背景:巴豆霜能够激活ERK通路、对神经元细胞具有抗凋亡作用,是否具有抑制JNK、p38通路的激活而发挥抗凋亡的协同效应尚不清楚。目的:探讨巴豆霜对脑缺血再灌注损伤大鼠缺血侧皮质区神经元损伤和凋亡的影响及机制。方法:①将90只SD大鼠... 背景:巴豆霜能够激活ERK通路、对神经元细胞具有抗凋亡作用,是否具有抑制JNK、p38通路的激活而发挥抗凋亡的协同效应尚不清楚。目的:探讨巴豆霜对脑缺血再灌注损伤大鼠缺血侧皮质区神经元损伤和凋亡的影响及机制。方法:①将90只SD大鼠随机分为假手术组,模型组,巴豆霜低、中、高剂量组,尼莫地平组,每组15只,除假手术组外,剩余各组均采取线栓法制备大脑中动脉栓塞大鼠模型,巴豆霜各组大鼠分别按剂量20,40,60 mg/kg灌胃;假手术组和模型组大鼠给予等量生理盐水,1次/d,连续给药7 d。运用神经功能缺损评分、TTC染色、脑组织含水量、苏木精-伊红染色及尼氏染色筛选出最佳浓度即巴豆霜高剂量。②将120只SD大鼠随机分为假手术组、模型组、巴豆霜组、JNK抑制剂组、巴豆霜+JNK抑制剂组、p38 MAPK抑制剂组、巴豆霜+p38 MAPK抑制剂组和尼莫地平组,每组15只,除假手术组外,剩余各组大鼠均制备大脑中动脉栓塞大鼠模型,在造模之前30 min,将10μL JNK抑制剂SP600125或10μL p38 MAPK抑制剂SB203580分别注射到大鼠侧脑室,巴豆霜各组大鼠灌胃60 mg/kg巴豆霜,7 d后,采用Western Blot、TUNEL染色及流式细胞术检测各组大鼠脑组织JNK/p38 MAPK信号通路及凋亡相关蛋白水平及细胞凋亡情况。结果与结论:①与假手术组相比,模型组神经功能缺损评分、脑含水量、脑梗死体积、细胞凋亡率显著升高(P<0.05),神经细胞呈散乱分布;与模型组相比,巴豆霜中、高剂量组和尼莫地平组大鼠神经功能缺损评分、脑组织含水量、脑梗死体积显著降低(P<0.05),神经细胞病理形态明显改善;②与JNK抑制剂组相比,巴豆霜+抑制剂组大鼠脑组织p-JNK/JNK、p-p38/p38、Bax表达显著降低(P<0.05),细胞凋亡率显著降低(P<0.05),而Bcl-2表达显著升高(P<0.05);③结果提示,巴豆霜可能通过抑制JNK/p38 MAPK信号通路的激活、减少神经元凋亡等途径,达到对脑缺血再灌注损伤大鼠的神经保护作用。 展开更多

芩蓟凉血合剂对痔病相近动物模型的影响

目的探讨芩蓟凉血合剂(Qinji Liangxue mixture,QLM)对抗痔作用的机制。方法分别使用巴豆油制剂(croton oil preparation,COP)、冰醋酸制备大鼠急性痔病动物模型,将80只SD雌性大鼠,随机分为COP组与冰醋酸组两组,每组40只。COP组,随机分... 目的探讨芩蓟凉血合剂(Qinji Liangxue mixture,QLM)对抗痔作用的机制。方法分别使用巴豆油制剂(croton oil preparation,COP)、冰醋酸制备大鼠急性痔病动物模型,将80只SD雌性大鼠,随机分为COP组与冰醋酸组两组,每组40只。COP组,随机分为对照组1(CA组),模型组1(CB组),QLM低剂量组1-1(CC组),QLM高剂量组1-2(CD组),地奥司明组1(CE组),每组8只;冰醋酸组,随机分为对照组2(GA组),模型组2(GB组),QLM低剂量组2-1(GC组),QLM高剂量组2-2(GD组),地奥司明组2(GE组),每组8只。除对照组,其余组均使用COP或冰醋酸处理,随后使用不同药物治疗7 d。使用苏木精-伊红(HE)染色观察COP及冰醋酸诱导的肛门直肠组织形态学,使用ELISA检测COP诱导的大鼠血清IL-6、TNF-α水平,记录冰醋酸诱导的大鼠肛周溃疡面积。将24只C57BL/6小鼠随机分为对照组(A组),QLM低剂量组(B组),QLM高剂量组(C组),肾上腺色腙片组(D组),每组6只,连续治疗7 d,使用毛细管法及断尾法评价QLM的止血疗效。结果QLM能够显著改善痔病相近动物模型的病理损伤,减轻COP诱导的病变程度评分(P<0.05),降低血清IL-6、TNF-α水平(P<0.05),减少冰醋酸诱导的肛周溃疡面积(P<0.05),缩短凝血及出血时间(P<0.05)。结论QLM具有良好的抗痔活性,其作用可能是通过抗炎、止血、减少组织损伤实现的。 展开更多

