Ligliptin for treatment of type 2 diabetes mellitus with early renal injury: Efficacy and impact on endogenous hydrogen sulfide and endothelial function 被引量:4

BACKGROUND Diabetes is a clinically common chronic disease,and its incidence has been increasing in recent years.Diabetes is believed to accelerate the process of atherosclerosis in patients,and abnormal endothelial function is an important factor leading to diabetic kidney damage.AIM To investigate the efficacy of ligliptin in the treatment of type 2 diabetes mellitus(T2DM)with early renal injury and its effect on serum endogenous hydrogen sulfide(H2S),endothelial cell particles,and endothelial function.METHODS From January 2018 to April 2019,110 patients with T2DM and early kidney injury treated at our hospital were divided into an observation group(receiving ligliptin treatment,n=54)and a control group(receiving gliquidone therapy,n=56).Blood glucose and renal function before and after treatment were compared between the two groups.RESULTS The differences in fasting blood glucose,2 h blood glucose,and glycated hemoglobin were not statistically significant between the two groups after treatment.The urinary albumin excretion rate after treatment in the ligliptin group was 70.32±11.21μg/min,which was significantly lower than that of the gliquidone group(P=0.000).Serum endogenous H2S and endothelial cell microparticles of the ligliptin treatment group were 40.04±8.82 mol/L and 133.40±34.39,respectively,which were significantly lower than those of the gliquidone treatment group(P=0.000 for both);endothelin-dependent diastolic function and nitric oxide after treatment in the ligliptin group were 7.98%±1.22%and 190.78±30.32 mol/L,significantly higher than those of the gliquidone treatment group(P=0.000 for both).CONCLUSION Ligliptin treatment of T2DM with early renal injury has the same glucoselowering effect as gliquidone treatment.Ligliptin treatment has a better effect and it can significantly improve the renal function and vascular endothelial function of patients,and reduce serum endogenous H2S and endothelial cell particle levels. 展开更多

Hydrogen sulfide (H2S) is the third gaseous signaling molecule discovered in recent years, and plays an important physiological role in the cardivascular system. To explore the effects of different doses of exogenous H2S on the electrocardiogram (ECG) of rats generally anesthetized by zoletil, different doses of NariS solution were used for the intervention of intraperitoneal injection 20 rain before the zoletil anesthesia. The ECGs of rats from each treatment group during the time range of 10^th-50^th min were determined under general anesthesia, and then were compared with those from the control group. The results showed that exogenous H2S could significantly reduce the Q-T interval time limit, thus played a role in slowing tachycardia or arrhythmia and other anomalies, thereby protecting the heart. S-T segment and T segment evaluation values were significantly reduced, which might be associated with bradycardia. 展开更多

Neuroinflammation has been recognized to play a critical role in the pathogenesis of Alzheimer's disease (AD), which is pathologically characterized by the accumulation of senile plaques containing activated microglia and amyloid β-peptides (Aβ). In the present study, we examined the neuroprotective effects of hydrogen sulfide (H2S) on neuroinflammation in rats with Aβ1-40 hippocampal injection. We found that Aβ-induced rats exhibited a disorder of pyramidal cell layer arrangement, and a decrease of mean pyramidal cell number in the CA1 hippocampal region compared with those in sham operated rats. NaHS (a donor of H2S, 5.6 mg/kg/d, i.p.) treatment for 3 weeks rescued neuronal cell death significantly. Moreover, we found that H2S dramatically suppressed the release of TNF-α, IL-1β and IL-6 in the hippocampus. Consistently, both immunohistochemistry and Western blotting assays showed that H2S inhibited the upregulation of COX-2 and the activation of NF-κB in the hippocampus. In conclusion, our data indicate that H2S suppresses neuroinflammation via inhibition of the NF-κB activation pathway in the Aβ-induced rat model and has potential value for AD therapy. 展开更多

