AIM: To clone and characterize the porcine aquaporins (AQPs) in the gastrointestinal system.METHODS: A PCR-based cloning strategy and RACE were used to clone full-length AQP coding sequence from reversely transcri...AIM: To clone and characterize the porcine aquaporins (AQPs) in the gastrointestinal system.METHODS: A PCR-based cloning strategy and RACE were used to clone full-length AQP coding sequence from reversely transcribed pig liver cDNA. Stopped-flow light scattering and a YFP-based fluorescence method were used to measure the osmotic water permeability of erythrocytes and the stably transfected CHO cells. RT-PCR, Northern blot, and immunohistochemistry were used to determine the gastrointestinal expression and localization of cloned AQPs. Protein expression in transfected cells and red blood cells was analyzed by Western blot.RESULTS: An 813 bp cDNA encoding a 271 amino acid porcine aquaporin (designated pAQP1) was cloned from liver mRNA (pAQP1 has a 93% identity with human AQP1 and contains two NPA motifs conserved in AQP family, one consensus sequence for N-linked glycosylation, and one mercury-sensitive site at cysteine 191). RT-PCR analysis revealed extensive expression of pAQP1 mRNA in porcine digestive glands and gut. Northern blot showed a single 3.0 kb transcript in selected digestive organs, pAQP1 protein was localized at central lacteals of the small intestine, microvessles of salivary glands, as well as epithelium of intrahepatic bile ducts by immunoperoxydase. High osmotic water permeability that is inhibitable by HgCI2 was detected in porcine erythrocytes and CHO cells stably transfected with pAQP1 cDNA. Immunoblot analysis of porcine erythrocytes and pAQP-transfected CHO cells revealed an unglycosylated 28 ku band and larger glycosylated proteins.CONCLUSION: pAQP1 is the first porcine aquaporin that can be molecularly identified so far. The broad distribution of pAQP1 in epithelium and endothelium of porcine digestive organs may suggest an important role of channel-mediated water transport in fluid secretion/absorption as well as in digestive function and pathophysiology of the gastrointestinal system.展开更多
Background:The objective of this study was to study the effect of Chinese patent drug Jian-Pi-Xiao-Shi formula on rats with infantile anorexia.Methods:The Sprague Dawley rats with 4 weeks old were divided into 5 group...Background:The objective of this study was to study the effect of Chinese patent drug Jian-Pi-Xiao-Shi formula on rats with infantile anorexia.Methods:The Sprague Dawley rats with 4 weeks old were divided into 5 groups:normal group,model group,large,medium,and small doses of sample group.Twelve rats were included in each group with half males and half females.In addition to the normal group,the rats in each group were free to take the special feed during 28 consecutive days,which results in the pretended anorexia model,being the rats in the normal group free to take the ordinary feed.Except for the normal group and the model group,the other groups were given Chinese patent drug Jian-Pi-Xiao-Shi formula by a gavage of 1 mL/100g once a day for 28 days.The distilled water was given to the normal group and the model group under the same conditions.After 28 days,the contents of theβ-endorphin,cholecystokinin octapeptide,gastrin,motilin and pepsin were measured.Results:The results demonstrated that the Chinese patent drug Jian-Pi-Xiao-Shi formula could significantly increase the body weight and the food intake of the infantile rats with anorexia,increase the pepsin activity,the content of motilin and the expression ofβ-endorphin,and in the other hand decrease the expression of the cholecystokinin.Conclusion:The Chinese patent drug Jian-Pi-Xiao-Shi formula can promote the appetite and the digestive function of infantile rats with anorexia.This study suggested that the Chinese patent drug Jian-Pi-Xiao-Shi formula can enhance the appetite of the children and to improve the overall digestive function.展开更多
This study evaluated the effects of barley inclusion and glucanase supplementation on the productive performance and digestive function in laying ducks.The experiment used a randomized design with a 5×2 factorial...This study evaluated the effects of barley inclusion and glucanase supplementation on the productive performance and digestive function in laying ducks.The experiment used a randomized design with a 5×2 factorial arrangement of 5 graded levels of barley(0%,15%,30%,45%and 60%)with or without 1.5 g/kgβ-1,3-1,4-glucanase(15,000 U/kg).During the experimental period of 120 d,the weight and total number of eggs within each pen were recorded daily,and egg quality was determined every 4 wk.At the end of the experiment,3 randomly selected ducks within each replicate were sacrificed,then duodenal digesta and jejunal mucosa was collected.Dietary inclusion of barley had no effects on egg production,daily egg mass or FCR,but supplementation with glucanase improved