目的研究佛波双酯(TPA)对照品的制备方法及质量标准,为佛波双酯原料和相关制剂的质量控制提供依据。方法利用硅胶柱制备色谱和反相制备色谱对巴豆油的甲醇-水提取物进行分离纯化,分别以高效液相色谱法(HPLC)和气相色谱对佛波双酯对照品... 目的研究佛波双酯(TPA)对照品的制备方法及质量标准,为佛波双酯原料和相关制剂的质量控制提供依据。方法利用硅胶柱制备色谱和反相制备色谱对巴豆油的甲醇-水提取物进行分离纯化,分别以高效液相色谱法(HPLC)和气相色谱对佛波双酯对照品进行纯度检查和含量测定,用紫外光谱(UV)、红外图谱(IR)、质谱(MS)、核磁共振氢谱(1H-NMR)、碳-13核磁共振(13C-NMR)等方法进行图谱分析及结构确证,确认分子式为C36H56O8。结果纯化制备得到的佛波双酯对照品,紫外最大吸收波长为232 nm,MS结果为高分辨电喷雾质谱(HRESI-MS)m/z 615.4120(M-H)-,HPLC检查纯度>99.5%。结论本制备方法科学有效,制备的对照品纯度符合中药化学对照品的相关要求,可作为佛波双酯原料及相关制剂质量控制的化学对照品。 展开更多

Croton sylvaticus(C. sylvaticus) is widely used as herbal medicine by the indigenous people of tropical Africa. The potential of C. sylvaticus as herbal medicine,the phytochemistry and pharmacological properties of its parts used as herbal medicines are reviewed. The extensive literature survey revealed that C. sylvaticus is traditionally used to treat or manage at least 24 human and animal diseases and ailments. The species is used as herbal medicine for diseases and ailments such as abdominal pains,boils,fever,inflammation,malaria,rheumatism,swellings and tuberculosis and as ethnoveterinary medicine. Multiple classes of phytochemicals such as alkaloids,anthraquinones,essential oils,flavonoids,lignan,phenolics,sterols,tannins and terpenoids have been isolated from the species. Scientific studies on C. sylvaticus indicate that it has a wide range of pharmacological activities which include antibacterial,antifungal,anti-inflammatory,antioxidant,larvicidal and effects on the central nervous system. Although studies have confirmed that C. sylvaticus has a wide range of bioactives,further research on the exact bioactive molecules and mechanisms of action are required. 展开更多

The antioxidant and cytotoxic activities of Methanolic and Dichloromethane extracts of Croton bonplandianum Baill were evaluated in a bid to provide better scientific basis for the isolation of bioactive compounds from the plant. The DPPH and hydroxyl radical scavenging activities of the Methanolic extract were 59.62%, and Dichloromethane extract was not toxic. Gallic acid used as reference compounds showed higher antioxidant activities to the plant extracts. Both methanolic and dichloromethane extracts were evaluated for their brine shrimp lethality. Methanolic extract showed cytotoxic activity in vitro with a LD50 value of 115.76 (0.0048-3.76) μg/ml. Etoposide was used as standard drug. Put together, these results confirm that Croton bonplandianum extracts possess appreciable natural antioxidant and cytotoxic potentials, thereby providing good justification for the isolation of pure bioactive compounds. 展开更多

In consideration of biomass, phytoremediation is a very useful techniques above all, can be exploit by identification of hyperaccumulator, which accumulates the heavy metal under metal stress condition. In view of constraints of efficient plant species in present study thirty seven different plant species were screened for the identification of heavy metal accumulators. Croton bonplandianum, sedges and Balanites aegyptiaca amongst the all exhibit superior potential of heavy metal accumulation. This is the first report to unravel the heavy metal accumulation property of three different plant species which can be exploited for the bioremediation of heavy metals. 展开更多

Water Quality, Contamination, and Wetlands in the Croton Watershed, New York, USA

The Croton Watershed (New York State, USA) is a semi-urban region that provides 10% of the drinking water for the City of New York. Nonpoint source contamination in the watershed is a major concern for managers because the water supply is currently unfiltered water. Results are reported from three synoptic studies of surface water quality from 98 wetland-containing sub-catchments in the Croton Watershed designed to broadly characterize, at a reconnaissance level, the geochemical controls on water quality, in particular as it relates to wetlands. Total dissolved organic carbon concentrations in surface waters draining wetlands correlated well (average R2 of 0.93) with standard Gelbstoff (g440) color measurements, although there is very little correlation between dissolved organic carbon concentrations and wetland areas in the sub-catchments. This may be a potential indication of other sources of colored organic material. Concentrations of dissolved sodium and chloride, while related to road length, stochiometrically had more chloride than expected for pure road-salt dissolution. This offset is likely due to cation exchange and sorbtion of sodium by wetlands in the Croton watershed. The results show contamination in the Croton hydrologic system that should addressed in ongoing management policies and decision-making. 展开更多

