The H9N2 subtype of avian influenza virus(AIV)is widely prevalent in poultry and wild birds globally,and has become the predominant subtype circulating in poultry in China.The H9N2 AIV can directly or indirectly(by serving as a"donor virus")infect humans,posing a significant threat to public health.Currently,there is a lack of in-depth research on the prevalence of H9N2 viruses in Shanxi Province,central China.In this study,we isolated 14 H9N2 AIVs from October 2020 to April 2022 in Shanxi Province,and genetic analysis revealed that these viruses belonged to 7 different genotypes.Our study on animals revealed that the H9N2 strains we identified displayed high transmission efficiency among chicken populations,and exhibited diverse replication abilities within these birds.These viruses could replicate efficiently in the lungs of mice,with one strain also demonstrating the capacity to reproduce in organs like the brain and kidneys.At the cellular level,the replication ability of different H9N2 strains was evaluated using plaque formation assays and multi-step growth curve assays,revealing significant differences in the replication and proliferation efficiency of the various H9N2 viruses at the cellular level.The antigenicity analysis suggested that these isolates could be classified into 2 separate antigenic clusters.Our research provides crucial data to help understand the prevalence and biological characteristics of H9N2 AIVs in central China.It also highlights the necessity of enhancing the surveillance of H9N2 AIVs. 展开更多

Allicin, an antioxidant, is known for providing garlic with its unique fragrance and taste, as well as for its antimicrobial properties. Black garlic, a fermented form of garlic, contains higher levels of antioxidants than fresh garlic. Antioxidants play a vital role in alleviating cellular stress during viral infections. Viral infections result in oxidative stress through the production of reactive oxidative species (ROS). A prolonged state of oxidative stress can result in cell death, DNA damage, and disease progression. In this study, black garlic extract (BGE) is evaluated for its ability to mitigate cytopathic effects and oxidative stress caused by herpes simplex virus-2 (HSV-2) infections in vitro. Antiviral assays were performed to determine the percent of viral inhibition resulting from treatment with the BGE. ROS-Glo<sup>TM</sup> H<sub>2</sub>O<sub>2</sub> assays were then completed to measure the post-infection ROS levels of BGE-treated virus and cells. The results thus far suggest that BGE may inhibit viral infection and decrease levels of oxidative stress. 展开更多

BACKGROUND The co-occurrence of Anti-phospholipase A2 receptor-associated membranous nephropathy(anti-PLA2R-MN)and human immunodeficiency virus(HIV)infection is a rare clinical scenario,presenting significant challenges in terms of management and treatment.CASE SUMMARY A 32-year-old Chinese male diagnosed with HIV infection presented with a clinical history of proteinuria persisting for over two years.A kidney biopsy demonstrated subepithelial immune complex deposition and a thickened glomerular basement membrane,indicative of stage I-II membranous nephro-pathy.Immunofluorescence staining revealed granular deposition of PLA2R(3+)along the glomerular capillary loops,corroborated by a strongly positive anti-PLA2R antibody test(1:320).Initial treatment involving losartan potassium,rivaroxaban,tacrolimus,and rituximab was discontinued due to either poor effec-tiveness or the occurrence of adverse events.Following a regimen of weekly subcutaneous injections of telitacicept(160 mg),a marked decline in the 24 h urine protein was observed within a three-month period,accompanied by a rise in serum albumin level.No significant reductions in peripheral blood CD3+CD4+T and CD3+CD8+T cell counts were detected.The patient's physical and psychological conditions showed significant improvements,with no adverse events reported during the treatment course.CONCLUSION Telitacicept might offer a potential therapeutic avenue for patients diagnosed with anti-PLA2R-MN concomitant with HIV infection. 展开更多

BACKGROUND The presence of two distinct hepatitis B virus(HBV)Pol RT polymorphisms,rt269L and rt269I,could contribute to the unique clinical or virological phenotype of HBV genotype C2.Therefore,a simple and sensitive method capable of identifying both types in chronic hepatitis B(CHB)patients infected with genotype C2 should be developed.AIM To develop a novel simple and sensitive locked nucleic acid(LNA)-real timepolymerase chain reaction(RT-PCR)method capable of identifying two rt269 types in CHB genotype C2 patients.METHODS We designed proper primer and probe sets for LNA-RT-PCR for the separation of rt269 types.Using synthesized DNAs of the wild type and variant forms,melting temperature analysis,detection sensitivity,and endpoint genotyping for LNA-RT-PCR were performed.The developed LNA-RT-PCR method was applied to a total of 94 CHB patients of genotype C2 for the identification of two rt269 polymorphisms,and these results were compared with those obtained by a direct sequencing protocol.RESULTS The LNA-RT-PCR method could identify two rt269L and rt269I polymorphisms of three genotypes,two rt269L types[‘L1’(WT)and‘L2’]and one rt269I type(‘I’)in single(63 samples,72.4%)or mixed forms(24 samples,27.6%)in 87(92.6%sensitivity)of 94 samples from Korean CHB patients.When the results were compared with those obtained by the direct sequencing protocol,the LNA-RT-PCR method showed the same results in all but one of 87 positive detected samples(98.9%specificity).CONCLUSION The newly developed LNA-RT-PCR method could identify two rt269 polymorphisms,rt269L and rt269I,in CHB patients with genotype C2 infections.This method could be effectively used for the understanding of disease progression in genotype C2 endemic areas. 展开更多