目的探讨慢性阻塞性肺疾病(COPD)合并肺动脉高压(PH)患者血清内源性气体分子一氧化氮(NO)和硫化氢(H2S)含量变化及其临床意义。方法回顾性分析2015年6月至2017年6月我院接诊的40例单纯COPD患者及40例COPD合并PH患者临床资料,另选择同期... 目的探讨慢性阻塞性肺疾病(COPD)合并肺动脉高压(PH)患者血清内源性气体分子一氧化氮(NO)和硫化氢(H2S)含量变化及其临床意义。方法回顾性分析2015年6月至2017年6月我院接诊的40例单纯COPD患者及40例COPD合并PH患者临床资料,另选择同期在我院接受体检的健康志愿者40例作为对照组。比较三组血清NO、H2S及肺动脉平均压(MPAP)水平的表达,并分析血清NO、H2S及MPAP的相关性。结果 COPD合并PH组、COPD组、对照组血清NO分别为(37. 90±4. 49)、(55. 55±7. 05)、(72. 94±10. 35)μmol/L(P <0. 05); COPD合并PH组、COPD组、对照组H2S水平分别为(21. 92±1. 65)、(28. 29±2. 25)、(34. 53±2. 55)μmol/L(P <0. 05); COPD合并PH组、COPD组、对照组MPAP水平分别为(30. 54±2. 54)、(20. 12±2. 31)、(15. 78±1. 94) mm Hg(P <0. 05);在Pearson相关分析结果中显示,血清NO、H2S和MPAP之间呈负相关(r=-0. 768、-0. 808,P <0. 01),而血清NO和H2S之间呈正相关(r=0. 681,P <0. 01);联合检测NO+H2S的诊断效能(AUC=0. 934,95%CI:0. 882-0. 987)高于NO(AUC=0. 876,95%CI:0. 800-0. 952)和H2S(AUC=0. 946,95%CI:0. 757-0. 934)。结论血清内源性气体分子NO、H2S在COPD合并PH患者中的表达明显降低,参与着疾病的发生、发展。 展开更多

Analysis of H_2S Tolerance of Pd-Cu Alloy Hydrogen Separation Membranes

The presence of a limited amount of H2S in H2-rich feed adversely affects the Pd-Cu membrane permeation performance due to the sulphidization of the membrane surface. A theoretical model was proposed to predict the S-tolerant performance of the Pd-Cu membranes in presence of H2S under the industrial water-gas-shift(WGS) reaction conditions. The ideas of surface coverage and competitive adsorption thermodynamics of H2S and H2 on Pd-Cu surface were introduced in the model. The surface sulphidization of the Pd-Cu membranes mainly depended on the pressure ratio of H2S to H2, temperature and S-adsorbed surface coverage, i.e., the occurrence of sulphidization on the surface was not directly related with the bulk compositions and structures [body centered cubic and face centered cubic(bcc or fcc)] of Pd-Cu alloy membranes because of the surface segregation phenomena. The resulting equilibrium equations for the H2S adsorption/sulphidization reactions were solved to calculate the pressure ratio of H2S to H2 over a wide range of temperatures. A validation of the model was performed through a comparison between lots of literature data and the model calculations over a rather broad range of operating conditions. An extremely good agreement was obtained in the different cases, and thus, the model can serve to guide the development of S-resistant Pd alloy membrane materials for hydrogen separation. 展开更多

With the aid of hydrogen permeating devices, the hydrogen permeation behaviors of X52 pipeline steel in NACE A solution with saturated H2S/CO2 were studied under the conditions of different ambient temperatures and pH values, and the hydrogen permeation behaviors of X52 pipeline steel in weld seam zone were comparatively studied. The experimental results show that the hydrogen permeation coefficient value is directly proportional to the time required for reaching the saturation anode current and inversely proportional to the saturation anode current, and the hydrogen permeation coefficient is influenced by the corrosion scales; the temperature is directly proportional to the saturation anode current, and the hydrogen permeation coefficient is influenced by the temperature and corrosion scales, heat-affected zone and matrix zone in NACE A solution with saturated H2S/CO2 at normal temperature. The hydrogen permeation coefficient in weld seam zone is larger than that in heat-affected zone which is further larger than that in matrix zone. 展开更多

