In the...This study examined the effects of combined administration of tyrosine, lecithin, L-glutamine and L-5-hydroxytryptophan (5-HTP) on heroin withdrawal syndromes and mental symptoms in detoxified heroin addicts. In the cluster-randomized placebo-controlled trial, 83 detoxified heroin addicts were recruited from a detoxification treatment center in Wuhan, China. Patients in the intervention group (n=41) were given the combined treatment with tyrosine, lecithin, L-glutamine and 5-HTP and those in the control group (n=42) were administered the placebo. The sleep status and the withdrawal symptoms were observed daily throughout the study, and the mood states were monitored pre- and post-intervention. The results showed that the insomnia and withdrawal scores were significantly improved over time in participants in the intervention group as compared with those in the control group. A greater reduction in tension-anxiety, depression-dejection, anger-hostility, fatigue-inertia and total mood disturbance, and a greater increase in their vigor-activity symptoms were found at day 6 in the intervention group than in the control group (all P〈0.05). It was concluded that the neurotransmitter-precursor-supplement intervention is effective in alleviating the withdrawal and mood symptoms and it may become a supplementary method for patients' recovery from heroin addiction.展开更多
Curcumin exerts a neuroprotective effect on Alzheimer’s disease;however,it is not known whether microRNAs are involved in this protective effect.This study was conducted using swAPP695-HEK293 cells as an Alzheimer’s...Curcumin exerts a neuroprotective effect on Alzheimer’s disease;however,it is not known whether microRNAs are involved in this protective effect.This study was conducted using swAPP695-HEK293 cells as an Alzheimer’s disease cell model.swAPP695-HEK293 cells were treated with 0,0.5,1,2,5,and 10μM curcumin for 24 hours.The changes in miR-15b-5p,miR-19a-3p,miR-195-5p,miR-101-3p,miR-216b-5p,miR-16-5p and miR-185-5p expression were assessed by real-time quantitative polymerase chain reaction.The mRNA and protein levels of amyloid precursor protein,amyloid-β40 and amyloid-β42 were evaluated by quantitative real-time polymerase chain reaction,western blot assays and enzyme-linked immunosorbent assays.swAPP695-HEK293 cells were transfected with miR-15b-5p mimic,or treated with 1μM curcumin 24 hours before miR-15b-5p inhibitor transfection.The effects of curcumin on amyloid precursor protein,amyloid-β40 and amyloid-β42 levels were evaluated by western blot assays and enzyme-linked immunosorbent assay.Luciferase assays were used to analyze the interaction between miR-15b-5p and the 3′-untranslated region of amyloid precursor protein.The results show that amyloid precursor protein and amyloid-βexpression were enhanced in swAPP695-HEK293 cells compared with HEK293 parental cells.Curcumin suppressed the expression of amyloid precursor protein and amyloid-βand up-regulated the expression of miR-15b-5p in swAPP695-HEK293 cells.In addition,we found a negative association of miR-15b-5p expression with amyloid precursor protein and amyloid-βlevels in the curcumin-treated cells.Luciferase assays revealed that miR-15b-5p impaired the luciferase activity of the plasmid harboring the 3′-untranslated region of amyloid precursor protein.These findings indicate that curcumin down-regulates the expression of amyloid precursor protein and amyloid-βin swAPP695-HEK293 cells,which was partially mediated by miR-15b-5p via targeting of the 3′-untranslated region of amyloid precursor protein.展开更多

目的:观察还脑益聪方复方组分早期干预对阿尔茨海默病(Alzhei mer disease,AD)模型β淀粉样前体蛋白(β-amyloid precursor protein,APP)转基因小鼠行为学及脑胆碱能系统的影响。方法:将60只3月龄APP695V717I转基因小鼠随机分为4组:模...目的:观察还脑益聪方复方组分早期干预对阿尔茨海默病(Alzhei mer disease,AD)模型β淀粉样前体蛋白(β-amyloid precursor protein,APP)转基因小鼠行为学及脑胆碱能系统的影响。方法:将60只3月龄APP695V717I转基因小鼠随机分为4组:模型组、大剂量还脑益聪方组[2.80g/(kg.d)]、小剂量还脑益聪方组[1.40g/(kg.d)]及盐酸多奈哌齐阳性对照组[0.65mg/(kg.d)],每组15只。15只遗传背景相同的非转基因C57BL/6J小鼠作为正常对照组。所试药物稀释至相同体积灌胃给药,正常组和模型组给以等体积蒸馏水灌胃,连续灌胃6个月。采用Morris水迷宫实验和避暗实验进行小鼠行为学测试,采用酶联免疫吸附测定法分别检测小鼠大脑皮层和海马组织的乙酰胆碱(acetylcholine,ACh)、乙酰胆碱酯酶(acetylcholinesterase,AChE)含量及胆碱乙酰转移酶(choline acetyltransferase,ChAT)的活性,采用考马斯亮蓝法测定脑组织中蛋白的含量。结果:与正常对照组比较,模型组小鼠学习记忆成绩均显著下降(P<0.05,P<0.01),海马和皮层组织的ACh含量和ChAT活性显著降低,AChE含量显著增加(P<0.01,P<0.05)。与模型组比较,盐酸多奈哌齐组、还脑益聪方组小鼠的学习记忆能力均有明显提高(P<0.05);盐酸多奈哌齐组、还脑益聪方大剂量组小鼠海马ACh含量明显提高(P<0.05),盐酸多奈哌齐组、还脑益聪方组小鼠海马AChE含量显著降低,ChAT活性显著升高(P<0.01,P<0.05);盐酸多奈哌齐组和还脑益聪方两组小鼠皮层ACh含量均明显提高(P<0.05),AChE含量有不同程度的降低,还脑益聪方两组小鼠皮层ChAT活性均明显提高(P<0.05)。结论:还脑益聪方能明显改善APP转基因小鼠的学习记忆能力,其作用机制可能与调控小鼠海马与皮层胆碱能系统相关酶的含量有关。展开更多
