Brownfield redevelopment is a new urban land use strategy to pursue the ideal of sustainable development. It relieves environmental pressure and resolves the glaring contradiction between urban land idleness and urban...Brownfield redevelopment is a new urban land use strategy to pursue the ideal of sustainable development. It relieves environmental pressure and resolves the glaring contradiction between urban land idleness and urban sprawl. It addresses issues of urban land use and farmland protection in China. This paper compares brownfield with greenfield, and defines brownfield and its redevelopment. It elaborates general and special government policies for this sustainable land use strategy in the context of China. General policies contain two aspects: clarifying brownfield redevelopment's priority in Chinese urban planning and land use and setting a general quantitative redevelopment target. The extent of the government's special policies, which aim at coordinating three factors in the redevelopment process, namely govern- ments, developers and users, are divided into five areas: statistics and categorization, assessment and remediation, fi- nancial supports and tax incentives, environmental and legal liability, and publicity and guidance. The paper concludes the major functions of different levels of Chinese government in the redevelopment process, i.e. making general strategy, formulating legislation, spectifying regulations and technical standards, estimating and categorizing brownfield sites, drawing up brownfield planning, providing information and financial support, guiding developers, publicizing the strategy to the public, and checking reuse results.展开更多

It consists of a 5’-UTR of 52bp, an open reading frame (ORF) of 1374bp and a 3’-UTR of 483bp and has more than 80% homology with that of other mammal oviductins. N-terminal peptide (NTP) (384 residues) and C-terminal peptide (CTP) (73 residues) of deduced protein precursor has about 80% and 50% identity with that of other mammals respectively. Fusion proteins GST-NTP 368(1R-368N)and GST-CTP73 (369F-441A) were expressed and purified. NH2-terminal of CTP sequencing reveals that the purified protein is consistent with the deduced one. In order to study the function of NTP and CTP the mouse anti-NTP and rabbit anti-CTP antisera were prepared. Tissue-specific (skeleton muscle, oviduct, uterus, ovary, liver, heart and brain) analysis indicated that rabbit oviductin was only found in oviduct. The conditioned medium derived from the rabbit oviduct mucosa epithelial cells has a function of overcoming the early embryonic development block of Kunming mous e cultured in vitro. Anti-CTP antiserum could totally inhibit the early embryo development at 2-cell stage cultured in the conditioned culture medium, but anti-NTP antiserum couldn’t. There was a positive relationship between the ratio of early embryos at development block and the dosage of anti-CTP antiserum added in the conditioned culture medium. These results suggest that oviductin has a function not only on fertilization, but also on the release of early embryonic development block, and the later function domain of rabbit oviductin may be situate in its C-terminal.展开更多
The COVID-19 pandemic has stalled and rolled back progress on Sustainable Development Goals(SDGs).Ecosys-tem services(ESs),defined as the contributions of ecosystems to human well-being,underpin the achievement of SDG...The COVID-19 pandemic has stalled and rolled back progress on Sustainable Development Goals(SDGs).Ecosys-tem services(ESs),defined as the contributions of ecosystems to human well-being,underpin the achievement of SDGs.To promote SDG achievement in post-pandemic era,we teased out the links between ESs and SDGs while examining the impact of COVID-19.We found that ESs benefited all SDGs,yet man-made pressures led to degra-dation of ecosystems and their services.There is broad consensus that the virus lurks in degraded ecosystems and generates spillover due to human interference.The pandemic and global lockdown/restriction disrupted the flow of ESs and altered human ESs demand,threatening the efforts for the SDGs.We suggested:1)to study the association and traceability of ESs-SDGs under the pandemic;2)to prioritize pressing issues such as health care,livelihood,and resource security and in the long run,we should promote human-nature harmony to achieve the SDGs;and 3)to enhance ESs and to promote the SDGs through local community efforts,ESs accounting,and ecosystem restoration.This paper provides insights into the importance of ESs to the SDGs and the ways to integrate ESs into socio-economic development to promote the SDG achievement after the pandemic.展开更多

