BACKGROUND Colorectal cancer(CRC)is the third most common cancer and a significant cause of cancer-related mortality globally.Resistance to chemotherapy,especially during CRC treatment,leads to reduced effectiveness of drugs and poor patient outcomes.Long noncoding RNAs(lncRNAs)have been implicated in various pathophysiological processes of tumor cells,including chemotherapy resistance,yet the roles of many lncRNAs in CRC remain unclear.AIM To identify and analyze the lncRNAs involved in oxaliplatin resistance in CRC and to understand the underlying molecular mechanisms influencing this resistance.METHODS Gene Expression Omnibus datasets GSE42387 and GSE30011 were reanalyzed to identify lncRNAs and mRNAs associated with oxaliplatin resistance.Various bioinformatics tools were employed to elucidate molecular mechanisms.The expression levels of lncRNAs and mRNAs were assessed via quantitative reverse transcription-polymerase chain reaction.Functional assays,including MTT,wound healing,and Transwell,were conducted to investigate the functional implications of lncRNA alterations.Interactions between lncRNAs and trans-cription factors were examined using RIP and luciferase reporter assays,while Western blotting was used to confirm downstream pathways.Additionally,a xenograft mouse model was utilized to study the in vivo effects of lncRNAs on chemotherapy resistance.RESULTS LncRNA prion protein testis specific(PRNT)was found to be upregulated in oxaliplatin-resistant CRC cell lines and negatively correlated with homeodomain interacting protein kinase 2(HIPK2)expression.PRNT was demonstrated to sponge transcription factor zinc finger protein 184(ZNF184),which in turn could regulate HIPK2 expression.Altered expression of PRNT influenced CRC cell sensitivity to oxaliplatin,with overexpression leading to decreased sensitivity and decreased expression reducing resistance.Both RIP and luciferase reporter assays indicated that ZNF184 and HIPK2 are targets of PRNT.The PRNT/ZNF184/HIPK2 axis was implicated in promoting CRC progression and oxaliplatin resistance both in vitro and in vivo.CONCLUSION The study concludes that PRNT is upregulated in oxaliplatin-resistant CRC cells and modulates the expression of HIPK2 by sponging ZNF184.This regulatory mechanism enhances CRC progression and resistance to oxaliplatin,positioning PRNT as a promising therapeutic target for CRC patients undergoing oxaliplatin-based chemotherapy. 展开更多

Stigma color is a critical agronomic trait in watermelon that plays an important role in pollination.However,there are few reports on the regulation of stigma color in watermelon.In this study,a genetic analysis of the F2 population derived from ZXG1553(P1,with orange stigma)and W1-17(P2,with yellow stigma)indicated that stigma color is a quantitative trait and the orange stigma is recessive compared with the yellow stigma.Bulk segregant analysis sequencing(BSA-seq)revealed a 3.75 Mb segment on chromosome 6 that is related to stigma color.Also,a major stable effective QTL Clqsc6.1(QTL stigma color)was detected in two years between cleaved amplified polymorphic sequencing(CAPS)markers Chr06_8338913 and Chr06_9344593 spanning a~1.01 Mb interval that harbors 51 annotated genes.Cla97C06G117020(annotated as zinc finger protein CONSTANS-LIKE 4)was identified as the best candidate gene for the stigma color trait through RNA-seq,quantitative real-time PCR(qRT-PCR),and gene structure alignment analysis among the natural watermelon panel.The expression level of Cla97C06G117020 in the orange stigma accession was lower than in the yellow stigma accessions with a significant difference.A nonsynonymous SNP site of the Cla97C06G117020 coding region that causes amino acid variation was related to the stigma color variation among nine watermelon accessions according to their re-sequencing data.Stigma color formation is often related to carotenoids,and we also found that the expression trend of ClCHYB(annotated asβ-carotene hydroxylase)in the carotenoid metabolic pathway was consistent with Cla97C06G117020,and it was expressed in low amounts in the orange stigma accession.These data indicated that Cla97C06G117020 and ClCHYB may interact to form the stigma color.This study provides a theoretical basis for gene fine mapping and mechanisms for the regulation of stigma color in watermelon. 展开更多

