Marine benthic dinoflagellates in the genus Coolia were reported in both tropical and temperate regions.We conducted a morphological and phylogenetic characterization of Coolia species from Hainan Island,South China Sea.The morphologies of three Coolia species were similar to those of the original descriptions of Coolia canariensis,C.tropicalis,and C.malayensis.In phylogenetic analyses based on the LSU rDNA and ITS regions(ITS1-5.8 S-ITS2),the Hainan strains of C.canariensis,C.tropicalis,and C.malayensis clustered within the clades of these species with other isolates from different areas.No differences(p-distance)in LSU rDNA sequences were found between the Hainan C.malayensis strains and the strains from New Zealand,Florida of USA,Malaysia,Japan,Dominican Republic and Guangxi and Hong Kong of China.For C.canariensis and C.tropicalis,no differences(p-distance)in the LSU rDNA sequences were found between the Hainan strains(D1 C2 and DS5 F4,respectively)and the Australian strains(NQAIF252 and NQAIF90,respectively).Our study reveals the morphological and genetic diversity of Coolia species from Hainan Island,South China Sea,which provides a detailed understanding of Coolia species of this area. 展开更多

Ganoderma is a genus of medicinally and economically important mushrooms in the family Ganodermataceae.Ganoderma species are popular medicinal mushrooms and their health benefits are well-documented.Ganoderma is a cosmopolitan genus that is widely distributed in both tropical and temperate regions.This genus is characterized by its unique laccate or non-laccate species with double-walled basidiospores.Here,we report on eight collections of G.gibbosum collected during surveys in Kunming,Yunnan Province,China.The specimens are described and illustrated based on macro-and micro-morphological characteristics.Total DNA of the eight G.gibbosum strains were extracted using the Biospin Fungal Extraction Kit following manufacturer protocol.Amplification of the Internal Transcribed Spacer(nrITS)region was carried out using ITS5/ITS4 primers and LROR/LR5 for the nuclear ribosomal large subunit 28S rDNA gene(LSU).Phylogenetic analysis with closely related species to G.gibbosum showed that all eight collections grouped with G.gibbosum with 100%bootstrap support.Phylogenetic similarity and morphological variations within the eight collections of G.gibbosum are discussed. 展开更多

Three Diophrys species,D.quadrinucleata n.sp.,D.oligothrix Borror,1965 and D.scutum(Dujardin,1841)Kahl,1932,were identified from the subtropical brackish waters in China,their morphology was investigated using live observation and protargol impregnation.D.quadrinucleata n.sp.can be distinguished from the congeners by four ellipsoidal macronuclear nodules.Phylogenetic analyses show that the three species with single marginal cirrus are closely related,namely D.quadrinucleata n.sp.,D.apoligothrix Song et al.,2009 and D.japonica Hu,2008.Our population of D.oligothrix corresponds very well with previous populations in both living morphology and ciliature.However,the SSU rDNA sequences of three well-described populations differ from each other by 11 to 18 nucleotides,indicating that there might be cryptic species in D.oligothrix.Moreover,we provided a detailed description of the symbiotic bacteria/archaeobacteria(?)concentrated on the back surface of D.scutum based on the present population,which was either overlooked or misinterpreted as cortical granules previously. 展开更多

Ciliates of the heterotrich family Folliculinidae are characterized by their transparent lorica,conspicuous peristomial lobes pair and dimorphic life cycle,a sessile trophont and a motile swarmer.However,understanding their biodiversity and systematics is relatively poor.In the present study,two species representing two genera of folliculinids,which were collected from marine and brackish habitats in China,were investigated with morphological and molecular methods.The genus Diafolliculina n.gen.is established for D.longilobata n.sp.Detailed morphological redescriptions for another species Eufolliculina is provided.Moreover,phylogenetic analysis based on small subunit ribosomal DNA(SSU rDNA)sequences showed that 1)all two genera locate in the Folliculinidae clade;2)Metafolliculina clusters with Eufolliculina;and 3)two genera(Diafolliculina and Ampullofolliculina)with a lorica closure device have close affinities with Folliculina.The lorica of Diafolliculina and Ampullofolliculina lacks a closure device,thereby the phylogeney contradicts the morphology-based classification. 展开更多