G...This paper discusses participatory processes in wildland fire management (WFM). Participation is an essential element of both the European Sustainable Development (SD) Strategy and the White Paper on Governance. Governance end SD have thus become an interconnected challenge to be applied to WFM (as a sub-area in forest policy), amongst other policies. An overspread weakness in WFM is lack of real participation of stakeholders. Absence of (or deficient) participation can seriously impair contribution of this group to WFM in high-risk areas and runs counter governance and the SDS. Further, this weakness might prevent an efficient use of fire as a land management tool (prescribed burning, PB) and as a technique for fighting wildfire (suppression fire, SF). Even though these fire practices have been well known in many different places, they have been increasingly neglected or Prohibited over time in Southern Europe. At present, forest and fire fighting administrations are turning their eyes back on them and analyzing the benefits of using fire in relation to preventive and suppressive actions. Therefore, participatory and diffusion mechanisms (the latter adopting the shape of national and international experts' networks) are required in order to solve the so- called fire paradox: that is, the need to move from a one-dimensinnal perception of the negative impacts of fire to a more sophisticated one that also stresses its positive effects. Governance, based on broad social participation, and diffusion, through fire networks, are of utmost importance in order, first, to diminish long-standing suspicion amongst different interests as to the use of fire and, secondly, to diffuse best practices associated with PB and SF. Most importantly, the ELI should exercise its environmental leadership so that these new fire practices and sustainable WFM are diffused across the international arena.展开更多

In this case the conventional policy of “Sustainable Development” is not sufficient to avert the impact of diminution of precipitation on Israel’s water supply. Instead, a policy of “Progressive Development” is proposed, which aims to exploit, stage by stage, first the water resources most available and at lowest cost until they are ex-hausted. Once this stage is under execution, progressive steps are taken to develop more costly resources by advanced methods of exploitation. A case study of such a development plan is the controlled deterioration of the water quality of the coastal plain aquifer of Israel.展开更多

Human brain development is a complex process,and animal models often have significant limitations.To address this,researchers have developed pluripotent stem cell-derived three-dimensional structures,known as brain-li...Human brain development is a complex process,and animal models often have significant limitations.To address this,researchers have developed pluripotent stem cell-derived three-dimensional structures,known as brain-like organoids,to more accurately model early human brain development and disease.To enable more consistent and intuitive reproduction of early brain development,in this study,we incorporated forebrain organoid culture technology into the traditional unguided method of brain organoid culture.This involved embedding organoids in matrigel for only 7 days during the rapid expansion phase of the neural epithelium and then removing them from the matrigel for further cultivation,resulting in a new type of human brain organoid system.This cerebral organoid system replicated the temporospatial characteristics of early human brain development,including neuroepithelium derivation,neural progenitor cell production and maintenance,neuron differentiation and migration,and cortical layer patterning and formation,providing more consistent and reproducible organoids for developmental modeling and toxicology testing.As a proof of concept,we applied the heavy metal cadmium to this newly improved organoid system to test whether it could be used to evaluate the neurotoxicity of environmental toxins.Brain organoids exposed to cadmium for 7 or 14 days manifested severe damage and abnormalities in their neurodevelopmental patterns,including bursts of cortical cell death and premature differentiation.Cadmium exposure caused progressive depletion of neural progenitor cells and loss of organoid integrity,accompanied by compensatory cell proliferation at ectopic locations.The convenience,flexibility,and controllability of this newly developed organoid platform make it a powerful and affordable alternative to animal models for use in neurodevelopmental,neurological,and neurotoxicological studies.展开更多
Lily(Lilium spp.)is an important horticultural crop,but its use is limited due to serious pollen contamination problems.There are many studies on pollen development in model plants,but few on flower crops such as lili...Lily(Lilium spp.)is an important horticultural crop,but its use is limited due to serious pollen contamination problems.There are many studies on pollen development in model plants,but few on flower crops such as lilies.Gibberellin(GA)is a large class of hormones and plays an important role in plant vegetative growth and reproductive development.GAMYB is a group of the R2R3-MYB family upregulated by gibberellin,and plays an important role in anther development.Here,we isolated a novel GAMYB,named LoMYB65,from lily,which was closely related to the AtMYB65 and AtMYB33 in Arabidopsis.Fluorescence quantitative PCR results showed that LoMYB65 was mainly expressed in lily anthers.LoMYB65 could be activated by 288μmol·L^(-1)GA3treatment and the LoMYB65 protein was located in the nucleus and cytoplasm,and had transactivation in yeast and tobacco leaf cells.The conserved motif within 226 amino acids of the C-terminal of LoMYB65 contributed to its transactivation.Overexpression of LoMYB65 caused dwarf phenotype,unnormal tapetum development,less seeds of siliques in transgenic Arabidopsis plants,the transgenic plants showed partly male sterile.Simultaneously,silencing of LoMYB65 with VIGS(Virus Induced Gene Silencing)in lily anthers caused unnormal pollen development and reduced the pollen amount.Overexpression of LoMYB65 in Arabidopsis and silencing of LoMYB65 in lily resulted in decreased pollen counts,so we speculate that LoMYB65 may be dose-dependent.Overall,these findings suggest that LoMYB65 may play an important role in anther development and pollen formation in lily.LoMYB65 may provide a useful candidate gene for pollenless breeding of lily.展开更多

