On the basis of the knowledge of Erwinia amylovora's biophysical characteristics and environmental data, the geographic information system (GIS) has been applied to determine areas where Erwinia amylovora can potentially invade China. Temperature and precipitation, during the blossoming period, are considered to be two critical factors affecting the Erwinia amylovora's suitable climatic zones. This spatial modeling approach was validated from a case study in Europe, where the occurrence of Erwinia amylovora has been proven. The model prediction agreed with the occurrence of the bacteria recorded in Europe, and the same procedure has been applied to produce a potential establishment area in China's two preferential apple cultivation regions, Bohai Bay region and Huangtu Altiplano region. It has been found that areas belonging to the high-risk category are more or less the main apple producing areas, accounting for their great economic importance in China. This methodology provides an initial baseline for assessment, prevention, and management of alien species that may become invasive under certain environmental conditions. In addition, this modeling approach provides a tool for policy makers to use, in making decisions on management practices where alien species are involved.展开更多

Biochemical a...Erwinia amylovora species were isolated from the blossoms, exudates, infected fruits, leaves and bent branches of diseased apple, pear and hawthorn trees, selected in the Chy, Osh and Jalal Abad regions. Biochemical and pathogenicity tests, alongside PCR analyses, were conducted to identify the local isolates of Erwinia amylovora. The alternative antagonistic microorganisms which combat bacterium E. amylovora were tested within in vitro and in vivo conditions. The results revealed the ability of Streptomyces antagonistic bacteria to decrease fire blight severity on pear and apple trees during the first stage of the fire blight disease in leaf tissues. Streptomyces strain C1-4 suppressed E. amylovora disease symptoms in the leaf tissues and excised apple and pear shoots. The incidence of fire blight on leaves was reduced by about 70% with two applications of bacterial antagonists. Further studies at different locations in Kyrgyzstan, using large scale application, would allow for stronger recommendations to be made, including studies and recommendations on their ability to prevent disease and to use them as main components in an integrated pest management program.展开更多

Lanthanum inhibited the growth of Ech in solid culture at the concentration of 50, 100, 150, 200, 250, 300 and 350 mgL-1, and the inhibition increased with the increase of the RE concentration. The formation of colony delayed and the diameter of colony decreased. Lanthanum stimulated the growth of Ech within 24h when the lanthanum concentration was less than 200 mgL-1 in liquid medium, but significantly inhibited the growth of Ech with the increase of lanthanum concentration and culture time. When the concentration was higher than 350 mgL-1, the growth of Ech was inhibited completely both in solid and liquid culture. The extracellular enzyme acitivity of Ech was affected by lanthanum at 200 mgL-1. The activity of cellulose was promoted more than that of protease but the activity of pectinase was decreased. Its viscosity behavior was investigated by a low-shear-rate multiball viscometer and a rotational viscometer. Its weight-average molecular weight M-w and intrinsic viscosity [eta] in 0.2 mol/L NaCl aqueous solution were measured by light scattering method at 35 degrees C and viscometry at 25 degrees C and found to be 1.06 x 10(6) g/mol and 1050 mL/g, respectively, and its aggregates in aqueous solution were proved by gel permeation chromatography (GPC). These results indicated that E gum in water has exceedingly high viscosity and exhibits Binham fluid behavior, owing to its aggregation. The viscosity of E gum decreased with increasing temperature, and the turning point appeared at 38 degrees C for dilute solution and 80 degrees C for concentrated solution suggesting that the aggregates of E gum in water started to disaggregate under these temperatures. In addition, the aggregates can be disrupted by adding either acid or base. The experimental results indicated that the E gum is a good thickening agent, and its fluid behavior is similar to xanthan.展开更多

The experimental results prove that E gum formed aggregates in the aqueous solution at 25 ℃ and the aggregates were broken gradually with increasing temperature. The dissociation of the aggregates of E gum in the aqueous solution started at 36 ℃, and was completed at around 90 ℃. The [η] values of E gum and its fractions are much higher than those of the conventional polymers with the similar molecular weights, and decrease with increasing NaCl concentration.展开更多

Isolates were identified as Erwinia carotovora ssp. carotovora (Ecc) by standard bacteriological techniques and pathogenicity tests on tubers and also by PCR analyses. Tests on the pathogenicity of E. carotovora ssp. carotovora (Ecc) strains to host plants by artificial inoculation have shown a high sensibility of the Picasso variety. As a result, five isolates were chosen, three isolates (EcPo1, EcPo2, and Eco3) were highly pathogenic, while two isolates (Eco4 and Eco5) were weakly pathogenic. The antagonistic bacteria, Streptomyces diastatochromogenes strain sk-6, and Streptomyces graminearuss strain sk-2, have a highly significant effect on soft rot bacteria isolates (Ecc), more than the other tested antagonistic organisms in vitro screening biotests. The Streptomyces diastatochromogenes sk-6 was selected for the control assay of storage potatoes against the most common soft rot bacterial strain in Kyrgyzstan, Erwinia carotovora sp. carotovora EcPo2. The pretreatment of potato tubers with antagonistic bacteria successfully prevented the initial infection multiplication of soft rot bacteria and reduced soft rot disease of potatoes in storage. These results justify selection of the dose 10<sup>6</sup> cells/ml of bacteria Streptomyces diastatochromogenes sk-6 for use in powdering the infected or non-infected potato tubers to suppress the development soft rot during storage. Streptomyces diastatochromogenes sk-6 as a biological disinfectant could destroy surface and internal infections, protect the tubers from the growth of phytopathogenic bacteria in the early period of their reproduction, and improve the overwintering of winter crops.展开更多

Lignin biosynthesis has been implicated in defensive reactions to injury and pathogen infection in plants. In this work, variations of lignin content and gene expression in the molecular interaction between Chinese cabbage and Ecc were investigated. H2O2 accumulation and peroxidase activity were detected by 3, 3'- Dimethoxybenzidine staining at mocked and Ecc-inoculated sites of Chinese cabbage leafstalks. Klason lignin content in inoculated plants increased by about 7.84%, 40.37%, and 43.13% more than that of the mocked site at 12, 24 and 72 h after inoculation, respectively. Gas chromatography detected more p-coumaryl (H) and less coniferyl (G) and sinapyl (S) monolignins in leafstalks of Chinese cabbage. All three monomers increased in Ecc-infected leafstalks, and the Ecc-induced "defense lignin" were composed of more G and H monolignins, and less S monolignin. After searching the expressed sequence tags (EST) data of Chinese cabbage, 12 genes putatively encoding enzymes involved in lignin biosynthesis were selected to study their expression. All of these genes could be induced by mock inoculation and Ecc infection, while the gene expression lasted for several more hours in the infected samples than in mocked and untreated plants. Our results indicated that "defense lignin" was different from the developmental lignin in composition; G and S monolignins were significantly induced in plants in response to the soft rot Ecc; thus, lignin biosynthesis was differentially regulated and played a role in plant response to the soft rot Ecc.展开更多
