Background Reducing Salmonella infection in broiler chickens by using effective and safe alternatives to antibiotics is vital to provide safer poultry meat and minimize the emergence of drug-resistant Salmonella and the spread of salmonellosis to humans. This study was to first evaluate the protective efficacy of feeding coated essential oils and organic acids mixture(EOA)on broiler chickens infected with Salmonella Enteritidis(S.Enteritidis,SE),and then its action mechanism was further explored. Methods A total of 4801-day-old Arbor Acres male chickens were randomly assigned into five treatments with six replicates, including non-challenged control fed with basal diet(A),SE-challenged control(B),and SE-infected birds fed a basal diet with 300 mg/kg of EOA(BL),500 mg/kg of EOA(BM)and 800 mg/kg of EOA(BH),respectively. All birds on challenged groups were infected with Salmonella Enteritidis on d 13.Results Feeding EOA showed a reversed ability on negative effects caused by SE infection,as evidenced by decreasing the feed conversion rate(FCR)and the ratio of villus height to crypt depth(VH/CD)(P<0.05),obviously decreasing intestinal and internal organs Salmonella load along with increasing cecal butyric acid-producing bacteria abundance(P<0.05).Moreover,supplemental different levels of EOA notably up-regulated claudin-1(CLDN-1),occludin(OCLN),zonula occludens-1(ZO-1),mucin-2(MUC-2),fatty acid binding protein-2(FABP-2),nuclear factor kappa-light-chainenhancer of activated B cells(NF-κB),myeloid differential protein-88(MyD88)and interleukin-6(IL-6)mRNA levels in the ileum of the infected chickens after challenge, whereas down-regulated toll-like receptor-4(TLR-4)mRNA levels(P<0.05).Linear discriminant analysis combined effect size measurements analysis(LEfSe)showed that the relative abundance of g_Butyricicoccus,g_Anaerotruncus and g_unclassified_f_Bacillaceae significantly was enriched in infected birds given EOA.Also, phylogenetic investigation of communities by reconstruction of unobserved states(PICRUSt)analysis showed that alpha-linolenic acid metabolism, fatty acid metabolism and biosynthesis of unsaturated fatty acids were significantly enriched in the EOA group. Conclusion Our data suggest that the essential oils and organic acids mixture can be used as an effective strategy to ameliorate and alleviate Salmonella Enteritidis infection in broilers. 展开更多

Background:The poultry industry is in need of effective antibiotic alternatives to control outbreaks of necrotic enteritis(NE)due to Clostridium perfringens. In the present study,we investigated the effects of dietary supplementation with a blend of encapsulated essential oils and organic acids(BLJ)on growth performance and gut health using a coinfection model of NE in broiler chickens. Methods:Two hundred and eighty-eight one-day-old male Arbor Acres broiler chicks were randomly assigned using a 2×2 factorial design into two groups fed either 0 or 500 mg/kg dietary BLJ and co-challenged(or not challenged for the control)with Eimeria spp./C.perfringens.Results:Infected birds fed the BLJ-supplemented diet exhibited an improved feed conversion ratio throughout the trial(P<0.01),a higher villus height and villus height/crypt depth ratio, and reduced intestinal C.perfringens counts, liver C.perfringens carriage, gut lesion scores and serum fluorescein isothiocyanate dextran(FITC-D)concentrations at 7 d postinfection compared with those of birds without BLJ supplementation(P<0.05).NE-infected birds fed BLJ exhibited significantly upregulated claudin-1 and IGF-2 mRNA levels(P<0.05),increased A20 mRNA expression and significantly downregulated TRAF-6,TNFSF15 and TOLLIP mRNA levels in the jejunum at 7 d post-infection compared with those in birds without BLJ supplementation(P<0.05).Compared with the uninfected and untreated birds, the uninfected birds fed BLJ displayed increased relative abundances of Lactobacillus and Coprococcus but reduced Rikenellaceae levels.Compared with the unsupplemented NE-challenged birds, infected birds fed BLJ showed an increased relative abundance of Unclassified_Lachnospiraceae and a significantly decreased relative abundance of Erysipelotrichaceae. Conclusion:BLJ supplementation improved growth performance and gut health in NE-infected broiler chickens by strengthening the intestinal barrier function, positively modulating the gut microbiota community and differentially regulating intestinal immune responses. Our results also suggested that adding BLJ effectively controlled NE infections after experimental Eimeria and Clostridium perfringens coinfection. 展开更多