Due to the extremely high heterogeneity of shale, the factors controlling the formation of internal fractures, especially horizontal fractures, remain controversial. In this study, we integrate thin section analysis and microcomputed tomography(CT) data from several lacustrine shale samples from the third member(Es3) of the Shahejie Formation, Qikou Sag, Bohai Bay Basin, to assess the fractures in detail. The goal is to reveal the development characteristics, controlling factors, and geological significance for evaluating sweet spots in a shale oil play. The fractures in the Es3contain high-angle structural and horizontal bed-parallel fractures that are mostly shear and extensional. Various factors influence fracture development,including lithofacies, mineral composition, organic matter content, and the number of laminae. Structural fractures occur predominantly in siltstone, whereas bed-parallel fractures are abundant in laminated shale and layered mudstone. A higher quartz content results in higher shale brittleness, causing fractures, whereas the transformation between clay minerals contributes to the development of bedparallel fractures. Excess pore pressure due to hydrocarbon generation and expulsion during thermal advance can cause the formation of bed-parallel fractures. The density of the bed-parallel and structural fractures increases with the lamina density, and the bed-parallel fractures are more sensitive to the number of laminae. The fractures are critical storage spaces and flow conduits and are indicative of sweet spots. The laminated shale in the Es3with a high organic matter content contains natural fractures and is an organic-rich, liquid-rich, self-sourced shale play. Conversely, the siltstone, massive mudstone, and argillaceous carbonate lithofacies contain lower amounts of organic matter and do not have bed-parallel fractures. However, good reservoirs can form in these areas when structural fractures are present and the source, and storage spaces are separated.展开更多
Tyrosine phosphorylation is crucial for controlling normal cell growth,survival,intercellular communication,gene transcription,immune responses,and other processes.protein tyrosine phosphatase(PTP)and protein tyrosine...Tyrosine phosphorylation is crucial for controlling normal cell growth,survival,intercellular communication,gene transcription,immune responses,and other processes.protein tyrosine phosphatase(PTP)and protein tyrosine kinases(PTK)can achieve this goal by regulating multiple signaling pathways.Oedaleus decorus asiaticus is an important pest that infests the Mongolian Plateau grassland.We aimed to evaluate the survival rate,growth rate,overall performance,and ovarian developmental morphology of the 4th instar nymphs of O.decorus asiaticus while inhibiting the activity of protein tyrosine phosphatase-1B(PTP1B)and PTK.In addition,the expression and protein phosphorylation levels of key genes in the MAPK signaling pathway and antioxidant enzyme activity were assessed.The results showed no significant differences in survival rate,growth rate,or overall performance between PTP1B inhibitor treatment and control.However,after PTK inhibitor treatment,these indexes were significantly lower than those in the control.The ovarian size of female larvae after 15 days of treatment with PTK inhibitors showed significantly slower development,while female larvae treated with PTP1B exhibited faster ovarian growth than the control group.In comparison to controls and nymphs treated with PTK inhibitors,the expression and phosphorylation levels of key genes in the MAPK signaling pathway under PTP1B inhibitor treatments were significantly higher in 4th instar nymphs.However,reactiveoxygen(ROS)species levels and the activities of NADPH oxidase and other antioxidant enzymes were considerably reduced,although they were significantly greater in the PTK inhibitor treatment.The results suggest that PTP1B and PTK feedback inhibition in the mitogen-activated-protein kinases(MAPK)signal transfer can regulate the physiological metabolism of the insect as well as its developmental rate.These findings can facilitate future uses of PTP1B and PTK inhibitors in controlling insect development to help control pest populations.展开更多
Enhancing photosynthetic efficiency is a major goal for improving crop yields under agricultural field conditions and is associated with chloroplast biosynthesis and development.In this study,we demonstrate that Golde...Enhancing photosynthetic efficiency is a major goal for improving crop yields under agricultural field conditions and is associated with chloroplast biosynthesis and development.In this study,we demonstrate that Golden2-like 1a(BnGLK1a)plays an important role in regulating chloroplast development and photosynthetic efficiency.Overexpressing BnGLK1a resulted in significant increases in chlorophyll content,the number of thylakoid membrane layers and photosynthetic efficiency in Brassica napus,while knocking down BnGLK1a transcript levels through RNA interference(RNAi)had the opposite effects.A yeast two-hybrid screen revealed that BnGLK1a interacts with the abscisic acid receptor PYRABACTIN RESISTANCE 1-LIKE 1-2(BnPYL1-2)and CONSTITUTIVE PHOTOMORPHOGENIC 9 SIGNALOSOME 5A subunit(BnCSN5A),which play essential roles in regulating chloroplast development and photosynthesis.Consistent with this,BnGLK1a-RNAi lines of B.napus display hypersensitivity to the abscisic acid(ABA)response.Importantly,overexpression of BnGLK1a resulted in a 10%increase in thousand-seed weight,whereas seeds from BnGLK1a-RNAi lines were 16%lighter than wild type.We propose that BnGLK1a could be a potential target in breeding for improving rapeseed productivity.Our results not only provide insights into the mechanisms of BnGLK1a function,but also offer a potential approach for improving the productivity of Brassica species.展开更多

