超临界CO_2萃取精馏制取高纯度EPA乙酯和DHA乙酯

Fish oil ethyl esters complexed with aqueous silver nitrate solution were extracted and rectified by supercritical CO 2 to obtain DHA ester and EPA ester with high purity. The effects of some independent variables,such as extraction pressure, temperature gradient of rectifying column and programmed pressure,on rectification were investigated.The results showed that programmed pressure is suitable for purification of EPA and DHA esters. Increase of column temperature gradient from bottom to top is one of the key elements in rectification. Furthermore, higher temperature gradient leads to better separation effect.

鱼油中二十碳五烯酸乙酯和二十二碳六烯酸乙酯的超临界流体色谱分离技术

建立鱼油中二十碳五烯酸乙酯(EPA-EE)和二十二碳六烯酸乙酯(DHA-EE)的超临界流体色谱纯化方法。比较Luna 5u Silica、Hedera ODS-BP、Hypersil ODS2、ZORBAX SB-C8和Hypersil C8等色谱柱对EPA-EE和DHA-EE的分离效果,确定以Hedera ODS-BP作为优选色谱柱,进一步以容量因子、选择性和分离度为考核指标,考察改性剂、流动相比例、背压及温度对分离效果的影响。得到的优化分离条件为:以Hedera ODS-BP为色谱柱,10%甲醇为改性剂,总流速2mL/min、柱温40℃、背压100bar。在此条件下,鱼油中EPA-EE和DHA-EE可以达到基线分离。结论:本法简便、快速、重现性好,可用于鱼油中EPA-EE和DHA-EE的分离。

鱼油制剂中过氧化值的降低方法

二十碳五烯酸(EPA)和二十二碳六烯酸(DHA)是一些鱼油制剂中的有效成分,临床上用于降血脂和其他一些疾病治疗,有关其新的药用价值,国内外也屡有报道。但由于EPA及DHA的高度不饱和性,在制剂和存放过程中,与空气接触后易被氧化形成过氧化物,使油脂的过氧化值(POV)超过指标而不能使用。目前,一般采用加入抗氧剂的方法抑制过氧化物的形成。
