Investigation of Biological Characteristics of Parasitic Fusarium Fungi from Tibetan Cordyceps sinensis

[Objective] This study aimed to investigate the biological characteristics of parasitic Fusarium fungi from tibetan Cordyceps sinensis. [Method] A Fusarium strain was isolated from fresh tibetan Cordyceps sinensis as the experimental material, to observe the morphologies of Fusarium mycelium and spores. [Result] The isolated Fusarium fungus strain grew lushly on rice medium, with white and villiform myceliurn, and the villous stroma appeared in colors from beige to pink. After cultivated at (18 ± 4) ℃ for 14 d, the Fusarium strain produced milky white protrusions containing pale yellow chlamydospores, which can be divided into microconidiums and macroconidiums based on the different sizes. According to Ainsworth's classification system, the isolated strain belongs to Fusarium, Tuberculariales, Hyphomycetes, adelomycete subphylum. Further observation identified three types of macroconidium, and two of them showed distinct morphology, which was not described in Ainsworth's classification system. [Conclusion] The structure of the strain isolated from tibetan Cordyceps sinensis is significantly different from that of Fusariurn fungi recorded in relevant literatures, which requires further investigation. 展开更多

莲腐败病是白莲种植区发生最普遍、为害最严重的病害之一。本试验对发病白莲上的病原茵进行分离鉴定。该病原分离物在PSA培养基上气生茵丝由白色渐变成深紫色,并产生大小两种孢子。大型分生孢子镰刀形,无色,稍呈弯曲,两端尖细,大... 莲腐败病是白莲种植区发生最普遍、为害最严重的病害之一。本试验对发病白莲上的病原茵进行分离鉴定。该病原分离物在PSA培养基上气生茵丝由白色渐变成深紫色,并产生大小两种孢子。大型分生孢子镰刀形,无色,稍呈弯曲,两端尖细,大小为(39.1-55.2)μm×(2.8~4.6)p,m,多为3个隔膜,少部分为1-2隔或4~5隔;小型分生孢子椭圆形。无色,无隔膜,大小为(5.0~9.2)μm×(2.0~3.5)μm。致病性测定表明分离物能100%引起白莲出现典型的白莲腐败病的症状。ITSl/ITS4的PCR扩增产物与GenBank中Fusariumoxysporum£sp.nelumbicola的序列同源性为99%。研究结果表明。分离到的病原物为尖孢镰刀茵莲专化型Foxysporum f.sp.ndumbicola。 展开更多

24种杀菌剂及其相关配比对人参根腐病菌的毒力测定及田间防效 被引量:8

[目的]生产上缺乏防治人参根腐病的有效药剂,为有效防治人参根腐病提供理论依据。[方法]采用生长速率法测定了24种杀菌剂及7种药剂的34种配比对人参根腐病菌的毒力,并进行田间防效试验。[结果]8%井冈·32%蜡芽菌WP、50%多菌灵WP抑菌效果较好,其EC90值分别为0.56、6.65 mg/L。具有增效作用的药剂配比有5个,2亿/g木霉菌WG+97.8%多菌灵TC 2∶3配比增效作用最为明显,其共毒系数为822.26。50%多菌灵WP和2亿/g木霉菌WG+97.8%多菌灵TC 2∶3配比在田间表现出较好的防效,防效均在65%以上。[结论]筛选出防治人参根腐病菌的有效药剂及配比,为更有针对性地防治人参根腐病奠定基础。 展开更多

In vitro Selection for Fusarium Wilt Resistance in Gladiolus 被引量:2

Cormels pieces of four Fusarium susceptible Gladiolus cultivars (Friendship, Peter Pears, Victor Borge and Novalux) formed friable calli when cultured in vitro on Murashige and Skoog basal medium containing various concentrations of auxin and cytokinin. The friable calli established cell suspensions. Plantlet regeneration was obtained from the control callus, control cell suspension derived callus and in vitro selected Fusarium oxysporum Schlecht. resistant cell-lines of Friendship. The in vitro cormlets showed 85-95% germination after breaking dormancy of 8 weeks at 4℃. Cell suspensions of all four Gladiolus cultivars were found to be highly sensitive to fusaric acid. Gradual increase in fusaric acid concentrations to the cell-suspension cultures decreased cell growth considerably. One albino plant was found from the second generation of the in vitro selected cell line of Friendship. The albino plant was found to be highly susceptible to F. oxysporum. The cormlets of all in vitro selected cell lines of Friendship were inoculated with a conidial suspension of the F. oxysporum before planting and were also sprayed with the same spore suspension for further characterization when the height of plants was about 6cm. The four selected cell lines showed the same response whether or not they were inoculated with conidia of the F. oxysporum. Plantlets of all of the selected cell lines exhibited significant growth as compared with the control after application of conidia of the F. oxysporum. 展开更多