Objective:Hepatocellular carcinoma(HCC)is a prevalent malignancy with poor survival.Different cell types in the tumor microenvironment participate in the tumorigenesis and progression of HCC.This study aimed to analyze the immune microenvironment of HCC and its relationship with clinical outcomes.Methods:We analyzed HCC RNA-seq for cell type identification and prognosis by estimating relative subsets of RNA transcripts using CIBERSORTx.The interaction between B cells and macrophages in HCC was analyzed using a Hepa1-6 orthotopic transplantation mouse model and flow cytometry.The effect of Zinc finger protein 296(ZNF296)on the interaction of B cells and macrophages was verified using human HCC tissues analyzed through western blot,quantitative real-time polymerase chain reaction(qPCR),and multiplex immunofluorescence.A comparative analysis of immune cells associated with HCC prognosis was performed using RNA-seq data from The Cancer Genome Atlas(TCGA),bulk multimodal data,and single-cell transcriptomic data from existing HCC single-cell transcriptomic data employing the Single Cell Inferred Site Specific Omics Resource for Tumor Microenvironments(SCISSOR).Results:Liver hepatocellular carcinoma(LIHC)RNA-seq analysis of TCGA showed that high eosinophil infiltration promoted HCC progression.The proportion of B cells correlated with that of macrophages(r=−0.24)and affected the infiltration and programmed death ligand 1(PD-L1)expression of macrophages in HCC.ZNF296 may participate in the interaction between B cells and macrophages to accelerate the HCC progression by regulating PAFAH1B3 and H2AFX.Moreover,ZNF296 expression positively correlated with LAG3(r=0.27)and CTLA4(r=0.31)expression levels.Among the immune cell phenotypes related to survival and death identified by SCISSOR analysis,T cells correlated with an excellent prognosis of HCC.The normal function of liver and dendritic cells was also associated with a good prognosis in HCC.Conclusions:This study analyzed the interaction of the immune microenvironment with HCC prognosis,identifying ZNF296 as a promising diagnostic and therapeutic target for HCC. 展开更多

This paper proposes an explicit scheme to analyze the failure of a subsea polyhedral tunnel-liner system with an inverted arch under mechanical loading and fire fields.The thin-walled liner is made of Functionally Graded Materials(FGMs),which may improve the stability behavior of the tunnel-liner system.Hydrostatic pressure is inevitable in the liner since underground water may penetrate the cracks of the tunnel,and reach the outer surface of the liner.In addition,an elevated temperature loading is taken into account,considering that fire may occur in the tunnel-liner system.Under the combination of mechanical loading and thermal loading,the liner deforms into a single-lobe shape,which is depicted by a trigonometric function.The total potential energy is expressed quantitatively after the energy approach and thin-walled shell theory are used.The minimum potential energy is obtained when the critical buckling occurs.The critical buckling pressure is calculated,which considers the effect of the thermal field.The present analytical prediction is subsequently compared precisely with other closed-form solutions.Finally,the effects of several parameters,such as the geometric shapes,temperature variations,and volume fraction indices,are discussed to further survey the buckling performance of the nonlinear buckling of an FGM polyhedral liner with an inverted arch.One may address a polyhedral liner with fewer polyhedral sides,and a lower volume fraction index is recommended to rehabilitate cracked tunnels in engineering applications. 展开更多

