BACKGROUND MicroRNAs(miRNAs)regulate gene expression and play a critical role in cancer physiology.However,there is still a limited understanding of the function and regulatory mechanism of miRNAs in gastric cancer(GC...BACKGROUND MicroRNAs(miRNAs)regulate gene expression and play a critical role in cancer physiology.However,there is still a limited understanding of the function and regulatory mechanism of miRNAs in gastric cancer(GC).AIM To investigate the role and molecular mechanism of miRNA-145-5p(miR145-5p)in the progression of GC.METHODS Real-time polymerase chain reaction(RT-PCR)was used to detect miRNA expression in human GC tissues and cells.The ability of cancer cells to migrate and invade was assessed using wound-healing and transwell assays,respectively.Cell proliferation was measured using cell counting kit-8 and colony formation assays,and apoptosis was evaluated using flow cytometry.Expression of the epithelial-mesenchymal transition(EMT)-associated protein was determined by Western blot.Targets of miR-145-5p were predicated using bioinformatics analysis and verified using a dual-luciferase reporter system.Serpin family E member 1(SERPINE1)expression in GC tissues and cells was evaluated using RT-PCR and immunohistochemical staining.The correlation between SERPINE1 expression and overall patient survival was determined using Kaplan-Meier plot analysis.The association between SERPINE1 and GC progression was also tested.A rescue experiment of SERPINE1 overexpression was conducted to verify the relationship between this protein and miR-145-5p.The mechanism by which miR-145-5p influences GC progression was further explored by assessing tumor formation in nude mice.RESULTS GC tissues and cells had reduced miR-145-5p expression and SERPINE1 was identified as a direct target of this miRNA.Overexpression of miR-145-5p was associated with decreased GC cell proliferation,invasion,migration,and EMT,and these effects were reversed by forcing SERPINE1 expression.Kaplan-Meier plot analysis revealed that patients with higher SERPINE1 expression had a shorter survival rate than those with lower SERPINE1 expression.Nude mouse tumorigenesis experiments confirmed that miR-145-5p targets SERPINE1 to regulate extracellular signal-regulated kinase-1/2(ERK1/2).CONCLUSION This study found that miR-145-5p inhibits tumor progression and is expressed in lower amounts in patients with GC.MiR-145-5p was found to affect GC cell proliferation,migration,and invasion by negatively regulating SERPINE1 levels and controlling the ERK1/2 pathway.展开更多

It considers family as partners and collaborators in care of children. It aims at involving family in all aspects of child care. Family centered care also mentions involvement of child. However, emphasis is given more on family than child and does not take into account the older child’s capacity for independent decision making and right to privacy. As such, child’s needs are missed out. With child centered care, children are involved and supported at all levels of care based on their age and developmental stage. This paper aims to stress the importance of involving children within family centered care. Involving children in their care, makes them feel less threatened by the health care professional and their self esteem is promoted. Currently, no studies have been identified in Malawi that demonstrates full partnership between the family, child and the nurse. Furthermore, Family Centered Care and Child Centered Care as models are not fully practiced. It is therefore important to practice both family and child centered care in child health care if the needs of both family and children are to be addressed concurrently.展开更多
MicroRNAs(miRNAs)have been demonstrated to control chicken skeletal muscle growth,however,the potential function of the miR-181-5p family in chicken myogenesis remains largely unknown.Here,our study identified the two...MicroRNAs(miRNAs)have been demonstrated to control chicken skeletal muscle growth,however,the potential function of the miR-181-5p family in chicken myogenesis remains largely unknown.Here,our study identified the two chicken(Gallus gallus;Gga)miR-181-5p family members widely expressed in various tissues,specifically miR-181a-5p and miR-181b-5p.Besides,the breast muscles of fast-growing broilers expressed higher levels of miR-181a-5p and miR-181b-5p than those of slow-growing layers.Functionally,miR-181a-5p and miR-181b-5p both promote the expression level of myogenic factors including myogenin(MyoG),myogenic differentiation 1(MyoD1),and myosin heavy chain(MyHC),meanwhile accelerating the myotube formation of skeletal muscle satellite cells(SMSCs).Mechanistically,miR-181a-5p and miR-181b-5p directly bind to the 3′untranslated region(UTR)of the transforming growth factor beta receptor 1(TGFBR1)mRNA,further reducing the expression of TGFBR1.TGFBR1 is a key Transforming growth factor beta(TGF-β)signaling transduction receptor and had a negative function in muscle cell differentiation.Furthermore,knockdown of TGFBR1 facilitated the expression of chicken myogenic factors,boosted myotube formation,and decreased the SMAD family member 2/3(SMAD2/3)phosphorylation in chicken SMSCs.SMAD2/3 are downstream of TGF-βsignaling,and miR-181a-5p and miR-181b-5p could reduce the expression of TGFBR1 to further diminish the SMAD2/3 phosphorylation.Our findings revealed that the miR-181-5p family targets TGFBR1 to break the TGF-βsignaling transduction,which resulted in promoting chicken skeletal muscle development.展开更多
Background: This study, it was aimed to determine the level of knowledge, attitudes, and behaviors of patients who applied to a family health center about the rational use of drugs. Irrational use of drugs is a major ...Background: This study, it was aimed to determine the level of knowledge, attitudes, and behaviors of patients who applied to a family health center about the rational use of drugs. Irrational use of drugs is a major problem worldwide. WHO reports that half of the patients do not use their drugs correctly. Methods: This cross-sectional study was conducted between 1 October 2017 and 30 November 2018 at Istanbul Fatih Family Health Center No 7. The sample size was determined as 301 people selected by systematic sampling method from patients aged 18 and older. Results: The mean age of the participants was 53.8% ± 16.8%, 59.8% were female, 62.5% were married, and 52.5% were primary school graduates. 63.9% of the participants knew the names of the drugs they used, and 79.9% of them knew the indication for use. 89% of individuals check the expiry date of the drugs before using them, and 83.1% read the drug prospectus. 84% of the participants support the prohibition of using over-the-counter drugs. Conclusion: The level of knowledge of the participants about the rational use of drugs is above the societal average. However, some issues need more awareness. The high level of rational use of drugs awareness of physicians and pharmacists will also strengthen the rational use of drugs and indirectly increase the awareness of the public.展开更多