[目的]试验旨在优化兔出血症病毒2型(Rabbit hemorrhagic disease virus 2,RHDV2)病毒样颗粒(virus-like particle, VLP)疫苗的制备策略,探究RHDV2 VLP疫苗对家兔的免疫原性,为低成本、高产量RHDV2新型疫苗研发提供新思路。[方法]根据昆虫细胞的密码子偏好性优化合成RHDV2 VP60全基因,将双VP60基因插入真核载体pFastBacTMDual,转化携带Bacmid质粒的大肠杆菌DH10Bac感受态细胞,构建含双VP60基因的重组杆粒Bacmid-VP60-VP60,转染Sf9昆虫细胞,通过Western blotting、间接免疫荧光试验(IFA)及透射电镜对重组杆状病毒Bacmid-VP60-VP60进行表达验证;将优化策略制备的重组蛋白抗原与氢氧化铝佐剂按照9∶1比例制备VLP灭活疫苗,通过安全性检验、最小免疫剂量、免疫持续期等评估优化策略制备的RHDV2 VLP疫苗的保护效果。[结果]试验成功构建重组杆粒Bacmid-VP60-VP60。Western blotting鉴定结果显示,重组杆状病毒转染Sf9细胞表达出大小约60 ku的RHDV2 VP60蛋白。IFA鉴定结果显示,感染重组杆状病毒的Sf9细胞产生了大量的黄绿色荧光,表明重组杆状病毒在Sf9细胞中大量表达VP60蛋白。透射电镜观察结果显示,VP60蛋白折叠成VLP,大小为40 nm左右,呈现球形结构,表面光滑。优化策略制备的RHDV2 VLP疫苗对家兔具有良好的安全性和免疫原性,最小免疫剂量为0.5 mL/只,免疫持续期可达210 d以上。[结论]试验构建了含有密码子优化的双VP60基因的重组杆状病毒,并在昆虫细胞中成功表达RHDV2 VP60蛋白,该蛋白制备的VLP疫苗对家兔具有良好的免疫原性。 展开更多

目的观察EBNA2免疫组化在不同EB病毒阳性淋巴组织疾病中的表达差异、分析其在病理诊断与鉴别诊断中的价值。方法收集344例EB病毒阳性的淋巴组织疾病,总结EBNA2免疫组化在病变中的表达特点。结果不同的EBV阳性淋巴组织病变,EBNA2的阳性... 目的观察EBNA2免疫组化在不同EB病毒阳性淋巴组织疾病中的表达差异、分析其在病理诊断与鉴别诊断中的价值。方法收集344例EB病毒阳性的淋巴组织疾病,总结EBNA2免疫组化在病变中的表达特点。结果不同的EBV阳性淋巴组织病变,EBNA2的阳性表达不同:IM中阳性39例(39/46),EBV阳性的弥漫大B细胞淋巴瘤3例(3/30),淋巴瘤样肉芽肿1例(1/5),纤维素相关弥漫大B细胞淋巴瘤2例(2/2),浆母细胞淋巴瘤1例(1/14),免疫相关淋巴组织增殖性疾病32例(32/58)(包括移植后淋巴组织增殖性疾病21例,HIV相关Burkitt淋巴瘤1例,HIV相关弥漫大B细胞淋巴瘤2例,原发免疫缺陷疾病相关淋巴组织疾病6例,医源性免疫相关淋巴组织增殖性疾病2例)。其余均显示EBNA2阴性。②不同病变中的EBNA2阳性细胞表达明显不同:传染性单核细胞增多症中的EBNA2阳性细胞相对较少,并且多呈散在分布,几乎很少见到簇状聚集的情况。EBNA2阳性细胞多表现为≤10个/HPF,少见11~50个/HPF,仅有个别病例显示大于50个/HPF。免疫相关淋巴组织增殖性疾病中,出现大量EBNA2阳性细胞,阳性细胞较弥漫分布,大部分为强阳性,并且阳性细胞多显示大于50个/HPF。结论EBNA2阳性在疾病的诊断及鉴别诊断提供了新的依据,具有一定的价值;当病变表达EBNA2时,提示患者可能存在免疫缺陷或处于免疫失调状态,对临床诊断及治疗具有一定的指导意义。 展开更多

