Although PD presents primarily with motor dysfunction, non-motor symptoms including cognitive decline, mood disorders, reduced olfaction and constipation are also of- ten present, with some of these non-motor symptoms even pre- senting prior to the onset of motor symptoms. It is well known that PD is largely caused by the gradual degeneration of dopa- minergic neurons within the substantia nigra pars compacta (SNc), along with the presence of protein aggregates called Lewy bodies, which consist primarily of ct-synuclein and are found in the cytoplasm of surviving neurons. This ongoing cell loss and Lewy body pathology is not confined to the SNc, but is also seen in other brain regions implicated in PD pathogenesis such as the locus ceruleus. 展开更多

Huntington's disease (HD) is the most common dominantly inherited neurodegenerative disorder, mainly characterized by the progressive striatal and cortical neurodegeneration and as- sociated motor, cognitive and behavioural disturbances (Zuccato et al., 2010). The disease-causing mutation is an expansion of a GAG trinucleotide repeat (〉 36 repeats) encoding a polygluta- mine stretch in the N-terminal region ofhuntingtin (Htt) (Zuc- cato et al., 2010), a ubiquitous protein whose function is still unclear (Zuccato et al., 2010). Expansion of the polyQ stretch endows mutant Htt (mHtt) with toxic properties, and results in the development of a broad array of undesirable effects in both neuronal and non-neuronal cells (Zuccato et al., 2010). Among all cellular dysfunctions and biochemical imbalances classically associated with HD, perturbed metabolism of (glyco) sphingolipids appears to play a crucial role in the pathogenesis of the disease. 目的:观察泪道内窥镜下泪小管切开联合新型R-S管置入治疗泪小管炎的临床疗效。方法:泪道内窥镜下泪小管切开联合泪道置管术治疗泪小管炎患者31例32眼,置入新型R-S管15例15眼为A组,置入环形泪道引流管16例17眼为B组,所有患者均术后2~3mo... 目的:观察泪道内窥镜下泪小管切开联合新型R-S管置入治疗泪小管炎的临床疗效。方法:泪道内窥镜下泪小管切开联合泪道置管术治疗泪小管炎患者31例32眼,置入新型R-S管15例15眼为A组,置入环形泪道引流管16例17眼为B组,所有患者均术后2~3mo拔管并观察至术后1a,比较两组手术疗效、手术置管时间、术后并发症、术后鼻腔异物感和鼻腔分泌物增多发生率。结果:A组治疗有效率为93%,B组为94%,两组无差异(P>0.05)。A组置管时间为2.27±1.335min,B组为5.29±1.404min;A组术后并发症发生率为7%,B组为47%;A组术后鼻腔异物感发生率为7%,B组为41%;A组术后鼻腔分泌物增多发生率为0,B组为29%。手术置管时间、术后并发症、鼻腔异物感及鼻腔分泌物增多发生率均有差异(P<0.05)。结论:泪道内窥镜下泪小管切开联合新型R-S管置入治疗泪小管炎效果良好,且新型R-S管具有操作简便、术后并发症少和患者舒适度高等优点。 展开更多

自交不亲和是植物避免近亲繁殖保持优良种性的重要途径,梅是典型的配子体(GSI)自交不亲和植物。本研究采用等位基因特异性PCR(AS-PCR,allele-specific PCR)和序列测定等技术,分别以梅S-RNase基因与SFB基因核苷酸序列保守区设计引物Pru-C... 自交不亲和是植物避免近亲繁殖保持优良种性的重要途径,梅是典型的配子体(GSI)自交不亲和植物。本研究采用等位基因特异性PCR(AS-PCR,allele-specific PCR)和序列测定等技术,分别以梅S-RNase基因与SFB基因核苷酸序列保守区设计引物Pru-C2和PCE-R、SFB-C1F和Pm-Vb,然后以11个品种的基因组DNA为模板进行AS-PCR扩增和PCR产物测序。经过序列对比分析明确了11个梅品种的S-RNase基因型与SFB基因型,分别为软条红梅(S14S13)(F-box2/SFB14)、小叶猪肝(S3S33)(SFB51/SFB53)、中红(S3S24)(SFB2/SFB24)、青佳915(S3S15)(SFB2/SFB43)、美林红(S1S37)(SFB1/SFB24)、赤凤红梅(S14S38)(SFB12/SFB49)、大龙梅(S20S30)(SFB7/SFB41)、红光梅(S3S33)(SFB24/SFB55)、南农丰羽(S3S5)(SFB31/SFB43)、南农丰娇(S3S6)(SFB2/SFB47)、南农龙霞(S15S37)(SFB41/SFB43)。对获得的S-RNase基因与SFB基因序列进一步分析发现,除了已知的S-RNase基因与SFB基因外,还鉴定到S37-RNase、S38-RNase 2个未登录的新S-RNase基因与SFB47、SFB49、SFB51、SFB53、SFB555个未登录的新SFB基因,并将其序列登录在Gen Bank数据库,其登录号分别为MT950061、MT950062,以及MW186463、MW186465、MW186467、MW186469、MW186471。研究结果为梅生产栽培、授粉树的配置和提高育种效率提供了依据。 展开更多

