Methods Diabetic mouse model was established by intraperitioneal injection of alloxan. Low, middle, and high doses of Cu2(Thr)4 at 0.002%, 0.02% and 0.1% were given respectively to diabetic mice following lavage. The fasting blood glucose was determined after the diabetic mice were given Cu2 (Thr)4 for 0, 30, and 45 d. The diabetic mice were killed on the 45th day. Then glycosylated hemoglobin (HbAlc) and blood lipids were assayed, and pathologic changes in hearts and kidneys stained with HE were observed. Results Compared with the control group in which the diabetic mice were given distilled water, the value of blood glucose reduced significantly in middle dose group (P 〈 0.01 ), followed by that in low dose group (P 〈 0.05). TC level reduced markedly and HDL level increased significantly in all three treatment groups (P 〈 0.05). Especially in middle dose group, cardiac muscle fibers were neatly arranged, nucleus and cytoplasm well distributed, glomeruli showing normal structure, cells well distributed and staining being normal. Conclusion Cu2 (Thr)4 reduces blood glucose, regulates blood lipids, and play protective action on the vessels of hearts and kidneys in diabetic mice. The effects of it in middle dose were better than those of other doses.展开更多

文摘Reaction of copper powder, dibenzoyl peroxide and 2-amino-4methyl-5-ethoxycarbonyl thiazole (amet) by refluxing in acetone yielded the dinuclearcopper(Ⅱ) complex Cu2 (μ-PhCOO)4 (amet)2- 2CH3COCH3. Crystal structure determination showed that each copper atom is coordinated by four bridged benzoxy and oneamet molecule with an M-M bond (Cu-Cu (A) = 2.6899 (7 ) ), therefore the coor-dination environment around the copper center is a distorted square pyramid with N(1)at the apical position. Blocks of Cu2 (μ-PhCOO)4 (amet)2·2CH3COCH3 crystallize inthe triclinic space group Ph, with Mr= 1100.14, a= 9. 631 (1), b=10. 446 (2), c=12. 758(2) ; a=91. 31 (1), β=95. 50(1),γ= 94. 87(1)°, V=1272. 4(3), Z=1, Dc= 1. 436 g/cm3, F(000) = 570, μ=0. 986 mm-1 and R=0.033, Rw=0.049.The IR spectra, which are of great interest since the molecule has ketone, ester andcarboxylate group simultaneously, are also discussed.
文摘Aim To study the effects of binuclear copper (Ⅱ) threonine complex (Cu2 (Thr)4) as analogue of superoxide dismutase (SOD) on blood glucose, blood lipids and vessels of hearts and kidneys in diabetic mice. The effects of it in middle dose were better than those of other doses.

文摘A novel copper(Ⅱ) complex derived from 1,4,7-triazacyclononane[CuL]_2(PF_6)_3×MeCN×H_2 O was synthesized and crystallographically characterized {L = 1,4-bis(2-carbamoylethyl)-7-benzimidazole-2-yl-methyl-1,4,7-triazacyclononane}. It crystallizes in triclinic, space group P1, with a = 13.2425(13), b = 14.0807(15), c = 17.6798(18), α = 86.296(2), β = 72.773(2), γ= 68.905(2)o, V = 2934.5(5)A^3, Z = 2, D_c = 1.611 g/m^3, F(000) = 1456, M_r = 1423.09, m = 0.920 mm^-1. The final R = 0.0671 and wR = 0.1874 for 6501 observed reflections with I 〉 2σ(I). The structural analysis shows that the complex cation([CuL]_2^3+) was formed by two complex cations, namely([CuL^3]^2+) and [CuL_(-H)~3]~+) through a hydrogen bond. In each complex cation, the Cu(Ⅱ) lies in a distorted square pyramidal geometry. The redox behavior was studied by cyclic voltammetry(CV) in aqueous solution which indicates a reversible one electron redox reaction. The result of UV absorption, ethidium bromide(EB) fluorescence spectra indicated that the complex binds to CT-DNA in an intercalative mode. Superoxide dismutase(SOD) activity of the complex was determined by photoreduction of NBT, and the value of IC_(50) is 5.22 μmol·L^-1.

文摘Six novel pentanuclear complexes [Cu 3Ln 2(ClCH 2CO 2) 12 (H 2O) 8]·2H 2O (Ln=La, Pr, Sm, Gd, Dy, or Er) were synthesized by the reaction of chloroacetic acid with Cu and Ln ions in aqueous solutions. The X ray crystallography established the structure of the Gd complex, which contains a pair of quadruply acetato bridged CuGd dinuclear subunits, and each of them is linked to the central Cu atom by a single acetato bridge. The coordination sphere of each Gd atom is completed by three aqua ligands [Gd-O=0.242 8(5)-0.244 4(4) nm] to form an 8 coordinated distorted square antiprism. The central Cu atom is coordinated by two oxygen atoms from the two bridging acetato groups and two oxygen atoms from the two monodentate acetato groups in a slightly distorted square planar fashion, while each terminal Cu atom is coordinated by the four acetato oxygen atoms at the basal plane and an aqua ligand at the apical position to form a square pyramid. The variable temperature (4 320 K) magnetic susceptibility data of the Gd complex were measured, and fitted to the Curie Weiss law with C =16.38 K cm 3·mol -1 and θ =-1.55 K, indicating very weak antiferromagnetic interaction.