目的构建表达单纯疱疹病毒2型感染细胞蛋白27(infected cells protein 27,ICP27)的重组质粒pcDNA3.1-ICP27377-513并观察白细胞介素(IL)-15核酸疫苗联合HSV-2-ICP27377-513核酸疫苗的免疫效果。方法采用分子克隆技术构建pcDNA3.1-ICP273... 目的构建表达单纯疱疹病毒2型感染细胞蛋白27(infected cells protein 27,ICP27)的重组质粒pcDNA3.1-ICP27377-513并观察白细胞介素(IL)-15核酸疫苗联合HSV-2-ICP27377-513核酸疫苗的免疫效果。方法采用分子克隆技术构建pcDNA3.1-ICP27377-513重组质粒。将BALB/c雌性小鼠随机分为pcDNA3.1-ICP27377-513联合pcDNA3.1-IL-15(pIL-15)组、pcDNA3.1-ICP27377-513组、pcDNA3.1组和pIL-15组,共免疫3次,每次间隔2周。末次免疫后28 d取血,用微量中和实验法检测血清中特异性中和抗体,ELISA法检测血清中IL-4、IL-2和干扰素-γ(IFN-γ)水平。阴道给予致死量HSV-2攻击小鼠,分别于接种后3 d、7 d和14 d收集阴道冲洗液,用荧光定量PCR法检测生殖道病毒载量。结果pcDNA3.1-ICP27377-513联合pIL-15组和pcDNA3.1-ICP27377-513组中和抗体效价分别为40.00±8.16、28.67±4.47,但两组间差异无统计学意义(P>0.05)。pcDNA3.1-ICP27377-513联合pIL-15组IFN-γ、IL-4水平明显高于pcDNA3.1-ICP27377-513组(P<0.05),但IL-2水平两组间差异无统计学意义(P>0.05)。阴道给予致死量HSV-2攻击后,pcDNA3.1-ICP27377-513联合pIL-15组和pcDNA3.1-ICP27377-513组小鼠阴道冲洗液中病毒载量随着时间延长逐渐减低,pcDNA3.1-ICP27377-513联合pIL-15组和pcDNA3.1-ICP27377-513组相比差异有统计学意义(P<0.05)。结论IL-15核酸疫苗联合HSV-2-ICP27377-513核酸疫苗对小鼠有较好的免疫保护作用。 展开更多

胃癌患者EB病毒感染情况及其对癌组织p53、Bcl-2表达的影响

目的探究胃癌患者人类疱疹(EB)病毒感染情况及其对癌组织p53、B淋巴细胞瘤-2(Bcl-2)表达的影响。方法选取新乡市中心医院2019年8月至2023年1月收治的76例胃癌患者,采用原位杂交法检测患者癌组织及癌旁组织EB病毒编码小分子RNA表达,采用... 目的探究胃癌患者人类疱疹(EB)病毒感染情况及其对癌组织p53、B淋巴细胞瘤-2(Bcl-2)表达的影响。方法选取新乡市中心医院2019年8月至2023年1月收治的76例胃癌患者,采用原位杂交法检测患者癌组织及癌旁组织EB病毒编码小分子RNA表达,采用逆转录聚合酶链式反应(RT-PCR)法检测患者癌组织及癌旁组织p53、Bcl-2 mRNA表达,分析EB病毒感染与胃癌临床病理特征及癌组织p53、Bcl-2表达的关系。结果76例胃癌患者癌组织标本中EB病毒阳性率为27.63%(21/76),邻近癌旁组织标本中EB病毒阳性率为4.29%(3/76),胃癌患者癌组织EB病毒阳性率高于癌旁组织(P<0.05);与EB病毒阴性的胃癌患者比,EB病毒阳性患者中病灶位于近端胃、组织浸至浆膜层、有淋巴结转移的占比较多(P<0.05);胃癌患者癌组织p53及Bcl-2 mRNA表达均高于癌旁组织(P<0.05);EB病毒感染胃癌患者癌组织p53及Bcl-2 mRNA表达均高于未感染胃癌患者(P<0.05)。结论EB病毒感染与胃癌患者近端胃病变、组织浸至浆膜层及有淋巴结转移有关,EB病毒可能通过驱动宿主p53基因甲基化来上调p53表达,与Bcl-2协同促进癌细胞生长,这些可能为临床提供胃癌诊疗评估因子及免疫治疗新靶点。 展开更多

