As a crucial channel for understanding traditional settlements culture,traditional settlement landscape gene is of vital importance to figure out status quo of traditional Chinese culture,to protect cultural heritage of settlements,and to help residents establish cultural confidence.This paper adopts diversity index,schema analysis,and qualitative analysis methods to identify cultural diversity and construct diversity map of cultural landscape genes.It shows that cultural diversity of landscape genes refers to the various forms of cultural factors in architecture,environment,culture,and layout,including type diversity,combination diversity,and functional diversity. 展开更多

As the National Conference on New Urbanization proposed, "(new urbanization) should enable residents to see mountains and waters, and remember nostalgia". Nostalgia is a kind of memories and yearning deep in the heart for one's homeland or the place once lived in, it is the softest emotion and also a spiritual need. To keep nostalgia in new urbanization is to preserve history, culture, memories, and humanistic spirits of the homeland, and also to maintain local cultural genes and carry forward rural civilization. The foundation of remembering the nostalgia is to intensify culture protection. And by means of protecting cultural heritage and style of the traditional settlement practically, preserving cultural gene, inheriting cultural memory, creating cultural brand and characteristic landscape etc., the nostalgia can meet the spiritual demands of the residents, such as homesickness, life emotion, historical emotion and cultural emotion in the process of new urbanization. The key of remembering nostalgia is to promote the construction of small characteristic towns. Relying on on-site urbanization, the signifi cant ways of achieving new urbanization are star-studded layout of small towns and the construction of small characteristic tourism town. The construction of Hunan-style Tourism Town is a positive attempt and exploration of remembering nostalgia, and it contributes to the construction of a beautiful and poetic homeland for the human-centered new urbanization. 展开更多

To investigate whether a reporter LacZ gene could be transferred into cultured ocular cells of human eyes in vitro Methods Fibroblast cells of Tenon's capsule, trabecular meshwork cells, and muscle celms in the ciliary body of human eyes were cultured and the pcDNA3 LacZ gene was transferred into these cells using a cationic liposome delivery system The cells were subsequently fixed with 4% paraformaldehyde, mixed with X gal, then observed under a microscope Results Blue stain was seen in the cytoplasm of the cultured cells under the microscope, demonstrating the successful transfer of the LacZ gene into these cells Conclusion Reporter LacZ gene was easily transferred into the cultured ocular cells in vitro This provides insights into the transfer of the genes into these cells to study the pathogenesis and therapy of glaucoma 展开更多

Comparison of inositol phosphates formation and gene expression of Gq alpha subunit in cultured aortic smooth muscle cells from spontaneously hypertensive and Wistar Kyoto rats

Methods The VSMCs ... Objecitve To explore whether phosphoinositide specific phospholipase C (PLC) activation via G protein in vascular smooth muscle cells (VSMCs) is altered in spontaneously hypertensive rats (SHR). Methods The VSMCs derived from aortae of SHR and Wistar Kyoto (WKY) rats were loaded for 48 hours with myo inositol. Inositol phosphate release was initiated by the addition of 10 5 mol/L norepinephrine in intact cells or by guanosine 5' 0 (3 thio tri sphosphate) (GTP gamma S) in permeabilized cells. In the meantime, growth arrested VSMCs were stimulated by 10% calf serum for 0, 30, 45, or 60 min, then gene expressions of Gq alpha subunit (G alph a q) were observed. Results There were no significant differences in inositol 1, 4,5 triphosphate (IP 3) level and expression of G alpha q mRNA between quiescent VSMCs from SHR and that from WKY. When stimulated by norepinephrine, IP 3 production increased transiently with a peak level at 10 s in VSMCs from WKY, and a rapid biphasic IP 3 response, which was significantly higher than that of WKY, in VSMCs from SHR had been observed. G proteins activated by GTP gamma S significantly raised IP 3 production in VSMCs from SHR compared to WKY (SHR vs WKY: 234.8%±29.2% vs 142.4%±12.0% of basal IP 3, P<0.05). In addition, the serum effect showed an significant increase in expression of G alpha q mRNA in VSMCs from SHR. Conclusions The hereditary factors are not the only variable regulating IP 3 metabolism and G alpha q gene expression. Influences of multi environmental factors such as vasoactive compounds, together with genetic predisposition, palys an important role in the highly sensitive response of IP 3 production and G alpha q gene over expression in SHR. 展开更多

During the reign of Emperor Wu of Han Dynasty,the ethnic relations in China entered a new historical stage.Records of the Historian written during this period gives a basically factual account of the ethnic history in China since ancient epochs.It is the first specialized work on ethnic history.The national consciousness that Sima Qian presents in this work is distinctively different from the Confucian tradition,which draws a strict line between Hua(华,China)and Yi(夷,the barbarians).His attention on the commonality of the cultural genes between the farming peoples of the Central-Plains and the nomadic hunting peoples of the borderlands,his recognition of the integrated impact that multiple ethnic groups have on the rise and development of Huaxia(华夏,Chinese)civilization,his respect for"differences among barbarians of the four directions"(四夷各异),and his approval of the“merits of Hu attire”(胡服之功)are all embodiment of his liberal views.To explore the formation and development of the cultural community of the Chinese nation and to understand and interpret this historical process of ethnic integration,we should attach great importance to the positive significance of Sima Qian's national consciousness. 展开更多

Over past decades plant tissue culture has emerged as an alternative of whole plant cultivation in the production of valuable secondary metabolites.Adventitious roots culture of Panax ginseng and Echinacea purpure has reached the scale of 1-10 kL.Some molecular biological techniques,such as transgenic technology and genetic stability are increasingly used in the studies on plant tissue cultures.The studies on elicitors have deepened into the induction mechanism,including signal molecules,functional genes,and so on.More and more biological elicitors,such as A.niger and yeast are used to increase the active compounds in plant tissue cultures.We also discussed the application of synthetic biology in the studies on biosynthesis of artemisinin,paclitaxel,and tanshinon.The studies on active ingredients biosynthesis of medicinal plants provide unprecedented possibilities to achieve mass production of active ingredients.Plant tissue cultures can not only produce active ingredients but also as experimental materials for biosynthesis.In order to improve the contents of active compounds in medicinal plants,following aspects could be carried out gene interference or gene silencing,gene overexpression,combination with chemical synthesis,application of elicitors,and site-directed mutagenesis of the key enzymes. 展开更多