文摘The COVID-19 pandemic has stalled and rolled back progress on Sustainable Development Goals(SDGs).Ecosys-tem services(ESs),defined as the contributions of ecosystems to human well-being,underpin the achievement of SDGs.To promote SDG achievement in post-pandemic era,we teased out the links between ESs and SDGs while examining the impact of COVID-19.We found that ESs benefited all SDGs,yet man-made pressures led to degra-dation of ecosystems and their services.There is broad consensus that the virus lurks in degraded ecosystems and generates spillover due to human interference.The pandemic and global lockdown/restriction disrupted the flow of ESs and altered human ESs demand,threatening the efforts for the SDGs.We suggested:1)to study the association and traceability of ESs-SDGs under the pandemic;2)to prioritize pressing issues such as health care,livelihood,and resource security and in the long run,we should promote human-nature harmony to achieve the SDGs;and 3)to enhance ESs and to promote the SDGs through local community efforts,ESs accounting,and ecosystem restoration.This paper provides insights into the importance of ESs to the SDGs and the ways to integrate ESs into socio-economic development to promote the SDG achievement after the pandemic.

文摘This paper discusses participatory processes in wildland fire management (WFM). Participation is an essential element of both the European Sustainable Development (SD) Strategy and the White Paper on Governance. Governance end SD have thus become an interconnected challenge to be applied to WFM (as a sub-area in forest policy), amongst other policies. An overspread weakness in WFM is lack of real participation of stakeholders. Absence of (or deficient) participation can seriously impair contribution of this group to WFM in high-risk areas and runs counter governance and the SDS. Further, this weakness might prevent an efficient use of fire as a land management tool (prescribed burning, PB) and as a technique for fighting wildfire (suppression fire, SF). Even though these fire practices have been well known in many different places, they have been increasingly neglected or Prohibited over time in Southern Europe. At present, forest and fire fighting administrations are turning their eyes back on them and analyzing the benefits of using fire in relation to preventive and suppressive actions. Therefore, participatory and diffusion mechanisms (the latter adopting the shape of national and international experts' networks) are required in order to solve the so- called fire paradox: that is, the need to move from a one-dimensinnal perception of the negative impacts of fire to a more sophisticated one that also stresses its positive effects. Governance, based on broad social participation, and diffusion, through fire networks, are of utmost importance in order, first, to diminish long-standing suspicion amongst different interests as to the use of fire and, secondly, to diffuse best practices associated with PB and SF. Most importantly, the ELI should exercise its environmental leadership so that these new fire practices and sustainable WFM are diffused across the international arena.

