Effect of morin against DOCA-salt induced hypertension was evaluated by measuring blood pressure and performing biochemical estimations and histopathological examination of renal tissues.Results:DOCA-salt hypertensive rats showed considerably increased systolic and diastolic blood pressure,serum hepatic marker enzyme activities such as aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP) and gamma-glutamyl transpeptidase(GGT)and renal function markers(urea, uric acid and creatinine) in plasma. Oral administration of morin(25, 50 and 75 mg/kg bw) brought back all the above parameters to near normal level.Histopathology of kidney also confirmed the biochemical findings of this study. The effect at a dose of 50 mg/kg bw of morin was more pronounced than that of the other two doses(25 and 75 mg/kg bw).Conclusions:These findings indicate that morin exhibits strong antihypertensive effect against DOCA-salt induced hypertension.展开更多
Objective:To investigate the antihyperlipidemic effect of crude ethanolic extract of Melothria maderaspatana(M.maderaspatana) leaf(CEEM) on deoxycorticosterone acetate(DOCA)-salt hypertensive rats.Methods:A midscapula...Objective:To investigate the antihyperlipidemic effect of crude ethanolic extract of Melothria maderaspatana(M.maderaspatana) leaf(CEEM) on deoxycorticosterone acetate(DOCA)-salt hypertensive rats.Methods:A midscapular incision was made on each rat and the left kidney was excised after ligation of the renal artery.The surgical wound was closed using an absorbable suture.After one week recovery period,hypertension was induced by subcutaneous injection of DOCA-salt solution,twice a week,and the rats received a 1%sodium chloride solution as drinking water throughout the experimental period.CEEM or nifedipine was administered orally once a day for 6 weeks.Results:In DOCA-salt hypertensive rats,the level of plasma and tissues of total cholesterol(TC),triglycerides(TG),free fatty acids(FFA) and phospholipids (PL) significantly increased and administration of CEEM significantly reduced these parameters towards normality.Further,the levels of low density lipoprotein-cholesterol(LDL-C) and very low density lipoprotein-cholesterol(VLDL-C) significantly increased while high density lipoprotein-cholesterol (HDL-C) decreased in hypertensive rats and administration of CEEM brought these parameters to normality which proved their antihyperlipidemic action.Histopathology of liver, kidney and heart on DOCA-salt induced rats treated with CEEM showed reduced the damages towards normal histology.Conclusions:These findings provided evidence that CEEM was found to be protecting the liver,kidney and heart against DOCA-salt administration and the protective effect could attribute to its antihyperlipidemic activities.展开更多

On the other hand, there is accumulating literature describing some so...Hypertension is a serious problem that is recently thought to be associated with damaging effects on target organs partially via oxidative stress. On the other hand, there is accumulating literature describing some sort of therapeutic interaction between antioxidant enzymes in vital organs and hypertension. Therefore, the aim of this study is to investigate the possible effect of a direct renin inhibitor, aliskiren, used in treatment of hypertension via renin-angiotensin-aldosterone system (RAAS), on selected anti-oxidant enzymes in hepatic homogenates in DOCA salt-induced hypertesnive albino rats. Thirty male wister albino rats were assigned randomly into 3 groups (n = 10/ group). Group 1 received no treatement and serves as control. Group 2 received 0.5% carboxymethylcellulose sodium ip as a solvent of aliskiren, as a direct renin inhibitor (DRI). Group 3 received aliskiren 100 mg/kg/day ip for 4 weeks through gastric tube. Systolic blood pressure (SBP) was measured every week and its mean was recorded at the end of the study. Superoxide dismutase (SOD) enzyme in RBCs lysates, activities of catalase (CAT) and glutathione peroxidase enzymes and thiobarbituric acid reactive substance (TBARS), as a marker of lipid peroxidation, in hepatic homogenates were measured at the end of the study. DRI produced a marked reduction in mean SBP of hypertensive rats. It also significantly (p < 0.05) increased the activities of measured anti-oxidant enzymes while it significantly (p < 0.05) reduced TBARS in liver homogenates. These results indicated that renin possesses an oxidative effect in the liver in hypertensive rats. Aliskiren, in addition to its powerful anti-hypertensive effect, it could induce a great anti-oxidant effect in liver homogenates of DOCA salt-hypertensive rats.展开更多