SARS-CoV-2合并感染患者的病原微生物分布特征及耐药性分析

目的分析严重急性呼吸综合征冠状病毒2(severe acute respiratory syndrome coronavirus 2,SARS-CoV-2)合并感染患者病原微生物分布特点及耐药情况,为临床诊治提供依据。方法收集2022年12月至2023年2月延安大学附属医院收治的2019冠状... 目的分析严重急性呼吸综合征冠状病毒2(severe acute respiratory syndrome coronavirus 2,SARS-CoV-2)合并感染患者病原微生物分布特点及耐药情况,为临床诊治提供依据。方法收集2022年12月至2023年2月延安大学附属医院收治的2019冠状病毒病(corona virus disease 2019,COVID-19)患者637例的临床资料,回顾性分析其临床特征及合并感染病原微生物情况。结果COVID-19患者以老年人为主,合并基础疾病患者占79.59%,好转及痊愈患者高达91.05%。培养阳性标本来源于呼吸道、血液和尿液,合并感染的病原微生物分别以肺炎克雷伯杆菌、凝固酶阴性葡萄球菌(coagulase negative staphylococci,CNS)和真菌为主。药敏试验结果显示,肺炎克雷伯杆菌对β内酰胺类抗生素及三代、四代头孢菌素类抗生素耐药率较低(14%~20%),鲍曼不动杆菌对氨基糖苷类抗生素和β内酰胺类复合物较为耐药(均>45%),大肠埃希菌对β内酰胺类抗生素及其复合物耐药率较低(14%~19%)。金黄色葡萄球菌、CNS、屎肠球菌、粪肠球菌均未发现对万古霉素、利奈唑胺、替加环素耐药。结论COVID-19合并感染病原体以革兰阴性菌及真菌为主,感染部位以呼吸道为主,主要病原菌为肺炎克雷伯杆菌。鲍曼不动杆菌对氨基糖苷类抗生素和β内酰胺类复合物耐药率高于全国细菌耐药平均水平,提示临床上应根据COVID-19患者的微生物鉴定和药敏试验结果,选择对症药物治疗,以改善患者预后。 展开更多

All 10 investigated mushroom species inhibited the reproduction of influenza virus strain A/FM/1/47(H1N1) in MDCK cells reducing the infectious titer by 2.0–6.0 lg ID50. Four species, Pleurotus ostreatus, Fomes fomentarius, Auriporia aurea, and Trametes versicolor, were also determined to be effective against HSV-2 strain BH in RK-13 cells, with similar levels of inhibition as for influenza. For some of the investigated mushroom species—Pleurotus eryngii, Lyophyllum shimeji, and Flammulina velutipes—this is the first report of an anti-influenza effect. This study also reports the first data on the medicinal properties of A. aurea, including anti-influenza and antiherpetic activities. T. versicolor 353 mycelium was found to have a high therapeutic index(324.67), and may be a promising material for the pharmaceutical industry as an anti-influenza and antiherpetic agent with low toxicity. Mycelia with antiviral activity were obtained in our investigation by bioconversion of agricultural wastes(amaranth flour after CO2 extraction), which would reduce the cost of the final product and solve some ecological problems. 展开更多

Hepatitis C virus(HCV) infection and diabetes mellitus are two major public health problems that cause devastating health and financial burdens worldwide. Diabetes can be classified into two major types: type 1 diabetes mellitus(T1DM) and T2 DM. T2 DM is a common endocrine disorder that encompasses multifactorial mechanisms, and T1 DM is an immunologically mediated disease. Many epidemiological studies have shown an association between T2 DM and chronic hepatitis C(CHC) infection. The processes through which CHC is associated with T2 DM seem to involve direct viral effects, insulin resistance, proinflammatory cytokines, chemokines, and other immunemediated mechanisms. Few data have been reported on the association of CHC and T1 DM and reports on the potential association between T1 DM and acute HCV infection are even rarer. A small number of studies indicate that interferon-α therapy can stimulate pancreatic autoim-munity and in certain cases lead to the development of T1 DM. Diabetes and CHC have important interactions. Diabetic CHC patients have an increased risk of developing cirrhosis and hepatocellular carcinoma compared with nondiabetic CHC subjects. However, clinical trials on HCV-positive patients have reported improvements in glucose metabolism after antiviral treatment. Further studies are needed to improve prevention policies and to foster adequate and cost-effec-tive programmes for the surveillance and treatment of diabetic CHC patients. 展开更多