文摘Poverty reduction is a world-wide concern.At the end of 2017,according to the rural poverty standard in China,there were 30.46 million poor in China.However,complete poverty alleviation by the end of 2020 had been achieved.This is significant and complicated,especially as poverty-stricken areas and ecologically fragile areas overlap.During the process of poverty alleviation,the development of forestry projects was not only conducive to improving the environment but also an important way to reduce poverty.Therefore,based on an analysis of the causes of poverty-stricken areas,this study examined successful cases in different regions and proposed ways to promote economic growth:providing state subsidies for tree planting and forest maintenance;developing undergrowth economy;and/or initiating an industrial chain.It also introduces principles to promote forestry progress,according to local conditions,keeping a balance between economic development and the environment.This study provides effective ways to promote forestry development and rural poverty alleviation.

文摘Yanji is the biggest China Korean inhabited city. Based on a newly finished research project, this paper discusses strategic problems about Yanji towards 21st century. The general point of the authors is that at the beginning of new century, Yanji will play a more important role in Tumen River Delta development, regional economy cooperation in Korea Peninsula as well as Northeast Asia, and it will develop to be an important central city in this area. This paper puts forwards Yanji City’s developing goal, urban nature, and urban size, probes its regional orientation, and further probes how to establish The Yanji Urban Economy Cooperation Zone. The authors think that Yanji should depend on its intellect resources and other characteristic resources. Industry development need to pay much attention to modern agriculture, tertiary industry (especially commerce, trade and tourism) and dominant industries, and high-tech industry must give a special care, in order to establish a light industrial structure.
文摘The last ten years of moderate to low precipitation rates in Israel may be a confirmation of the forecast, based on historical data, that recent, repeated droughts are a function of global warming. In this case the conventional policy of “Sustainable Development” is not sufficient to avert the impact of diminution of precipitation on Israel’s water supply. Instead, a policy of “Progressive Development” is proposed, which aims to exploit, stage by stage, first the water resources most available and at lowest cost until they are ex-hausted. Once this stage is under execution, progressive steps are taken to develop more costly resources by advanced methods of exploitation. A case study of such a development plan is the controlled deterioration of the water quality of the coastal plain aquifer of Israel.
文摘The role of public investment and its substantial contribution to economic growth and the quality of life of citizens is now being reaffirmed internationally,creating a trend towards enhancing the relative public spending.This paper analyses this role at international and European level,while it examines especially the case of Greece;having undergone a significant period of shrinkage over the years of the memorandums,the Public Investment Program(PIP)has again shown a tendency to increase its resources and its importance.Greece makes an important reform effort,which must take into account that,in order to keep track of current global trends and prospects,Public Investment Programs are now called for to be reconciled with new development models as well as new,internationally accepted rules of proper governance and financial management.In this context,they should also act as catalysts for further supporting and strengthening private investment.