egg production and FCR(P<0.01).Barley did not affect feed intake of laying ducks,but glucanase tended to increase feed intake(P=0.09).Neither barley norβ-glucanase had effects on the egg quality variables,except for yolk color score,which was decreased with increasing barley supplementation.Glucanase,but not barley,increased the activity of chymotrypsin and amylase in duodenal digesta.Barley inclusion affected the activity of alkaline phosphatase and maltase in jejunal mucosa(P<0.05),butβ-glucanase had no effects on the activity of these brush border enzymes.Barley inclusion increased the glucan content in duodenal digesta,but supplementation of glucanase to barley-based diet reduced digesta glucan content and reduced total volatile fatty acids and increased the proportion of acetic acid in cecal contents.The results indicate that,without glucanase,the optimal dietary barley level in the diets of laying ducks is about 13%for maximal production performance;glucanase supplementation of the barley diets improved production perfor-mance,probably through enhancing digestive function.展开更多

Methods: 25 patients undergoing thoracic and abdominal aortic...Objective: To explore the nursing experience of digestive function after thoracic and abdominal aortic replacement in order to promote the better recovery. Methods: 25 patients undergoing thoracic and abdominal aortic replacement were managed after operation. First, the fluid volume and blood pressure were controlled, and the use of vasoactive drugs was strictly regulated to ensure the perfusion of abdominal organs. And the enteral nutrition is carried out as early as possible to accelerate the recovery of gastrointestinal function of patients;And multimode analgesia is adopted to promote patients to get out of bed as early as possible to recover gastrointestinal peristalsis;At the same time, anticoagulation management is strengthened to prevent microthrombosis. Results: 23 of 25 patients after thoracic and abdominal aortic replacement recovered well and discharged smoothly. 2 Cases died,1 case died of hemorrhagic shock due to rupture of abdominal great vessels, and 1 case died of multiple organ failure due to septic shock. Conclusion: To strengthen the maintenance of digestive function, ensure the blood perfusion of gastrointestinal tract, get out of bed early, promote the gastrointestinal peristalsis and implement enteral nutrition as early as possible, can promote the recovery of gastrointestinal function, accelerate the rehabilitation process and shorten the hospitalization time.展开更多
Objective To evaluate the efficacy and safety of administration of the formulated Persian herbal syrup on improving the symptoms of patients with constipation-predominant irritable bowel syndrome(IBS-C).Methods This s...Objective To evaluate the efficacy and safety of administration of the formulated Persian herbal syrup on improving the symptoms of patients with constipation-predominant irritable bowel syndrome(IBS-C).Methods This study was conducted in 70 patients with IBS-C,who were recruited from 3 medical centers in Mashhad,Iran,from November 2017 to August 2018.Seventy patients were randomly assigned to 2 groups including treatment and placebo groups by block randomization,35 cases in each group.Patients in the treatment group received 15 mL of anti-IBS syrup,thrice daily for 6 weeks and followed up for 4 weeks.Placebo syrup was also prepared through similar instruction,BP syrup without plant extract was used.Primary outcome induding IBS Symptom Severity Scale(IBS-SSS)questionnaire and secondary outcomes in terms of Hospital Anxiety and Depression(HADS)questionnaires,the Bristol Stool Form Scale(BSFS)were completed and evaluated at weeks 6 and 10,respectively.Safety indices were collected at the end of the treatment and Common Terminology Criteria for Adverse Events v4.0(CTCAE)was used to evaluate the adverse events.Results The response to treatment was 84.4%(27/32)in the treatment group and 46.4%(13/28)in the placebo group,respectively(P=0.002).Compared with pre-treatment,a significant decrease was found on the IBS-SSS and BSFS scores after 6-week intervention in both groups(P<0.001).Moreover,IBS-SSS and BSFS scores in the treatment group were lower than the placebo group after the intervention(P=0.041).There was no significant difference in the anxiety and depression scores after treatment in both groups(P>0.05).Side effects reported in the treatment group included 2 cases of headache during the first week of the onset of the treatment,1 case of drowsiness,1 case of increase in menstrual bleeding,which did not result in discontinuation of the treatment.In the placebo group,1 case of exacerbation of the disease was reported.Conclusions Anti-IBS syrup significantly reduced the severity of IBS symptoms compared to placebo.However,there was a need for further investigation regarding the anxiety and depression scores.(Registration No.IRCT2017061034446N1).展开更多