Objective:Uterine corpus endometrial carcinoma(UCEC),a kind of gynecologic malignancy,poses a significant risk to women’s health.The precise mechanism underlying the development of UCEC remains elusive.Zinc finger protein 554(ZNF554),a member of the Krüppel-associated box domain zinc finger protein superfamily,was reported to be dysregulated in various illnesses,including malignant tumors.This study aimed to examine the involvement of ZNF554 in the development of UCEC.Methods:The expression of ZNF554 in UCEC tissues and cell lines were examined by qRT-PCR and Western blot assay.Cells with stably overexpressed or knocked-down ZNF554 were established through lentivirus infection.CCK-8,wound healing,and Transwell invasion assays were employed to assess cell proliferation,migration,and invasion.Propidium iodide(PI)staining combined with fluorescence-activated cell sorting(FACS)flow cytometer was utilized to detect cell cycle distribution.qRT-PCR and Western blotting were conducted to examine relative mRNA and protein levels.Chromatin immunoprecipitation assay and luciferase reporter assay were used to explore the regulatory role of ZNF554 in RNA binding motif 5(RBM5).Results:The expression of ZNF554 was found to be reduced in both UCEC samples and cell lines.Decreased expression of ZNF554 was associated with higher tumor stage,decreased overall survival,and reduced disease-free survival in UCEC.ZNF554 overexpression suppressed cell proliferation,migration,and invasion,while also inducing cell cycle arrest.In contrast,a decrease in ZNF554 expression resulted in the opposite effect.Mechanistically,ZNF554 transcriptionally regulated RBM5,leading to the deactivation of the Wingless(WNT)/β-catenin signaling pathway.Moreover,the findings from rescue studies demonstrated that the inhibition of RBM5 negated the impact of ZNF554 overexpression onβ-catenin and p-glycogen synthase kinase-3β(p-GSK-3β).Similarly,the deliberate activation of RBM5 reduced the increase inβ-catenin and p-GSK-3βcaused by the suppression of ZNF554.In vitro experiments showed that ZNF554 overexpression-induced decreases in cell proliferation and migration were counteracted by RBM5 knockdown.Additionally,when RBM5 was overexpressed,it hindered the improvements in cell proliferation and migration caused by reducing the ZNF554 levels.Conclusion:ZNF554 functions as a tumor suppressor in UCEC.Furthermore,ZNF554 regulates UCEC progression through the RBM5/WNT/β-catenin signaling pathway.ZNF554 shows a promise as both a prognostic biomarker and a therapeutic target for UCEC. 展开更多

BACKGROUND Although the occurrence of multiple primary cancers(MPC)is not exceedingly common,it is not rare in clinical practice.In recent years,there has been a notable increase in its incidence.The frequent confusion between MPC and tumor metastasis or recurrence often leads to delays in diagnosis and treatment.This study aimed to enhance understanding of MPC,improve diagnostic accuracy,guide precise clinical treatment,and implement a case management nursing model(CMNM)to facilitate quick patient recovery.CASE SUMMARY A 61-year-old female patient presented with persistent upper abdominal pain lasting over 2 months.Gastroscopy revealed the presence of both gastric and duodenal cancers.Following a thorough evaluation,the patient underwent pancreaticoduodenectomy,cholecystectomy,and total gastrectomy.Post-surgery,an individualized case management nursing approach was applied,leading to a successful recovery.Three months after the surgery,follow-up examinations showed no signs of recurrence.CONCLUSION The CMNM effectively promoted rapid patient recovery,enhanced the quality of orthopedic nursing services,and accelerated postoperative recovery,ultimately leading to increased patient satisfaction with nursing care. 展开更多

Line Patterns Segmentation in Blurred Images Using Contrast Enhancement and Local Entropy Thresholding