文摘Human brain development is a complex process,and animal models often have significant limitations.To address this,researchers have developed pluripotent stem cell-derived three-dimensional structures,known as brain-like organoids,to more accurately model early human brain development and disease.To enable more consistent and intuitive reproduction of early brain development,in this study,we incorporated forebrain organoid culture technology into the traditional unguided method of brain organoid culture.This involved embedding organoids in matrigel for only 7 days during the rapid expansion phase of the neural epithelium and then removing them from the matrigel for further cultivation,resulting in a new type of human brain organoid system.This cerebral organoid system replicated the temporospatial characteristics of early human brain development,including neuroepithelium derivation,neural progenitor cell production and maintenance,neuron differentiation and migration,and cortical layer patterning and formation,providing more consistent and reproducible organoids for developmental modeling and toxicology testing.As a proof of concept,we applied the heavy metal cadmium to this newly improved organoid system to test whether it could be used to evaluate the neurotoxicity of environmental toxins.Brain organoids exposed to cadmium for 7 or 14 days manifested severe damage and abnormalities in their neurodevelopmental patterns,including bursts of cortical cell death and premature differentiation.Cadmium exposure caused progressive depletion of neural progenitor cells and loss of organoid integrity,accompanied by compensatory cell proliferation at ectopic locations.The convenience,flexibility,and controllability of this newly developed organoid platform make it a powerful and affordable alternative to animal models for use in neurodevelopmental,neurological,and neurotoxicological studies.

文摘Lily(Lilium spp.)is an important horticultural crop,but its use is limited due to serious pollen contamination problems.There are many studies on pollen development in model plants,but few on flower crops such as lilies.Gibberellin(GA)is a large class of hormones and plays an important role in plant vegetative growth and reproductive development.GAMYB is a group of the R2R3-MYB family upregulated by gibberellin,and plays an important role in anther development.Here,we isolated a novel GAMYB,named LoMYB65,from lily,which was closely related to the AtMYB65 and AtMYB33 in Arabidopsis.Fluorescence quantitative PCR results showed that LoMYB65 was mainly expressed in lily anthers.LoMYB65 could be activated by 288μmol·L^(-1)GA3treatment and the LoMYB65 protein was located in the nucleus and cytoplasm,and had transactivation in yeast and tobacco leaf cells.The conserved motif within 226 amino acids of the C-terminal of LoMYB65 contributed to its transactivation.Overexpression of LoMYB65 caused dwarf phenotype,unnormal tapetum development,less seeds of siliques in transgenic Arabidopsis plants,the transgenic plants showed partly male sterile.Simultaneously,silencing of LoMYB65 with VIGS(Virus Induced Gene Silencing)in lily anthers caused unnormal pollen development and reduced the pollen amount.Overexpression of LoMYB65 in Arabidopsis and silencing of LoMYB65 in lily resulted in decreased pollen counts,so we speculate that LoMYB65 may be dose-dependent.Overall,these findings suggest that LoMYB65 may play an important role in anther development and pollen formation in lily.LoMYB65 may provide a useful candidate gene for pollenless breeding of lily.

文摘Utilizing energy storage in depleted oil and gas reservoirs can improve productivity while reducing power costs and is one of the best ways to achieve synergistic development of"Carbon Peak–Carbon Neutral"and"Underground Resource Utiliza-tion".Starting from the development of Compressed Air Energy Storage(CAES)technology,the site selection of CAES in depleted gas and oil reservoirs,the evolution mechanism of reservoir dynamic sealing,and the high-flow CAES and injection technology are summarized.It focuses on analyzing the characteristics,key equipment,reservoir construction,application scenarios and cost analysis of CAES projects,and sorting out the technical key points and existing difficulties.The devel-opment trend of CAES technology is proposed,and the future development path is scrutinized to provide reference for the research of CAES projects in depleted oil and gas reservoirs.