Lung cancer has been the most common cancer in the world for several decades,and it was also the most common cause of death from cancer.Despite the several advances in the last few years into treatment of lung cancer,the survival remains extremely low.There is an urgent need to discover viable leads that can be developed into new clinically therapeutic drugs with good treatment effects.The analysis of current reported literatures suggest that natural products possess strong inhibitory efficacy against lung cancer.The plants of Croton genus belonging to Euphorbiaceae family,are widely used as herbal medicine in the folk.Which are reported to have abilities to treat several cancer cell lines especially lung cancer,a successive study was carried out to identify selective anticancer agents from these plants specie.Herein we employed a network pharmacological approach to understand its multi-therapeutics potentiality in Croton genus against lung cancer.In total,the database of the compound was established based on the methods of network pharmacology prediction and the compound-disease target network was constructed.We believed that our study would provide important clues for finding novel drug inhibitors for lung cancer. 展开更多

目的探究巴豆油对寒积便秘大鼠的治疗作用及毒性,阐明巴豆油治疗寒积便秘的“毒-效”关系。方法大鼠连续3天灌胃2℃冰水的10%活性炭溶液复制寒积便秘模型,于第4天灌胃巴豆油,观察记录大鼠首次排便时间和3 h内排便次数;HE染色评价大鼠结... 目的探究巴豆油对寒积便秘大鼠的治疗作用及毒性,阐明巴豆油治疗寒积便秘的“毒-效”关系。方法大鼠连续3天灌胃2℃冰水的10%活性炭溶液复制寒积便秘模型,于第4天灌胃巴豆油,观察记录大鼠首次排便时间和3 h内排便次数;HE染色评价大鼠结肠组织一般病理情况;检测大鼠血清中白介素(Interleukin,IL)-6、IL-10、肿瘤坏死因子(Tumor necrosis factor,TNF)-α、环氧合酶(Cyclooxygenase,COX)-2、巨噬细胞炎性蛋白(Macrophage inflammatory protein 1,MIP-1)、谷丙转氨酶(Alanine aminotransferase,ALT)、谷草转氨酶(Aspartate aminotransferase,AST)及总蛋白(Total protein,TP)水平;免疫组化检测大鼠结肠组织中髓样分化因子(Myeloid differentiation factor 88,MyD88)和核转录因子(Nuclear transcription factor,NF)-κB p65的表达。结果巴豆油在各剂量下均能不同程度地缩短寒积便秘大鼠首次排便时间并增加3 h内排便次数,其中3.78、2.83和1.89 g·kg^(-1)巴豆油组具有显著统计学意义(P<0.05);HE染色结果显示,与模型组相比,巴豆油在各剂量下均能导致大鼠结肠组织黏膜出现不同程度的炎症及灶性坏死,杯状细胞缺失和固有层水肿,且以3.78 g·kg^(-1)巴豆油组大鼠结肠组织损伤最为严重;此外,与模型组相比,巴豆油各组大鼠血清中COX-2、TNF-α、IL-6水平均有不同程度的提高(P<0.05),且以3.78 g·kg^(-1)巴豆油组效果最为明显;巴豆油在各剂量下均能使大鼠血清中MIP-1水平上升,且3.78 g·kg^(-1)巴豆油组效果明显(P<0.01);但巴豆油各组大鼠血清中IL-10水平的变化不明显;免疫组化结果显示,除0.47 g·kg^(-1)剂量组外,巴豆油各剂量均能造成大鼠结肠组织中MyD88蛋白水平表达增加,其中3.78、2.83和1.89 g·kg^(-1)剂量组差异具有显著性(P<0.05);巴豆油各剂量均能增加大鼠结肠组织中NF-κB p65蛋白水平,其中3.78 g·kg^(-1)巴豆油组差异具有显著性(P<0.05)。此外,与模型组相比,巴豆各剂量组均未造成大鼠心、脾、肺、肾和胸腺的脏器指数变化(P>0.05),而1.89、0.94 g·kg^(-1)巴豆油组大鼠肝脏器指数明显增加(P<0.05);肝脏生化指标检测结果显示,巴豆油各组并未造成大鼠血清中ALT、AST及TP含量升高,差异不具有统计学意义(P>0.05)。结论巴豆油在3.78、2.83和1.89 g·kg^(-1)浓度下能够剂量依赖性地发挥泻下作用,但也能通过调控MyD88/NF-κB通路介导的炎症反应,诱导大鼠结肠黏膜损伤。在0.94和0.47 g·kg^(-1)剂量下,巴豆油几乎不引起大鼠结肠组织的损伤,但在此剂量下,巴豆油的泻下作用较弱,这提示巴豆油治疗寒积便秘最佳的浓度范围可能为0.94-1.89 g·kg^(-1)。 展开更多