With 5 rice varieties( Yifeng 3321,Yifeng 3315,Yifeng 3317,Yifeng 45,Yifeng 14-24) as the experimental objects, the aseptic seedling method was used to culture the rice seeds, and the HPLC was used to determine the content of amino acids in aseptic rice seedlings as well as the total content of 8 kinds of essential amino acids and the content of protein. And the content of protein and amino acids in the polished rice of 5 varieties was detected, and the analysis of variance was performed for screening of the varieties containing high essential amino acids. The rice with high essential amino acids could be produced to meet people's rice quality requirements. The analysis of variance indicated that there were significant differences( F = 39. 262,p = 0. 01) in the content of essential amino acids among 5 kinds of rice varieties. By the Duncan's multiple comparison, the differences among rice varieties were calculated to obtain screening results. The aseptic seedlings:Yifeng 3321(7. 268),Yifeng 3315(6. 395),Yifeng 3317(5. 698),Yifeng 45(5. 567),Yifeng 14-24(5. 190). Rice: Yifeng 3321(2.998),Yifeng 14-24(2. 627),Yifeng 3317(2. 611),Yifeng 3315(2. 503),Yifeng 45(2. 281). Based on the estimated human daily requirement of essential amino acids and amino acids requirement pattern,it was found that the polished rice of 5 varieties was up to standard. The variety with the highest content of essential amino acids was Yifeng 3317(3830 mg/100g),followed by Yifeng 3321(3770 mg/100 g).The results showed that the high quality rice variety in this experiment was Yifeng 3321. 展开更多

Dietary lipid supplementation benefits the prolific and high-producing modern lactating sow. A comprehensive review of recent studies showed that lipid supplementation increases average daily energy intake, which is partitioned for lactation as indicated by greater milk fat output and improved litter growth rate. Recent compelling findings showed that addition of particular lipids during lactation improved the subsequent reproductive outcome of sows. Such benefits were related to the level of dietary essential fatty acids(EFA, linoleic acid, C18:2n-6; and α-linolenic acid, C18:3n-3) during lactation. Lactation diets without supplemental EFA resulted in a pronounced negative balance(intake minus milk output) of linoleic(-25.49 g/d) and α-linolenic acid(-2.75 g/d); which compromised sow fertility(farrowing rate 〈 75 % and culling rates 〉 25 % of weaned sows). This phenomenon seems to be increasingly important with advancing sow age because of a progressive reduction of body EFA pool over successive lactations. The net effect of supplemental EFA during lactation was to create a positive EFA balance, which improved the subsequent reproduction of sows. Adequate linoleic acid intake improved the proportion of sows that farrowed in the subsequent cycle(Farrowing rate(%) = [(-1.5 × 10^-3× linoleic acid intake(g/d)^2) +(0.53 × linoleic acid intake(g/d)) +(45.2)]; quadratic P = 0.002, R^2= 0.997, RMSE = 0.031). In addition, increasing linoleic acid intake increased the number of pigs born in the subsequent cycle(total pigs born(n) = [(9.4 × 10^-5× linoleic acid intake(g/d)^2) +(0.04 × linoleic acid intake(g/d)) +(10.94)]; quadratic P = 0.002, R^2= 0.997, RMSE = 0.031). Supplemental α-linolenic acid resulted in a rapid return to estrus(sows bred: sows weaned = 94.2 %; wean-to-estrus interval = 4.0 d) and achieved a high retention of pregnancy(sows pregnant: sows bred = 98 %). Collectively, we conclude that a minimum dietary intake of10 g/d of α-linolenic acid, simultaneous with a minimum of 125 g/d of linoleic acid should be provided to ≥ 95 % of the sows; thereby, achieving a maximum sow reproductive efficiency through multiple mechanisms that include rapid return to estrus, high maintenance of pregnancy and large subsequent litter size in mature sows, that appear to be susceptible to EFA deficiency. 展开更多

Both Omega-6 and Omega-3 fatty acids are essential nutrients important for cellular structure and function but cannot be synthesized by the human body and must be obtained from food. These two classes of fatty acids are metabolically and functionally different and antagonistically regulate many physiological and pathological processes. Thus, the relative balance between Omega-6 and Omega-3 fatty acids are critical for good health. However, today’s diet contains too much Omega-6 but too little Omega-3,leading to a severe imbalance with a very high Omega-6/Omega-3 ratio(>10)in most people. Many lines of evidence suggest that this imbalance is a key factor contributing to the development of modern chronic diseases.A growing number of studies ranging from laboratory research to clinical trials have shown that balancing the tissue ratio of Omega-6/Omega-3 fatty acids by increasing tissue levels of Omega-3 fatty acids and/or decreasing the content of Omega-6 fatty acids are very beneficial for the prevention and treatment of many life-threating chronic diseases as well as for health promotion of the general public. Therefore, development and production of foods rich in Omega-3-fatty acids should be considered as a key health program to balance essential fat intake and thereby improve the nutritional and health status of all people. 展开更多

