目的研究生长休止蛋白(Growth arrest-specific protein 7,Gas7)在大鼠梨状皮质发育过程中的表达。方法采用逆转录聚合酶链反应(RT-PCR)方法和免疫组织化学方法检测Gas7核酸与蛋白在SD大鼠胚胎第18.5天(E18.5)、E20.5、出生当天(P0)、...目的研究生长休止蛋白(Growth arrest-specific protein 7,Gas7)在大鼠梨状皮质发育过程中的表达。方法采用逆转录聚合酶链反应(RT-PCR)方法和免疫组织化学方法检测Gas7核酸与蛋白在SD大鼠胚胎第18.5天(E18.5)、E20.5、出生当天(P0)、生后第7天(P7)、P14、P21和成年(Adult)各时期梨状皮质中的表达。结果 RT-PCR结果显示Gas7核酸在大鼠梨状皮质各发育时期均有表达,在P14时表达最强;免疫组织化学方法显示梨状皮质在E18.5时即出现Gas7免疫阳性产物,至P7时出现清晰的Gas7免疫阳性细胞,至P14时细胞数达到峰值,免疫阳性反应最强,P21细胞数少于P14(P<0.05),Adult细胞数少于P21(P<0.05)。结论 Gas7在梨状皮质的表达具有时间上的差异性,提示Gas7可能在梨状皮质结构形成和功能成熟方面起着重要的调控作用。展开更多

Methods: Immunohistochemistry SABC method was applied to detect GAS7 expression in specimens of 54 osteosarcoma and 15 osteochondroma cases. Results: The positive expression rate of GAS7 was 74.7% (40/54) in the group of osteosarcoma and 0% (0/15) in the group of osteochondroma. There was a significant difference in the comparison of GAS expression in these two groups (P 〈 0.05). GAS7 was higher expressed in the samples which complicated with relapse or pulmonary metastasis than the control group (P 〈 0.05). There was no significant association between GAS7 expression and the size of tumor, the gender or the age of the patients (P 〉 0.05). Conclusion: The hyper-expression ot GAST may play an important role in the initiation and development of human osteosarcoma.展开更多

Methods: Twenty-four male Swiss albino mice were used for the...Objective: The aim of the present study was to check the potential interaction of two neurodevelopmental proteins, Disc1 and Gas7, in the adult mice brain. Methods: Twenty-four male Swiss albino mice were used for the study. The mice were 12 weeks old in the beginning of the experiment. Immunohistochemistry and co-immunofluorescence were performed on the coronal sections of mice brain and immunoblotting and co-immunoprecipitation were done on the whole brain lysate. Results: Data from immunohistochemistry and co-immunofluorescence indicate the occurrence and co-localization of Disc1 and Gas7 proteins in soma and projections of the brain cells. Immunostaining was observed in cerebral cortex, hypothalamus, midbrain, pons, medulla oblongata and CA3 of hippocampus of the brain. The data from Immunoblotting and co-immunoprecipitation validates the presence and interaction of Disc1 and Gas7 protein in whole brain lysate. Conclusion: Data indicates the potential interaction of Disc1 and Gas7 protein in adult brain. The study highlights the need for further research on Disc1-Gas7 protein interaction in brain development and neuro-disorders.展开更多
Objective Circular RNAs(circRNAs)participate in several important pathological processes and have been used in the diagnosis and treatment of various diseases.This study aimed to investigate the role of circRNAs in ne...Objective Circular RNAs(circRNAs)participate in several important pathological processes and have been used in the diagnosis and treatment of various diseases.This study aimed to investigate the role of circRNAs in neural tube defects(NTDs).Method We characterized circRNA-associated competitive endogenous RNA(ceRNA)networks in brain tissue of low folate-induced NTDs mouse at embryonic day 13.5 by high-throughput sequencing.The expression levels of Circzfp644,miR-20-5p and Gas7 were detected by RT-PCR.Gas7 and Circzfp644functions were determined by miRNA-mimics and inhibitors in mouse teratocarcinoma cells(F9 cells),and luciferase gene reporter assay was assessed in the F9 cells.In addition,the expression levels of Circzfp644,miR-20-5p and Gas7 were determined by Nanostring in human NTDs tissues.Results We detected 57 circRNA transcripts,16 miRNAs,and 148 mRNAs that were significantly dysregulated in NTDs brain tissues compared with their expression levels in control(normal)tissues.Circzfp644 shared miRNA response elements with the growth arrest specific 7(Gas7)gene and competitively bound with miR-20-5p to increase the expression of Gas7.Downregulation of Circzfp644and Gas7 and upregulation of miR-20-5p were found in human NTD tissue.Conclusion This study provides new perspectives on the role of circRNAs in nervous system development and the pathogenesis of NTDs.展开更多