Treatment repurposing for inflammatory bowel disease using literature-related discovery and innovation 被引量:2

Inflammatory bowel disease(IBD)incidence has been increasing steadily,most dramatically in the Western developed countries.Treatment often includes lifelong immunosuppressive therapy and surgery.There is a critical need to reduce the burden of IBD and to discover medical therapies with better efficacy and fewer potential side-effects.Repurposing of treatments originally studied in other diseases with similar pathogenesis is less costly and time intensive than de novo drug discovery.This study used a treatment repurposing methodology,the literature-related discovery and innovation(LRDI)text mining system,to identify potential treatments(developed for non-IBD diseases)with sufficient promise for extrapolation to treatment of IBD.By searching for desirable patterns of twenty key biomarkers relevant to IBD(e.g.,inflammation,reactive oxygen species,autophagy,barrier function),the LRDI-based query retrieved approximately 9500 records from Medline.The most recent 350 records were further analyzed for proof-of-concept.Approximately 18%(64/350)met the criteria for discovery(not previously studied in IBD human or animal models)and relevance for application to IBD treatment.Many of the treatments were compounds derived from herbal remedies,and the majority of treatments were being studied in cancer,diabetes,and central nervous system disease,such as depression and dementia.As further validation of the search strategy,the query identified ten treatments that have just recently begun testing in IBD models in the last three years.Literature-related discovery and innovation text mining contains a unique search strategy with tremendous potential to identify treatments for repurposing.A more comprehensive query with additional key biomarkers would have retrieved many thousands more records,further increasing the yield of IBD treatment repurposing discovery. 展开更多

冠状病毒在系统分类上属冠状病毒科(Coronaviridae)冠状病毒属(Coronavirus),是一种具有球形颗粒和包膜结构的单链RNA病毒。从2003年在中国暴发的重症急性呼吸综合征(SARSCoV),到2012年在沙特出现的中东呼吸综合征(MERS-CoV),再到2019... 冠状病毒在系统分类上属冠状病毒科(Coronaviridae)冠状病毒属(Coronavirus),是一种具有球形颗粒和包膜结构的单链RNA病毒。从2003年在中国暴发的重症急性呼吸综合征(SARSCoV),到2012年在沙特出现的中东呼吸综合征(MERS-CoV),再到2019年开始报道的新型冠状病毒感染(2019-nCoV),21世纪冠状病毒已经3次严重危害到人类的健康。对冠状病毒的序列特征与进化关系进行分析对病毒的起源与防控具有重要意义。本研究从GenBank数据库中筛选到30条冠状病毒刺突蛋白(S)、核衣壳蛋白(N)的蛋白序列,用邻接法构建进化树,分析冠状病毒进化关系,并对蛋白保守基序进行了分析。结果显示:2019-nCoV与从云南省的中菊头蝠中检测到的冠状病毒分离株RaTG13进化关系最近,并且与SARS-CoV具有较近的亲缘关系,与其他哺乳类和禽类动物感染的冠状病毒遗传进化关系较远。同时发现,相比S蛋白,N蛋白更适合用于构建冠状病毒进化树。保守基序分析表明:N蛋白中,选择的30个蛋白中有9个只含有2个保守元件,21个成员含有3个保守元件,保守元件在蛋白中的分布相对均匀;S蛋白中,选择的30个蛋白均含有3个保守元件,且集中在C端。本研究结果将为冠状病毒的进化关系及防治措施提供一定的理论依据。 展开更多