One of these γ-ray sources, the 10 TeV -γ-ray radiation reported by HESS, has been suggested as having a hadronic origin when relativistic protons are injected into and interact with the dense ambient gas. Assuming that such relativistic protons required by the hadronic model come from the tidal disruption of a star by the massive black hole of Sgr A*, we explore the spectrum of the relativis- tic protons. In the calculations, we investigate cases where different types of stars are tidally disrupted by the black hole of Sgr A*, and we consider that different diffusion mechanisms are used for the propagation of protons. The initial energy distribution of the injected spectrum of protons is assumed to follow a power-law with an exponential cut-off, and we derive the different indices of the injected spectra for the tidal disruption of different types of stars. For the best fit to the spectrum of photons detected by HESS, the spectral index of the injected relativistic protons is about 2.05 when a red giant is tidally disrupted by the black hole of Sgr A* and the diffusion mechanism is the Effective Confinement of Protons.展开更多

Increasing evidence indicates that aberrant expressions of some microRNAs are associated with cancer progression.However,the roles and biological mechanisms of miRNA-16-5p in human non-small cell lung cancer(NSCLC)are...Increasing evidence indicates that aberrant expressions of some microRNAs are associated with cancer progression.However,the roles and biological mechanisms of miRNA-16-5p in human non-small cell lung cancer(NSCLC)are not to be well studied.Here,we validated that the expression of miR-16-5p was decreased significantly in NSCLC samples and cell lines.The correlation between the clinicopathological features of NSCLC and the miR-16-5p expression showed that the expression of miR-16-5p in non-small cell lung cancer was linked with the advanced TNM stage,positive lymph node metastasis,with short overall survival(OS).Also,a negative correlation between miR-16-5p and Fermitin family member 2(FERMT2)was observed,implying there may be a potential link about their regulation.The hypothesis was further confirmed by in-silico analysis and dual-luciferase reporter assay.Moreover,we demonstrated that the transfections of miR-16-5p mimics could alter some biological characteristics of NSCLC cells remarkably accomplished by the expression variance of FERMT2 in vitro and in vivo assays.Summarily,this study demonstrated that miR-16-5p,as a tumor suppression factor in NSCLC by targeting FERMT2,could serve as one promising biomarker in the prediction for NSCLC patients.展开更多

