Objective:To investigate the therapeutic effects and mechanisms of Feng-Liao-Chang-Wei-Kang combined with 5-fluorouracil administration in mice with colitis-associated colon cancer.Methods:To establish a colitis-associated colon cancer mouse model and observe the behavior and activity of mice after Feng-Liao-Chang-Wei-Kang and 5-fluorouracil administration;HE staining to observe the pathological changes of mouse colonic tissue;Western blot was used to detect the expression of mouse colon tissue in IL-6/STAT3 pathway-related proteins.Results:The survival rate of mice in the co-administered group was significantly increased,and the intestinal wall thickening and interstitial inflammation of mice were significantly reduced.Western blot results showed that the expression levels of P-STAT3 and IL-6 were significantly increased in the colonic tissue of mice after modeling,and the combined administration inhibited the expression of Cyclin D1,CDK4 and Bcl-2 protein in the IL-6/STAT3 pathway and upregulated the expression of Bax(P<0.05).Conclusion:Feng-Liao-Chang-Wei-Kang combined with 5-fluorouracil inhibits IL-6/STAT3 pathway to exert inhibition of colitis-associated colon cancer inhibition of colitis-associated colon cancer. 展开更多

[Objectives]To analyze the clinical effect of Feng-Liao-Chang-Wei-Kang Granule combined with mesalazine on chronic colitis.[Methods]120 patients with chronic enteritis admitted to Qionghai Hospital of Traditional Chinese Medicine from October 2020 to March 2022 were selected as the research objects,and were randomly divided into combined group(62 cases)and conventional group(58 cases).Patients in the conventional group were treated with oral mesalazine on the basis of conventional treatment,while patients in the combined group were treated with Feng-Liao-Chang-Wei-Kang Granule combined with mesalazine.The clinical efficacy,inflammatory factor level,colonic mucosal lesion degree,quality of life and TCM syndrome scores of the two groups were compared.[Results]The total clinical effective rate in the combined group was higher than that in the conventional group(P<0.05).After treatment,the levels of inflammatory factors in the two groups were lower than those before treatment,and the levels of interleukin-6(IL-6),C-reactive protein(CRP)and tumor necrosis factor-α(TNF-α)in the combined group were lower than those in the conventional group(P<0.001).After treatment,the Baron and Geboes scores of the two groups were lower than those before treatment,and the Baron and Geboes scores of the combined group were lower than those of the conventional group(P<0.001).After treatment,the scores of quality of life in the two groups were higher than those before treatment,while the scores of TCM syndromes were lower than those before treatment.The scores of quality of life in the combined group were higher than those in the conventional group(P<0.001).[Conclusions]Feng-Liao-Chang-Wei-Kang Granule combined with mesalazine can obviously improve the curative effect,reduce the level of inflammatory factors,improve the degree of colonic mucosal lesions and improve the quality of life of patients with chronic colitis,which is worthy of popularization and application in clinical practice. 展开更多

目的 探讨枫蓼肠胃康颗粒对肥胖诱导的2型糖尿病大鼠糖脂代谢及胰高血糖素样肽-1(GLP-1)的影响。方法 7周龄SD大鼠70只,随机取10只作为分为空白组,给予正常饲料喂养;其余大鼠采用高脂高糖饲料喂养结合腹腔注射链脲佐菌素方法建立2型糖... 目的 探讨枫蓼肠胃康颗粒对肥胖诱导的2型糖尿病大鼠糖脂代谢及胰高血糖素样肽-1(GLP-1)的影响。方法 7周龄SD大鼠70只,随机取10只作为分为空白组,给予正常饲料喂养;其余大鼠采用高脂高糖饲料喂养结合腹腔注射链脲佐菌素方法建立2型糖尿病模型。将造模成功2型糖尿病大鼠随机分为模型组、西格列汀组及枫蓼肠胃康低、中、高剂量组,每组10只。西格列汀组给予西格列汀16.7 mg/kg灌胃,枫蓼肠胃康低、中、高剂量组分别给予2.65 g/kg、5.3 g/kg、10.6 g/kg枫蓼肠胃康颗粒灌胃,空白组和模型组给予生理盐水灌胃,均1次/d,连续4周。测量各组大鼠造模结束和灌胃4周后体重,检测各组大鼠灌胃4周后血糖、血清胰岛素、血脂[胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)]及血清GLP-1水平,免疫组织化学法检测回肠组织中GLP-1表达情况。结果 灌胃4周后,枫蓼肠胃康中、高剂量组及西格列汀组大鼠体重与造模后比较差异无统计学意义(P均>0.05);模型组与枫蓼肠胃康低剂量组大鼠体重与造模后比较均明显降低(P均<0.05)。枫蓼肠胃康中、高剂量组和西格列汀组大鼠空腹血糖、负荷后30 min血糖、负荷后60 min血糖、负荷后120 min血糖和空腹胰岛素水平均明显低于模型组(P均<0.05);枫蓼肠胃康高剂量组和西格列汀组大鼠负荷后60 min和120 min血糖均明显低于枫蓼肠胃康低剂量组(P均<0.05);枫蓼肠胃康中剂量组和西格列汀组大鼠负荷后60 min胰岛素水平均明显高于枫蓼肠胃康低剂量组(P均<0.05)。枫蓼肠胃康高剂量组和西格列汀组大鼠血清TC、TG、LDL-C水平及枫蓼肠胃康中剂量组大鼠血清TC、TG水平均明显低于模型组(P均<0.05),枫蓼肠胃康中剂量组大鼠HDL-C水平明显高于模型组(P均<0.05)。枫蓼肠胃康中、高剂量组和西格列汀组血清GLP-1水平均明显高于模型组(P均<0.05),枫蓼肠胃康高剂量组和西格列汀组均明显高于枫蓼肠胃康低剂量组(P均<0.05)。枫蓼肠胃康颗粒中、高剂量组和西格列汀组大鼠回肠组织中GLP-1平均光密度值均明显高于模型组(P均<0.05)。结论 枫蓼肠胃康颗粒能够降低肥胖诱导的2型糖尿病大鼠血糖,改善血脂代谢,机制可能与其调节肠道分泌GLP-1功能有关。 展开更多

目的:探讨枫蓼肠胃康联合5⁃氟尿嘧啶给药对结肠炎相关性结肠癌小鼠的治疗作用及机制。方法:建立结肠炎相关性结肠癌小鼠模型,经枫蓼肠胃康和5⁃氟尿嘧啶给药后,观察小鼠的行为及活动情况;HE染色观察小鼠结肠组织的病理学变化;采用Western... 目的:探讨枫蓼肠胃康联合5⁃氟尿嘧啶给药对结肠炎相关性结肠癌小鼠的治疗作用及机制。方法:建立结肠炎相关性结肠癌小鼠模型,经枫蓼肠胃康和5⁃氟尿嘧啶给药后,观察小鼠的行为及活动情况;HE染色观察小鼠结肠组织的病理学变化;采用Western blot检测小鼠结肠组织在IL⁃6/STAT3通路相关蛋白的表达。结果:联合给药组小鼠的存活率明显提高,且小鼠肠壁增厚及间质炎症情况明显减轻。Western blot结果显示经造模后小鼠结肠组织中P⁃STAT3及IL⁃6的表达水平明显升高,联合给药后,抑制了IL⁃6/STAT3通路中Cyclin D1、CDK4和Bcl⁃2蛋白表达,上调了Bax的表达(P<0.05)。结论:枫蓼肠胃康联合5⁃氟尿嘧啶可抑制IL⁃6/STAT3通路来发挥对结肠炎相关性结肠癌的抑制作用。 展开更多