文摘Natural fractures are critical for shale oil and gas enrichment and development. Due to the extremely high heterogeneity of shale, the factors controlling the formation of internal fractures, especially horizontal fractures, remain controversial. In this study, we integrate thin section analysis and microcomputed tomography(CT) data from several lacustrine shale samples from the third member(Es3) of the Shahejie Formation, Qikou Sag, Bohai Bay Basin, to assess the fractures in detail. The goal is to reveal the development characteristics, controlling factors, and geological significance for evaluating sweet spots in a shale oil play. The fractures in the Es3contain high-angle structural and horizontal bed-parallel fractures that are mostly shear and extensional. Various factors influence fracture development,including lithofacies, mineral composition, organic matter content, and the number of laminae. Structural fractures occur predominantly in siltstone, whereas bed-parallel fractures are abundant in laminated shale and layered mudstone. A higher quartz content results in higher shale brittleness, causing fractures, whereas the transformation between clay minerals contributes to the development of bedparallel fractures. Excess pore pressure due to hydrocarbon generation and expulsion during thermal advance can cause the formation of bed-parallel fractures. The density of the bed-parallel and structural fractures increases with the lamina density, and the bed-parallel fractures are more sensitive to the number of laminae. The fractures are critical storage spaces and flow conduits and are indicative of sweet spots. The laminated shale in the Es3with a high organic matter content contains natural fractures and is an organic-rich, liquid-rich, self-sourced shale play. Conversely, the siltstone, massive mudstone, and argillaceous carbonate lithofacies contain lower amounts of organic matter and do not have bed-parallel fractures. However, good reservoirs can form in these areas when structural fractures are present and the source, and storage spaces are separated.

文摘Tyrosine phosphorylation is crucial for controlling normal cell growth,survival,intercellular communication,gene transcription,immune responses,and other processes.protein tyrosine phosphatase(PTP)and protein tyrosine kinases(PTK)can achieve this goal by regulating multiple signaling pathways.Oedaleus decorus asiaticus is an important pest that infests the Mongolian Plateau grassland.We aimed to evaluate the survival rate,growth rate,overall performance,and ovarian developmental morphology of the 4th instar nymphs of O.decorus asiaticus while inhibiting the activity of protein tyrosine phosphatase-1B(PTP1B)and PTK.In addition,the expression and protein phosphorylation levels of key genes in the MAPK signaling pathway and antioxidant enzyme activity were assessed.The results showed no significant differences in survival rate,growth rate,or overall performance between PTP1B inhibitor treatment and control.However,after PTK inhibitor treatment,these indexes were significantly lower than those in the control.The ovarian size of female larvae after 15 days of treatment with PTK inhibitors showed significantly slower development,while female larvae treated with PTP1B exhibited faster ovarian growth than the control group.In comparison to controls and nymphs treated with PTK inhibitors,the expression and phosphorylation levels of key genes in the MAPK signaling pathway under PTP1B inhibitor treatments were significantly higher in 4th instar nymphs.However,reactiveoxygen(ROS)species levels and the activities of NADPH oxidase and other antioxidant enzymes were considerably reduced,although they were significantly greater in the PTK inhibitor treatment.The results suggest that PTP1B and PTK feedback inhibition in the mitogen-activated-protein kinases(MAPK)signal transfer can regulate the physiological metabolism of the insect as well as its developmental rate.These findings can facilitate future uses of PTP1B and PTK inhibitors in controlling insect development to help control pest populations.

文摘Enhancing photosynthetic efficiency is a major goal for improving crop yields under agricultural field conditions and is associated with chloroplast biosynthesis and development.In this study,we demonstrate that Golden2-like 1a(BnGLK1a)plays an important role in regulating chloroplast development and photosynthetic efficiency.Overexpressing BnGLK1a resulted in significant increases in chlorophyll content,the number of thylakoid membrane layers and photosynthetic efficiency in Brassica napus,while knocking down BnGLK1a transcript levels through RNA interference(RNAi)had the opposite effects.A yeast two-hybrid screen revealed that BnGLK1a interacts with the abscisic acid receptor PYRABACTIN RESISTANCE 1-LIKE 1-2(BnPYL1-2)and CONSTITUTIVE PHOTOMORPHOGENIC 9 SIGNALOSOME 5A subunit(BnCSN5A),which play essential roles in regulating chloroplast development and photosynthesis.Consistent with this,BnGLK1a-RNAi lines of B.napus display hypersensitivity to the abscisic acid(ABA)response.Importantly,overexpression of BnGLK1a resulted in a 10%increase in thousand-seed weight,whereas seeds from BnGLK1a-RNAi lines were 16%lighter than wild type.We propose that BnGLK1a could be a potential target in breeding for improving rapeseed productivity.Our results not only provide insights into the mechanisms of BnGLK1a function,but also offer a potential approach for improving the productivity of Brassica species.
