Long-term application of levodopa (L-3, 4-dihydroxyphenylalanine, L-DOPA) for Parkinson's disease can lead to adverse effects and reduce the amount of dopamine transporter (DAT) in the corpus striatum. The present study attempted to vedfy whether increasing the amount of DAT can reduce the adverse effects of L-DOPA. The specific radioactive uptake value of DAT in the corpus striatum of the lesioned hemisphere was significantly decreased, but was significantly increased following administration of compound rehmannia formula [Radix rehmanniae preparata (prepared rehmannia root), Concha margantifera usta (nacre), Radix paeoniae alba (white peony alba), Radix salviae miltiotThizae (Danshen root), Scorpio (scorpion), green tea] for 4 weeks. The changes in DAT 1251-beta-carbomethoxy-3 beta-(4-iodophenyl) tropane autoradiography were consistent with those in radioactivity. The results revealed that the compound rehmannia formula can reduce the adverse effects of L-DOPA in treating Parkinson's disease, possibly by increasing the amount of DAT. 展开更多

Compound Formula Rehmannia has been shown to be clinically effective in treating Parkinson's disease and levodopa-induced dyskinesia; however, the mechanisms remain unclear. In this study, we established a model of Parkinson's disease dyskinesia in rats, and treated these animals with Compound Formula Rehmannia. Compound Formula Rehmannia inhibited the increase in mRNA expression of N-methyl-D-aspartate receptor subunits 1 and 2 and excitatory amino acid neurotransmitter genes, and it inhibited the reduction in expression of γ-aminobutyric acid receptor B1, an inhibitory amino acid neurotransmitter gene, in the corpus striatum. In addition, Compound Formula Rehmannia alleviated dyskinesia symptoms in the Parkinson's disease rats. These experimental findings indicate that Compound Formula Rehmannia alleviates levodopa-induced dyskinesia in Parkinson's disease by modulating neurotransmitter signaling in the corpus striatum. 展开更多

目的探讨阴虚动风证帕金森病LID大鼠脑内氨基酸类神经递质的动态变化及复方地黄方的干预作用。方法采用6-羟基多巴胺(6-OHDA)偏侧损毁黑质制备帕金森病(PD)大鼠模型,成功的PD模型大鼠腹腔注射左旋多巴/苄丝阱[50/12.5(mg.kg)],制备阴虚... 目的探讨阴虚动风证帕金森病LID大鼠脑内氨基酸类神经递质的动态变化及复方地黄方的干预作用。方法采用6-羟基多巴胺(6-OHDA)偏侧损毁黑质制备帕金森病(PD)大鼠模型,成功的PD模型大鼠腹腔注射左旋多巴/苄丝阱[50/12.5(mg.kg)],制备阴虚动风证帕金森病LID病证的大鼠模型。将大鼠分为复方地黄方组、LID组、正常对照组、假手术组,并在治疗4周、6周后取纹状体,应用高效液相法(HPLC)测定各组大鼠纹状体内氨基酸的含量。结果阴虚动风证帕金森病LID模型大鼠纹状体内兴奋性氨基酸显著升高;复方地黄方能显著降低兴奋性氨基酸的含量,抑制性氨基酸含量则明显升高。结论复方地黄方缓解帕金森病LID的机制可能是通过调节脑内氨基酸的水平、减轻其兴奋毒性来实现的。 展开更多

目的探讨复方地黄方对帕金森病(PD)左旋多巴毒副反应大鼠淋巴细胞(PBL)表面多巴胺转运体(DAT)表达的影响。方法在经典注射6-羟基多巴胺损毁法制作PD大鼠模型的基础上,进一步腹腔注射50 mg·kg-1·d-1左旋多巴(LD)/苄丝肼2 w,制... 目的探讨复方地黄方对帕金森病(PD)左旋多巴毒副反应大鼠淋巴细胞(PBL)表面多巴胺转运体(DAT)表达的影响。方法在经典注射6-羟基多巴胺损毁法制作PD大鼠模型的基础上,进一步腹腔注射50 mg·kg-1·d-1左旋多巴(LD)/苄丝肼2 w,制备LD大鼠模型。采用随机区层分组法,分为正常对照组、假手术组、LD模型组及复方地黄方组四组,每组12只,分别干预4 w、6 w,应用RT-PCR法测定大鼠PBL表面DAT的表达。结果 LD模型组大鼠PBL表面DAT的表达较正常及假手术组明显减少(P<0.001),且与病程成负相关;复方地黄方组在不同时间点均明显高于LD模型组(P<0.001)。结论复方地黄方可能通过增加PBL表面DAT表达来发挥减毒抗PD的作用。 展开更多