Tumor cells have an increased demand for glucose and amino acids to support their rapid growth, and also exhibit alterations in biochemical pathways that metabolize these nutrients.Transport across the plasma membrane is essential to feed glucose and amino acids into these tumor cell-selective metabolic pathways.Transfer of amino acids across biological membranes occurs via a multitude of transporters;tumor cells must upregulate one or more of these transporters to satisfy their increased demand for amino acids.Among the amino acid transporters,SLC6A14 stands out with specific functional features uniquely suited for the biological needs of the tumor cells. This transporter is indeed upregulated in tumors of epithelial origin, including colon cancer, cervical cancer, breast cancer, and pancreatic cancer.Since normal cells express this transporter only at low levels, blockade of this transporter should lead to amino acid starvation selectively in tumor cells, thus having little effect on normal cells. This offers a novel, yet logical, strategy for the treatment of cancers that are associated with upregulation of SLC6A14.In addition,a variety of amino acid-based prodrugs are recognized as substrates by SLC6A14,thus raising the possibility that anticancer drugs can be delivered into tumor cells selectively via this transporter in the form of amino acid prodrugs. This strategy allows exposure of SLC6A14-positive tumor cells to chemotherapy with minimal off-target effects. In conclusion, the amino acid transporter SLC6A14 holds great potential not only as a direct drug target for cancer therapy but also for tumor cell-selective delivery of anticancer drugs. 展开更多

The transition period of dairy cattle is characterized by a number of metabolic, endocrine, physiologic, and immune adaptations, including the occurrence of negative energy balance, hypocalcemia, liver dysfunction, overt systemic inflammatory response, and oxidative stress status. The degree and length of time during which these systems remain out of balance could render cows more susceptible to disease, poor reproductive outcomes, and less efficient for milk production and quality. Studies on both monogastrics and ruminants have reported the health benefits of nutraceuticals(e.g. probiotics, prebiotics, dietary lipids, functional peptides, phytoextracts) beyond nutritional value, interacting at different levels of the animal's physiology. From a physiological standpoint, it seems unrealistic to disregard any systemic inflammatory processes. However, an alternate approach is to modulate the inflammatory process per se and to resolve the systemic response as quickly as possible. To this aim, a growing body of literature underscores the efficacy of nutraceuticals(active compounds) during the critical phase of the transition period. Supplementation of essential fatty acids throughout a 2-month period(i.e. a month before and a month after calving) successfully attenuates the inflammatory status with a quicker resolution of phenomenon. In this context, the inflammatory and immune response scenario has been recognized to be targeted by the beneficial effect of methyl donors, such as methionine and choline, directly and indirectly modulating such response with the increase of antioxidants GSH and taurine. Indirectly by the establishment of a healthy gastrointestinal tract, yeast and yeast-based products showed to modulate the immune response, mitigating negative effects associated with parturition stress and consequent disorders. The use of phytoproducts has garnered high interest because of their wide range of actions on multiple tissue targets encompassing a series of antimicrobial, antiviral, antioxidant, immune-stimulating, rumen fermentation, and microbial modulation effects. In this review, we provide perspectives on investigations of regulating the immune responses and metabolism using several nutraceuticals in the periparturient cow. 展开更多

To investigate the effect of low light (LL, 50% natural light) during grain filling (GF) stage on rice transamination, amino acid (AA) accumulation, nutritional value, and cooking quality in three different rice genotypes, transaminase activities and AA levels in grains during GF stage and the traits that significantly affected rice quality (physical appearance, cooking quality, and nutritional value) were analyzed. LL did not disturb transamination in rice grains during GF stage, as minimal impact was found on alanine and aspartate transaminase activities. Nevertheless, most AAs in caryopses, including lysine and threonine, increased in response to LL, except for sulfur-containing AAs. These results suggest that AA metabolism and accumulation in rice grains were rarely suppressed by LL during GF stage. Rice nutritional ingredients at harvest, such as major protein components including glutelin and most important essential amino acids (EAAs) including lysine and threonine, increased significantly in response to LL, whereas most protein and EAA ratios were rarely affected. However, LL markedly affected physical appearance of rice grains by reducing brown rice rate, milled rice rate, and 1000-grain weight and increasing the chalkiness rate. In addition, cooking qualities decreased in response to LL, while breakdown values and amylose levels decreased and setback values increased. We concluded that LL during GF stage decreased the cooking quality of rice, but could potentially improve the nutritional value of rice. 展开更多