The mechanism of action of ribavirin(RBV) as an immunomodulatory and antiviral agent and its clinical significance in the future treatment of patients with hepatitis C virus(HCV) infection are reviewed.RBV up-regulates type 1 and/or 2 cytokines to modulate the T helper(Th) 1/2 cell balance to Th1 dominance.Examination of co-stimulatory signaling indicated that RBV down-modulates inducible co-stimulator on Th cells,which contributes to differentiating na?ve Th cells into Th2 cells while reducing their interleukin-10 production.The effects on T-regulatory(Treg) cells were also investigated,and RBV inhibited the differentiation of na?ve Th cells into adaptive Treg cells by downmodulating forkhead box-P3.These findings indicate that RBV mainly down-regulates the activity of Th2 cells,resulting in the maintenance of Th1 activity that contributes to abrogating HCV-infected hepatocytes.Although an interferon-free treatment regimen exhibits almost the same efficacy without serious complications,regimens with RBV will be still be used because of their ability to facilitate the cellular immune response,which may contribute to reducing the development of hepatocellular carcinogenesis in patients infected with HCV. 展开更多

Both global warming and influenza trouble humans in varying ways, therefore it is important to study the trends in both global warming and evolution of influenza A virus, in particular, proteins from influenza A virus. Recently, we have conducted two studies along this line to determine the trends between global warming and polymerase acidic protein as well as matrix protein 2. Although these two studies reveal some interesting findings, many studies are still in need because at least there are ten different proteins in influenza A virus. In this study, we analyze the trends in global warming and evolution of polymerase basic protein 2 (PB2) from influenza A virus. The PB2 evolution from 1956 to 2008 was defined using the unpredictable portion of aminoacid pair. Then the trend in this evolution was compared with the trend in the global temperature, the temperature in north and south hemispheres, and the temperature in influenza A virus sampling site and species carrying influenza A virus. The results show the similar trends in global warming and in PB2 evolution, which are in good agreement with our previous studies in polymerase acidic protein and matrix protein 2 from influenza A virus. 展开更多

Mature porcine interleukin-2 (pIL-2) gene was amplified by PCR from the plasmid pGEM-T-pIL2 and cloned into the baculovirus pFastBacTM Dual vector of the Bac-to-Bac baculovirus expression system under the control of the PH promoter. Recombinant plL-2 (rpIL-2) expressed in Sf9 insect cells was detected by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and immunofluorescence assay. Western blot analysis confirmed that the rpIL-2 protein had a molecular mass of 20 kDa, which was larger than the molecular mass of the mature protein predicted based on its peptide sequence. The rpIL-2 protein induced in vitro proliferation of ConA-stimulated porcine splenocytes and enhanced in vivo protective immune responses induced by vaccinating the pigs with inactivated oil emulsion vaccine against swine influenza virus. The results showed that the rpIL-2 expressed in Sf9 insect cells has immunoenhancement effects; the finding lays the foundation for the preparation of a specific recombinant IL-2 protein and the development of a novel immune adjuvant of vaccines against various infectious porcine pathogens to increase the immunoprotective efficacy of vaccines. 展开更多

AIM: To investigate the role of mi R-125 b in regulating monocyte immune responses induced by hepatitis C virus(HCV) core protein.METHODS: Monocytic THP-1 cells were treated with various concentrations of recombinant HCV core protein, and cytokines and mi R-125 b expression in these cells were analyzed. The requirement of Tolllike receptor 2(TLR2) or My D88 gene for HCV core protein-induced immune responses was determined by the transfection of THP-1 cells with gene knockdown vectors expressing either TLR2 si RNA or My D88 si RNA. The effect of mi R-125 b overexpression on TLR2/My D88 signaling was examined by transfecting THP-1 cells with mi R-125 b mimic RNA oligos.RESULTS: In response to HCV core protein stimulation, cytokine production was up-regulated and mi R-125 b expression was down-regulated in THP-1 cells. The modulatory effect of HCV core protein on cellular events was dose-dependent and required functional TLR2 or My D88 gene. Forced mi R-125 b expression abolished the HCV core protein-induced enhancement of tumor necrosis factor-α, interleukin(IL)-6, and IL-10 expression by 66%, 54%, and 66%, respectively(P < 0.001), by inhibiting My D88-mediated signaling, including phosphorylation of NF-k Bp65, ERK, and P38.CONCLUSION: The inverse correlation between mi R-125 b and cytokine expression after HCV core challenge suggests that mi R-125 b may negatively regulate HCVinduced immune responses by targeting TLR2/My D88 signaling in monocytes. 展开更多