Streptococcus mutans is a well-known cause of dental caries,due to its acidogenicity,aciduricity,and ability to synthesize exopolysaccharides in dental plaques.Intriguingly,not all children who carry S.mutans manifest...Streptococcus mutans is a well-known cause of dental caries,due to its acidogenicity,aciduricity,and ability to synthesize exopolysaccharides in dental plaques.Intriguingly,not all children who carry S.mutans manifest caries,even with similar characteristics in oral hygiene,diet,and other environmental factors.This phenomenon suggests that host susceptibility potentially plays a role in the development of dental caries;however,the association between host genetics,S.mutans,and dental caries remains unclear.Therefore,this study examined the influence of host gene-by-S.mutans interaction on dental caries.Genome-wide association analyses were conducted in 709 US children (<13 years old),using the dbGap database acquired from the center for oral health research in appalachia (COHRA) and the Iowa Head Start programmes (GEIRS).A generalized estimating equation was used to examine the gene-by-S.mutans interaction effects on the outcomes (decayed and missing/filled primary teeth due to caries).Sequentially,the COHRA and GEIRS data were used to identify potential interactions and replicate the findings.Three loci at the genes interleukin 32 (IL32),galactokinase 2 (GALK2),and CUGBP,Elav-like family member 4 (CELF4) were linked to S.mutans carriage,and there was a severity of caries at a suggestive significance level among COHRA children (P < 9×10?5),and at a nominal significance level among GEIRS children (P = 0.047–0.001).The genetic risk score that combined the three loci also significantly interacted with S.mutans (P < 0.000 1).Functional analyses indicated that the identified genes are involved in the host immune response,galactose carbohydrate metabolism,and food-rewarding system,which could potentially be used to identify children at high risk for caries and to develop personalized caries prevention strategies.展开更多

文摘Increasing evidence indicates that aberrant expressions of some microRNAs are associated with cancer progression.However,the roles and biological mechanisms of miRNA-16-5p in human non-small cell lung cancer(NSCLC)are not to be well studied.Here,we validated that the expression of miR-16-5p was decreased significantly in NSCLC samples and cell lines.The correlation between the clinicopathological features of NSCLC and the miR-16-5p expression showed that the expression of miR-16-5p in non-small cell lung cancer was linked with the advanced TNM stage,positive lymph node metastasis,with short overall survival(OS).Also,a negative correlation between miR-16-5p and Fermitin family member 2(FERMT2)was observed,implying there may be a potential link about their regulation.The hypothesis was further confirmed by in-silico analysis and dual-luciferase reporter assay.Moreover,we demonstrated that the transfections of miR-16-5p mimics could alter some biological characteristics of NSCLC cells remarkably accomplished by the expression variance of FERMT2 in vitro and in vivo assays.Summarily,this study demonstrated that miR-16-5p,as a tumor suppression factor in NSCLC by targeting FERMT2,could serve as one promising biomarker in the prediction for NSCLC patients.
文摘Tunable color center lasers making use of electron trapping defects such as F A and F + 2 like centers in alkali halide crystals can deliver broadly tunable power over the near infrared 0.8—4.0μm range.The tunable range and their impressive feature of narrow linewidth make these solid state lasers important to molecular spectroscopy,pollution detection,fiber optics communication and other scientific research fields.The( F + 2) H center consisting of an F + 2 center and an neibouring O 2- ion,is one of the most important color centers.In this paper,we will study the preparation,absorption and emission spectra of the ( F + 2) H center in mixed crystal KCl KBr∶OH -. Single crystals of KCl,KBr and different compositions of KCl x Br 1-x solid solutions are growtn with 0.1mol%—0.3mol% KOH in the melt by the Czochralski technique in air.

文摘Streptococcus mutans is a well-known cause of dental caries,due to its acidogenicity,aciduricity,and ability to synthesize exopolysaccharides in dental plaques.Intriguingly,not all children who carry S.mutans manifest caries,even with similar characteristics in oral hygiene,diet,and other environmental factors.This phenomenon suggests that host susceptibility potentially plays a role in the development of dental caries;however,the association between host genetics,S.mutans,and dental caries remains unclear.Therefore,this study examined the influence of host gene-by-S.mutans interaction on dental caries.Genome-wide association analyses were conducted in 709 US children (<13 years old),using the dbGap database acquired from the center for oral health research in appalachia (COHRA) and the Iowa Head Start programmes (GEIRS).A generalized estimating equation was used to examine the gene-by-S.mutans interaction effects on the outcomes (decayed and missing/filled primary teeth due to caries).Sequentially,the COHRA and GEIRS data were used to identify potential interactions and replicate the findings.Three loci at the genes interleukin 32 (IL32),galactokinase 2 (GALK2),and CUGBP,Elav-like family member 4 (CELF4) were linked to S.mutans carriage,and there was a severity of caries at a suggestive significance level among COHRA children (P < 9×10?5),and at a nominal significance level among GEIRS children (P = 0.047–0.001).The genetic risk score that combined the three loci also significantly interacted with S.mutans (P < 0.000 1).Functional analyses indicated that the identified genes are involved in the host immune response,galactose carbohydrate metabolism,and food-rewarding system,which could potentially be used to identify children at high risk for caries and to develop personalized caries prevention strategies.
