A procedure to evaluate the quality consistency of generic drugs based on the impurity profile and the similarity analysis methods was presented in this paper. Nifedipine extended-release tablets from six generic fact...A procedure to evaluate the quality consistency of generic drugs based on the impurity profile and the similarity analysis methods was presented in this paper. Nifedipine extended-release tablets from six generic factories of China were used to evaluate the uniformity with the original drug in the study. The procedure includes: choice of chromatographic methods, data collection and conformity test, evaluation of intra-batch similarity of drugs, evaluation of generic drugs with the original drug and weighted similarity evaluation of generic drugs. The data were collected via high-performance liquid chromatography (HPLC), and then calculated by correlation coefficient, cosine, principal component analysis (PCA) and hierarchical clustering analysis (HCA). It is more suitable to use peak areas as the vector when calculating the similarity of impurity profile. After weighting the peak areas of the unspecified impurities in further evaluation of the generic quality, the generic level of different factories was differentiated and the best generic factory was picked out.展开更多
A tablet matrix system was developed for ibuprofen and the influence of the polymer blend and concentration on the release rate of the drug was evaluated.Tablets containing different concentrations of calcium silicate...A tablet matrix system was developed for ibuprofen and the influence of the polymer blend and concentration on the release rate of the drug was evaluated.Tablets containing different concentrations of calcium silicate and PVP(polyvinyl pyrrolidone)were prepared using direct compression and the weight uniformity,crushing strength,friability,drug content uniformity,dissolution profile,and in vitro release kinetics were examined.Formulated tablets were found to be within the official acceptable limits of physical and chemical parameters except for the thickness test that was below the conformation of extended-release tablets.The crushing strength of the tablets was in the range of 2.5 to 5.6 kg/f,the weight variations of the tablets of all the formulation was less than±5%.The friability of all the formulations was in the range of 0.6%to 1.83%.Tablet thickness and diameter was in range of 3.18 mm to 4.48 mm and 12.53 mm to 12.64 mm respectively.Absolute drug contents of all the formulations were found to be in range of 83.50%to 98%.The release kinetic of F3 containing 20 mg of calcium silicate,40 mg of PVP as matrix formers showed the best linearity(r^2=0.6975)with%drug release of 96 showing that combination of the two polymers(20 mg calcium silicate and 40 mg PVP)for use as a matrix former is best for extended-release formulation of ibuprofen.展开更多

According to the property of the moment generating function, the distribution characteristics...A multichannel noncoherent integration detection method based on high range resolution profile was presented in this paper. According to the property of the moment generating function, the distribution characteristics of the noncoherent integrated signals with or without target presence were derived under the circumstance with noncorrelated Gaussian distribution noises. The loss of noncoherent integration was due to improper selection of integration range of cell numbers. A multi channel noncoherent integration detection scheme where the integration number in each channel va ries was proposed to solve this problem. The quality of this method for detection of various targets was evaluated. A comparison of fixed integration range cell number detection and multichannel inte gration detection for a high range resolution profile was presented. Simulation results indicated that the principle of the method was correct and performed well for unknown physical dimension targets. The method required little prior knowledge about target and was convenient for practical implementa tion.展开更多