Finger vein extraction and recognition hold significance in various applications due to the unique and reliable nature of finger vein patterns. While recently finger vein recognition has gained popularity, there are still challenges associated with extracting and processing finger vein patterns related to image quality, positioning and alignment, skin conditions, security concerns and processing techniques applied. In this paper, a method for robust segmentation of line patterns in strongly blurred images is presented and evaluated in vessel network extraction from infrared images of human fingers. In a four-step process: local normalization of brightness, image enhancement, segmentation and cleaning were involved. A novel image enhancement method was used to re-establish the line patterns from the brightness sum of the independent close-form solutions of the adopted optimization criterion derived in small windows. In the proposed method, the computational resources were reduced significantly compared to the solution derived when the whole image was processed. In the enhanced image, where the concave structures have been sufficiently emphasized, accurate detection of line patterns was obtained by local entropy thresholding. Typical segmentation errors appearing in the binary image were removed using morphological dilation with a line structuring element and morphological filtering with a majority filter to eliminate isolated blobs. The proposed method performs accurate detection of the vessel network in human finger infrared images, as the experimental results show, applied both in real and artificial images and can readily be applied in many image enhancement and segmentation applications. 展开更多

In this study, an improved analytical solution for evaluating the safety of vertically exposed faces in backfilling was proposed. Based on a differential slice method, the proposed solution emphasizes the arching effect as having the advantages of more rigor and wider scalability. Feasibility of the proposed solution was validated with classic centrifuge results. Good agreement between compared results indicated that the proposed solution skillfully predicts the behavior of the paste centrifuge model. Additionally, calculation of exposed face safety in sequential filling was presented. The proposed solution has practical significance in mine backfill design. 展开更多

It is significant to develop a robot hand with high rigidity by a 6-DOF parallel manipulator(PM).However,the existing6-DOF PMs include spherical joint which has less capability of pulling force bearing,less rotation range and lower precision under alternately heavy loads.A novel 6-DOF PM with three planar limbs and equipped with three fingers is proposed and its kinematics and statics are analyzed systematically.A 3-dimension simulation mechanism of the proposed manipulator is constructed and its structure characteristics is analyzed.The kinematics formulae for solving the displacement,velocity,acceleration of the platform,the active legs and the fingers are established.The statics formulae are derived for solving the active forces of PM and the finger mechanisms.An analytic example is given for solving the kinematics and statics of proposed manipulator and the analytic solved results are verified by the simulation mechanism.It is proved from the error analysis of analytic solutions and simulation solutions that the derived analytic formulae are correct and provide the theoretical and technical foundations for its manufacturing,control and application. 展开更多

To assist in managing the stability of mining-related voids and improving the recovery of orebodies, tailing-based backfill technology has been widely used around the world. In the design of tailing-based backfill strategy, the specific function and engineering requirements of the filling are intimately dependent on the stress distribution within the backfilled stope. In this paper, the main mechanics involved in tailing-based backfill in underground mines, which include arching, consolidation, hydration process, and movement of surrounding rocks, are reviewed. Research on the safety of a barricade and stability of an exposed fill face are also presented. In conclusion, the backfilling process should be performed on the basis of a better understanding of the complicated interactions of the mechanisms of filling, consolidation, and hydration process(when cement is added). 展开更多

Even though a large number of large-scale arch dams with height larger than 200 m have been built in the world, the transient groundwater flow behaviors and the seepage control effects in the dam foundations under difficult geological conditions are rarely reported. This paper presents a case study on the transient groundwater flow behaviors in the rock foundation of Jinping I double-curvature arch dam, the world's highest dam of this type to date that has been completed. Taking into account the geological settings at the site, an inverse modeling technique utilizing the time series measurements of both hydraulic head and discharge was adopted to back-calculate the permeability of the foundation rocks,which effectively improves the uniqueness and reliability of the inverse modeling results. The transient seepage flow in the dam foundation during the reservoir impounding was then modeled with a parabolic variational inequality(PVI) method. The distribution of pore water pressure, the amount of leakage, and the performance of the seepage control system in the dam foundation during the entire impounding process were finally illustrated with the numerical results. 展开更多