Hydrogen sulfide is synthesized endogenously in mammals and has been shown to have both physiological and pathological functions. So far there has been little agreement as to the actual levels of endogenous sulfide under physiological or pathological conditions;this is partly due to the complexity involved in measuring free sulfides due to H2S volatility, oxidation, reactivity and the presence of bound labile sulfur in tissues. In this report we describe a method of measuring free tissue sulfides using a zinc sulfide precipitation and wash method. It is an indirect method that measures the sulfide difference between samples prepared at pH 9 and pH 6, assuming that at pH 9 free sulfides would be retained in solution, while at pH 6 free sulfides would volatilize during sample preparation. Using this approach we were able to measure appreciable amounts of free sulfides in mouse: lung, pancreas, liver and kidney at 0.036 + 0.006, 0.082 + 0.009, 0.215 + 0.016 and 0.323 + 0.031 nmole per mg of tissue respectively (n = 6). 展开更多

光伏电催化硫化氢分解制氢脱硫经济性分析

近年来,以太阳能驱动电催化硫化氢(H2S)制氢脱硫,不仅能够生产绿氢与硫磺,还能高效利用环境资源,推动能源结构转型,是绿色可持续利用H2S资源的变革性新技术。光伏电催化H2S制氢技术具有以可再生能源供能、常温常压、产物易分离等优点,... 近年来,以太阳能驱动电催化硫化氢(H2S)制氢脱硫,不仅能够生产绿氢与硫磺,还能高效利用环境资源,推动能源结构转型,是绿色可持续利用H2S资源的变革性新技术。光伏电催化H2S制氢技术具有以可再生能源供能、常温常压、产物易分离等优点,展现出了极大的应用前景,研究其经济性对于促进技术创新与规模应用具有重要意义。为此,以柴达木盆地某光伏发电项目为例,根据光伏电解H2S制氢脱硫系统各模块运行特性,运用线性规划模型,计算了制氢脱硫系统内部收益率(IRR)和平准化制氢成本(LCOH)等经济指标,最后从经济性和敏感性两个方面,剖析了年辐射量、光伏规模、设备成本和制氢能耗等多个因素对关键指标的影响程度。研究结果表明:①储能成本的增加会降低系统经济效益,离网不储能模式下的LCOH优于电解水制氢技术;②年辐射量大于1396 kW·h/m^(2)的油气田布局光伏电催化H2S制氢项目将具有一定经济性;③技术进步率对LCOH的降低效果优于光伏装机规模的增加,未来5年,加速的技术进步率可降低LCOH至15.10元/kg,接近于工业副产提纯制氢技术;④LCOH对电解槽制氢能耗的敏感性程度最高,当电解槽制氢能耗降低20%后,LCOH低至22.37元/kg,基本具备市场竞争条件。结论认为,随着设备制备水平的提升和制氢能耗的进一步降低,光伏电催化H2S制氢脱硫技术将具有较大的经济优势与发展潜力,应加大对该技术的研究和公关力度,以推动油气田的绿色低碳转型。 展开更多

