It has been reported that Mycobacterial cell walls are characterized by high DAP (diaminopimelic acid) content—an intermediate of the (S)-lysine biosynthetic pathway. Hence, the Lysine/DAP biosynthetic pathway is a promising target because of its role in cell wall and amino acid biosynthesis. In this study we performed a molecular docking analysis of a novel antibacterial isolated from Streptomyces sps. 201 against dihydrodipicolinate synthase (DHDPS) enzyme of Mycobacterium tuberculosis. The docking studies suggest that the novel molecule binds at active site LYS 171 forming a cleft and at other potential ligand binding site exhibiting all the major interactions such as hydrogen bonding, hydrophobic interaction and electrostatic interaction with (THR55, TYR143, ARG148, LYS171, VAL257 and GLY256) residues.展开更多
We address a state-of-the-art reinforcement learning(RL)control approach to automatically configure robotic pros-thesis impedance parameters to enable end-to-end,continuous locomotion intended for transfemoral amputee...We address a state-of-the-art reinforcement learning(RL)control approach to automatically configure robotic pros-thesis impedance parameters to enable end-to-end,continuous locomotion intended for transfemoral amputee subjects.Specifically,our actor-critic based RL provides tracking control of a robotic knee prosthesis to mimic the intact knee profile.This is a significant advance from our previous RL based automatic tuning of prosthesis control parameters which have centered on regulation control with a designer prescribed robotic knee profile as the target.In addition to presenting the tracking control algorithm based on direct heuristic dynamic programming(dHDP),we provide a control performance guarantee including the case of constrained inputs.We show that our proposed tracking control possesses several important properties,such as weight convergence of the learning networks,Bellman(sub)optimality of the cost-to-go value function and control input,and practical stability of the human-robot system.We further provide a systematic simulation of the proposed tracking control using a realistic human-robot system simulator,the OpenSim,to emulate how the dHDP enables level ground walking,walking on different terrains and at different paces.These results show that our proposed dHDP based tracking control is not only theoretically suitable,but also practically useful.展开更多

Jinghua No.l, Jing411, You899 and Yang...Expression vector pBPC102, which carries winged bean lysine-rich protein (wblrp) gene and dihydropicolinate synthase (DHDPS) gene, was transferred into hexaploid winter wheat cv. Jinghua No.l, Jing411, You899 and Yangnongl5 explants of immature inflorescence and immature embryos by particle bombardment. More than 100 transgenic plants were obtained under the selection of s-(2-aminoethyl)-L-cysteine (AEC). Confirmed transgenic plants of To and TI generation by PCR and PCR-Southern blotting analyses showed successful integration of wblrp gene into wheat genome. Analysis of transgenic plant lines of T2 by Northern dot-blotting showed good expression of wblrp gene in offspring seed. The content of free lysine in leaves, contents of bound lysine and total proteins in seeds of T2 transgenie wheat lines were determined and analyzed. Among 34 tested transgenic lines, levels of free lysine content in leaves of 9 transgenic lines are 2~3times higher than un-trans-formed wild-type cultivars. Among 17 analyzed transgenic lines, bound lysine content of 4 transgenic lines is more than 10% higher than that of wild-type cultivars. Our research suggests that introducing wblrp gene into wheat is an effective way to improve its nutrition quality.展开更多

As one of online learning algorithm of app...In this paper, an improved PID-neural network (IPIDNN) structure is proposed and applied to the critic and action networks of direct heuristic dynamic programming (DHDP). As one of online learning algorithm of approximate dynamic programming (ADP), DHDP has demonstrated its applicability to large state and control problems. Theoretically, the DHDP algorithm requires access to full state feedback in order to obtain solutions to the Bellman optimality equation. Unfortunately, it is not always possible to access all the states in a real system. This paper proposes a solution by suggesting an IPIDNN configuration to construct the critic and action networks to achieve an output feedback control. Since this structure can estimate the integrals and derivatives of measurable outputs, more system states are utilized and thus better control performance are expected. Compared with traditional PIDNN, this configuration is flexible and easy to expand. Based on this structure, a gradient decent algorithm for this IPIDNN-based DHDP is presented. Convergence issues are addressed within a single learning time step and for the entire learning process. Some important insights are provided to guide the implementation of the algorithm. The proposed learning controller has been applied to a cart-pole system to validate the effectiveness of the structure and the algorithm.展开更多