The mass mortality of pond-reared scattered mirror carp, Cyprinus carpio, caused by Thelohanellus kitauei, occurred at fish farms in Sichuan Province, southwest China. Morphological and molecular analyses were supplemented with histological evaluation of infected tissues to better understand the route of infection and the pathological effects of T. kitauei on the fish host. The intestine of the diseased host was full of large cysts of the myxosporean. The cysts range from 2 cm to 3.6 cm in diameter. Histopathology indicated that T. kitaue i first invaded the submucosa of the host intestine and then moved into the mucosa layers with the development of their spores, finally entering into the enteric cavity of the hosts after the disruption of mucosa layers. The pyriform spores of T. kitaue i were surrounded by the transparent spore sheath, measuring 25.98 μm±0.95 μm in body length, 8.72 μm±0.51 μm in body width, and 7.86 μm±0.26 μm in body thickness. The single polar capsule was pyriform, measuring 14.73 μm±0.92 μm in length and 6.82 μm±0.45 μm in width, with eight to 10 turns of filament coils winding inside. Phylogenetic analysis based on the 18 S small-subunit ribosomal DNA sequences indicated that minimal genetic differences were present between T. kitauei samples from South Korea and from China. Close affinity was found between the genus Thelohanellus and Myxobolus. Additionally, two polar capsule nuclei were found at the anterior end of the single polar capsule in spores of T. kitaue i stained with hematoxylin and eosin, which suggested the separation of the genus Thelohanellus from Myxobolus. 展开更多

The marine and freshwater Hildenbrandia samples were collected from diverse locations of China and Japan,and mor-phological characterization and molecular phylogenetic analysis were conducted on these samples.Morphological measurements of freshwater specimens were consistent with the results of H.jigongshanensis,whereas cell dimensions of marine specimens were slightly larger than those of widely distributed H.rubra and H.crouanii.Phylogenetic trees based on rbcL,psbA and UPA sequences were consistent.Freshwater specimens collected in this study formed an independent clade with H.jigongshanensis supported by high values.Species attribution was confirmed further by the similar ITS2 secondary structures among the samples of H.jigongshan-ensis.It was found that the intraspecific divergence of rbcL gene in H.jigongshanensis was smaller than other two freshwater species H.rivularis and H.angolensis.Phylogenetic trees showed that marine specimens in this study grouped together with H.rubra samples of North America,and were different with the H.rubra samples of Europe.Combining the comparison results of CBC numbers in ITS2 secondary structure,we propose the marine samples collected in this study to be a new species Hildenbrandia qingdaoensis sp.nov.The separation of the marine and freshwater specimens was supported by the rbcL,UPA and psbA phylogeny as well as ITS2 se-condary structure inference.Biogeographical reconstruction showed the ancestor of freshwater Hildenbrandia might derive from South America and dispersed from North America to Europe and then to Asian countries,which needs to be verified further with more sam-ples and molecular evidences from South America. 展开更多

During faunal studies of psammophilic ciliates along the coast of Qingdao, China, several marine karyorelictean species were isolated. Among them,four species within the genus Remanella were investigated, including two species new to science:i.e., R. rugosa, Remanella elongata sp. nov.,Remanella aposinica sp. nov.,and R.unicorpusculata. Remanella rugosa has been reported several times, but this study is the first to provide detailed morphological characters and phylogenetics. Remanella elongata sp. nov. can be distinguished from its congeners by the presence of complex cortical granules, fewer macronuclei, and longer body size. Remanella aposinica sp. nov.differs from its congeners by having 14–17 right lateral ciliary rows and 24–37 dikinetids of intrabuccal kinety. Poorly known Remanella rugosa var. unicorpusculata(Kahl, 1933) Foissner, 1996should be elevated from subspecies to species level,Remanella unicorpusculata(Foissner, 1996) stat.nov., based on detailed redescriptions with statistical data, living morphology, infraciliature, and species definitions. Small subunit(SSU) rDNA was sequenced for the four species, and phylogenetic analysis revealed that all known taxa in Remanella formed the outline branch to the genus Loxodes with moderate to high bootstrap support among Remanella lineages. 展开更多

The morphology and phylogeny of two hypotrichous ciliates, Metaurostylopsis parastruederkypkeae n. sp. and Neourostylopsis flavicana(Wang et al., 2011) Chen et al., 2013 were investigated based on morphology, infraciliature and the small subunit(SSU) ribosomal RNA gene(r RNA) sequence. The new species, M. parastruederkypkeae n. sp. was identified according to its characteristics: body shape ellipsoidal, size about(165–200) ×(45–60) μm in vivo, cell color reddish; two types of cortical granules including wheat grain-like and yellow-greenish larger ones along the marginal cirri rows and dorsal kineties and dot-like and reddish smaller ones, grouped around marginal cirri on ventral side and arranged in short lines on dorsal side; 26–41 adoral membranelles; three frontal and one parabuccal, five to seven frontoterminal, one buccal, and three to six transverse cirri; seven to thirteen midventral pairs; five to nine unpaired ventral cirri, five to seven left and three to five right marginal rows; and three complete dorsal kineties. Phylogenetic analysis based on SSU r DNA sequences showed that both Metaurostylopsis and Neourostylopsis are monophyletic. As the internal relationship between and within both genera are not clear, further studies on the species in these two genera are necessary. The key characteristics of all known twelve Metaurostylopsis-Apourostylopsis-Neourostylopsis species complex were updated. 展开更多

