potential and reactive oxygen species.展开更多

Cobalt chloride was used to establish a model of hypoxia. Western blotting indicated that cytoglobin was overexpressed and there was low expression of hypoxia-inducible factor-la in SH-SY5Y cells after transfection. Following cobalt chloride-induced hypoxia, cytoglobin and hypoxia-inducible fac- tor-la expression gradually increased in SH-SY5Y cells. Flow cytometry showed that with increas- ing duration of hypoxia, the proportion of normal cells significantly diminished in the transfected and non-transfected groups. The proportion of cells in the early stages of apoptosis increased. However, the proportion of apoptotic cells was significantly lower in the transfected group compared with the non-transfected group. These results demonstrate that cytoglobin and hypoxia-inducible factor-la are strongly up-regulated by hypoxia, and that there is a strong relationship between hy- poxia-inducible factor-la and cytoglobin during hypoxic injury.展开更多

Objective:To investigate to the expression effect of hypoxia and hypoxia/reoxygenation on ROS,MAPKs and cell apoptosis in H9c2 cardiomyocytes.Methods:H9c2 cells were treated with cobalt chloride(CoCl2)to establish the...Objective:To investigate to the expression effect of hypoxia and hypoxia/reoxygenation on ROS,MAPKs and cell apoptosis in H9c2 cardiomyocytes.Methods:H9c2 cells were treated with cobalt chloride(CoCl2)to establish the chemical hypoxia and hypoxia/reoxygenation-induced cardiomyocyte injury model.CoCl2 was used to process cells at different concentrations from 150-2400μmol/L and different time from4-24 h;H9c2 cells viability was detected by MTT,and the intracellular ROS level was measured by 2’,7’-dichlorflμoresceindiacetate(DCFH-DA)and dihydroethidiμm(DHE)staining and photoflurography.The active expression level of mitogen-activated protein kinases(MAPKs)(including JNK,ERK and p38)and caspase-3.Results:At the concentration from 300 to 1200μmol/L,CoCl2 does/time-dependently inhibited the cell viability in H9c2 cells(P<0.01).Compared with control group,the ROS levels in hypoxia group were significantly increased(P<0.05).In hypoxia group,the active expression levels of p-JNK,p-p38 and caspase-3 was higher than those in control group(P<0.05).However,the expression of p-ERK wasn’t significant differernce.Furthermore,all the expression levels of ROS,p-JNK,p-ERK,p-p38 and caspase-3 in H/R group were significantly raised compared with hypoxia group(P<0.01).Conclusions:Reoxygenation further aggravate chemical hypoxia induced cardiomyocyte oxidative stress injury by activating ROS/MAPKs signals,suggesting the role of myocardial ischemia/reperfusion injury in the pathogenesis of ischemic heart disease.展开更多