Modern strains of broiler chickens are selected for fast growth and are marketed anywhere from 36 to 49 days after a21-day incubational period. For a viable healthy chick, all the necessary nutrients required for growth and development must be provided by the hen through the fertilized egg. The current feeding strategies for improved growth, health and productivity are targeted towards chicks after hatching. Considering the fact that developing chick embryo spends over 30 % of its total life span inside the hatching egg relying on nutrients deposited by the breeder hen, investigations on nutritional needs during pre-hatch period will improve embryonic health, hatchability and chick viability. In this context, investigations on hatching egg lipid quality is of utmost importance because, during incubation, egg fat is the major source of energy and sole source of essential omega-6(n-6) and omega-3(n-3) fatty acids to the chick embryo.Due to the unique roles of n-3 and n-6 fatty acids in growth, immune health, and development of central nervous system, this review will focus on the role of early exposure to essential fatty acids through maternal diet and hatching egg and its impact on progeny in meat-type broiler chickens. 展开更多

The macronutrient composition of food supply in China has altered dramatically in the past 70 years.Fat(oil)has increased more than 4.2-times while the carbohydrate content has declined by 34%.Vegetable oils are the major component of the fat intake and since these oils are rich in linoleic acid, there has been a significant rise in the consumption of this fatty acid(as much as a 4-fold rise).Linoleic acid has essential functions in the body in skin and as a precursor of prostaglandins and related compounds. The current intakes of linoleic acid are well in excess of the minimum requirements. In this review, the effects of a food supply rich in linoleic acid on pain in arthritis and headache,non-alcoholic fatty liver and neural function are explored, with emphasis on lipid mediators derived from linoleic acid and other long chain polyunsaturated fatty acids. The current world food systems have created an imbalance of dietary linoleic acid in relation to n-3 polyunsaturated fatty acids, and an imbalance in the lipid mediators derived from these polyunsaturated fatty acids which may be contributing to sub-optimal health status. 展开更多

In some species of growing mammals glutamine is an essential amino acid that,if inadequate in the diet,is needed for normal growth and development. It is thus sometimes considered to be a conditionally essential amino acid in some species.A review of studies that have measured L-glutamine concentrations([glutamine])in horses demonstrates that plasma[glutamine]has routinely been reported to be much lower(~330μmol/L)than in other mammals(>600μmol/L).Plasma[glutamine]represents the balance between intestinal transport into the blood after hepatic first pass, tissue synthesis and cellular extraction. The hypothesis is proposed that sustained low plasma[glutamine]represents a chronic state of sub-optimal glutamine intake and glutamine synthesis that does not meet the requirements for optimum health. While this may be without serious consequence in feral and sedentary horses, there is evidence that provision of supplemental dietary glutamine ameliorates a number of health consequences, particularly in horses with elevated metabolic demands. The present review provides evidence that glutamine is very important(and perhaps essential)for intestinal epithelial cells in mammals including horses, that horses with low plasma[glutamine]represents a sub-optimal state of well-being,and that horses supplemented with glutamine exhibit physiological and health benefits. 展开更多

Colibacillosis caused by avian pathogenic Escherichia coli(APEC)is a very prevalent disease in poultry farms in China. The exploration of effective non-antibiotic substances is of great significance for the control of APEC infections. This experiment evaluated the efficacy of coated essential oil and organic acid(EOA)supplementation to prevent E.coli O78 infection in broiler chickens.A total of 288 one-day-old male broiler chicks were randomly distributed into 4 groups with 6 replicates per group.Chickens were fed a diet either supplemented with EOA(500 mg/kg feed)or not,and either uninfected or infected with E.coli O78 intratracheally. Results showed that E.coli O78 infection reduced body weight gain, increased mortality and the ratio of feed to gain along with cecal and liver E.coli load, damaged gut mucosa, induced local and systemic inflammation, and altered cecal microbial composition, diversity and function(P<0.05).Supplemental EOA improved feed conversion efficiency, lowered gross lesion scores and cecal E.coli population, enhanced intestinal goblet cells and serum IgG concentration, and tended to decrease serum IL-12 production(P<0.05).Essential oil and organic acid addition downregulated IFN-γmRNA,tended to decrease mucin-2 mRNA levels while upregulating IL-10 mRNA,and tended to increase ZO-1 gene expression in the jejuna of infected birds at 7 d after E.coli O78 challenge(P<0.05).The 16S rRNA gene sequencing indicated that both EOA addition and E.coli O78 challenge altered the diversity and composition of the cecal microbiota community.Furthermore, infected birds fed EOA showed decreased Bacteroidetes and genus Lactobacillus abundance compared with the infected control.LEfSe analysis showed that Firmicutes,Ruminococcaceae,Clostridiales,Clostridia,Lactobacillus,Lactobacilaceae, and cc-115 were enriched in the non-infected but EOA-treated group(P<0.05).Collectively,dietary EOA supplementation could mildly alleviate E.coli-induced gut injury and inflammation. 展开更多