Engelhardia,a genus of Juglandaceae(the walnut family),is endemic to tropical and subtropical Asia.The rich Cenozoic fossil records and distinctive morphological characters of the living plants have been used to explore the evolutionary history and geographic distribution of Juglandaceae.However,the taxonomy of this genus has been suffered from a lack of in-depth investigation and good specimens across its distribution ranges.Species delimitation of Engelhardia was defined with seven species in 2020,but detailed information on the circumscription of the species still remains poorly understood.In this study,two new species are described from Sulawesi and Borneo,Engelhardia anminiana and E.borneensis.We also revised and reconstructed the phylogeny within Engelhardia using morphological,molecular(plastid and ribosomal),and distribution data.We sampled 787 individuals in 80 populations,and all the samples were genotyped using plastid regions,trnS-trnG,rps16,trnL-trnF,psbA-trnH,and rpl32-trnL;one ribosomal region,nuclear ribosomal internal transcribed spacer(nrITS).The all datasets were used to reconstruct the phylogenetic relationships.Then,the molecular analyses were combined for 738 sheets of specimens with 15 morphological characteristics to further explore the morphological clusters of Engelhardia.Cluster analysis using morphological data confirmed the delimitation of nine Engelhardia species.Also,phylogenetic analysis based on molecular data(i.e.,plastid and ribosomal)supported the monophyly of Engelhardia and generated phylogenetic trees that included E.fenzelii,E.roxburghiana,E.borneensis,E.hainanensis,E.anminiana,E.serrata,E.villosa,E.apoensis and the varieties of E.spicata(i.e.,E.spicata var.spicata,E.spicata var.rigida,E.spicata var.aceriflora,and E.spicata var.colebrookeana).Our comprehensive taxonomic revision of Engelhardia will provide an insight into understanding the plant diversity in tropical and subtropical Asia. 展开更多

Hypocreomycetidae is a highly diverse group with species from various habitats. This subclass has been reported as pathogenic,endophytic, parasitic, saprobic, fungicolous, lichenicolous, algicolous, coprophilous and insect fungi from aquatic andterrestrial habitats. In this study, we focused on freshwater fungi of Hypocreomycetidae which resulted 41 fresh collectionsfrom China and Thailand. Based on morphological and phylogenetic analyses, we identified 26 species that belong to twoorders (Hypocreales and Microascales) and six families (Bionectriaceae, Halosphaeriaceae, Microascaceae, Nectriaceae,Sarocladiaceae and Stachybotryaceae). Ten new species are introduced and 13 new habitats and geographic records arereported. Mariannaea superimposita, Stachybotrys chartarum and S. chlorohalonatus are recollected from freshwater habitatsin China. Based on phylogenetic analysis of combined LSU, ITS, SSU, rpb2 and tef1-α sequences data, Emericellopsisis transferred to Hypocreales genera incertae sedis;Pseudoacremonium is transferred to Bionectriaceae;Sedecimiella isplaced in Nectriaceae;Nautosphaeria and Tubakiella are excluded from Halosphaeriaceae and placed in Microascalesgenera incertae sedis;and Faurelina is excluded from Hypocreomycetidae. Varicosporella is placed under Atractium as asynonym of Atractium. In addition, phylogenetic analysis and divergence time estimates showed that Ascocodina, Campylospora,Cornuvesica and Xenodactylariaceae form distinct lineages in Hypocreomycetidae and they evolved in the family/order time frame. Hence, a new order (Xenodactylariales) and three new families (Ascocodinaceae, Campylosporaceae andCornuvesicaceae) are introduced based on phylogenetic analysis, divergence time estimations and morphological characters.Ancestral character state analysis is performed for different habitats of Hypocreomycetidae including freshwater, marineand terrestrial taxa. The result indicates that marine and freshwater fungi evolved independently from terrestrial ancestors.The results further support those early diverging clades of this subclass, mostly comprising terrestrial taxa and freshwaterand marine taxa have been secondarily derived, while the crown clade (Nectriaceae) is represented in all three habitats. Theevolution of various morphological adaptations towards their habitual changes are also discussed. 展开更多

