Objective: To evaluate the in vitro and in vivo inhibitory effects of two commonly used herbs, Aframomum melengueta(A. melengueta) and Dennettia tripetala(D. tripetala) on CYP 3A enzymes. Methods: In vitro inhibition ...Objective: To evaluate the in vitro and in vivo inhibitory effects of two commonly used herbs, Aframomum melengueta(A. melengueta) and Dennettia tripetala(D. tripetala) on CYP 3A enzymes. Methods: In vitro inhibition of the enzymes were assessed with microsomes extracted from female albino rats using erythromycin-N-demethylation assay(EMND) method while their in vivo effects were measured by estimating simvastatin plasma concentrations in rats. Pharmacokinetic parameters were determined using non-compartmental anaysis as implemented in Win Nonlin pharmacokinetic program. Results: EMND assay with intestinal microsomes indicated that aqueous extracts of D. tripetala and A. melengueta significantly(P < 0.05) inhibited intestinal CYP 3A activity at both 50 μg and 100 μg concentrations. Petroleum ether extract of D. tripetala and ethanol extracts of A. melengueta inhibited intestinal CYP3 A activity at 100 μg but not at 50 μg concentrations. All the extracts showed an in vitrodose dependent CYP 3A inhibition with liver microsomes. In vivo analysis showed that pretreatment with the extracts enhanced systemic absorption of simvastatin with reductions in metabolizing enzymes activity as indicated in significant increases in maximal concentration, area under curve, area under moment curve and mean resident time of simvastatin(P < 0.05). Conclusions: Herbal preparations containing these plants' extracts should be used with caution especially in patients on CYP450 3A substrate medications.展开更多

The process coupled the extraction chromatography method to obtain the decontamination of Pd(II) from HLLW with the electrochemical method to recover metallic palladium from the concentrated solution.Separation of Pd(II) from a nitric acid medium by extraction chromatography using iso Bu-BTP/SiO_2-P adsorbent and the electrochemical behavior of Pd(II) in nitric acid solution in the presence of thiourea(TU) were investigated.iso Bu-BTP/SiO_2-P exhibited a high selectivity for Pd(II)over other fission products(FPs), and Pd(II) could be desorbed by TU from loaded BTP/SiO_2-P. The adsorbent performed good stability against HNO_3 because the adsorption performance kept Pd(II) after extended contact with HNO_3 solution. The column experiment achieved the separation of Pd(II) from simulated HLLW successfully.The electrochemical behavior of Pd(II) in palladium desorption solution containing TU and nitric acid was investigated at a platinum electrode by cyclic voltammetry. The effective separation factors c...Solvent extraction experiments of La(III) with di-(2-ethylhexyl)phosphoric acid (P204) from chloride solution in the presence of a complexing agent (lactic acid) have been performed. The effective separation factors can be achieved when the complexing agent is added to the aqueous phase of the extraction system. The complexing agent lactic acid can be effectively recycled using tributyl phosphate (TBP) as extractant, by the use of a countercurrent extraction process, and the chemical oxygen demand (COD) value in the raffinate is 57.7 mg/L, which meets the emission standards of pollutants from rare earths industry. Thus, the simple and environment-friendly complexing method has been proved to be an effective strategy for separating light rare earths, and provides a positive influence on the purification of La(III).展开更多

Within the experimental contact time range of 0.5–24 h, iso Hex-BTP/Si O2-P showed high selectivity for241Am(Ⅲ) compared to152Eu(Ⅲ) in3 mol/dm3 nitric acid solution. However, the adsorption kinetics of241Am(Ⅲ) and152Eu(Ⅲ) was slow. Eu(Ⅲ)adsorption followed the pseudo-second-order kinetic model, indicating chemical adsorption as the rate-limiting step of the adsorption process. And the adsorption agreed well with the Langmuir adsorption model at various temperatures. The adsorption kinetics and isotherm data indicated that the equilibrium adsorption capacity, the adsorption rate, the maximum adsorption capacity and the adsorption affinity, increased with temperature. The thermodynamic parameters, negative change in Gibbs free energy, and positive change in enthalpy and entropy,suggested that the adsorption of Eu(Ⅲ) was spontaneous and endothermic process with an increase of entropy.展开更多