Cemented paste backfill(CPB) is largely used in underground mines worldwide.A key issue associated with application of CPB is to estimate the stresses in backfilled stopes and on barricades.Recent numerical and experimental results show that arching effect is absent shortly after the placement of CPB in stopes.However,stress decreases in barricade drift with increasing distance between the measurement points and drawpoint have also been observed,demonstrating arching effect shortly after the pouring of CPB.To explain these paradoxes,CPB is considered as Bingham fluid having a yield shear stress.Three dimensional analytical solutions are proposed to evaluate the short-term total stresses in backfilled stopes and on barricades,accounting for the CPB's yield shear stress-induced arching effect.Stress diminution due to such arching effect in the backfilled stopes and on barricades is indeed obtained.But the reduction becomes insignificant using typical yield shear stress and stope geometry.More analyses indicate that the typical yield shear stress values do not fully correspond to field conditions where the yield shear stress would increase exponentially due to apparent consolidation(loss of water by drainage,a phenomenon similar to the desiccation of overly saturated fine-grained materials). 展开更多

To determine the distribution of active earth pressure on retaining walls, a series of model tests with the horizontally translating rigid walls are designed. Particle image velocimetry is used to study the movement and shear strain during the active failure of soil with height H and friction angle φ. The test results show that there are 3 stages of soil deformation under retaining wall translation: the initial stage, the expansion stage and the stability stage. The stable sliding surface in the model tests can be considered to be composed of two parts. Within the height range of 0.82 H-1.0 H, it is a plane at an angle of π/4+φ/2 to the horizontal plane. In the height range of 0-0.82 H, it is a curve between a logarithmic spiral and a plane at an angle of π/4+φ/2 to the horizontal. A new method applicable to any sliding surface is proposed for active earth pressure with the consideration of arching effect. The active earth pressure is computed with the actual shape of the slip surface and compared with model test data and with predictions obtained by existing methods. Deep underground projects(e.g., coal mines), are often faced with complex conditions such as high stress and extremely soft rock. The strength and rigidity of the traditional support system are often insufficient,which makes it difficult to meet the requirements of ground control under complex conditions. As a new support form with high strength and rigidity, the confined concrete arch plays an important role in controlling the rock deformation under complex conditions. The section shape of the tunnel has an important impact on the mechanical properties and design of the support system. However, studies on the mechanical properties and influence mechanism of the new confined concrete arch are rarely reported. To this end, the mechanical properties of traditional U-shaped steel and new confined concrete arches are compared and comparative tests on arches of circular and straight-leg semicircular shapes in deep tunnels are conducted. A large mechanical testing system for underground engineering support structure is developed. The mechanical properties and influence mechanism of confined concrete arches with different section shapes under different loading modes and cross-section parameters are systematically studied. Test results show that the bearing capacity of the confined concrete arch is 2.10 times that of the U-shaped steel arch, and the bearing capacity of the circular confined concrete arch is 2.27 times that of the straight-leg semicircular arch. Among the various influencing factors and their engineering parameters,the lateral stress coefficient has the greatest impact on the bearing capacity of the confined concrete arch,followed by the steel pipe wall thickness, steel strength, and core concrete strength. Subsequently, the economic index of bearing capacity and cost is established, and the optimization design method for the confined concrete arch is proposed. Finally, this design method is applied to a high-stress tunnel under complex conditions, and the deformation of the surrounding rock is effectively controlled. 展开更多

In this case report, we present a patient (PID: 6925981) who was seen in our outpatient department with a first episode of tophaceous gout in the finger, an infrequently seen condition that mostly affects older men with long-term gout. We report a case of blast injury to the left hand which resulted in fractures of the fingers with exposure of bones and joints of the phalanges. We used three reverse adipofascial cross finger flaps raised at the same time from 2 fingers to reconstruct adjacent fingers of the patient. The patient recovered well postoperatively and had good range of movement of the fingers. This avoided the complications of the use of regional or distal flaps. To our knowledge, this is the first case reported in which three reverse adipofascial cross fingers flaps are raised at the same time, two of them from an injured finger, to cover three raw areas on two fingers of a patient. 展开更多