The inland saltwater lakes harbor exceptional biodiversity.Here,two new species of solitary sessile peritrich ciliates were isolated from Qinghai Lake,the largest inland saltwater lake in China.Their morphology,ciliature,silverline system,and molecular phylogeny were investigated based on live observation,silver staining,and analysis of the small subunit ribosomal DNA(SSU rDNA).Vorticella paraglobosa sp.n.is characterized mainly by its obconical or elongate bell-shaped zooid,C-shaped macronucleus,single ventrally located contractile vacuole,two-rowed infundibular polykinety 3,and 28-38 silverlines between peristome and aboral tro-chal band and 10-15 between aboral trochal band and scopula.Vorticella cotyliformis sp.n.differs from its congeners mainly by its double-layered peristomial lip,cup-shaped zooid,J-shaped macronucleus,single ventrally positioned contractile vacuole,three-rowed infundibular polykinety 3,and 70-85 silverlines between peristome and aboral trochal band and 21-25 between aboral trochal band and scopula.The SSU rDNA sequences of the two new species were obtained,and the subsequent molecular phylogenetic analysis supported their taxonomic classification. 展开更多

The characteristic signs and s... Maize(Zea mays L.) is an economically vital grain crop that is cultivated worldwide. In 2011, a maize foliar disease was detected in Lingtai and Lintao counties in Gansu Province, China. The characteristic signs and symptoms of this disease include irregular chlorotic lesions on the tips and edges of infected leaves and black punctate fruiting bodies in dead leaf tissues. Given favourable environmental conditions, this disease spread to areas surrounding Gansu. In this study, infected leaves were collected from Gansu and Ningxia Hui Autonomous Region between 2018and 2020 to identify the disease-causing pathogen. Based on morphological features, pathogenicity tests, and multilocus phylogenetic analysis involving internal transcribed spacer(ITS), 18S small subunit rDNA(SSU), 28S large subunit rDNA(LSU), translation elongation factor 1-alpha(TEF), and β-tubulin(TUB) sequences, Eutiarosporella dactylidis was identified as the causative pathogen of this newly discovered leaf blight. Furthermore, an in vitro bioassay was conducted on representative strains using six fungicides, and both fludioxonil and carbendazim were found to significantly inhibit the mycelial growth of E. dactylidis. The results of this study provide a reference for the detection and management of Eutiarosporella leaf blight. 展开更多

The genus Scincella Mittleman,1950,within the family Scincidae,is a diverse group comprising 39 described species.The taxonomy of the genus is complex due to morphological similarities among species,which often obscures the true extent of biodiversity,resulting in several undescribed species requiring further exploration.In China,particularly within the unique ecosystems of the Hengduan Mountains,the diversity of Scincella is notably underestimated,with only 12 species currently recognized.In the present study,we describe a new species of the genus Scincella,Scincella liangshanensis sp.nov.,from Sichuan Province,Southwest China.Morphological characteristics and phylogenetic analyses revealed that the putative new species is distinct from all other known congeners.This discovery not only expands the known diversity of Scincella but also emphasizes the inadequacies in our current understanding of herpetological diversity in China.The addition of this species to the scientific record highlights the necessity for ongoing biodiversity assessments to uncover hidden species and enhance our understanding of the intricate web of life in one of the world’s biodiversity hotspots. 展开更多

Species of the ciliate genus Spirostomum Ehrenberg,1834 are distributed worldwide and have a research history spanning more than two centuries.However,species delimitation and phylogenetic relationships within this genus are still uncertain due to the paucity of stable morphologic characters for species separation and the unavailability of accompanying morphological data for most molecular sequences in public databases.In the present study,S.yagiui Shigenaka,1959(three populations)and S.caudatum(Müller,1786)Delphy,1939(one population)were investigated using morphological and molecular methods for the first time in China.Detailed morphological data for the two species were documented,and improved diagnoses were supplied based on a combination of previous studies and the current work.It should be highlighted that there were three different atypical morphotypes identified in a Ningbo population of S.yagiui which may represent various stages in conjugative reproduction.Molecular phylogenies based on 18S,ITS1-5.8S-ITS2,and 28S rRNA gene sequences show that the genus Spirostomum is monophyletic,however,the internal relationships inferred from different genes were poorly resolved but suggest that the species with a moniliform macronucleus comprise an early-diverging clade within this genus.Finally,the global distribution of Spirostomum is summarized based on previous and present studies. 展开更多