为揭示锅炉水冷壁H_(2)S在不同负荷时的浓度分布、H_(2)S与炉膛组分CO和SO_(2)的关系、结焦区域H_(2)S分布情况以及运行调整对H_(2)S和NO_(x)的影响情况,指导锅炉运行,在某300 MW等级电站锅炉上安装了水冷壁气氛在线监测装置,系统研究... 为揭示锅炉水冷壁H_(2)S在不同负荷时的浓度分布、H_(2)S与炉膛组分CO和SO_(2)的关系、结焦区域H_(2)S分布情况以及运行调整对H_(2)S和NO_(x)的影响情况,指导锅炉运行,在某300 MW等级电站锅炉上安装了水冷壁气氛在线监测装置,系统研究了炉内近壁面H_(2)S、CO等还原性组分的生成规律及运行调整对炉内氛围的影响。结果表明:(1)在高负荷时炉内H_(2)S、CO整体浓度较高,并且负荷越高,腐蚀倾向越严重。(2)负荷在由高向低下降的过程中,存在一临界负荷(40%BMCR),当降至此临界负荷时,壁面气氛由还原性气氛开始转化为氧化性气氛。该炉易发生高温腐蚀的负荷范围为60%~100%。(3)总体上壁面H_(2)S与CO表现出正相关性,而H_(2)S与SO_(2)呈负相关性,但同时壁面各组分间又呈现出一定的独立性。(4)结焦区域H_(2)S平均浓度比非结焦区域高出537×10^(-6)(体积分数,下同),这与结焦区域水冷壁相较于非结焦区域水冷壁腐蚀更严重的现象一致。(5)关闭燃尽风投运层数、增加运行氧量,可以显著降低H_(2)S浓度,最高降幅可达91.4%,但H_(2)S浓度降低会引起NO_(x)增加,关一层燃尽风,NO_(x)平均上涨109 mg/m^(3)。建议在实际运行中,需要平衡好这对矛盾。 展开更多

硫化氢(hydrogen sulfide, H_(2)S)是继一氧化氮和一氧化碳之后的第3种内源性气体信号分子,通过影响细胞信号通路调节机体各个系统,具有广泛的生理和病理作用。近年的研究发现,H2S的作用主要通过对靶蛋白进行S-巯基化修饰,改变其结构,... 硫化氢(hydrogen sulfide, H_(2)S)是继一氧化氮和一氧化碳之后的第3种内源性气体信号分子,通过影响细胞信号通路调节机体各个系统,具有广泛的生理和病理作用。近年的研究发现,H2S的作用主要通过对靶蛋白进行S-巯基化修饰,改变其结构,影响其活性、稳定性以及蛋白质之间互相作用,进而调控细胞内信号通路及相关生物学过程。本文基于国内外对S-巯基化修饰的研究,主要综述了:S-巯基化修饰的研究现状,详细总结了目前已揭示S-巯基化修饰及其具体半胱氨酸(cysteine, Cys)位点的研究,并进一步总结了S-巯基化修饰与S-亚硝基修饰之间的关系;S-巯基化修饰的反应类型,3种化学检测(马来酰亚胺法、改良生物素转换法和标记转换法)方法的检测原理和特点,通过S-巯基化修饰检测的条件和应用范围,将这3种方法进行对比,并对各方法在检测过程中应注意的事项进行了讨论。根据作者在S-巯基化修饰研究方面积累的经验,重点阐述马来酰亚胺法检测过程中,细胞和组织样品的制备及其较详细的实验步骤。此外,本文还讨论了当前S-巯基化修饰检测方法存在的不足及该领域未来亟待解决的问题,为S-巯基化修饰的相关研究提供参考。 展开更多