Artemia is deficient in polyunsaturated fatty acids(PUFAs),particularly in arachidonic(ARA,20:4n-6),eicosapentaenoic(EPA,20:5n-3),and docosahexaenoic acid(DHA,22:6n-3).The aim of this study was to determine the optimal time in which the higher contents of PUFAs in juveniles of Artemia franciscana were obtained by the effect of enrichment with the tuna orbital oil emulsion.Six enrichment periods were evaluated:3,6,9,12,15 and 18 h,in addition to a control treatment(0 h).The most abundant fatty acids in A.franciscana were monounsaturated(43.10%±4.35–52.92%±5.82%),followed by saturated(33.83%±1.71–42.33%±2.31%)and PUFAs(8.86%±2.83%–21.32%±2.38%).ARA decreased over the enrichment time;the maximum content was 5.74±0.37%at 3 h,which was not statistically different with respect to the content recorded at 0 h.The highest content of EPA was at 3 h(6.47%±1.44%),without significant differences with the content registered at 0 h,while that from 6 h and until 15 h tended to decrease significantly. At 6 h,the content of DHA(8.84%±2.72%)was significantly higher compared to the rest of the treatments, which did not differ among themselves,or with the control. After to the 6 h and until 15 h,the content of PUFAs tended to decrease, which could indicate the metabolization of them by A.franciscana, coupled with the possible oxidation of these fatty acids in the enrichment solution. 展开更多

Rosa roxburghii Tratt., is an important commercial horticultural crop in China due to its nutritional and medicinal value. To date, the economic production of a single predominant cultivar of R. roxburghii(‘Guinong 5') consumes 30 000 hm^2 in China. Here, we characterize the amino acid composition of ‘Guinong 5' and two novel genotypes(AL11, GP6) that prove valuable and deserve wide application. Here, 17 amino acids(acid hydrolysis of proteins) were detected in ‘Guinong 5', AL11 and GP6 fruits, with Glutamine and Asparagine being abundant, and L-cystine being absent. The essential amino acids and total amino acids were the highest in AL11, while Asparagine was most abundant in GP6. For the free amino acid, 18 amino acids and 9 metabolites were detected in ‘Guinong 5', AL11 and GP6 fruits. γ-aminobutyric acid, L-arginine and ornithine were the most abundant in ‘Guinong 5', while phosphoserine and taurine were the highest in AL11. In comparison, GP6 had the highest levels of L-asparagine, an essential amino acid, and the highest total N containing compounds. 展开更多

Emerging aquatic insects, including mosquitoes, are known to transfer to terrestrial ecosystems specific essential biochemicals, such as polyunsaturated fatty acids (PUFA). We studied fatty acid (FA) composition and contents of dominant mosquito populations (Diptera: Culicidae), that is, Anopheles messeae, Ochlerotatus caspius, Oc. flavescens, Oc. euedes, Oc. subdiversus, Oc. cataphylla, and Aedes cinereus, inhabited a steppe wetland of a temperate climate zone to fill up the gap in their lipid knowledge. The polar lipid and triacylglycerol fractions of larvae and adults were compared. In most studied mosquito species, we first found and identified a number of short-chain PUFA, for example, prominent 14:2n-6 and 14:3n-3, which were not earlier documented in living organisms. These PUFA, although occurred in low levels in adult mosquitoes, can be potentially used as markers of mosquito biomass in terrestrial food webs. We hypothesize that these acids might be synthesized (or retroconverted) by the mosquitoes. Using FA trophic markers accumulated in triacylglycerols, trophic relations of the mosquitoes were accessed. The larval diet comprised green algae, cryptophytes, and dinoflagellates and provided the mosquitoes with essential n-3 PUFA, linolenic, and eicosapentaenoic acids. As a result, both larvae and adults of the studied mosquitoes had comparatively high content of the essential PUFA. Comparison of FA proportions in polar lipids versus storage lipids shown that during mosquito metamorphosis transfer of essential eicosapentaenoic and arachidonic acids from the reserve in storage lipids of larvae to functional polar lipids in adults occurred. 展开更多
