Screening of QHF formula for effective ingredients from Chinese herbs and its anti-hepatic cell cancer effect in combination with chemotherapy 被引量:17

Background Recent studies have shown that effective ingredients of Chinese herbs are used more and more widely in the treatment or co-treatment of cancers, however, they are usually used separately and there has been limited research about joint application of Chinese herbs in multi-modal treatment. The aim of this study was to screen a QHF (Q: Qingrejiedu, H: Huoxuehuayu and F: Fuzhengguben) formula for effective ingredients from Chinese medicines and assess its anti-hepatic cell cancer (HCC) effect in combination with chemotherapy.Methods Six effective ingredients from Chinese medicine were selected based on the previous literature and used in the study. The QHF formula and the best ratio of ingredients were evaluated in H22 mouse (KM) models with solid tumors and ascites tumors by uniform design and monitoring inhibition of tumor growth and survival. We then observed the anti-hepatic cell cancer (HCC) effect of QHF when combined with cisplatin (DDP) in H22 mouse (Balb/c) models with solid tumors and ascites tumors. Evaluating of the therapeutic effect included the general condition of the mice, inhibition of tumor growth, survival, changes in body weight, thymus index, spleen index and WBC counts.Results The optimal QHF dose ratio for anti-hepatic cell cancer treatment was: 800 mg/kg Cinobufotalin, 14 mg/kg Ginsenosides Rg3, 5.5 mg/kg PNS and 100 mg/kg Lentinan. Treatment was more efficient in inhibiting the growth of transplanted tumors in H22 mice when using the QHF formula (55.91%) than using Cinobufotalin (33.25%), Ginsenosides Rg3 (35.11%), PNS (27.12%) or Lentinan (4.97%) separately. QHF also prolonged the life of H22 ascites hepatic cancer mice more efficiently (38.13%) than Cinobufotalin (25.00%), Ginsenosides Rg3 (27.27%), PNS (23.30%) or Lentinan (24.43%). QHF combined with DDP could reduce DDP-induced leucopenia, spleen and thymus atrophy and other toxic reactions. Combining QHF with DDP the tumor growth inhibition reached 82.54% with a 66.83% increase in survival. Conclusions QHF is more efficient in anti-hepatic cell caner treatment than the single drugs that constitute the formula. QHF combined with DDP can attenuate tumor growth and suppresses the DDP-induced toxic reactions. 展开更多

[ Method ] Through potting efficacy experiment of Chinese cabbage, the control effects of two preparations(20% EC and 10% EW)of methanol extract of Blumea megacephala Chang et Tseng against turnip aphid, as well as leaf-protection effect on Chinese cabbage were determined. [ Result] 20% EC of B. megacephala had the highest efficacy in 10-fold dilution, and its control effects against turnip aphid were 57.78%, 77.19% and 84.79% after pesticide spraying for 24, 48 and 72 h; 10% EW of B. megacephala had the highest efficacy in 5-fold dilution, and its control effects against turnip aphid were 74.44%, 84.79% and 89.14% after pesticide spra- ying for 24, 48 and 72 h. The leaf-protection rates of 20% EC orB. megacephala in 10-fold dilution were 89.31% and 94.80% after pesticide spraying for 3 and 5 d; the leaf-protection rates of 10% EW of B. megacephala in 5-fold dilution were 93.50% and 97.83% after pesticide spraying for 3 and 5 d. Investigation of paeonol-geniposide on acute alcoholic liver injury based on uniform design method

Objective:To explore the optimal ratio and compatibility effect of paeonol-geniposide combination on acute alcoholic liver injury by uniform design.Methods:Lieber-DeCarli alcoholic liquid feed was used to induce acute alcoholic liver injury in mice.Uniform design was used to select the best dosage combination of paeonol and geniposide,and the related indexes of liver injury and oxidative stress were detected by kit.Serum inflammatory factors were detected by ELISA,and the expressions of p38 MAPK,JNK and NF-κB P65 related proteins in liver were detected by Western-blot.Results:The regression equation suggested that paeonol:geniposide=220:20 was the best ratio of paeonol and geniposide to resist alcoholic liver injury.Compared with the model group,the liver injury indexes and oxidation products of the paeonol+geniposide group decreased significantly,the antioxidant activity of liver tissue increased significantly,and the expression levels of p-p38 MAPK,p-JNK and NF-κB P65 protein decreased significantly.Conclusion:The optimal dosage of paeonolgeniposide was effectively optimized by uniform design and pharmacodynamic analysis.The combination of the two drugs could reduce the alcoholic liver injury by reducing the oxidative stress injury and inflammatory response in the liver tissue of mice,and its effect might be related to the targeting of p38 MAPK/JNK/NF-κB channel. 展开更多