高热惊厥患儿外周血NLR和H_(2)S水平变化及与脑损伤的相关性

目的探讨高热惊厥患儿外周血中性粒细胞/淋巴细胞比值(NLR)和硫化氢(H_(2)S)水平变化及与脑损伤的相关性。方法选取2021年1月至2023年1月期间本院收治的高热惊厥患儿共220例,其中单纯性153例纳入单纯性组,复杂性67例纳入复杂性组,同期... 目的探讨高热惊厥患儿外周血中性粒细胞/淋巴细胞比值(NLR)和硫化氢(H_(2)S)水平变化及与脑损伤的相关性。方法选取2021年1月至2023年1月期间本院收治的高热惊厥患儿共220例,其中单纯性153例纳入单纯性组,复杂性67例纳入复杂性组,同期选取高热无惊厥患儿109例纳入对照组。所有患儿均检测外周血NLR和H_(2)S水平,采用受试者工作特征(ROC)曲线鉴别单纯性及复杂性高热惊厥患儿。根据高热惊厥患儿有无脑损伤发生情况分为脑损伤组及无脑损伤组,收集两组高热惊厥患儿临床资料,采用多因素Logistic回归模型分析影响高热惊厥患儿脑损伤的独立危险因素。结果与对照组比较,单纯性组及复杂性组外周血NLR水平升高,H_(2)S水平降低(P<0.05),与单纯性组比较,复杂性组外周血NLR水平升高,H_(2)S水平降低(P<0.05)。ROC分析结果显示,外周血NLR、H_(2)S单独检测及二者联合检测鉴别单纯性及复杂性高热惊厥患儿的AUC值(95%CI)分别为0.615(0.547~0.680)、0.731(0.667~0.788)、0.827(0.771~0.875),NLR、H_(2)S联合检测的预测效能高于单独检测(Z=4.488、2.161,P<0.05)。脑损伤组惊厥时体温≥40℃、复杂性高热惊厥、惊厥发作次数≥2次、惊厥持续时间≥15 min例数占比、外周血NLR水平均高于无脑损伤组,H_(2)S水平低于无脑损伤组(P<0.05)。多因素Logistic回归模型分析结果显示,复杂性高热惊厥、惊厥发作次数≥2次,惊厥持续时间≥15 min、外周血NLR高水平及H_(2)S低水平是影响高热惊厥患儿发生脑损伤的独立危险因素(P<0.05)。结论高热惊厥患儿外周血NLR水平升高,H_(2)S水平降低,外周血NLR、H_(2)S水平对于高热惊厥患儿分型具有重要的鉴别价值,且与高热惊厥患儿脑损伤的发生密切相关。 展开更多

化合物(Et4N)2[Pd2(mp)2(Hmp)2](1)(H2mp=2-巯基苯酚)的1HNMR谱中芳氢H16的峰出现在δ8.79处,与预测值(δ7.1)相差1.7,这是由于H16与配位硫S2原子之间存在弱相互作用,... 化合物(Et4N)2[Pd2(mp)2(Hmp)2](1)(H2mp=2-巯基苯酚)的1HNMR谱中芳氢H16的峰出现在δ8.79处,与预测值(δ7.1)相差1.7,这是由于H16与配位硫S2原子之间存在弱相互作用,即新型的sp2C—H…S之间的氢键作用.原子间距离C16…S2(0.361pm)和H16…S2(0.295pm)分别小于相应原子的范德华半径之和(0.370pm和0.305pm),C16—H16…S2角为128.1°,证明了这种新型氢键的存在.此外,在分子振动光谱中观察到这种氢键的作用使得芳氢νC—H出现在低于3000cm-1处.通过与类似物[Pd2(PPh3)2(Hmp)2Cl2](2)的比较,以及1在DMSO溶剂中仍然保持O—H…O和C—H…S这2种氢键的事实,表明C—H…S之间的作用,对1的配体Hmp中的苯环与Pd2S2平面之间的夹角偏离90°可能是有贡献的. 展开更多

为研究底板排气、通风及挡烟垂壁对狭长密闭空间高硫天然气泄漏控制效果,利用ANSYS Fluent 19.2软件模拟多种工况下硫化氢摩尔含量为5%的高硫天然气持续泄漏60 s后硫化氢及甲烷分布特征。分析底板排气速度、通风风速及挡烟垂壁下垂高度... 为研究底板排气、通风及挡烟垂壁对狭长密闭空间高硫天然气泄漏控制效果,利用ANSYS Fluent 19.2软件模拟多种工况下硫化氢摩尔含量为5%的高硫天然气持续泄漏60 s后硫化氢及甲烷分布特征。分析底板排气速度、通风风速及挡烟垂壁下垂高度对泄漏高硫天然气分布影响规律。结果表明:增设挡烟垂壁使泄漏高硫天然气形成涡流,挡烟垂壁下垂高度应设置为0.5 m;当底板排气速度为4.0 m/s,采用0.63 m/s通风能将泄漏高硫天然气限制在泄漏口所在挡烟垂壁区间,为最优组合;通风及底板排气系统能在高硫天然气云团表面形成空气膜,可防止高硫天然气大范围扩散。 展开更多