Due to the loose structure,high porosity and high permeability of soil-rock mixture slope,the slope is unstable and may cause huge economic losses and casualties.The h-type anti-slide pile is regarded as an effective means to prevent the instability of soilrock mixture slope.In this paper,a centrifuge model test was conducted to investigate the stress distribution of the h-type anti-slide pile and the evolution process of soil arching during the loading.A numerical simulation model was built based on the similar relationship between the centrifuge model and the prototype to investigate the influence factors of the pile spacing,anchored depth,and crossbeam stiffness,and some recommendations were proposed for its application.The results show that the bending moment distribution of the rear pile exhibits Wshaped,while for the front pile,its distribution resembles V-shaped.The soil arching evolution process during loading is gradually dissipated from bottom to top and from far to near.During the loading,the change of bending moment can be divided into three stages,namely,the stabilization stage,the slow growth stage,and the rapid growth stage.In engineering projects,the recommended values of the pile spacing,anchored depth,and crossbeam stiffness are 4.0d,2.0d,and 2.0EI,where d and EI are the diameter and bending stiffness of the h-type anti-slide pile respectively. 展开更多

Placement of geosynthetic is effective in reducing the relative displacement between pile and subsoil. When the mobilized shear stress is less than the shear strength, partially developed arching will occur. Consequently, existing analytical methods, adopting the ultimate shear strength failure criterion, need to be improved. This study developed a simplified 2 D analytical method, which is based on the developing arching effect, to evaluate the load redistribution of the PGR embankment. Then, the influences of embankment height and internal friction angle, subsoil depth, ratio of pile cap width to pile clear spacing(RPC) and geosynthetic tensile stiffness on the critical height ratio, stress concentration ratio, soil arching ratio, geosynthetic tension and axial strain were investigated. This study suggests that a RPC of 1:1.0 and a one-way of single-layer geosynthetic tensile stiffness of 2000 kN/m should be considered as the sensitivity thresholds for the PGR embankment. 展开更多

A huge number of old arch bridges located in rural regions are at the peak of maintenance.The health monitoring technology of the long-span bridge is hardly applicable to the small-span bridge,owing to the absence of technical resources and sufficient funds in rural regions.There is an urgent need for an economical,fast,and accurate damage identification solution.The authors proposed a damage identification system of an old arch bridge implemented with amachine learning algorithm,which took the vehicle-induced response as the excitation.A damage index was defined based on wavelet packet theory,and a machine learning sample database collecting the denoised response was constructed.Through comparing three machine learning algorithms:Back-Propagation Neural Network(BPNN),Support Vector Machine(SVM),and Random Forest(R.F.),the R.F.damage identification model were found to have a better recognition ability.Finally,the Particle Swarm Optimization(PSO)algorithm was used to optimize the number of subtrees and split features of the R.F.model.The PSO optimized R.F.model was capable of the identification of different damage levels of old arch bridges with sensitive damage index.The proposed framework is practical and promising for the old bridge’s structural damage identification in rural regions. 展开更多

Seismic pile–slope stability analysis and the formation mechanism of soil arching have not been well studied. This study used a threedimensional(3D) finite difference to determine soil and pile parameter changes in the static and seismic stability of the pile–slope caused by the interaction between stabilizing piles. Pile–slope stability analysis was performed to determine the optimal design of piles along a slope and the corresponding failure mode involving the formation of soil arching around two adjacent piles. The Factor of Safety(FS) of the slope was evaluated using the shear strength reduction method for static and seismic analyses. The results of the analysis show that suitable pile spacing(S) and a suitable pile diameter(D) in the middle of a slope result in the maximum FS for the pile–slope system and the formation of soil arching around two adjacent piles. FS of the pile–slope increased negligibly in the seismic analysis of piles located at the slope crest and toe. An optimized pile diameter and installation location afforded the maximum FS for the slope that corresponded to a specified slope failure mode for different pile locations. A pile spacing S ≤ 2.5D for piles installed in the middle of the slope is suggested for increasing the static and seismic pile–slope stability. 展开更多