以十二胺(DA)和聚乙烯吡咯烷酮(PVP)为模板剂,制备具有不同孔径的多孔二氧化硅微球,然后采用聚乙烯亚胺(PEI)对微球进行改性制得复合材料,探究其对卷烟主流烟气中巴豆醛的选择性吸附性能。通过扫描电镜(SEM)、透射电镜(TEM)、X射线衍射(... 以十二胺(DA)和聚乙烯吡咯烷酮(PVP)为模板剂,制备具有不同孔径的多孔二氧化硅微球,然后采用聚乙烯亚胺(PEI)对微球进行改性制得复合材料,探究其对卷烟主流烟气中巴豆醛的选择性吸附性能。通过扫描电镜(SEM)、透射电镜(TEM)、X射线衍射(XRD)、紫外-可见吸收光谱(UV-Vis)、热失重(TG)和低温氮气吸脱附等方法探究材料的组成、形貌和结构,并结合动态重量法(DVS)、吸附动力学模拟以及卷烟抽吸实验对其吸附性能进行研究。研究结果表明,合成的二氧化硅微球具有均匀的介孔结构,且随着PVP分子量的增大,微球孔径逐渐增大;经PEI改性后,二氧化硅载体材料表面及部分孔道被PEI所覆盖,导致复合材料的孔容、比表面积以及孔径均有所减小;巴豆醛在复合材料上的吸附符合拟二级动力学,且受颗粒内扩散作用影响。与对照品相比,复合材料对主流烟气中的巴豆醛的选择性去除率达到18.5%。 展开更多

目的探讨巴豆叶对脑缺血再灌注模型大鼠海马CA1区c-Jun-氨基末端激酶(JNK)通路的影响。方法将216只SD雄性大鼠按随机数表法分为假手术组、模型组、尼莫地平组、巴豆叶低、中、高浓度组,各组按取材时间点不同又分为1 d、3 d和7 d亚组,尼... 目的探讨巴豆叶对脑缺血再灌注模型大鼠海马CA1区c-Jun-氨基末端激酶(JNK)通路的影响。方法将216只SD雄性大鼠按随机数表法分为假手术组、模型组、尼莫地平组、巴豆叶低、中、高浓度组,各组按取材时间点不同又分为1 d、3 d和7 d亚组,尼莫地平组灌胃尼莫地平混悬液1.08 g·L^(-1),巴豆叶低、中、高浓度组灌胃药物浓度分别为60 g·L^(-1)、120 g·L^(-1)、180 g·L^(-1),假手术组和模型组灌胃等体积蒸馏水,2次/d。以Garcia JH评分法进行神经功能评测,HE观察神经元形态;TUNEL法检测大鼠海马CA1区细胞凋亡;Western blot检测蛋白表达。结果1)神经功能评分:同一处理时间下,模型组和各干预组评分较假手术组均降低(P均<0.01);各干预组评分较模型组均升高,随着干预浓度的增加,评分也逐渐升高,尼莫地平组评分高于低、中浓度组(P均<0.01);除假手术组外,各组评分均随着干预时间的延长升高(P均<0.01)。2)HE染色:模型大鼠神经细胞出现不同程度的损伤;各药物干预组均能不同程度改善损伤;随着干预浓度的增加,修复程度也逐渐明显,尼莫地平组效果优于中、低浓度组;各干预组细胞形态均随着干预时间的延长明显改善。3)凋亡细胞计数:在各干预时间下,模型组及各干预组凋亡计数较假手术组均升高(P均<0.01);各干预组较模型组,计数均下降(P均<0.01);随着巴豆叶浓度的增加凋亡计数下降,且尼莫地平组高于中浓度组(P均<0.01)。4)Western blot检测:在各干预时间下,模型组及各干预组蛋白表达较假手术组均显著升高(P均<0.01);各干预组较模型组,蛋白表达下降(P均<0.01);随着浓度的增加蛋白表达下降,尼莫地平组低于中浓度组(P均<0.01);除假手术组外,各组大鼠蛋白表达均随着时间变化在1~7 d逐渐降低(P<0.01),且各时间点差异均有统计学意义(P均<0.01)。结论巴豆叶能下调JNK通路蛋白的表达,有效改善大鼠脑缺血再灌注损伤后神经功能缺损、抗海马CA1区细胞凋亡。 展开更多
